DEVELOPMENT OF 15 PSI SAFE HAVEN POLYCARBONATE WALLS FOR USE IN UNDERGROUND COAL MINES

Following three major mining accidents in 2006, the MINER Act of 2006 was enacted by MSHA and required every underground coal mine to install refuge alternatives to help prevent future fatalities of trapped miners in the event of a disaster. The following research was performed in response to NIOSH’s call for the investigation into new refuge alternatives. A 15 psi safe haven polycarbonate wall for use in underground coal mines was designed and modeled using finite element modeling in ANSYS Explicit Dynamics. The successful design was tested multiple times in both half-scale and small scale using a high explosive shock tube to determine the walls resistance to blast pressure. The safe haven wall design was modeled for an actual underground coal mine environment to determine any responses of the wall within a mine. A full scale design was fabricated and installed in an underground coal mine to determine any construction constraints and as a final step in proof of concept for the safe haven design.

We're Not in Kansai Anymore: Designing for Reverse Culture Shock

I developed a resource to aid people going through reentry and reverse culture shock after returning from long stays abroad. Based on my experience with the JET Programme, I used North American JET alumni as my case study, but aimed to develop a solution that would be scalable and useful for people in other programs and situations. While JET and other similar programs assist and support participants in the initial journey abroad and during their stay, they tend to not provide much support when the participants return. This is a real issue because culture shock upon returning from an extended trip is stronger and generally unexpected than what’s experienced at the initial departure. Because of the increasing number of people going abroad for extended stays, this highlights a very real need for support, either from the organizing groups or from alumni of those groups and programs. I began my exploratory research with an extensive survey, where I gathered information on people’s situations prior to joining JET, various qualitative and quantative aspects of their time in Japan, and their experiences upon their return. I continued to conduct exploratory research to gather people’s personal experiences, and I worked closely with JET alumni during my generative and evaluative research phases as well. From the beginning, I leaned towards an online solution that would connect JET alumni regardless of distance or location. Though I considered other options, this approach was vindicated by the strong needs for “community” and “shared experiences” voiced by JET alumni throughout all my research. Ultimately I developed a plan for an online platform that enables people to share their experiences through writing and other media, as well as to communicate and connect easily with others. The name of this platform is okaeri, which means both “return” (verb) and “welcome” (greeting) in Japanese. Beyond the site’s structure and function, the key element is the visual-verbal rhetorical strategy throughout the content and layout, which will set an empathetic tone and perpetuate the sense of community that already exists among JET alumni.

A defining moment : Malaysian nurses' perspectives of transnational higher education

Transnational Higher Education (TNHE) post-registration top-up nursing degree programmes are relatively new in Malaysia and their impact in clinical settings is unknown. This research interprets Malaysian nurses’ experiences of such programmes and their perspectives of the extent TNHE theoretical knowledge has been applied in clinical settings. The contextual framework was established by drawing on a range of relevant dominant discourses, i.e. TNHE, nurse education, continuous professional development, theory-practice link in nursing, and culture and its influences, including coping with and adjusting to new ways of learning. Hermeneutic phenomenology and the ethnographic principle of cultural interpretation were used to explore the views of eighteen Malaysian nurses from two UK and one Australian TNHE universities (determined by convenience and snowball sampling methods) to enable data saturation. Semi-structured interviews were conducted to enable the nurses’ voices to define, describe and evaluate their TNHE experiences that were focused on personal and professional development, implementation and reaction of others towards change. In addition to the interviews, three threads of my own personal, professional and researcher experiences were reflected upon, to provide the contextual lens to shape the research process and situate the work firmly in the practice context. Data was analysed using thematic analysis. Four pre-determined key areas drawn from the literature were investigated and eight new sub-themes emerged. Findings indicated nurses’ improved self-confidence, knowledge, questioning skills and professionalism. The extent to which TNHE theory was applied in clinical practice was unable to be determined due to conflicting perceptions, contradicting views and restricted number of nurse-led examples. The main contribution this thesis offers to practice is what the voices of nurses tell about their experiences in TNHE programmes and in applying the taught theory in clinical settings. This study indicates enhanced application of theoretical knowledge in practice for improved quality and culturally competent patient care is unlikely to occur under current TNHE arrangements. Nurses’ motives for enrolling were mainly to obtain the high status western degree and the extrinsic benefits of a financial incentive and promotion. However, drawing on their resilience, nurses developed self and professional perspective transformation. The research provides new insights to inform continuous professional education policy for nurses, employers and the Malaysian Nursing Board, and can assist TNHE provider institutions to improve their programme delivery.

