Cultivar Selection Trials of Navel Orange in Arizona for 2006-07

Two orange cultivar trials have been established in Arizona, one at the Yuma Mesa Agricultural Center, Yuma, AZ and one at the Citrus Agriculture Center, Waddell, AZ. For the navel orange trial in Yuma, all the selections had reduced yields in 2006-07. 'Fisher' navel continues to have the greatest yield. Of the rest in the Yuma trial, 'Lane Late' had the best quality and yield. For the Waddell trial, the fourth year data has been collected, and suggests that 'Fisher', 'Beck-Earli' and 'Lane Late' are outperforming the other cultivars tested to date.

For the year, 'Fina Sodea' had the greatest yield, and average fruit size, while 'Gold Nugget' had the smallest yield, and ‘Fremont’ had the smallest fruit size.

The Effects of Rootstock, Scion, Grafting Method and Plant Growth Regulators on Flexural Strength and Hydraulic Resistance of Apple

The apple rootstock ‘Geneva® 41’ (‘G.41’), is desirable for its resistance to fire blight, Erwinia amylovora, while producing high yields. However, ‘G.41’ and other Geneva® selections tend to form weak graft unions in the nursery that are susceptible to breaking in the wind, at harvest, or during shipment. In order to understand and remedy this graft union weakness, six scions (‘Fuji’, ‘Gala’, ‘Honeycrisp’, ‘McIntosh’, ‘Pink Lady’, and ‘Scilate’) by seven rootstocks (‘G.41’, ‘G.935’, ‘G.214’, ‘G.11’, ‘M.9-NIC 29’, ‘B.9’ and ‘EMLA 26’), by two grafting methods (chip bud and saddle graft) in a factorial design were tested for graft union flexural strength and flexibility. Additionally, plant growth regulators (PGR) were applied to trees with ‘G.41’ as rootstock as a solution to overcome graft union weakness. Transpiration rate and hydraulic resistance were measured for comparison of a weak graft forming rootstock (‘G.41’) to a strong graft forming rootstock (‘M.9-NIC 29’). ‘G.41’ consistently formed a weaker graft union regardless of scion or grafting method. Scions such as ‘McIntosh’ and ‘Pink Lady’ formed stronger graft unions, while ‘Scilate’ and ‘Honeycrisp’ form weaker graft unions. Saddle grafting did not improve graft union strength. Nurseries that graft on ‘G.41’ should choose scions that form stronger graft unions. Benzyl adenine (BA) in a latex paint increased the strength and flexibility of the graft union relative to scion cross sectional area (SCSA), but further research is needs to identify more efficient methods of application. Rootstock effect on transpiration rate was different between years. No detectable differences were found for hydraulic resistance through the graft union of different rootstocks, suggesting weaker graft unions did not limit hydraulic conductance or transpiration rate. Thus graft union weakness is not an indicator of poor vascular connection.

O experimento foi realizado nos ciclos produtivos de 2014 e 2015 no município de São Manuel-SP (22° 44' S e 48° 34' O e 740 m). As plantas foram sustentadas em espaldeira baixa no espaçamento de 2,0 x 0,80 m. O delineamento experimental foi em blocos casualizados em esquema fatorial 2 x 2, correspondendo aos dois porta-enxertos ('IAC 766' e 'IAC 572') e dois sistemas de condução do cordão principal (unilateral e bilateral). Foram avaliados o número de gemas do cordão principal e o número de ramos por planta, fertilidade de gemas, comprimento, diâmetro e massa fresca dos mesmos. Durante o ciclo foram avaliadas a duração dos estádios fenológicos e a evolução da maturação das uvas. Na colheita, avaliaram-se: número de cachos por planta, produção; produtividade; massa fresca, comprimento e largura de cachos, bagas e engaços; número de bagas por cacho e relação entre comprimento e largura da baga. O mosto da uva foi avaliado quanto ao teor de sólidos solúveis (SS), acidez titulável (AT), relação SS/AT, pH e açúcares redutores. The experiment was conducted in the productive cycles of 2014 and 2015 at the Experimental Farm of the Faculty of Agricultural Sciences/UNESP, located in São Manuel, SP (22º 44' S and 48º 34' W and 740 m). The plants were spaced 2,0 x 0,80 m, in espalier support system low. The treatments consisted of the combination of two rootstocks ('IAC 766' and 'IAC 572') and two training systems of the main cord (unilateral and bilateral). Were avaluated the bud number of the main cord and the number of branches per plant, the fertility evaluation of bud, length, diameter and fresh weight thereof. During the cycle were evaluated, the duration of phenological stages and the evolution of maturation the grapes. At harvest, were evaluated: number of bunches per plant; production; productivity; fresh weight, length and width of bunches, berries and stems; number of berries per cluster and the relationship between length and width of berry. Influence du génotype de porte-greffe dans la signalisation azotée et le développement du greffon chez la vigne / Rootstock genotype impact on nitrogen signalling and development of the scion in Grapevine

