O tecido ósseo tem função importante no suporte, proteção e locomoção e está sob o controle de fatores sistêmicos como hormônios e fatores locais, entre eles os fatores de crescimento e citocinas. A Fosfatase Ácida Tartarato Resistente (TRAP) é uma enzima que faz parte da família das fosfatases ácidas e apresenta localização intracelular; mais especificamente dentro do compartimento lisossomal de osteoclasto, macrófagos e células dendríticas, tem sido utilizada como um marcador histoquímico da atividade osteoclástica. Objetivos: Avaliar a expressão da proteína TRAP em alvéolos dentários de ratos hipertensos (SHR) e normotensos tratados ou não com atenolol. Métodos: Neste estudo foram utilizados 4 grupos de ratos sendo: 1) W (wistar sem tratamento), 2) WT (wistar tratado com atenolol), 3) S (SHR sem tratamento) e 4) ST (SHR tratado com atenolol), submetidos a exodontia do incisivo superior direito, com eutanásia no 7º, 14º, 21 e 28º dia pós-operatório. A análise dos mecanismos biológicos envolvidos no processo de reparo alveolar foi obtida pela análise da expressão de proteínas TRAP por meio da técnica de imunoistoquímica. Os resultados foram analisados pela média e erro padrão da média e aplicado o teste paramétrico ANOVA, com pos-test de Tukey para avaliar os períodos dentro de cada grupo e entre os grupos, sendo consideradas as diferenças significativas quando p<0,05. Resultados: Os resultados mostraram que a marcação TRAP aumenta em alvéolo dentais de ratos Wistar durante todos os períodos pós – operatórios. A marcação TRAP aumenta apenas ao 14o nos dias de reparação alveolar em alvéolo dental de SHR não tratados. O atenolol não altera o processo de reparo alveolar em ratos Wistar, porém o atenolol promoveu a redução da marcação de TRAP em SHR ao 14º dia. Conclusão: A hipertensão aumenta a expressão da proteína TRAP no 14o dia pós-cirúrgico de reparação alveolar e o atenolol promove redução da marcação aumentada de TRAP ao 14º dia pós-cirúrgico em alvéolos de SHR. / Introduction: Arterial hypertension has been one of the world’s biggest health problems, with considerable alterations for cardiovascular and renal diseases. The bone tissue has an important role in support, protection and locomotion and is controlled by systemic factors like hormones and local factors, such as growth factors and cytokines. The Tartrate-resistant Acid Phosphatase (TRAP) is an enzyme that belongs to the Acid Phosphatases family and has an intracellular location, more specifically inside the lysosomal compartment of osteoclasts, macrophages and dendritic cells. It has been used as a histochemical marker of the osteoclast activity. Objectives: Evaluate TRAP protein’s expression in the dental alveoli of normotensive and hypertensive rats (SHR) treated or not treated with Atenolol. Methods: In this study, four groups of rats were used: 1) W (with no treatment), 2) WT (wistar treated with Atenolol), 3) S (SHR without treatment) and 4) ST (SHR treated with Atenolol), all of which underwent exodontia of the upper right incisor with euthanasia on the 7th, 14th, 21st and 28th day after the operation. The analysis of the biological mechanisms involved in the process of alveolar repair was obtained by the expression of TRAP proteins in the alveolar process through an immunohistochemistry technique. The results were analyzed through the average and its standard error. The parametric test ANOVA was applied with Tukey’s post-test were applied to evaluate the periods within each group and between the groups, considering the significant differences when p< 0,05. Results: The results demonstrated that TRAP staining increases in the dental alveoli of Wistar rats during all the post-surgical periods. TRAP staining increases only on the 14th day of alveolar recovery in the dental alveoli of non-treated SHR. Atenolol does not change the process of alveolar repair in Wistar rats, but Atenolol promoted the reduction of TRAP staining among SHR on the 14th day. Conclusion: Hypertension increases the expression of TRAP proteins on the 14th alveolar recovery postsurgical day and Atenolol promotes the reduction of the increased TRAP staining on the 14th postsurgical day in SHR’s alveoli.