Evolution of Reproduction and Stress Tolerance in Brachionid Rotifers

Stress can be a driving force for new evolutionary changes leading to local adaptation, or may be responded to with pre-existing, ancestral tolerance mechanisms. Using brachionid rotifers (microzooplankton) as a study system, I demonstrate roles of both conserved physiological mechanisms (heat shock protein induction) and rapid evolution of traits in response to ecologically relevant stressors such as temperature and hydroperiod. Rapid evolution of higher levels of sex and dormancy in cultures mimicking temporary waters represents an eco-evolutionary dynamic, with trait evolution feeding back into effects on ecology (i.e., reduced population growth). I also reveal that prolonged culture in a benign laboratory environment leads to evolution of increased lifespan and fecundity, perhaps due to reduction of extrinsic mortality factors. Potential mechanisms (e.g., hormonal signals) are suggested that may control evolvability of facets of the stress response. Due to prior studies suggesting a role of progesterone signaling in rotifer sex and dormancy, the membrane associated progesterone receptor is assayed as a candidate gene that could show positive selection indicating rapid divergence. Despite some sequence variation that may contribute to functional differences among species, results indicate this hormone receptor is under purifying selection. Detailed analyses of multiple stress responses and their evolution as performed here will be imperative to understanding current patterns of local adaptation and trait-environment correlations. Such research also is key to predicting persistence of species upon introduction to novel habitats and exposure to new stressors (e.g., warming due to climate change). Perhaps one of the most intriguing results of this dissertation is the rapid, adaptive change in levels of sex and dormancy in a metazoan through new mutations or re-arrangements of the genetic material. This suggests species may be able to rapidly evolve tolerance of new stressors, even if standing genetic variation does not currently encompass the suite of alleles necessary for survival.

Exploration of themes evolving from the experiences of third culture kids

The purpose of this study was to explore how living outside of the passport country during the developmental years affected the experiences of adult Third Culture Kids (TCKs). Individual interviews were conducted with 15 adult TCKs who had lived at least one year outside of their passport country prior to age 18 and had returned to their passport country for at least one year. They were asked questions about how their experiences living outside of their passport country affected them. Data analyzed from the interviews generated 13 themes: (a) broader understanding of the world and the differences of people, (b) developing a support system of genuine relationships based on support and understanding, (c) development of an identity that integrates experiences and cultures, (d) feeling misunderstood, isolated, and different, (e) experience resulted in opportunities and skills sets that are not traditional in the passport country, (f) challenges of returning to the passport country, (g) internal struggles in relating to others and developing effective coping skills, (h) ability to negotiate change and a lack of fear of change, (i) difference in understanding of wealth and financial management, (j) view experience as positive and life-changing, (k) challenges in adjusting to a different culture, (l) variation in relationships with family, and (m) desire to travel and be in other places. These themes and their repeating ideas generated a grounded theory about the experiences of TCKs. This theory represents the relationships between the themes and the research questions. The theory consisted of six elements: (a) development of a global perspective, (b) affirmation of the complexity of relationships, (c) struggle to achieve cultural efficacy, (d) confirmation of the value of the experience, (e) challenge of acculturation, and (f) change as a source of achieving equilibrium. Findings from this study demonstrated that the experience of growing up outside of the passport country during the developmental years was perceived as a valuable and beneficial experience that significantly affected the participants. The results also identified challenges of the TCK experience and the influences these had in participants’ lives. Research and practice implications are presented. Results of this study may assist mental health professionals to understand the term TCK and the challenges with which they may present for treatment. / Department of Counseling Psychology and Guidance Services

The role of trauma support work in peace-building.