Ce travail de recherche a eu pour objectif de caractériser l’influence du génotype de porte-greffes sur le développement du greffon notamment à travers l’étude des mécanismes relatifs à la signalisation azotée et hormonale. Dans ce contexte, deux porte-greffes, 1103 Paulsen (1103P) et Riparia Gloire de Montpellier (RGM), induisant respectivement une forte et faible vigueur du greffon, ont été étudiés. Une analyse du transcriptome racinaire a été effectuée chez des plants greffés impliquant ces deux génotypes dans un système split-root où la disponibilité en azote était hétérogène. Une réponse plus prononcée a été observée pour le génotype RGM, ainsi qu’une régulation temporelle différente entre les deux génotypes, notamment concernant des gènes clés de la réponse au nitrate intervenant dans la régulation de la croissance des racines ou dans la signalisation hormonale. D’un point de vue développemental, une régulation étroite de la production des racines latérales et des rameaux latéraux par la disponibilité en azote a été mise en évidence chez RGM. D’autre part, les propriétés intrinsèques de chaque porte-greffe semblent être conférées au greffon induisant notamment une ramification plus importante de celui-ci lorsque le porte-greffe 1103P est utilisé. La caractérisation fonctionnelle des gènes impliqués dans la voie de biosynthèse des strigolactones et la réalisation de bio-essais ont permis de mettre en évidence la production de composés de type strigolactones chez la Vigne. Ces travaux suggèrent aussi une balance contrastée entre les strigolactones et les cytokinines au sein des deux génotypes, corrélée à leur capacité de contrôle de la croissance du greffon. L’ensemble de ces résultats a permis de mettre en évidence une réponse transcriptomique et développementale à la disponibilité en azote toujours plus prononcée pour le génotype connu pour conférer le moins de vigueur au greffon, RGM. / This work aimed to characterize the impact of the rootstock genotype on the scion development especially through the study of the mechanisms related to nitrogen and hormonal signalling. In that context, two rootstocks were studied, 1103 Paulsen (1103P) and Riparia Gloire de Montpellier (RGM), inducing high and low scion vigour, respectively. A transcriptomic analysis was performed on roots of grafted plants involving both genotypes, cultivated in a split-root system where the nitrogen availability was heterogeneous. A more pronounced response was observed for RGM together with a different temporal regulation between both rootstocks, in particular concerning the expression of key genes of the nitrate response involved in root growth regulation or in hormonal signaling. Concerning plant development, a clear impact of the nitrogen availability on the production of lateral roots and shoot branching was highlighted for RGM. Moreover, the intrinsic properties of each rootstock seemed to be conferred to the scion inducing especially a higher shoot branching when the rootstock 1103P was used. The functional characterization of the genes involved in the strigolactone biosynthesis pathway and bioassays highlighted the production of strigolactone-like compounds in Grapevine. These experiments suggest also a contrasted balance between strigolactones and cytokinins within each rootstock genotype, correlated with their ability to control the scion growth. Groundcover, rootstock and root restriction effects on vegetative growth, crop yield components, and fruit composition of Cabernet Sauvignon

