(B.) microplus SBm7462 by oral and nasal routes and to study the induction of AICD in memory T cells were used 80 BALB/c mice, divided in four experimental groups with 20 animals each (group I: oral immunization; group II: nasal immunization; group III: animals not immunized; and a group with animals givers of peritoneal macrophages. Two immunizations had been made with an interval of 21 days. At nine and 15 days after the first and the second immunization the collects were realized, separately for each group, and the T cells isolated had been cultivated with 5% CO2 at 37ºC, per 10 days in the presence of 1μg of SBm7462 and APCs. Later, the different agents, inductive (mithramycin and cycloheximide) or inhibiting (benzamide riboside and cyclosporin A) of apoptosis had been added, subdivided in distinct treatments. After the specific incubation period for each one of the treatments, it had been determined the percentage of cellular viability by the exclusion for Tripan Blue and the percentage of apoptotic cells with the dye Acridine Orange and by TUNEL technique. The specific nature of the T cells was confirmed by the increase in the number of cells after re-stimulation in vitro with SBm7462, which had held as memory T cells. The induction of AICD occurred by the inhibition of the expression of c-myc and by the increase of the expression of Fas and the induced AICD for the synthetic peptide SBm7462 was mediated by an independent way of the calcineurina and the kinetic of regulation of FasL. The mucosa routes, oral and nasal, can be considered alternative ways for the immunization with the synthetic peptide SBm7462 however they require a bigger number of doses to induce a response with high levels of immunoglobulins. / Com o objetivo avaliar a resposta imune de camundongos BALB/c imunizados por via oral e nasal com o peptídeo sintético anti-carrapato R. (B.) microplus SBm7462 e estudar a indução da AICD dos linfócitos T de memória dos animais imunizados, utilizou-se 80 camundongos BALB/c, divididos em quatro grupos experimentais com 20 animais cada (grupo I: imunização por via oral; grupo II: imunização por via nasal; grupo III: animais não imunizados; e um grupo de animais doadores de macrófagos peritoneais). Foram feitas duas imunizações, com intervalo de 21 dias. Aos nove e 15 dias após a primeira e segunda imunização realizou-se as coletas, separadamente para cada grupo, e os linfócitos T isolados foram cultivados em estufa, 5% de CO2 a 37ºC, por 10 dias na presença de 1μg de SBm7462 e APCs. Posteriormente, adicionou-se os diferentes agentes indutores (mitramicina e CHX) ou inibidores (DBR e CsA) de apoptose, subdivididos em tratamentos distintos. Após o período de incubação específico para cada um dos tratamentos, determinou-se a porcentagem de viabilidade celular pela exclusão pelo Azul de Tripan e a porcentagem de células apoptóticas pela coloração com Laranja de Acridina e pela técnica de TUNEL. A natureza específica dos linfócitos T foi confirmada pelo aumento no número de células após re-estimulação in vitro com SBm7462, os quais se comportaram como linfócitos T de memória. Ocorreu indução da AICD por inibição da expressão do c-myc e por aumento da expressão de Fas e a AICD induzida pelo peptídeo sintético SBm7462 foi mediada por uma via independente da calcineurina e da cinética de regulação do FasL. As vias de mucosa, oral e nasal, podem ser consideradas vias alternativas para a imunização com o peptídeo sintético SBm7462, porém requerem um maior número de doses para induzir uma resposta com altos níveis de imunoglobulinas.

