Role of 26S Proteasome and Regulator of G-Protein Signaling 10 in Regulating Neuroinflammation in the Central Nervous System

Major histocompatibility complex molecules (MHCII) are cell surface glycoproteins that present extracellular antigens to CD4+ T lymphocytes and initiate adaptive immune responses. Apart from their protective role, overexpression of MHCII contributes to autoimmune disorders where the immune system attacks our own tissues. Autoimmune diseases are characterized by self-reactive responses to autoantigens, promoting tissue damage, inflammation mediated by proinflammatory cytokines, autoreactive lymphocytes, and autoantibodies. MHCII molecules are tightly regulated at the level of transcription by Class II transactivator (CIITA). CIITA associates with an enhanceosome complex at MHCII promoters and regulates the expression of MHCII. It is thus crucial to understand the regulation of CIITA expression in order to regulate MHCII in autoimmune diseases. Our lab has shown that the 19S ATPases of the 26S proteasome associate with MHCII and CIITA promoters and play important roles in gene transcription, regulate covalent modifications to histones, and are involved in the assembly of activator complexes in mammalian cells. The mechanisms by which the proteasome influences transcription remain unclear. Here, we define novel roles of the 19S ATPases Sug1, S7, and S6a in expression of CIITApIV genes. These ATPases are recruited to CIITApIV promoters and coding regions, interact with the elongation factor PTEFb, and with Ser5 phosphorylated RNA Pol II. Both the generation of CIITApIV transcripts and efficient recruitment of RNA Pol II to CIITApIV are negatively impacted by knockdown of 19S ATPases. Alternatively, inflammation is also suppressed via the Regulator of G-protein signaling 10 (RGS10) in microglial cells which express high levels of RGS10 and promote homeostasis in the central nervous system. However, chronic activation of microglial cells leads to release of cytokines which cause neuroinflammation. Our investigation of roles played by RGS10 in chronically activated microglial cells indicates that RGS10 binds to promoters of IL-1β, and TNF-α and regulates these genes, while the molecular mechanism remains to be investigated. Together, our observations indicate roles for the UPS in modulating gene expression and for RGS10 in regulating proinflammatory cytokines in microglial cells, each of which provides novel therapeutic targets to combat inflammation in autoimmune and neurodegenerative diseases.

The automation of industrial processes has been one of the greatest innovations in the industrial sector. It allows faster and accurate operations of production processes while producing more outputs than old manual production techniques. In the beverage industry, this innovation was also well embraced, especially to improve its bottling processes. However it has been proven that a continuous optimization of automation techniques using advanced and current trend of automation is the only way industrial companies will survive in a very competitive market. This becomes more challenging for small to medium scale enterprises (SMEs) which are not always keen in adopting new technologies by fear of overspending their little revenues. By doing so, SMEs are exposing themselves to limited growth and vulnerable lifecycle in this fast growing automation world. The main contribution of this study was to develop practical and affordable applications that will optimize the bottling process of a SME beverage plant by combining its existing production resources to basic principles of the current trend of automation, Industry 4.0 (I40). This research enabled the small beverage industry to achieve higher production rate, better delivery time and easy access of plant information through production forecast using linear regression, predictive maintenance using speed vibration sensor and decentralization of production monitoring via cloud applications. The existing plant Siemens S7-1200 programmable logic controller (PLC) and ZENON supervisory control and data acquisition (SCADA) system were used to program the optimized process with very few additional resources. The following diploma thesis deals with the management of asynchronous motor revolutions by a frequency converter MICROMASTER 440 via the programmable controller S7-200. At the beginning, I researched the company documentation of Siemens, which is the producer of the frequency converter and the programmable controller mentioned above. As my work proceeded, I also started to study the company documentation of Mitsubishi Electric, which produces the operating pannel. The first part of my thesis provides a general description of the stated devices and the second part deals with the solution of the task itself. My task was to design a way of communication between the converter and the controller. I chose a USS protocol via serial port RS-485. The operation of the controller was performed by using an operating terminal type GOT1000 which was connected to the controller. In summary, my diploma thesis primarily deals with the construction of an operating programme for the controller and with creating of a graphical control on the terminal. In the end, I evaluated the knowledge gained during the work on this subject and suggested further procedures.

This thesis deals with a technology of continuous packaging machine in the beginning from the point of risk assessment and design of necessary measures to reduce the risk as much as it is possible. Here the HRN method was used to quantify every risk factor. Next part documents the implementation of the safety measures and the realization of the safety circuits by using Simatic S7–1500F safety PLC and Shared device functionality. Safety function which is realized in the Sinamics S120 drive is Safe Stop 1. Next, it describes the drive dimensioning and the principle of the drive control. Simotion PLC is discussed in more details and then is used as a motion control master. An approach to the configuration of camming synchronous operation is dealt with in details. Cam design and their realization is documented, too. The thesis also demonstrates the state manager that was implemented in the user program and which is used according to the concept of OMAC state manager. In the end a simple HMI for packaging machine control is described.

This Diploma work is concerned with the design of project documentation for discharging shakeout machine electric installation. This work continues in the Semestral project 2 in which the background of a new machine was made. Then, this work deals with the replacement of a switch room with required devices including the design of a new connecting cables and power streams. In the first part, the work deals with the control part of the device and the design of control and signal cables including the product documentation of the device. In the second part, the work deals with the proposal of building electro installation in which a new lighting and its use is described. The last part focuses on the coordination during the realization of a suggested device. In this part, all problems coming from the particular working procedures including a proposal of possible solutions are described. According to the project documentation and following realization, the documentation of final concept reflecting the real status of the device is described.

It also deals with the project of mechanical construction, electrical system, with the computing procedure and the optimum choice of gear for galvanic line model. The work includes the design of a DC motor controller, the computing procedure and the construction of heating element for vats heater simulation. The last part of this thesis offers an insight to the control system and the assembled program. In the addendum, the photo of the realized model, the wiring scheme, and the programme transcript for control galvanic line model can be found.