Wine vineyards in humid environments like the mid-Atlantic United States are characterized by vines that develop too much vegetative growth for optimum quality wine production. Cover crops, rootstocks and rootzone restriction were evaluated for their effect on vegetative and reproductive growth on Cabernet Sauvignon. Treatments were arranged in a strip-split-split plot arrangement with under-trellis cover crops (UTCC) compared to row-middle only cover crop combined with 1-m weed-free strips in the vine row as main plots. Rootstocks riparia Gloire, 420-A, and 101-14 were sub-plots, while sub-sub-plots comprised two treatments: vines were either planted in root-restrictive (RR), fabric bags (0.016 m3) at vineyard establishment (2006), or were planted without root restriction. All three factors were effective in suppressing vegetative development as measured by rate and extent of shoot growth, lateral shoot development, trunk circumference, and dormant pruning weights. Canopies of vines with UTCC and RR had reduced leaf layer values by approximately 21% and 23% compared to conventional controls. The principal effect of the UTCC and the RR treatments was a sustained reduction in stem (xylem) water potential. UTCC and RR caused significant 7 and 10% reductions in berry weight, compared to their conventional controls. Berry weights of vines grafted to riparia were greater than those of vines grafted to other rootstocks. Wine made from UTCC and RR treatments increased red wine color compared to herbicide UTGC and NRR, respectively. This study identified treatments that improve vine balance while simultaneously improving grape composition and potential wine quality. / Master of Science

Effects of water stress and rootstock genotype on grape berry composition / Effet de la contrainte hydrique et du génotype du porte-greffe sur la composition de la baie de raisin

Au cours des prochaines années, il est prédit que le changement climatique va influencer la production des cultures et impacter négativement le secteur agricole. Parmi les cultures mondiales majeures, la vigne est cultivée pour ses baies, qui sont la base de produits à forte valeur ajoutée (vin, liqueurs, et métabolites secondaires utilisés dans les industries pharmaceutiques et cosmétologiques) et dont le métabolisme est fortement sensible au climat. Cependant, la réponse au niveau de la composition de la baie ainsi que les mécanismes moléculaires sous-jacents mettant en évidence la capacité de l’interaction porte-greffe/greffon à influencer le métabolisme de la baie dans des conditions de stress hydrique est encore très peu étudiée et compris. Dans ce contexte, cette thèse a eu pour objectifs de répondre à ces questionnements en combinant des approches ecophysiologique, biochimique et transcriptomique. Vitis vinifera cv. Pinot noir greffé soit sur le porte greffe 110R (tolérance à la sécheresse, vigueur conférée moyenne à forte) ou 125AA (sensible à la sécheresse, forte vigueur conférée) ont été étudié au cours de 3 années (2009, 2010 et 2011), au vignoble et dans des conditions témoin (pluviométrie normale) ou de stress hydrique provoqué. Différents paramètres physiologiques (statut hydrique et rendement) ainsi que le profil métabolique de la baie (sucres, acides organiques, acides aminés et anthocyanes) ont été caractérisés à quatre stades de développement (E-L 33, E-L 35, E-L 36, E-L 38). D’autre part, une analyse microarray sur génome complet a également été réalisée pour deux années (2009 et 2010) et deux stades de développement critiques et représentatifs (E-L 35 et E-L 36). Dans son ensemble, cette thèse fournit des nouveaux éléments concernant la réponse métabolique de la baie au porte-greffe et à la contrainte hydrique, et met en évidence des possibles mécanismes moléculaires impliqués dans cette réponse. / Climate change is expected to influence crop production and to impact negatively the agricultural sector in the future. Among the major crops cultivated worldwide, grapevine provides berries that are the basis of high added value products (wines, liquors, and secondary metabolites used in the pharmaceutical and cosmetological industry) and whose metabolism is strongly sensitive to climate (vintage effect). However, the response of berry composition and the molecular mechanisms underlying the ability of rootstock/scion interaction to influence grape berry metabolism under drought stress are still poorly understood. In this context, this work aimed to fill the gaps on the aforementioned questions by combining comprehensive ecophysiological measurements, detailed metabolite analysis, and whole-genome transcriptome analysis. Vitis vinifera cv. Pinot noir grafted on either rootstock 110R (drought tolerant, mid- to high vigor) or 125AA (drought sensitive, high vigor) were studied during three growing seasons (2009, 2010, and 2011) in the field under normal rainfall or water shortage conditions. We characterized different physiological parameters (water status and yield components) and berry metabolomic profiles (sugars, organic acids, free amino acids and anthocyanins) during four developmental stages (E-L 33, E-L 35, E-L 36, E-L 38). Besides we also performed a microarray analysis in two years (2009 and 2010) at two critical and representative developmental stages (E-L 35 and E-L 36). Overall, this work provides novel insights into the response of grape berry metabolites to rootstock and to drought and uncovers some possible molecular mechanisms underlying the berry response to different rootstock/water status combinations.

