Genetic characterization and utilization of multiple Aegilops tauschii derived pest resistance genes in wheat

Fritz / Aegilops tauschii, the D-genome donor of modern wheat, has served as an important source of genetic variation in wheat breeding. The objective of this research was to characterize and utilize multiple Ae. tauschii-derived pest resistance genes contained in the germplasm KS96WGRC40. Two Ae. tauschii-derived genes, H23 and Cmc4, provide resistance to the Hessian fly (HF) and wheat curl mite (WCM), respectively. A linkage analysis of a testcross population estimated 32.67% recombination between H23 and Cmc4 on chromosome 6DS in wheat indicating that the two genes are not tightly linked as previous mapping reports show. Haplotype data of recombinant lines and physical mapping of linked microsatellite markers located Cmc4 distal to H23. Haplotype data indicated that both KS89WGRC04 and KS96WGRC40 have the distal portion of 6DS derived from Ae. tauschii. Microsatellite primer pairs BARC183 and GDM036 were more useful than the previously published linked markers in identifying lines carrying Cmc4 and H23, respectively. Through phenotypic selection and advancement within the testcross population, three TC1F2:4 lines were identified as homozygous resistant for H23 and Cmc4 and have the complete terminal segment of 6DS from Ae. tauschii. Two lines are more desirable than the original germplasm releases and can serve as a source of resistance to both HF and WCM in an elite background. A linkage analysis of a segregating recombinant inbred line population identified an Ae. tauschii-derived gene of major effect conferring resistance to Septoria leaf blotch (STB) and another Ae. tauschii-derived gene of major effect conferring resistance to soil-borne wheat mosaic virus (SBWMV) in the germplasm KS96WGRC40. The STB resistance gene in KS96WGRC40 is located in the distal 40% of the short arm of chromosome 7D flanked by microsatellite markers Xgwm044 and Xbarc126. Two previously reported STB genes, Stb4 and Stb5, have been mapped to 7DS in the same region as the STB resistance gene in KS96WGRC40. The uniqueness of the STB resistance genes on 7DS is questionable. The SBWMV resistance gene in KS96WGRC40 is located on chromosome 5DL linked to microsatellite marker Xcfd010. The SBWMV resistance gene within KS96WGRC40 was derived from TA2397 via KS95WGRC33.

Etude phytochimique de quelques plantes extrêmophiles tunisiennes et exploration de leurs activités biologiques / Phytochimie study of some Tunisian extremophilic plants and exploration of their biological activities