Some TRP channels with altered expression in hypertensive rats may be new therapeutic targets for the control of hypertension. Aim: Compare the responses induced by Essential Oil of Mentha x villosa Hudson (OEMV), rotundifolone and menthol in Spontaneously Hypertensive rats (SHR) and normotensive Wistar Kyoto (WKY), evaluating the role of TRP channels. Methods and Results: In vivo (blood pressure measurement and heart rate), in vitro (measure of the frequency and force of contraction in the atria and the isometric tension in superior mesenteric arteries) and biochemical (PCR and Western blot) studies were used. The OEMV (3, 5, 10 and 20 mg/kg), the rotundifolone (10, 20 and 30 mg/kg) and the menthol (3, 5, 10 and 20 mg/kg) induced significant hypotensive and bradycardic response in non-anesthetized SHR and WKY rats. The reduction in the diastolic blood pressure was significantly greater than the decrease in the systolic blood pressure, suggesting a greater action on the vascular component of blood pressure. However, the significant bradycardic effect and reduction in the systolic blood pressure also suggest an action on the cardiac component. Furthermore, the decrease in the blood pressure and heart rate induced by rotundifolone and by menthol were significantly more potent in SHR. The action of OEMV, the rotundifolone and menthol in the right atrium (with spontaneous activity) and left (electrically stimulated) showed negative inotropic and chronotropic effects and culminating in complete inhibition of cardiac activity. Moreover, the negative inotropic effect was more potent in SHR and protein TRPM8 channel showed increased expression in the ventricles (left > right) and atria (left > right) of SHR rats. Also, OEMV, rotundifolone and menthol induced vasorelaxant response in superior mesenteric arteries of SHR and WKY rats, precontracted with PHE. The major mechanism involves the endothelium-independent route, which was more potent in SHR. The mechanism of the endothelium-independent vasorelaxant response induced by rotundifolone and menthol probably involves TRPM8 channels, which showed increased expression in SHR, and TRPC1, TRPC3 and TRPC6 channels. However, the response induced by menthol in WKY rats involves other TRP channels (probably TRPM6 and TRPM7). In addition, the flow cytometry showed an increase in [Ca2+]i induced by rotundifolone in SHR vascular myocytes, probably by activating of the TRPM8 channel. Conclusions: The hypotensive, bradycardia, negative inotropic and vasorelaxant responses induced by OEMV, rotundifolone and menthol were significantly more potent in SHR than in WKY rats. The mechanism of the endothelium-independent vasorelaxant response induced by rotundifolone and menthol involves TRPM8, TRPC (probably TRC1, TRPC3 and TRPC6), BKCa and CaV channels, but menthol may be acting in other TRP channels (probably TRPM6 and TRPM7) in WKY rats. The TRPM8 channel showed increased expression in SHR rats. Thus, the action of OEMV, rotundifolone and menthol on these channels can be related with the higher potency observed in SHR rats. / Os monoterpenos presentes em óleos essenciais de plantas atuam sobre canais Potencial Receptor Transiente (TRP). Alguns canais TRP com expressão alterada em ratos hipertensos podem ser novos alvos terapêuticos para o controle da hipertensão arterial. Objetivo: Comparar as respostas induzidas pelo Óleo Essencial de Mentha x-villosa Hudson (OEMV), pela rotundifolona e pelo mentol em Ratos Espontaneamente Hipertensos (SHR) e normotensos Wistar Kyoto (WKY), avaliando o papel de canais TRP. Métodos e Resultados: Estudos in vivo (medida de pressão arterial e freqüência cardíaca), in vitro (medida da freqüência e força de contração em átrios e da tensão isométrica em artérias mesentéricas superiores) e bioquímicos (PCR e Western blot) foram usados. O OEMV (3, 5, 10, 20 mg/kg), a rotundifolona (10, 20 e 30 mg/kg), e o mentol (3, 5, 10 e 20 mg/kg) induziram significativa resposta hipotensora e bradicárdica em ratos SHR e WKY não-anestesiados. A redução na pressão arterial diastólica foi significativamente maior do que a redução na pressão arterial sistólica, sugerindo uma maior ação sobre o componente vascular da pressão arterial. Entretanto, o significativo efeito bradicárdico e a redução na pressão arterial sistólica sugerem também