Characterization of a charge injection device detector for atomic emission spectroscopy.

A Charge Injection Device (CID) detector has been evaluated as a detector for simultaneous multielement atomic emission spectroscopy. The CID was incorporated into a special liquid nitrogen cooled, computer controlled camera system. Electro-optical characterization of the CID and camera system included determination of readout noise, quantum efficiency, spatial crosstalk, temporal hysteresis, spatial response uniformity, and linear dynamic range. The CID was used as a spectroscopic detector for an echelle grating spectrometer equipped with a direct current plasma emission source. The spectrometer was a standard commercial instrument modified to provide a reduced image format more suitable for use with the CID detector. The optical characteristics of this spectrometer, including wavelength coverage, and optical aberrations are described. The spectroscopic system was evaluated with respect to detection limits, linear dynamic range, and accuracy in both single element and simultaneous multielement modes. Detection limits compared well to literature values reported for photomultiplier tube detector based systems under similar conditions. CID detection limits were superior in the near infrared and visible wavelength region, comparable in the middle UV, and higher in the far UV. The detection limits were determined to be limited by background radiation shot noise. Several elements of a certified standard reference material were simultaneously determined in order to assess the accuracy of the spectroscopic system. The results were highly accurate, even when operating near or below the 3σ limits of detection. Spectral interferences for elements were avoided by using several analytical lines for each element. The results of these investigations indicate that the CID is a superior multichannel detector for analytical atomic emission spectrometry. The capability to simultaneously monitor a wide, continuous spectral range with high spatial resolution, high dynamic range, low readout noise, and insignificant signal crosstalk is now possible. Many analytical benefits of this approach, such as the potential capability to perform rapid qualitative and semiquantitative analysis and the ability to select the optimum spectral lines for highly accurate quantitative analysis are now readily achievable.

Measurement of the temperature dependence of the Buckingham effect (electric-field-gradient-induced birefringence) in gases

The aim of this research project was to assemble an apparatus to measure the electric quadrupole moments of gas molecules using the technique of electricfield- gradient-induced birefringence, or the Buckingham effect. Comprehensive research by various workers in the field has shown that this technique provides the only direct means of obtaining the quadrupole moment of a molecule. Theory has shown that the most accurate determination of the electric quadrupole moment is through a study of the temperature dependence of the effect. This not only allows for the quadrupole moment to be obtained but also enables the temperature-independent quadrupole hyperpolarisability term to be extracted. Both the quadrupole moment and the hyperpolarisabilty provide valuable information in a variety of applications, including intermolecular forces, electrostatic potentials and non-linear optical phenomena. This thesis fully describes the apparatus used in these measurements, including a description of the custom built oven that allowed for measurements to be performed over a temperature range spanning from 25"C up to 200"C. Results for the quadrupole moments and quadrupole hyperpolarisabilities of carbon dioxide, carbon monoxide, nitrous oxide and hydrogen are presented, together with a quadrupole moment for carbonyl sulphide from room-temperature measurements. Evaluates and assesses the probes and associated probeware of three manufacturers using a variety of criteria that are essential information for a consumer. The probes and probeware examined include Pasco Scientific's ScienceWorkshop, Venier's LabPro, and Onset's HOBO. Three experiments were used to test each of the criteria for comparison. Based on the data in the study no manufacturer proved themselves to be clearly the best, and taken collectively the benefits and drawbacks balance out.

While the impact of this microscale heterogeneity has been investigated in the laboratory with microbial isolates and theoretical models, microbial ecologists have lacked adequate tools to interrogate microscale processes directly in the natural environment. Within this thesis I introduce three new technologies that enable interrogation of microbial processes at the microscale in natural marine communities. The IFCB-Sorter acquires images and sorts individual phytoplankton cells, directly from seawater, allowing studies exploring connections between the diversity of forms present in the plankton and genetic variability at the single-cell level. / The In Situ Chemotaxis Assay (ISCA) is a field-going microfluidic device designed to probe the distribution and role of motility behavior among microbes in aquatic environments. By creating microscale hotspots that simulate naturally occurring ones, the ISCA makes it possible to examine the role of microbial chemotaxis in resource acquisition, phytoplankton-bacteria interactions, and host-symbiont systems. Finally, the Millifluidic In Situ Enrichment (MISE) is an instrument that enables the study of rapid shifts in gene expression that permit microbial communities to exploit chemical hotspots in the ocean. The MISE subjects natural microbial communities to a chemical amendment and preserves their RNA in a minute-scale time series. / Leveraging an array of milliliter-volume wells, the MISE allows comparison of community gene expression in response to a chemical stimulus to that of a control, enabling elucidation of the strategies employed by marine microbes to survive and thrive in fluctuating environments. Affordances of Instrumentation in General Chemistry Laboratories

The purpose of this study is to find out what students in the first chemistry course at the undergraduate level (general chemistry for science majors) know about the affordances of instrumentation used in the general chemistry laboratory and how their knowledge develops over time. Overall, students see the PASCO™ system as a useful and accurate measuring tool for general chemistry labs. They see the probeware as easy to use, portable, and able to interact with computers. Students find that the PASCO™ probeware system is useful in their general chemistry labs, more advanced chemistry labs, and in other science classes, and can be used in a variety of labs done in general chemistry. Students learn the affordances of the probeware through the lab manual, the laboratory teaching assistant, by trial and error, and from each other. The use of probeware systems provides lab instructors the opportunity to focus on the concepts illustrated by experiments and the opportunity to spend time discussing the results. In order to teach effectively, the instructor must know the correct name of the components involved, how to assemble and disassemble it correctly, how to troubleshoot the software, and must be able to replace broken or missing components quickly. The use of podcasts or Web-based videos should increase student understanding of affordances of the probeware.