L'approche écologique est une des stratégies de recherche de substances actives. Cette approche porte sur les plantes extrêmophiles, incluant les plantes halophytes et xérophytes, végétaux natifs des biotopes salins et arides. De par leur capacité à subsister dans un environnement contraignant, qui se traduit, entre autres, par l'acquisition d'un système antioxydant puissant, elles se révèlent être des réservoirs en biomolécules, notamment en composés phénoliques. Dans cette étude, nous nous sommes ainsi intéressés à huit plantes halophytes et xérophytes Tunisiennes pouvant avoir des activités biologiques en santé humaine et en santé du végétal. Limonium virgatum (Willd.) Fourr. (Plumbaginaceae), plante halophyte, a montré des activités antioxydantes et antibactériennes intéressantes qui se trouvent respectivement liées à un contenu en polyphénols élevé et à la présence de phénylpropanoïdes. Cirsium scabrum (Poir.) Bonnet & Barratte (Asteraceae), plante xérophyte, a montré une activité cytotoxique selective envers la lignée cancéreuse J774 mettant en avant l'efficacité de triterpènes de type cycloartane ; mais également un pouvoir antibactérien sur des souches de Dermabacter hominis, qui serait lié à la présence de flavones glycosylées. Juncus maritimus Lam. (Juncaceae), plante halophyte, s'est montrée être une source de dérivés phénanthréniques à activités originales. Le juncunol, a montré une activité prometteuse sur une souche de Streptococcus pyogenes résistante à l'amoxicilline. L'effusol actif sur le pathogène de la septoriose du blé (Zymoseptoria tritici), serait stimulé dans les rhizomes de la plante en conditions de salinité élevées. Enfin, le dehydrojuncusol s'est montré être un nouvel inhibiteur de la réplication du virus de l'hépatite C, capable d'agir sur des formes de résistance à certains traitement disponibles sur le marché. / Research strategies of active natural products include ecological approaches. Extremophile plants, and in particular, halophytes and xerophytes, salt and drought-resistant plants, are a case in point. As they develop an arsenal of adaptation responses, including a powerful antioxidant system, they prove to be a promising source of biomolecules, especially in phenolic compounds. In this study, an assessment of biological activities was carried out on eight Tunisian halophyte and xerophyte plants that would offer new applications in public health-care, as well as in agriculture. Limonium virgatum (Willd.) Fourr. (Plumbaginaceae), a halophyte plant, showed interesting antioxidant and antibacterial activities in accordance to a high polyphenol content and to the presence of phenylpropanoides, respectively. Cirsium scabrum (Poir.) Bonnet & Barratte (Asteraceae), a xerophyte plant, revealed a cytotoxic potential involving cycloartane-type triterpenes, and a promising activity against Dermabacter hominis strains which is linked to the presence of glycosylated flavones. Juncus maritimus Lam. (Juncaceae), a halophyte plant, should be referred to a source of phenanthrene derivativeswith original biological activities. Juncunol showed a high activity against an amoxicillin-resistant strain of Streptococcus pyogenes. Effusol was highly active against the wheat pathogen (Zymoseptoria tritici), for which high salinity conditions would stimulate its activity in the rhizomes of the plant. Most interestingly, dehydrojuncusol, has been introduced as a novel inhibitor of hepatitis C virus replication that is effective against resistant mutants for which some treatments available on the market became ineffective.

Plasticité de l'architecture du blé d'hiver modulée par la densité et la date de semis et son effet sur les épidémies de Septoria tritici

Les pratiques culturales modifient l'architecture des couverts de manière à augmenter ou diminuer le développement des épidémies mais les processus mis en jeu sont complexes ; des modèles mécanistes simulant l'interaction entre plante et pathogène devraient aider à les clarifier. Les modèles de Plantes Virtuelles, qui permettent de décrire explicitement la structure tridimensionnelle de la plante, semblent particulièrement prometteurs pour exprimer les effets de l'architecture de la plante sur le développement des épidémies. L'objectif de cette étude est d'examiner la possibilité de simuler l'effet de l'architecture des plantes sur le développement de la maladie en utilisant un modèle Plante Virtuelle. Dans ce travail, nous nous intéressons au pathosystème blé-Septoria tritici, dans lequel l'architecture joue un rôle important. En effet, les spores de Septoria tritici sont propagées par les éclaboussures de pluie depuis les feuilles infectées du bas du couvert vers les nouvelles feuilles saines. Notre travail s'est appuyé sur un modèle pré-existant d'épidémie de la septoriose, Septo3D. L'architecture du blé a été étudiée pour une gamme de densités et de date de semis. Les différences de phyllochrone entre traitements ont été dans une gamme susceptible de modifier le développement de la septoriose. Ces variations ont été représentées par un modèle descriptif qui tient compte du nombre de feuilles final et de la photopériode. Une description détaillée des variables d'architecture à l'échelle des organes et du couvert a fourni une documentation originale et complète sur la plasticité de l'architecture du blé. Ces données ont été utilisées pour paramétrer la description du blé dans Septo3D. Globalement, les traitements étudiés ont conduit à de fortes différences de la densité de végétation au cours du temps. Les dynamiques de développement de la septoriose ont été suivies pour trois traitements de densités contrastées. Les cinétiques de la maladie simulées par le modèle étaient conformes aux mesures expérimentales. Bien que, l'approche nécessite davantage de validation, les résultats confirment que l'approche Plante Virtuelle apporte un nouvel éclairage sur les processus et les caractéristiques des plantes qui impactent les épidémies. En conclusion, nous proposons quelques perspectives en vue de nouvelles applications et améliorations de l'approche.