Current studies in peace-building suggest that effective peace-building approaches comprise of intervention strategies that value the interconnectedness of trauma and peace-building processes. However current research reports suggest that there is limited evidence supporting this notion. In addition, there is little information as to how to effectively integrate the two themes to achieve lasting peace. The research study aims to articulate the role of an integrated approach to peace-building to inform current practice and it serves to encourage the donor community to support initiatives which recognise the link between trauma and peace building. The research took a close look at experiences of participants who attended trauma recovery workshops conducted by Sinani (an isiZulu word meaning “we are with you”), the KwaZulu-Natal Programme for Survivors of Violence, the name of the Non-Governmental Organisation (NGO) for which the researcher works. In particular, it explored how participants who had attended these workshops could serve as catalysts to peace by examining their experiences of violence in relation to trauma and peace-building. In addition it attempted to explore experiences of participants who did not attend Sinani trauma recovery workshops and the possible implication this would have for trauma and peace-building. Furthermore it analysed recent research papers and reports which addressed trauma and peace-building from a psycho-social perspective. The proposed hypothesis is that if trauma support work is ignored in post conflict peace-building processes, certain survivors of past violence are at risk of becoming perpetrators of future violence. Integrating trauma support work in peace-building interventions will yield lasting peace. The emerging findings suggest several factors contribute to violence and peace-building. Children’s exposure to violence, the extent of trauma and certain aspects of the criminal justice system have been described by participants as factors that contribute to violence. Equally participants suggested a competent leadership collective, functioning safety and security structures as valuable contributions to peace. Other valuable insights were shared by participants on the role of spirituality as well as indigenous cultural rituals valuable in the trauma and peace-building field. / Thesis (M.A.)-University of KwaZulu-Natal, Durban, 2013.

Examination of Cadmium-Induced Heat Shock Protein Gene Expression in Xenopus laevis A6 Kidney Epithelial Cells

Cadmium is a highly toxic chemical and has been classified by the International Agency for Research on Cancer as a human carcinogen. Cadmium is abundant in the environment, at specific work places, and in food and water. Toxicological responses to cadmium exposure include respiratory diseases, neurological disorders and kidney damage. The present study examined the effects of cadmium on heat shock protein (HSP) accumulation in Xenopus laevis A6 kidney epithelial cells. HSPs are molecular chaperones involved in protein folding and translocation. In response to environmental stress these proteins bind to unfolded protein and inhibit their aggregation. Stress-inducible hsp gene transcription is mediated by the heat shock promoter element (HSE), which interacts with heat shock transcription factor (HSF). In the present study, hsp30 and hsp70 mRNA and protein were induced by heat shock, as determined by northern and western blot analysis. Exposure of A6 cells to cadmium chloride also induced the expression of hsp genes. For example, northern and western blot analysis revealed that exposure of A6 cells to cadmium chloride induced the accumulation of hsp30 and hsp70 mRNA and their respective proteins. Western blot analysis also revealed that A6 cells recovering from a cadmium chloride treatment retained relatively high levels of HSP30 and HSP70 protein accumulation over 24 h after the removal of the stress. Treatments combining a mild heat shock and cadmium chloride resulted in a synergistic increase in hsp30 and hsp70 gene expression at mRNA and protein levels. Further experiments in which two stressors were combined revealed that synergistic effects occurred with varying cadmium concentrations and different temperatures. Immunocytochemistry and confocal microscopy were used to confirm the results attained from western blot analysis. Further, this technique allowed the determination of intracellular localization of HSP30 in A6 cells and the examination of cellular morphology and cytoskeletal structure during cadmium chloride treatments. A 2 h heat shock at 33??C resulted in the accumulation of HSP30 in the cytoplasm, whereas a 2 h heat shock at 35??C resulted in some HSP30 accumulation in the peripheral region of the nucleus. This is in contrast to cells treated with cadmium chloride, where HSP30 accumulation was restricted to the cytoplasm. A 14 h 50 ??M cadmium chloride treatment resulted in the accumulation of HSP30 in approximately 10% of cells. The proportion of cells displaying HSP30 accumulation increased to 80% and 95% in cells treated with 100 ??M and 200 ??M, respectively. HSP30 accumulation frequently occurred in large granular structures. High concentrations of cadmium chloride resulted in cell membrane ruffling at areas of cell-cell contact, as well as actin disorganization. This study characterized the pattern of hsp gene expression, accumulation and localization under various cadmium chloride conditions. These results suggest that hsp30 and hsp70 gene expression can be used as potential biomolecular markers for cadmium exposure.