The most commonly used forms of combat for this ectoparasite is mainly based on the use of chemical acaricides. Seeking an alternative way to control the parasite, the vaccine was developed synthetic anti R. (B.) microplus (SBm7462®). The synthetic vaccine SBm7462® contains three immunogenic epitopes (4822, 4824 and 4823) derived from the Bm86 protein. Genetic analysis showed that the fragment corresponding to SBm7462® is conserved across populations R. (B.) microplus from different regions in South America, however, the production of this vaccine on an industrial scale would cost very high. Therefore, alternative methods of production are being developed. The use of plants as bioreactors for production of heterologous proteins is a very attractive approach, since the transgenic plants are one of the most economical systems for protein production on industrial scale. The objective was to study the immune response of mice immunized orally with transgenic Arabidopsis thaliana expressing the recombinant peptide rBm7462. For this, seeds processed in a previous work were grown until the fifth generation. After confirmation of the production of the peptide, 0.5 g of leaves of transformed plants were administered orally to BALB/c, three times (interrupted by 3 weeks), as well as 200μL of total soluble protein extract, inoculated subcutaneously. After 9 weeks, during which blood samples were collected from animals, weekly, they were euthanized for collection of lymph nodes and ileum. The results showed that it was not possible to detect levels of IgG anti-rBm7462 in mice immunized orally with leaves of transgenic plants expressing the peptide rBm7462, as well as in mice immunized with total soluble protein extracts of these plants, while the indirect ELISA method was used. However, the lymph nodes of immunized animals showed microscopic changes related to immune response and markings, by immunohistochemistry, for the peptide rBm7462. These observations led to the conclusion that the antigen dose was given to the animals sub-antigenic which may have stimulated mechanisms of tolerance to the antigen. What makes a necessary adjustment in the doses given to animals. Seeds used strains derived from 2NF allowed for the preparation of the fifth generation of transformed plants, indicating that the gene seq2, responsible for the synthesis of peptide rBm7462 probably is homozygous. What it indicates the genomic stability and viability of producing the antigen in transgenic plants. / Os prejuízos causados pela infestação de Rhipicephalus (Boophilus) microplus tem sido estimada em bilhões de dólares nos países tropicais e subtropicais. As formas de combate mais utilizadas para este ectoparasita são baseadas principalmente no uso de químicos acaricidas. Buscando uma forma alternativa para o controle do parasita, foi desenvolvida a vacina sintética anti R. (B.) microplus (SBm7462®). A vacina sintética SBm7462® contém três epitopos imunogênicos (4822, 4824 e 4823) derivados da proteína Bm86. Análises genéticas demonstraram que o fragmento correspondente a SBm7462® é conservado em populações R. (B.) microplus de diferentes regiões na América do Sul. Contudo, a produção desta vacina em escala industrial tornaria seu custo muito elevado. Assim sendo, formas alternativas de produção estão sendo desenvolvidas. A utilização de plantas como bioreatores para produção de proteínas heterólogas é uma metodologia muito atrativa, visto que as plantas transgênicas constituem um dos sistemas mais econômicos para produção de proteínas em escala industrial. O objetivo deste trabalho foi estudar a resposta imunológica de camundongos, imunizados oralmente com Arabidopsis thaliana transgênicas, que expressam o peptídeo recombinante rBm7462. Para isso, sementes transformadas em um trabalho anterior foram cultivadas até a quinta geração. Após confirmação da produção do peptídeo, 0,5g de folhas das plantas transformadas foram administradas oralmente a camundongos BALB/c, por três vezes (intercaladas por 3 semanas); assim como também 200μL de extrato de proteínas totais solúveis, inoculado por via subcutânea. Após 9 semanas, durante as quais foram feitas coletas sanguíneas dos animais, semanalmente, estes foram eutanasiados para a coleta dos linfonodos e íleo. Os resultados demonstraram que não foi possível detectar níveis de IgGs anti-rBm7462 em camundongos imunizados oralmente com folhas de plantas transgênicas expressando o peptídeo rBm7462, como também em camundongos imunizados com extratos de proteínas totais solúveis das mesmas plantas, quando a metodologia ELISA indireto foi utilizada. Contudo, os linfonodos dos animais imunizados apresentaram alterações microscópicas relacionadas à resposta imunológica e marcações, através da técnica de imunohistoquímica, para o peptídeo rBm7462. Estas observações levaram a conclusão que a dose de antígeno oferecida aos animais foi sub-antigênica o que pode ter estimulado mecanismos de tolerância ao antígeno. O que torna necessário um ajuste nas doses oferecidas aos animais. Sementes utilizadas derivadas da linhagem 2FN possibilitaram a obtenção da quinta geração de plantas transformadas, indicando que o gene seq2, responsável pela síntese do peptídeo rBm7462, provavelmente encontra-se em homozigose. O que indica a estabilidade genômica dele e a viabilidade em se produzir o antígeno em plantas transgênicas.