Conselho Nacional de Desenvolvimento Científico e Tecnológico / Sunflower is an alternative specie to the system of no-tillage and crop rotation. His inclusion in this system provides soil improvements and interruption on cycle of pests and diseases, which are responsible for losses of productivity of the mainly sown crops. In addition to these benefits this specie has characteristics of increased tolerance to drought than traditional crops such as soybean and corn, maintaining satisfactory yields even in dry years considereds. For inclusion in the rotation and maintaining satisfactory yields the knowledge of the best sowing dates is necessary. As the weather has interannual variability, the risk analysis for the sowing dates is required, through a study of probability with a long series of weather data for better definition. This study aims to determine the sowing date of sunflower in which lower severity of alternaria and septoria leaf spots in the RS Central Region occurs. For it has adapted and calibrated an algorithm providing a forecasting system diseases, based on data from leaf wetness duration, air temperature during the leaf wetness period and rain, calculating the accumulated severity values (VSCA) throughout the cycle. In this adaptation and calibration data from six experiments performed at the Department of Plant Science since 2007. Hourly values of weather data from 1968 to 2011 was estimated from the default readings, and use this algorithm were simulated for 14 different dates of sowing spaced in 15 days, beginning on August 1 and ending on February 16 of each year. Due to the interannual variability of ENSO and their influence on weather conditions, crop seasons years was apart in Neutral, La Niña and El Niño. The data generated from the analysis of variance and the means of Scott- Knott test at 5% probability of error was taken. In addition, the VSCA were subjected to analysis of probability distribution, verifying that the Weibull, lognormal and normal distributions has the best represent the distribution of values of severity of leaf spots on sunflower, both for the entire cycle and for the subperiods emergency-end of anthesis and early reproductive-final stage of anthesis sunflower. It was also found that the period between early February and late April is more favorable to leaf spots, while sowing dates from August 1 to October 16, resulting in lower severity values, differing significantly from later sowings. In El Niño years suffer the greatest severities of disease, followed by La Niña years, and finally with the lowest values of VSCA for Neutrals years. / O girassol é uma espécie alternativa para o sistema de plantio direto e rotação de culturas. Sua inserção neste sistema proporciona melhorias edáficas e interrupção do ciclo de pragas e doenças causadoras de perdas de produtividade nas culturas agrícolas mais semeadas. Além destes benefícios, esta espécie possui características de maior tolerância ao déficit hídrico do que culturas tradicionais como a soja e o milho, mantendo produtividades satisfatórias mesmo em anos considerados secos. Para a sua inclusão no sistema de rotação e manutenção de produtividades satisfatórias é necessário o conhecimento das melhores datas semeadura. Como as condições meteorológicas tem variabilidade interanual, é necessária a análise de risco para uma mesma data de semeadura, através de um estudo de probabilidade com uma série longa de dados meteorológicos para sua melhor definição. Este trabalho objetiva determinar a data de semeadura do girassol na qual ocorre menor severidade das manchas foliares de alternária e septória, na Região Central do RS. Para isso adaptou-se e calibrou-se um algoritmo provendo um sistema de previsão de doenças, baseado em dados de duração do molhamento foliar, temperatura do ar durante o período de molhamento foliar, e chuva, obtendo-se os valores de severidade calculada acumulados (VSCA) durante todo o ciclo. Nesta adaptação e calibração utilizaram-se dados de seis experimentos realizados no Departamento de Fitotecnia desde o ano de 2007. Os dados meteorológicos estimados para valores horários de 1968 até 2011, a partir das leituras padrão, e a utilização deste algoritmo, foram simuladas para 14 diferentes datas de semeadura espaçadas em 15 dias, iniciando em 01 de agosto e finalizando em 16 de fevereiro de cada ano. Devido à variabilidade interanual do fenômeno ENOS e sua influência sobre as condições meteorológicas, os anos separados em Neutros, La Niña e El Niño. A partir dos dados gerados foi feita a análise de variância e o teste de médias de Scott-Knott a 5% de probabilidade de erro. Além disso, os VSCA obtidos foram submetidos a análise de distribuição de probabilidade, verificando-se que as distribuições Weibull, lognormal e normal representam melhor a distribuição dos valores de severidade máxima das manchas foliares no girassol, tanto para o ciclo completo quanto para os subperíodos emergência-final da antese e início da fase reprodutiva-final da antese do girassol. Verificou-se ainda que o período entre o início de fevereiro e o final de abril é mais favorável às manchas foliares, enquanto que as datas de semeadura de 01 de agosto a 16 de outubro resultam nos menores valores de severidade, diferindo significativamente das semeaduras posteriores. Em anos El Niño ocorrem as maiores severidades das doenças, seguido pelos anos de La Niña, e finalmente com os menores valores de VSCA para os anos Neutros.