Analysis of heat shock protein 30 gene expression and function in Xenopus laevis A6 kidney epithelial cells

Heat shock proteins (HSPs) are molecular chaperones that assist in protein synthesis, folding and degradation and prevent stress-induced protein aggregation. The present study examined the pattern of accumulation of HSP30 and HSP70 in cells recovering from heat shock as well as the effect of proteasome inhibition on cytoplasmic/nuclear and endoplasmic reticulum (ER) molecular chaperone accumulation, large multimeric HSP30 complexes, stress granule and aggresome formation in Xenopus laevis A6 kidney epithelial cells. Initial immunoblot analysis revealed the presence of elevated levels of HSP30 after 72 h of recovery. However, the relative levels of HSP70 declined to near control levels after 24 h. The relative levels of both hsp30 and hsp70 mRNA were reduced to low levels after 24 h of recovery from heat shock. Pretreatment of cells with cycloheximide, a translational inhibitor, produced a rapid decline in HSP70 but not HSP30. The cycloheximide-associated decline of HSP70 was blocked by the proteasomal inhibitor, MG132, but had little effect on the relative level of HSP30. Also, treatment of cells with the phosphorylation inhibitor, SB203580, in addition to cycloheximide treatment enhanced the stability of HSP30 compared to cycloheximide alone. Immunocytochemical studies detected the presence of HSP30 accumulation in a granular pattern in the cytoplasm of recovering cells and its association with aggresome-like structures, which was enhanced in the presence of SB203580. To verify if proteasome inhibition in A6 cells induced the formation of similar HSP30 granules, immunoblot and immunocytochemical analyses were performed. MG132, celastrol and withaferin A enhanced ubiquitinated proteins, inhibited chymotrypsin-like activity of the proteasome and induced the accumulation of cytoplasmic/nuclear HSPs, HSP30 and HSP70 as well as ER chaperones, BiP and GRP94 and heme oxygenase-1. Northern blot experiments determined that proteasome inhibitors induced an accumulation in hsp30, hsp70 and bip mRNA but not eIF1α. The final part of this study demonstrated that treatment of A6 cells with proteasome inhibitors or sodium arsenite or cadmium chloride induced HSP30 multimeric complex formation primarily in the cytoplasm. Moreover, these stressors also induced the formation of RNA stress granules, pre-stalled translational complexes, which were detected via TIA1 and polyA binding protein (PABP), which are known stress granule markers. These stress granules, however, did not co-localize with large HSP30 multimeric complexes. In comparison, proteasome inhibition or treatment with sodium arsenite or cadmium chloride also induced the formation of aggresome-like structures, which are proteinaceous inclusion bodies formed as a result of an abundance of aggregated protein. Aggresome formation was identified by monitoring the presence of vimentin and γ-tubulin, both of which are cytoskeletal proteins and serve as markers of aggresome detection. Aggresome formation, which was also verified using the ProteoStat assay, co-localized with large HSP30 multimeric complexes. Co-immunoprecipitation experiments revealed that HSP30 associated with γ-tubulin and β-actin in cells treated with proteasome inhibitors or sodium arsenite or cadmium chloride suggesting a possible role in aggresome formation. In conclusion, this study has shown that the relative levels of heat shock-induced HSP30 persist during recovery in contrast to HSP70. While HSP70 is degraded by the ubiquitin-proteasome system, it is likely that the presence of HSP30 multimeric complexes that are known to associate with unfolded protein as well as its association with aggresome-like structures may delay its degradation. Finally, proteasome inhibition, sodium arsenite and cadmium chloride treatment of A6 cells induced cytoplasmic/nuclear and ER chaperones as well as resulting in the formation stress granules and aggresome-like structures which associated with large HSP30 multimeric complexes.