uma ação sobre o componente cardíaco. Além disso, a diminuição na pressão arterial e freqüência cardíaca induzida por rotundifolona e por mentol foram significativamente mais potentes em ratos SHR. A ação do OEMV, da rotundifolona e do mentol em átrios direito (com atividade espontânea) e esquerdo (estimulado eletricamente) mostrou efeitos cronotrópico e inotrópico negativos e culminando na completa inibição da atividade cardíaca. Além disso, o efeito inotrópico negativo foi mais potente em ratos SHR e a proteína do canal TRPM8 mostrou expressão aumentada nos ventrículos (esquerdo > direito) e nos átrios (esquerdo > direito) de ratos SHR. O OEMV, a rotundifolona e o mentol também induziram resposta vasorrelaxante em artérias mesentéricas superiores de ratos SHR e WKY, pré-contraídos com FEN. O mecanismo majoritário envolve a via independente do endotélio, que foi mais potente em ratos SHR. O mecanismo da resposta vasorrelaxante independente do endotélio induzida por rotundifolona e mentol envolve provavelmente canais TRPM8, que apresentaram expressão aumentada em ratos SHR, e canais TRPC1, TRPC3 e TRPC6. Entretanto, a resposta induzida por mentol em ratos WKY envolve outros canais TRP (provavelmente TRPM6 e TRPM7). Além disso, a citometria de fluxo mostrou um aumento na [Ca2+]i induzido por rotundifolona em miócitos vasculares de ratos SHR, provavelmente por ativação de canais TRPM8. Conclusões: As respostas hipotensora, bradicárdica, inotrópica negativa e vasorrelaxante induzidas por OEMV, rotundifolona e mentol foram significativamente mais potentes em ratos SHR do que em ratos WKY. O mecanismo da resposta vasorrelaxante independente de endotélio induzida por rotundifolona e mentol envolve canais TRPM8, TRPC (provavelmente TRPC1, TRPC3 e TRPC6), BKCa e CaV, porém o mentol pode estar atuando em outros canais TRP (provavelmente TRPM6 e TRPM7) em ratos WKY. Os canais TRPM8 mostraram expressão aumentada em ratos SHR. Dessa forma, a ação do OEMV, da rotundifolona e do mentol sobre esses canais pode estar relacionada com a maior potência observada em ratos SHR.

β-Adrenergic signalization plays an important role in heart, regulating cardiac frequency and contractility. It is also involved in development of hypertension and heart hypertrophy. Spontaneous hypertensive rat strain is a common model for human essential hypertension, although the origin of blood pressure abnormalities in SHR remains unknown. Dysfunction in the regulation of fatty acid translocase Cd36 was suggested as a link to development of hypertension in SHR. Transgenic strain SHR-Tg19 (also known as SHR-Cd36) was obtained, harboring a wild type of FAT/Cd36. This thesis aimed to investigate key elements of β-adrenergic signaling in the heart of SHR-Tg19 compared to their SHR controls. Expression and distribution of β1- and β2-ARs were measured using radioligand binding and Western blot analysis along with expression of selected G proteins and expression and activity of adenylyl cyclase. Our experiments showed that there were no significant changes in the Gsα and Giα subunits expressions, along with the amount of β1-AR in both left and right ventricles, according to the Western immunoblotting, but radioligand binding showed an increase in the quantity of β-ARs, particularly β2 subtype. Characterisation of Grape and Grape pomace Polyphenolics : their Absorption and Metabolism and Potential Effects on Hypertension in a SHR Rat Model / Caractérisation d’Antioxydants Phénoliques de Raisins et de Marcs : leur Absorption, Métabolisme et Effets Potentiels sur l’Hypertension dans un Modèle de Rats SHR (Spontanément Hypertendus)

Cette étude examine les effets bénéfiques des marcs de raisins obtenus après vinification de différents cépages caractéristiques de la Vallée du Rhône, de leurs compositions phénoliques à leurs effets in vivo. Les raisins et leurs marcs respectifs des cépages Grenache (provenant de deux endroits différents [GRE1] et [GRE2]), Syrah (provenant de deux endroits différents [SYR1] et [SYR2]), Carignan (CAR), Mourvèdre (MOU), Counoise (COU) et Alicante (ALI) ont été étudiés. La comparaison des extraits de raisins et de leurs marcs respectifs montre que les marcs représentent une source importante d’antioxydants phénoliques malgré le processus de vinification. Les pépins et pellicules de marcs