Intérêt de la diversité architecturale des plantes cultivées pour limiter la progression épidémique de maladies foliaires à dispersion pluviale : cas de la septoriose au sein d'associations variétales de blé / Interest of architectural diversity of cultivated plants in order to limit the epidemic progression of splashed-dispersed leaf diseases : case of septoria tritici blotch in wheat cultivar mixtures

La culture d’associations de variétés sensibles et résistantes au sein d’une même parcelle permet de réduire la propagation des maladies fongiques foliaires aériennes. L’architecture des plantes a un impact sur la dispersion de spores et le microclimat, mais est rarement prise en compte dans la conception des associations. L’objectif de cette thèse était de comprendre le rôle joué par l’architecture dans des associations de variétés différant par leur niveau de résistance à une maladie dispersée par éclaboussement, la septoriose du blé, causée par Zymoseptoria tritici. Une expérimentation en conditions contrôlées a permis de quantifier des relations entre interception de spores et architecture des couverts. Des différences de densité entre couverts purs et associés ont donné lieu à une réduction du niveau de maladie sur les plantes sensibles cultivées en association par rapport à celles cultivées pures. Lors d’une expérimentation au champ, les associations de variétés de hauteurs de tiges contrastées étaient moins malades que celles ayant des hauteurs similaires. Ces résultats ont été mis en relation avec des effets de l’architecture sur la dispersion de spores et la durée d’humectation au sein des couverts. Une approche de modélisation spatialement explicite a permis d’identifier des mécanismes de dispersion par éclaboussement liés à l’architecture des couverts associés. Dans des associations de variétés de hauteurs différentes, la quantité d’inoculum éclaboussée dépendait de la surface foliaire présente au dessus des feuilles malades (effet parapluie). La quantité d’inoculum interceptée par un étage foliaire était liée à la différence de hauteur entre la source d’inoculum et l’étage foliaire (effet hauteur). Les différences de hauteur de plantes entre variétés d’une association ont modulé l’interception de spores par des feuilles résistantes (effet barrière). Nos résultats suggèrent qu’une prise en compte de l’architecture des variétés dans la conception des associations variétales permettrait de mieux maîtriser les maladies par éclaboussement. / Growing mixtures of susceptible and resistant cultivars in the same field makes it possible to reduce the propagation of airborne fungal plant diseases. Plant architecture has an impact on spore dispersal or microclimate, but is rarely taken into account in mixture design. The objective of this work was to understand the role of canopy architecture in mixtures of cultivar of different levels of resistance to a disease dispersed by rain-splash, septoria tritici blotch, caused by Zymoseptoria tritici. A controlled conditions experiment made it possible to quantify relationships between spore interception and canopy architecture. Differences of canopy density between pure stands and mixtures led to a reduction in disease on susceptible plants grown in mixture, compared to the susceptible pure stand. During a field experiment, mixtures of cultivars with contrasted stem height were less diseased than those with similar stem height. These results were related to the effect of canopy architecture on spore dispersal and leaf wetness duration. A spatially explicit modeling approach made it possible to identify splash dispersal mechanisms related to the architecture of mixed canopies. In mixtures of cultivar with diverse plant height, the amount of splashed inoculum depended on leaf area located above diseased leaves (umbrella effect). The amount of inoculum intercepted by a leaf layer was related to its difference of height between the inoculum sources (height effect). Differences of plant height between cultivars composinga mixture modulated the interception of spores by resistant leaves, providing an increased protection of susceptible leaves (barrier effect). Our results suggest that considering cultivar architecture in the design of cultivar mixtures could make it possible to improve the management of splash-dispersed diseases.
