The thesis deals with evaluation of finances drawing in operational programmes, especially in regional ones, and provides also description of project documentation that is needed to get a subsidy from regional operational programme. The diploma paper comprises basic data about regional policy of the European Union and about implementation of this policy in the Czech Republic. The thesis carries on with the system of operational programmes in the Czech Republic, gives insight into regional operational programmes and analyses the state of finances drawing. Last part focuses on project preparing for getting a subsidy from structural funds, especially on feasibility study and project application.

The aim is to analyze the drawing of funding from the Regional Operational Programme South-East during programming period 2007-2013, which is financed by the European Regional Development Fund. The theoretical part is focused on regional policy in the European Union, its principles, instruments, and objectives. The thesis also describes the regional policy of the Czech Republic and its connection to the EU. In the practical part I analyzed drawing of financial resources and unsuccessful projects. The analysis is observed from three aspects: the area of support, the geographical distribution and the type of applicant.

Nové postupy přípravy polymerů kyseliny mléčné / New Processes of Lactid Acid Polymers Preparation

The work focuses on new processes for the preparation of lactic acid derivatives. The main objective was to verify the feasibility of preparing high molecular weight polylactide using ethyl lactate as a precursor of lactide synthesis. Part of the work is devoted to the new ethyl lactate synthesis method. The experimental part of the thesis is divided into partial key steps on the way from the lactic acid to the high molecular polylactide. The preparation of anhydrous ethyl ester of lactic acid (EtLA) was solved in an innovative way using alcoholysis of the oligomeric lactic acid. A kinetic model for isothermal alcoholysis and equimolar reactants ratio was derived from this method. The ethyl lactate was oligomerized by transesterification into the low and high molecular weight oligomer with the help of newly found catalysts suitable for the reaction medium. Stannous lactate was used as catalyst for oligomerization of the low molecular weight polymer suitable for the preparation of lactide (Mn 1000 g.mol-1). Experimental polymerization of ethyl lactate into high molecular weight product in tens of kDa has been investigated with newly synthesized tetraethyllactoyl titanate, Ti(EtLA)4. The laboratory method was derived for the depolymerization of the oligomer into lactide. Optimal conditions found for lactidation are as follow: temperature 225 ° C, pressure 2 kPa, catalysis 0,05 mol% of stannous lactate (with respect to oligomer lactate units). The prepared lactide was refined to polymer grade purity by distillation and subsequent recrystallization from ethyl acetate and toluene. The method for the preparation of high molecular weight PLA through ROP polymerization of lactide has been optimized. By optimization, suitable catalyst concentration was found in combination with the polymerization temperature and the polymerization length. An equimolar mixture of Tin 2-ethylhexanoate and 1-decanol was used as the catalytic system. The optimal ROP conditions for achieving the maximum molecular weight and suppressed polymer coloration (yellowing) are: catalyst concentration 0,01 mol%, temperature 160 °C, and polymerization length 4 hours. PLA with molecular weight Mw= 447 ± 7,8 kg.mol-1 was prepared at these conditions and good repeatability of the result was achieved. The effect of naturally occurring lactide contaminants and their influence on the course and ROP result was experimentally verified on the optimized polymerization system. The purpose was to explore the effect of lactide contamination with water and ethanol as natural lactide contaminants. The results clearly confirm the orderly lower sensitivity of the polymerization system for the presence of ethanol as compared to water contamination. In the case of the presence of water, the course and the result of the polymerization in terms of both the conversion and achieved molecular weights are negatively affected, even when the water content is in the order of 0,001%. Conversely, the presence of ethanol has a positive effect on lactide conversion and polymer polydispersity. Lactides with an alcohol content of about one-tenth of a percent are suitable to reach molecular weights of PLA similar to commercially available products. This difference shoves a significant advantage in the proposed technology of preparing PLA from ethyl lactate, especially for easier purification of crude lactide into polymer grade purity.

