Demonstration of a link between seminal plasma proteins and male fertility in the domestic fowl (Gallus domesticus)Al-Aghbari, Abdulwali M. 11 March 1992 (has links)
The objective of this research was to clarify the basis
of subfertility in Delaware roosters. It was anticipated
that a sensitive method would be needed to compare seminal
plasma protein composition between subfertile and fertile
roosters. Consequently, the applicability of two-dimensional
electrophoresis was tested as a tool for the
analysis of chicken seminal plasma proteins. Two-dimensional
electrophoresis resolved 95 ± 4.4 derivative
polypeptides from seminal plasma proteins of fertile
roosters, whereas one-dimensional electrophoresis resolved
only 23 ± 0.4. Thus, two-dimensional electrophoresis was
found to be a useful tool for seminal plasma protein
analysis. Seminal plasma composition was compared between
subfertile Delaware and fertile roosters. Seminal plasma
from subfertile roosters was characterized by an imbalance
of proteins, electrolytes, and amino acids (P<0.05).
Neither type of seminal plasma contained proteolytic
activity. No difference (P>0.05) was observed in seminal
plasma osmolality. Differences in seminal plasma
composition were attributed to a dysfunction of the
excurrent ducts of the testis. This realization lead to
experiments designed to modulate subfertility.
Hemicastration exacerbated (P<0.001) subfertility, whereas
supplementation of spermatozoa with seminal plasma proteins
from fertile roosters ameliorated (P<0.001) subfertility.
Addition of seminal plasma proteins from subfertile roosters
to spermatozoa from fertile roosters had no effect (P>0.05)
on fertility. Therefore, subfertility was attributed to
protein deficiency in seminal plasma rather than the
presence of some agent that induces subfertility. The study
of subfertile Delaware roosters has helped establish a link
between seminal plasma proteins and fertility in the

This study was conducted at the installation of the Rural Academic Unit-Tiahuanaco of the BCU, located in the community of Achaca, third municipal section of Ingavi province, department of La Paz. It is 57 km from the La Paz-Desaguadero international highway, at 68 degrees 42 minutes 28 seconds latitude South by 16 degrees 35 minutes 41 seconds longitude West, at an altitude of 3856 meters above sea level. The study went from October 2000 to September 2001. The study consisted of determining the concentrations of the biochemical components in llama seminal plasma at three ages. Components studied were glucose, inorganic phosphate, creatinine, total protein, albumin, globulins, cholesterol, calcium, potassium, sodium, and magnesium. Twelve male llamas of 3, 4, and 5 years were selected and acquired from the Choquecota area of the Carangas province, department of Oruro. Four animals were chosen at each age and were subjected to a training period of semen collection during 2 months, using the artificial hindquarters designed for this effect. The 6 that best responded to the training were used for the investigation. Eight collections were obtained from each animal over the course of the study, and they were used for laboratory analysis. The results were analyzed using a hierarchical factorial design that involved a mixed analysis (nested and crossed) of the factors of age and collections. (The averages of two collections corresponding to each week were analyzed.) Each weekly collection average per age was an experimental unit. Four experimental units were obtained for each age, and the analysis of the data was done with the SAS statistics package version 6.12. From the analyses done the following results were obtained: the concentrations of glucose (6.246 [plus or minus] 0.716 mg/dl), creatinine (3.459 [plus or minus] 1.27 mg/dl), cholesterol (67.28 [plus or minus] 18.21 mg/dl), potassium (8.249 [plus or minus] 1.78 mEq/L), and sodium (123.187 [plus or minus] 18.39 mEq/L) did not show significant differences between ages or collections (p>0.05). The concentrations of calcium (12.138 [plus or minus] 3.64 mg/dl) and magnesium (1.943 [plus or minus] 0.52 mEq/L) showed significant (p<0.05) differences in age only and not in collections. Globulins (1.574 [plus or minus] 0.51 g/dl) showed differences between collections (p<0.05) but not between ages. Total protein (3.732 [plus or minus] 0.45 g/dl), albumin (2.158 [plus or minus] 0.46 g/dl), and inorganic phosphate (9.42 [plus or minus] 2.42 mg/dl) showed differences both between ages and between collections (p<0.05).

In animal reproduction, another relevant factor for a good reproductive performance is feeding, being the probiotic the goal of unnumbered researchs reporting the use of them for weight gain, and food conversion. The present work had the objective of evaluating the winter and summer effects on the seminal plasma proteins, using SDS - PAG electroforesis and investigating the probiotic influence on weight, scrotum girth and spermiogram in Tabapuã bred bulls on range breeding. The 6 Kda proteic bands (81,81% in winter and 90,90% in summer) 9 Kda (90,90% in winter and 72,72% in summer) and 125 kda (81,81% in winter and 90,90% in summer) showed, in both seasons, the largest percentages among all the identified bands, the same happening with the 26 Kda band (100%) in summer. It was concluded that the seasons of the year can influence the presence or absence of seminal plasma proteins, however this has no relationship with the spermatic quality. The probiotic has not caused deleterious effect in the bulls' spermograms, and also has not exhibited a pro-action for weight gain and increasing of scrotal perimeter. / Na década de 90, iniciou-se o estudo do perfil protéico do plasma seminal e sua relação com a fertilidade, cujo objetivo era estudar a função de cada proteína no quadro espermático, nas espécies domésticas. Na reprodução animal, outro fator relevante, para um bom desempenho reprodutivo, é a alimentação, sendo os probióticos alvo de várias pesquisas relatando o uso do mesmo para o ganho de peso e conversão alimentar. O presente estudo teve o objetivo de avaliar o efeito do inverno e verão, sobre as proteínas do plasma seminal, por meio da eletroforese SDS-PAGE e investigar a influência do probiótico no peso, perímetro escrotal e espermiograma de touros da raça Tabapuã criados extensivamente. As bandas protéicas de 6 Kda (81,81% no inverno e 90,90% no verão), 9 KDa (90,90 no inverno e 72,72% no verão) e 125 Kda (81,81% no inverno e 90,90% no verão), apresentaram, nas duas estações, as maiores percentagens dentre todas as bandas identificadas, o mesmo ocorrendo com a banda de 26 KDa (100%) no verão. Conclui-se que: as estações do ano podem influenciar na presença ou ausência de proteínas do plasma seminal, entretanto isso não tem relação direta com a qualidade espermática. O probiótico não ocasionou efeito deletério no espermograma dos touros, no entanto, não demonstrou ação promotora para o ganho de peso e aumento do perímetro escrotal.