renferment des quantités appréciables de flavan-3-ols et anthocyanes. La distribution qualitative et quantitative des polyphenols dans les marcs de raisin présentent des différences significatives au travers des variétés et millésimes allant de 15% à 70% de polyphénols extraits. Les pépins de Grenache (GRE1), Syrah (SYR1) et les pellicules de Syrah (SYR1), Carignan et Alicante sont les fractions les plus intéressantes dues a la présence d’importantes quantités de flavan-3-ols (monomères, dimères et trimère) jusqu’à 8.7 mg/g MS et d’anthocyanes (glycosylées, acétylées et coumaroylées jusqu’à 17.40 mg/g MS, 1.57 mg/g MS et 2.38 mg/g MS respectivement). L’analyse des extraits aqueux (EAQ) et hydro-alcoolique 70% (EA70) indique que les pépins de Carignan et Syrah (SYR1) et les pellicules de Carignan et d’Alicante contiennent les plus fort taux en composés phénoliques et activités antioxydantes. L’efficacité in vivo sur l’hypertension de certains extraits a été évaluée utilisant un modèle de rat SHR. Les résultats des expériences in vivo démontrent que certains extraits administrés seuls ou en association avec le vérapamil possèdent un effet anti-hypertenseur. Cette capacité a été mise en évidence une fois que les extraits de pépins de marcs de GRE1 (EA70) et SYR1 (EA70) et de pellicules de marcs d’ALI (EA70) ont été administrés seuls ou lorsque les extraits de pépins de marcs GRE1 (EA70) et SYR1 (EAQ) et les pellicules de marcs d’ALI (EA70) et SYR2 (EAQ) ont été administrés en association avec le vérapamil. Cette étude met en évidence la biodisponibilité des extraits de pépins et de pellicules de marcs des rats SHR incluant à la fois le métabolisme de phase II et de la microflore intestinale. Les extraits de marcs administrés seuls et en association au vérapamil sont absorbés en tant que métabolites de phase II dérivant du métabolisme des monomères de flavan-3-ols. La détection de métabolites microbiens dérivés de flavan-3-ols, d’hydroxyphényl-γ-valérolactones sous leurs formes glucuronidés et sulfatés confirme l'absorption des métabolites dérivés des flavan-3-ols monomères et polymères et des conjugaisons supplémentaires dans le foie. De nombreux métabolites issus de la dégradation microbienne des hydroxyvalerolactones ont également été détectés. L’excrétion urinaire de ces métabolites représente une plus grande proportion de polyphénols ingérés comparé à ceux issus de métabolisme de phase II des monomères de flavan-3-ols, indiquant un rôle important des bactéries intestinales dans le métabolisme des molécules hautement polymérisées. Ces métabolites peuvent avoir exercé leurs effets biologiques lors de leur passage dans la circulation sanguine. Cette étude constitue une première étape de valorisation de marcs de raisins après vinification, en tant qu’activateur de vérapamil, un médicament couramment utilisé contre l’hypertension. Des quantités suffisantes de composés phénoliques subsistent dans les marcs, en particulier en terme de flavan-3-ols et d’anthocyanes, pour exercer des effets antihypertenseurs. En effet, selon le type et la composition des extraits, il est possible de moduler les effets antihypertenseurs en amplifiant ou en diminuant l’absorption des polyphénols et/ou du vérapamil. / This study investigated the beneficial potential effects of grape pomaces obtained after winemaking of different Mediterranean grape varieties from crude materials to their in vivo effectiveness. Grapes and their respective grape pomaces from six different V. vinifera L. cultivar were studied namely Grenache (from two different locations [GRE1 and GRE2]), Syrah (from two different locations [SYR1 and SYR2]), Carignan (CAR), Mourvèdre (MOU), Counoise (COU) and Alicante (ALI) grape varieties from the Rhône Valley. The comparison of several wine industry by-products with their respective grapes provided evidence that pomace remaining at the end of the winemaking process can be very rich sources of antioxidants. The quantitative and qualitative distribution of polyphenols by HPLC-PDA-Fluo-MS in grape pomaces showed significant differences through varieties and vintages varying from 15% to 70% of polyphenols extracted. Seeds from Grenache (GRE1), Syrah (SYR1) and skins from Syrah (SYR1), Carignan and Alicante were of particular interest because of their higher polyphenol contents in terms of