Quantitative Metrics and Measurement Methodologies for System Security Assurance

Proactive approaches for preventing attacks through security measurements are crucial for preventing sophisticated attacks. However, proactive measures must employ qualitative security metrics and systemic measurement methodologies to assess security guarantees, as some metrics (e.g., entropy) used for evaluating security guarantees may not capture the capabilities of advanced attackers. Also, many proactive measures (e.g., data pointer protection or data flow integrity) suffer performance bottlenecks. This dissertation identifies and represents attack vectors as metrics using the knowledge from advanced exploits and demonstrates the effectiveness of the metrics by quantifying attack surface and enabling ways to tune performance vs. security of existing defenses by identifying and prioritizing key attack vectors for protection. We measure attack surface by quantifying the impact of fine-grained Address Space Layout Randomization (ASLR) on code reuse attacks under the Just-In-Time Return-Oriented Programming (JITROP) threat model. We conduct a comprehensive measurement study with five fine-grained ASLR tools, 20 applications including six browsers, one browser engine, and 25 dynamic libraries. Experiments show that attackers only need several seconds (1.5-3.5) to find various code reuse gadgets such as the Turing Complete gadget set. Experiments also suggest that some code pointer leaks allow attackers to find gadgets more quickly than others. Besides, the instruction-level single-round randomization can restrict Turing Complete operations by preventing up to 90% of gadgets. This dissertation also identifies and prioritizes critical data pointers for protection to enable the capability to tune between performance vs. security. We apply seven rule-based heuristics to prioritize externally manipulatable sensitive data objects/pointers. Our evaluations using 33 ground truths vulnerable data objects/pointers show the successful detection of 32 ground truths with a 42% performance overhead reduction compared to AddressSanitizer. Our results also suggest that sensitive data objects are as low as 3%, and on average, 82% of data objects do not need protection for real-world applications. / Doctor of Philosophy / Proactive approaches for preventing attacks through security measurements are crucial to prevent advanced attacks because reactive measures can become challenging, especially when attackers enter sophisticated attack phases. A key challenge for the proactive measures is the identification of representative metrics and measurement methodologies to assess security guarantees, as some metrics used for evaluating security guarantees may not capture the capabilities of advanced attackers. Also, many proactive measures suffer performance bottlenecks. This dissertation identifies and represents attack elements as metrics using the knowledge from advanced exploits and demonstrates the effectiveness of the metrics by quantifying attack surface and enabling the capability to tune performance vs. security of existing defenses by identifying and prioritizing key attack elements. We measure the attack surface of various software applications by quantifying the available attack elements of code reuse attacks in the presence of fine-grained Address Space Layout Randomization (ASLR), a defense in modern operating systems. ASLR makes code reuse attacks difficult by making the attack components unavailable. We perform a comprehensive measurement study with five fine-grained ASLR tools, real-world applications, and libraries under an influential code reuse attack model. Experiments show that attackers only need several seconds (1.5-3.5) to find various code reuse elements. Results also show the influence of one attack element over another and one defense strategy over another strategy. This dissertation also applies seven rule-based heuristics to prioritize externally manipulatable sensitive data objects/pointers – a type of attack element – to enable the capability to tune between performance vs. security. The thesis introduces the European Structural Funds, which are involved in co-financing of projects within different areas. The object is to analyze three selected projects which were realized in programming period 2007 to 2013 under the priority axis 3 Tourism. The analysis should outline the impacts of projects which were supported by European funds. Another objective is to determine whether these projects helped to meet the objectives which were set in strategic documents. The thesis includes detailed statistics about the size of the volume of funds invested in projects of priority axis 3 Tourism in Central Moravia Region for the past year.

Ce travail de thèse avait pour objectif d'élaborer un système vecteur ciblantl'endothélium pathologiquement activé dans les tissus enflammés, infectés ou tumoraux.Ce vecteur est sous forme de nanoparticules décorées de ligands glycosides capablesd'interagir avec l'E-sélectine, un récepteur exprimé sur les cellules endothélialesactivées.Nous avons mis au point une synthèse de copolymère amphiphile avec une architectureà bloc muni sur sa partie hydrophile d'un ligand glucidique. Ce copolymère a été par lasuite utilisé pour la préparation de nanoparticules de type coeur/couronne. La partiehydrophobe centrale est entourée d'une couronne hydrophile dont l'encombrementstérique et la mobilité limitent l'opsonisation de la particule. Le ciblage actif a été assurépar la présence d'un ligand du récepteur de l'E-sélectine aux extrémités des chaînes depolymères hydrophiles à la surface du vecteur.Avec ces nanoparticules dont les propriétés de surface sont prédéfinies, nous avonsmontré in vitro l'association efficace avec les cellules endothéliales activées, ce qui apermis de valider ce concept de ciblage moléculaire actif par l'intermédiaire du couplerécepteur/ ligand. Un tel système permettra d'améliorer l'indice thérapeutique et labiodistribution des principes actifs anti-inflammatoire et/ou anticancéreux.

The subject of my thesis "Possible utilisation of EU funds for reconstruction and development of a business real property" is to analyse the most relevant options of the support for a selected realty with regard to its future use. The theoretical part is focused on the description and analysis of small and medium-sized enterprises in the Czech Republic. There are outlined different types of support for SMEs at the same time. In the practical part is presented a selected real property intended for a reconstruction. An analysis of grant opportunities suitable for that business real property is made furthermore. At the end is drawn up a concept of a project of reconstruction and development for that realty. This concept of a project is based on a selected real property and the appropriate grant program.