As cochonilhas farinhentas (Hemiptera: Pseudococcidae) têm se tornado pragas importantes em diversas culturas. Na videira, são responsáveis por causar danos diretos, resultando em perdas na produção e danos indiretos como vetoras de vírus. Entretanto, poucas informações estão disponíveis sobre a biologia das espécies na cultura. Neste trabalho, foi estudado em laboratório, o efeito de diferentes estruturas vegetativas, cultivares e porta-enxertos de videira no desenvolvimento, sobrevivência e reprodução de Dysmicoccus brevipes (Cockerell, 1893) e Pseudococcus viburni (Signoret, 1875). As exigências térmicas de D. brevipes foram determinadas, sendo estimado o número anual de gerações da espécie nas principais regiões produtoras de uvas de mesa do Brasil. A biologia de D. brevipes foi estudada em folhas de videira das cultivares de uva de mesa Itália (Vitis vinifera) e Niágara Rosada (Vitis labrusca) e em raízes dos porta-enxertos Paulsen 1103 (Vitis berlandieri X Vitis rupestris) e IAC 572 (Vitis caribaea X 101-14 Mgt). As exigências térmicas foram estimadas com base nos dados de biologia obtidos em cinco temperaturas constantes (15, 20, 25, 28 e 30°C) em folhas de videira da cultivar Itália. A biologia de P. viburni foi estudada em folhas de videira das cultivares de uva de mesa Itália e Niágara Rosada. D. brevipes se desenvolveu em diferentes estruturas vegetativas da planta, bem como em diferentes cultivares e porta-enxertos de videira; no entanto, folhas da cultivar Itália foram as mais adequadas ao desenvolvimento da espécie, considerando-se a menor duração do período ninfa-adulto. A taxa líquida de reprodução (Ro) de D. brevipes foi maior em folhas da cultivar Itália (19,82) em relação à cultivar Niágara Rosada (13,78). As ninfas de D. brevipes que sobreviveram nas raízes do porta-enxerto Paulsen 1103, se desenvolveram melhor do que em folhas. Raízes do porta-enxerto IAC 572 foram inadequadas para o desenvolvimento de D. brevipes, originando fêmeas que não se reproduziram. A velocidade de desenvolvimento de D. brevipes aumentou com a elevação da temperatura, sendo a faixa de temperatura entre 20ºC e 30ºC a mais adequada para a espécie. O limiar térmico inferior e superior de desenvolvimento e a constante térmica para o período ninfa-adulto de D. brevipes em folhas de videira da cultivar Itália, foram de 8,2ºC, 34,5°C e 678,42 graus-dia, respectivamente. De acordo com as exigências térmicas de D. brevipes foi possível estimar a ocorrência de 3,4 gerações anuais em Bento Gonçalves (RS), 3,1 em Caxias do Sul (RS), 4,4 em Marialva (PR), 4,8 em Jales (SP) e 5,2 em Petrolina (PE). Através dos dados de longevidade dos adultos e de viabilidade das diferentes fases do ciclo biológico (ninfa-adulto) de fêmeas de P. viburni, verificou-se que a cultivar Niágara Rosada foi mais adequada para o desenvolvimento da espécie. Os resultados deste estudo demonstraram que a planta hospedeira afeta o desenvolvimento e a sobrevivência de D. brevipes e P. viburni e que a escolha da cultivar ou do porta-enxerto