flavan-3-ols (monomers, dimers and trimers) up to 8.7 mg/g DW and anthocyanins (glycosides, acetylated and coumaroylated derivatives up to 17.40, 1.57 and 2.38 mg/g DW, respectively). The investigation of aqueous and hydro-alcoholic 70% extracts of seeds from Carignan and Syrah (SYR1) and skins from Carignan and Alicante was carried out as they contained high levels of total phenols and antioxidant activity. Several extracts, were tested in order to evaluate their in vivo biological effects on hypertension using a spontaneously hypertensive rat (SHR) model. A series of different grape pomace extracts were tested in association with verapamil. All in vivo experiments demonstrated that some grape pomace extracts administrated with or without co-ingestion with verapamil possessed an anti-hypertensive activity. This was evident with GRE1 (EA70) seed pomace extract, SYR1 (EA70) seed pomace extract, ALI (EA70) skin pomace extract administrated alone and with GRE1 (EA70) seed pomace extract, SYR1 (EAQ) seed pomace extract, ALI (EA70) skin pomace extract and SYR2 (EAQ) skin pomace extract administrated in association with verapamil. Grape pomace extracts with or without co-ingestion with verapamil were absorb as phase II metabolites mainly including glucuronide, O-methyl glucuronide, sulfate, and O-methyl sulfate derivatives of (epi)catechin which arise from the metabolism of monomeric flavan-3-ols. The detection by HPLC-PDA-Fluo-MSn and GC-MS of microbial-derived metabolites of flavan-3-ols, hydroxyphenyl-γ-valerolactones in their glucuronide and sulfate forms confirmed the absorption of metabolites derived from both monomeric and polymeric flavan-3-ols from grape pomace extracts and subsequent post-absorption conjugation. Numerous metabolites derived from further microbial degradation of hydroxyvalerolactones were also detected. The urinary excretion of these metabolites accounted for a larger proportion of the total polyphenol ingested than phase II metabolites of monomeric flavan-3-ols, indicating the important role of intestinal bacteria in the metabolism of polymerized procyanidins. All these metabolites may have exerted biological effects during the period in which they circulated in the bloodstream. This study constitutes the first step of assessing grape pomace as an enhancer of the verapamil, an anti-hypertensive drug. Substantial levels of polyphenols, especially flavan-3-ols, procyanidins and anthocyanins, remain in pomace after the winemaking process in quantities sufficient to exert anti-hypertensive effects. In addition, according to the extract used and its composition, it is feasible to modulate anti-hypertensive effects by amplifying or decreasing polyphenols and/or verapamil absorption.

Avaliação histológica da reparação óssea em defeitos bicorticais no ângulo de mandíbula de ratos geneticamente hipertensos e de seus controles Wistar-Kyoto / Histological evaluation of bone repair in through-in-through defects of mandibular angle of the spontaneously hypertensive rats and controls Wistar-Kyoto

A presença da hipertensão arterial pode comprometer a qualidade da reparação óssea, pois a doença é caracterizada por alterações fisiopatológicas vasculares e do metabolismo mineral. Com o objetivo de avaliar a neoformação e o remodelamento ósseo, este trabalho investigou o processo da reparação óssea em ratos geneticamente hipertensos (SHR) e de seus controles Wistar-Kyoto (WKY). Defeitos bicorticais de 2mm de diâmetro no lado direito e de 5mm no lado esquerdo, foram realizados com trefinas na região do ângulo de mandíbula. Os animais, divididos em grupos de cinco indivíduos cada, foram sacrificados após 2, 3, 5, 10, 15, 30, 60 e 90 dias pós-operatórios; as mandíbulas foram removidas, fixadas em formol a 10%, descalcificadas em ácido fórmico a 20%, incluídas em parafina e as secções histológicas, de 7m de espessura, coradas com hematoxilina e eosina. As imagens foram capturadas com aumento de 40x e a área do defeito mensurada pelo programa de histometria digital Image J versão 1.4. A análise estatística revelou que não houve diferença significante na comparação entre as linhagens WKY e SHR (p = 0,884), independente dos períodos ou lados avaliados; entre períodos, nas linhagens WKY (p = 0,101) e SHR (p = 0,479), independente dos lados avaliados; entre períodos por linhagens