O estudo se realizou com as doses de 1,0mg e 2,0mg em 30 bovinos da raça holandesa com média de grau de sangue de 91,16% (H/Z), os quais foram mantidos sob o manejo rotineiro no Campo Experimental do Centro Nacional de Pesquisa em Gado de Leite-EMBRAPA/CNPGL em pastos altamente infestados com larvas dessa espécie de carrapato. Os animais foram separados em três grupos: vacinal 2,0mg, vacinal 1,0mg e controle, sendo os grupos vacinais imunizados nos dias 0, 30 e 60 a campo. Foram realizadas a contagem de carrapatos e a coleta de sangue semanalmente, a partir do dia 0. Duas semanas após a terceira inoculação, os animais foram levados para baias individuais e submetidos a uma infestação artificial com um total de 4.500 larvas/animal de B. microplus (amostra “Porto Alegre”). A partir do 19 o dia pós-infestação, foram coletadas as teleóginas desprendidas, contadas, pesadas, identificadas e incubadas em estufa B.O.D. por quinze dias, quando os respectivos ovos foram pesados. A análise da fertilidade desses ovos e a viabilidade das larvas provenientes foram também analisadas. A cinética da produção de anticorpos foi acompanhada através de ELISA, obtendo- se títulos maiores para os grupos vacinais, estatisticamente diferentes do grupo controle (p<0,01). Os parâmetros biológicos foram analisados levando-se em conta o número de teleóginas, peso dos ovos e fertilidade dos ovos. A eficácia foi de 15,8% para a dose de 1,0mg do peptídeo sintético SBm7462 e de 53,29% para 2,0mg, com diferenças estatísticas significativas entre os grupos 1,0mg, 2,0mg e controle (p<0,05). Quanto à avaliação do número de carrapatos a campo, não houve diferença estatística entre os grupos testados. Os resultados obtidos mostram que o peptídeo sintético SBm7462 é uma alternativa viável para o controle do carrapato comum dos bovinos, pela sua eficácia, segurança e baixo custo. / The synthetic peptide SBm7462 was evaluated as immunogen for the control of the tick Boophilus microplus in mixed test (stable and field). The study was done with the quantity of 1,0mg and 2,0mg of the peptide SBm7462 on 30 Holstein female cattles (91,16% H/Z), which were maintained under the routine handling in the experimental field of the Centro Nacional de Pesquisa em Gado de Leite- EMBRAPA/CNPGL on pastures highly infested with larvaes of that tick species. The animals were separate in three groups: vaccinated with 2,0mg, vaccinated with 1,0mg and control, being the vaccinated groups immunized on the days 0, 30 and 60 in field. The count of the adults females ticks and the blood collection were weekly done, starting from the day 0. Two weeks after the third inoculation, the animals were taken to individual stalls and submitted to an artificial infestation with 4.500 larvaes/animal of B. microplus (strain “Porto Alegre”). Starting on the 19 o . day post-infestation, the adult females ticks were collected, counted, identified, weighed and incubated in B.O.D. After 15 days, the respective eggs were weighed. The analysis of the eggs fertility and the viability of the larvaes were also analyzed. The kinetics of the production of antibodies was accompanied through of ELISA, being obtained larger titles for the vaccinated groups with statistic differences to the control group (p < 0,01). The biological parameters were analyzed through of the account the number of adult females ticks, eggs weight and eggs fertility. The efficacy was of 15,8% for the 1,0mg of the synthetic peptide SBm7462 and of 53,29% for the 2,0mg, of the same peptide, with significant statistical differences among the groups 1,0mg, 2,0mg and control (p < 0,05). When compared the number of ticks in field, there was not statistic difference among the tested groups. The obtained results show that the synthetic peptide SBm7462 is a viable alternative for the control of the cattle tick, regarding its efficacy, safety and low cost.