a ser utilizado na implantação do vinhedo, pode auxiliar na redução da infestação destas pragas, sendo um componente adicional a ser utilizado em programas de Manejo Integrado na cultura. / Mealybugs (Hemiptera: Pseudococcidae) have become increasingly important pests in many crops. In vineyards, they cause direct damage resulting in production losses and also indirect damages through virus transmission. However, little information is available about the species biology. The effects of different vegetative structures, cultivars and vine rootstocks on the development, survival and reproduction of Dysmicoccus brevipes (Cockerell, 1893) and Pseudococcus viburni (Signoret, 1875) were studied under laboratory conditions. The thermal requirements of D. brevipes were determined and the annual number of generations was estimated for the principal grape-growing regions of Brazil. Biological aspects of D. brevipes were studied using table grape leaves (Itália: Vitis vinifera and Niágara Rosada: Vitis labrusca) and grapevine rootstocks (Paulsen 1103: Vitis berlandieri X Vitis rupestris and IAC 572: Vitis caribaea X 101-14 Mgt). The thermal requirements were estimated using five constant temperatures (15, 20, 25, 28 and 30°C) on Itália table grape leaves. The biology of P. viburni was studied on Itália and Niágara Rosada table grape leaves. D. brevipes developed on different vegetative structures, cultivars and vine rootstocks. However, table grape leaves of V. vinifera Itália were the most suitable for D. brevipes development, considering the shortest developmental period. The net reproductive rate (Ro) of D. brevipes was higher on Itália table grape leaves (19.82) than on Niágara Rosada (13.78). D. brevipes nymphs that survived on Paulsen 1103 roots developed better than on the leaves. Roots of IAC 572 were unsuitable for the development of D. brevipes, resulting in non-reproductive females. The developmental rate of D. brevipes increased with increasing temperature and the range between 20 to 30°C was the most suitable for this species. The predicted upper lethal temperature was 34,5°C, with a minimum threshold for total development at 8,2°C. D. brevipes required 678,42 degree-days to complete development from first instar nymph to the adult stage. According to the thermal requirements, D. brevipes can complete 3.4 annual generations per year in Bento Gonçalves (RS), 3.1 in Caxias do Sul (RS), 4.4 in Marialva (PR), 4.8 in Jales (SP) and 5.,2 in Petrolina (PE). From the the data on adult longevity and the viability of the different female life stages of P. viburni, table grape leaves of V. labrusca Niágara Rosada were the most suitable for development of this species. We conclude that the host plant can affect the development and survival of D. brevipes and P. viburni and that the right choice of the cultivar or vine rootstock to be used in the vineyard can help to reduce pest infestation and is, therefore, an additional component to be included in the integrated pest management of the crop.