no lado direito; e entre linhagens por lados, esquerdo com p = 0,466 e direito com p = 0,689, independente do fator período. Houve diferença estatisticamente significante entre o lado esquerdo e o direito (p < 0,001), independente das linhagens e períodos avaliados; entre os lados por linhagens, WKY e SHR, ambas com p < 0,001; entre períodos por linhagens no lado esquerdo, no qual o grupo WKY de 15 dias apresentou área menor que o grupo WKY de 60 dias e o grupo SHR de 10 dias, e o grupo WKY de 60 dias apresentou área maior que o grupo SHR de 30 dias e SHR de 60 dias. With the objective of evaluating the bone neoformation and remodeling, this study investigated the process of bone repair in spontaneously hypertensive rats (SHR) and their match controls Wistar-Kyoto (WKY). Through-in-through defects were done with trephine burs in the mandibular angle area, of 2mm diameter on the right side and 5mm diameter on the left side. The animals were divided into groups of five individuals each one and killed after 2, 3, 5, 10, 15, 30, 60 and 90 postoperative days; the mandibles were removed, fixed in 10% formalin solution, decalcified with 20% formic acid, and embedded in paraffin; the histological sections of 7m thickness were stained with hematoxylin and eosin. The images were captured with 40x magnification and the defect area was measured by the image processing program Image J version 1.4. The statistical analysis showed that there is no significant difference in the comparison of WKY and SHR strains (p = 0,884), independent of periods or sides; among periods, in the WKY strain (p = 0,101) and SHR one (p = 0,479), independent of sides; among periods by strains on the right side; and among strains by sides, left side with p = 0,466 and right side with p = 0,689, independent of periods. There is a significant difference between left and right side (p < 0,001), independent of strains and periods; between sides by strains, WKY and SHR, both with p < 0,001; among periods by strains on the left side, which WKY 15 days group showed an area smaller than WKY 60 days and SHR 10 days groups, and WKY 60 days group showed an area bigger than SHR 30 days and SHR 60 days groups. Despite the changes founded on the left side that could be attributed to the functional remodeling of the mandibular bone, there were no differences in the bone repair of 5mm and 2mm diameter defects between spontaneously hypertensive rats and their match controls Wistar-Kyoto.

A presença da hipertensão arterial pode comprometer a qualidade da reparação óssea, pois a doença é caracterizada por alterações fisiopatológicas vasculares e do metabolismo mineral. Com o objetivo de avaliar a neoformação e o remodelamento ósseo, este trabalho investigou o processo da reparação óssea em ratos geneticamente hipertensos (SHR) e de seus controles Wistar-Kyoto (WKY). Defeitos bicorticais de 2mm de diâmetro no lado direito e de 5mm no lado esquerdo, foram realizados com trefinas na região do ângulo de mandíbula. Os animais, divididos em grupos de cinco indivíduos cada, foram sacrificados após 2, 3, 5, 10, 15, 30, 60 e 90 dias pós-operatórios; as mandíbulas foram removidas, fixadas em formol a 10%, descalcificadas em ácido fórmico a 20%, incluídas em parafina e as secções histológicas, de 7m de espessura, coradas com hematoxilina e eosina. As imagens foram capturadas com aumento de 40x e a área do defeito mensurada pelo programa de histometria digital Image J versão 1.4. A análise estatística revelou que não houve diferença significante na comparação entre as linhagens WKY e SHR (p = 0,884), independente dos períodos ou lados avaliados; entre períodos, nas linhagens WKY (p = 0,101) e SHR (p = 0,479), independente dos lados avaliados; entre períodos por linhagens no lado direito; e entre linhagens por lados, esquerdo com p = 0,466 e direito com p = 0,689, independente do fator período. Houve diferença estatisticamente significante entre o lado esquerdo e o direito (p < 0,001), independente das linhagens e períodos avaliados; entre os lados por linhagens, WKY e SHR, ambas com p < 0,001; entre períodos por linhagens no lado esquerdo, no qual o grupo WKY de 15 dias apresentou área menor que o grupo WKY de 60 dias e o grupo SHR de 10 dias, e o grupo WKY de 60 dias apresentou área maior que o grupo SHR de 30 dias e SHR de 60 dias.
