Conventional methods of body fluid identification use labor-intensive, technologically diverse techniques that are performed in a series, not parallel, manner and are costly in terms of time and sample. Furthermore, for some frequently encountered body fluids, such as saliva or vaginal secretions, no confirmatory technique exists. Terminally differentiated cells, such as blood lymphocytes or epithelial cells lining the oral cavity, have a unique pattern of gene expression, which is evinced by the presence and relative abundance of specific mRNA species. If the type and abundance of mRNAs can be determined in a stain or tissue sample recovered at the crime scene, it would be possible to definitively identify the tissue or body fluid in question. Advantages of an mRNA-based approach, compared to conventional biochemical analysis, include greater specificity, simultaneous and semi-automated analysis though a common assay format, improved timeliness, decreased sample consumption and compatibility with DNA extraction methodologies. In this report, we demonstrate that RNA is stable in biological stains and can be recovered in sufficient quantity and quality for analysis using reverse transcriptasepolymerase chain reaction assay (RT-PCR). We have identified sets of candidate tissuespecific genes for body fluids and tissues of forensic interest, namely blood, saliva, semen, vaginal secretions, menstrual blood, urine, skin, muscle, adipose, and brain. We also report the identification of a new housekeeping gene for use in mRNA based assays. Select body fluid-specific genes have been incorporated into multiplex PCR and real-time PCR assays. These assays allow for the positive identification of blood, saliva, semen,vaginal secretions, and/or menstrual blood in a stain. The final task of this work was the molecular characterization of mRNA degradation patterns in biological stains, which not only has fundamental importance in possibly revealing mRNA degradation pathways in dried biological stains, but may ultimately lead to better assay design strategies for mRNA markers for forensic use. An mRNA-based approach described in this report could allow the facile identification of the tissue components present in a body fluid stain and could conceivably supplant the battery of serological and biochemical tests currently employed in the forensic serology laboratory.

Many manufacturing processes require complex tooling which contributes significantly to the cost and time required to develop new products. Bead foam molding is often hampered by these limitations. This thesis presents an analysis of Additive Manufacturing (AM) applications in low cost bead foam molding, focusing on molding trials, economic analysis, and future potential.
Through molding trials, the thesis evaluates the efficacy of AM tooling in comparison to traditional aluminum tooling, specifically in evaluating tool life and cost. A key finding is a reduction in lead time up to 70% and cost of up to 63% compared to traditional tooling, particularly in low-volume production scenarios. This thesis includes a detailed cost analysis, which breaks down the cost components associated with AM processes such as pre-processing, production, material costs, post-processing, and overheads. This analysis reveals that AM tooling can offer substantial cost savings over conventional methods, making it a viable option for specific manufacturing contexts.
Findings suggest that while AM tooling shows significant promise in reducing costs and accelerating production in bead foam molding, further research is required. This research should focus on exploring the scalability of AM for larger tools and investigating the application of new and emerging AM processes and materials. / Master of Science / This thesis explores the use of Additive Manufacturing (AM), often known as 3D printing, in creating molds for bead foam molding—a process used in manufacturing a variety of foam products. Findings reveal that using AM for toolmaking can be faster and more cost-effective than traditional methods, especially for small-scale production. The thesis details experiments comparing AM with conventional tooling and presents a cost analysis showing the potential time and cost savings. While promising, further research is needed to fully harness the benefits of AM in this field. This study opens doors to more efficient and economical manufacturing techniques using emerging AM technology.

Framing the Olympics: RT’s Responses to Controversies Linked With Russia’s Presence at the Winter Games from 2014-2022Fritz, Colin January 2024 (has links)
The Russian Federation’s presence on the global stage of the Olympic Games has been linked with considerable controversy from 2014 onwards. The nation’s use of international sport for promoting its image, state-sponsored doping, and Russia’s illegal occupations of Ukraine staged in close time proximity with the Games are all controversies which have taken their toll on the country’s image, both within and beyond the context of sport. Using a news framing theory and analysis, this thesis examines how RT, a state-sponsored media outlet that operates in alignment with the Russian state, counters these image-affecting controversies through its framings of Russia at the Winter Olympics between 2014 and 2022. This research finds that, while RT has consistently functioned to defend and potentially improve Russia’s image, its framing tactics have shifted substantially over time, and vary significantly depending on the controversy addressed. RT’s articles linked with Russia’s use of sport to promote its image used a wide variety of frames to highlight the impact of Russia’s athletes, justify the funding of the Sochi Olympics, and accuse Western media of “Russia-bashing”. Articles related to Russian doping shifted over time from framing Russian athletes as powerless victims of sport governing body decisions to highlighting supposed Western interference in sport. Articles linked with Russia’s illegal occupation of Ukraine increased substantially in 2022, and most often framed the issue by focusing on the moral values associated with athletes from the two nations. This thesis found that RT’s controversy-linked content increased in publication frequency over time, which along with the above shifts in framing, showcase the increasingly aggressive politicisation of RT’s Olympic sport coverage, which lacks exploration in prior scholarly research.

This thesis explores the application of the Virtual Test Bed (VTB) and its real-time extension, VTB-RT, for protective relay modeling, simulation, and testing. An instantaneous overcurrent relay model was developed in VTB for a transmission line protection. The same relay model was built in Matlab/Simulink for validation purposes. Both models were tested for various fault conditions on a radial power system and results were compared. Moreover, a low cost real-time Hardware-In-the-Loop (HIL) simulation platform was implemented for relay model testing using VTB-RT and public domain software packages such as Real-Time Application Interface (RTAI), Comedi, and Comedilib, and notebook computer hardware. The applicability of VTB-RT was verified through an open-loop simulation and a HIL simulation of a simple dynamic system using dSPACE as the control hardware and NI DAQCard-6062E as the input/output interface. Simulation results are presented showing the effectiveness of the VTB-RT platform for model testing.