Obtenção de plantas autotetraplóides de variedades de porta-enxertos de citros / Obtention of autotetraploids plants of varieties of rootstocks of Citrus

A citricultura brasileira destaca-se como a de maior expressão em nível mundial, com cerca de 27,9% da produção mundial de laranjas e 59% da produção global de suco concentrado congelado de laranja. No entanto, apresenta uma vulnerabilidade que é o predomínio de uma única espécie de porta-enxerto, o limão \'Cravo\', com aproximadamente 85% das plantas cítricas plantadas sobre ele. Assim, uma maior diversificação dos porta-enxertos é necessária mas isto depende também da obtenção e da disponibilização de variedades superiores ao limão Cravo. O porta-enxerto é de fundamental importância na formação da muda cítrica, visto que ele interfere em vários aspectos da planta enxertada, dentre eles o vigor da copa e o tamanho das plantas. Atualmente, devido ao surgimento de novas doenças e a necessidade de maximização dos recursos alocados na propriedade citrícola, tem sido utilizado o plantio de pomares mais adensados, com consequentemente aumento no interesse da produção de plantas com menor porte. Uma das possibilidades de se obter plantas de porte baixo é por meio do uso de portaenxertos ananicantes ou semi-ananicantes, sendo que alguns autores já verificaram que plantas autotetraplóides, quando usadas como porta-enxertos, induzem à formação de plantas de porte baixo. O objetivo inicial deste trabalho foi obter e identificar plantas autopoliplóides de limão Cravo, tangerinas Cleópatra e Sunki, utilizando-se o método de cultivo in vitro de segmentos de epicótilo temporariamente em meio de cultura contendo colchicina, seguido de regeneração das brotações em meio sem o alcalóide. A colchicina causou toxidez aos explantes, nos diversos tempos de cultivo e concentrações testadas. Várias plantas foram obtidas e dentre estas, 27 foram selecionadas por possuir características morfológicas semelhantes à plantas poliplóides. A seguir, foram realizados experimentos com seis plantas selecionadas (duas de cada espécie) visando confirmar a ploidia e a origem nucelar, além de caracterizar a morfologia das folhas, a densidade estomática e alguns parâmetros fisiológicos. Dentre as seis plantas, apenas quatro foram confirmadas como sendo autotetraplóides pelo método de citometria de fluxo. Estas, geralmente apresentaram folhas com comprimento significativamente menor, maior espessura do limbo foliar e menor densidade estomática nas folhas. Todas as seis plantas apresentaram alguma modificação em parâmetros fisiológicos, mas ainda não se pode determinar se estas modificações poderão afetar as taxas de crescimento. / The Brazilian citriculture stands out as the greatest expression worldwide, with about 27.9% of world production of oranges and 59% of global production of frozen orange juice concentrate. However, it presents a vulnerability that is the predominance of a single species of rootstock, the \'Cravo\' lemon, with approximately 85% of citrus planted on it. Thus, greater diversification of rootstocks is necessary, which also depends on obtaining and provision of superior kind of lemon Cravo. The rootstock is of fundamental importance in shaping the citrus scion, since it interferes with various aspects of graft, such as the vigour of the graft and plant size. Currently, due the appearance of new diseases and the need to maximize resources allocated in the citrus farm, more dense citrus orchards have been used, increasing the interest in the production of smaller plants. One of the possibilities of obtaining plants of small size is through the use of dwarf or semi-dwarf rootstocks, and some authors have found that plants autotetraploids, when used as rootstock, induce the formation of small-sized plants. The initial objective of this study was to obtain and identify plants autopoliploids of \'Cravo\' lemon, \'Cleopatra\' and \'Sunki\' tangerines, using the method of in vitro culture of segments of epicotyl temporarily in the culture medium containing colchicine, followed by shoots regeneration in medium without the alkaloid. The colchicine caused toxicity in explants, at the different times of cultivation and concentrations tested. Several plants were obtained and among these, 27 were selected for morphological characteristics similar to polyploid plants. Afterwards, trials were conducted with six selected plants (two of each kind) to confirm the nucelar ploidy origin, and to characterize the morphology of the leaves, stomatal density and some physiological parameters. Among the six plants, only four were confirmed as autotetraploids by method of flow cytometry. These, generally presented leaves with small length, greater thickness of the leaf lamina and lower stomatal density in leaves. All six plants showed some change in physiological parameters, but data could not determine whether these changes could affect the plants growth rates