Effects of termination shock acceleration on cosmic rays in the heliosphere / U.W. Langner

The interest in the role of the solar wind termination shock (TS) and heliosheath in cosmic ray (CR) modulation studies has increased sigm6cantly as the Voyager 1 and 2 spacecraft approach the estimated position of the TS. For this work the modulation of galactic CR protons, anti-protons, electrons with a Jovian source, positrons, Helium, and anomalous protons and Helium, and the consequent charge-sign dependence, are studied with an improved and extended two-dimensional numerical CR modulation model including a TS with diffusive shock acceleration, a heliosheath and drifts. The modulation is computed using improved local interstellar spectra (LIS) for almost all the species of interest to this study and new fundamentally derived diffusion coefficients, applicable to a number of CR species during both magnetic polarity cycles of the Sun. The model also allows comparisons of modulation with and without a TS and between solar minimum and moderate maximum conditions. The modulation of protons and Helium with their respective anomalous components are also studied to establish the consequent charge-sign dependence at low energies and the influence on the computed p/p, e-/p, and e-/He. The level of modulation in the simulated heliosheath, and the importance of this modulation 'barrier' and the TS for the different species are illustrated. From the computations it is possible to estimate the ratio of modulation occurring in the heliosheath to the total modulation between the heliopause and Earth for the mentioned species. It has been found that the modulation in the heliosheath depends on the particle species, is strongly dependent on the energy of the CRs, on the polarity cycle and is enhanced by the inclusion of the TS. The computed modulation for the considered species is surprisingly different and the heliosheath is important for CR modulation, although 'barrier' modulation is more prominent for protons, anti-protons and Helium, while the heliosheath cannot really be considered a modulation 'barrier' for electrons and positrons above energies of ~150 MeV. The effects of the TS on modulation are more pronounced for polarity cycles when particles are drifting primarily in the equatorial regions of the heliosphere along the heliospheric current sheet to the Sun, e.g. the A < 0 polarity cycle for protons, positrons, and Helium, and the A > 0 polarity cycle for electrons and anti-protons. This study also shows that the proton and Helium LIS may not be known at energies <~ 200 MeV until a spacecraft actually approaches the heliopause because of the strong modulation that occurs in the heliosheath, the effect of the TS, and the presence of anomalous protons and Helium. For anti-protons, in contrast, these effects are less pronounced. For positrons, with a completely different shape LIS, the modulated spectra have very mild energy dependencies <~ 300 MeV, even at Earth, in contrast to the other species. These characteristic spectral features may be helpful to distinguish between electron and positron spectra when they are measured near and at Earth. Expression and function of heat shock factors in zebrafish (Danio rerio)

Heat shock proteins (hsp) and heat shock transcription factors (HSF) have important roles in the development of the eye lens. Our lab previously demonstrated that knockdown of hsp70 gene expression using morpholino antisense technology (MO) resulted in a small lens phenotype in zebrafish (Danio rerio) embryos. A less severe phenotype was seen with knockdown of hsf1, suggesting other factors that regulate hsp70 are involved during lens formation. Both HSF1 and HSF4 are known to play a role in mammalian lens development. An expressed sequence tag encoding zebrafish HSF4, named hsf4a, has been identified and a second splice variant, hsf4b, has been predicted in the Ensembl database. The objectives of this thesis were to characterize the zebrafish HSF4 and compare its expression to other HSFs as well as investigate its role in lens development. Analysis of zebrafish HSF4 sequence was performed using standard in silico analytical software. The deduced amino acid sequence of HSF4a shares structural similarities with mammalian HSF4 including the lack of an HR-C domain. This domain is absent due to a C-terminal truncation within zebrafish HSF4a relative to the mammalian protein. HSF4b is identical to the HSF4a sequence with the exception of an additional 155 amino acids at the carboxyl end of the protein which contains an HR-C domain, unlike mammalian HSF4. Surprisingly, electrophoretic mobility shift assays (EMSA) demonstrated that the binding affinity of zebrafish HSF4 to discontinuous HSEs is more similar to HSF1 than to other HSF4 proteins. The amino acid sequence of zebrafish HSF4 DNA binding domain was also more similar to HSF1 than other HSF4 proteins. These results, along with a phylogenetic analysis of HSF proteins from eleven species, suggest that HSF1 was an evolutionary precursor of HSF4 and that functions of this protein may differ between zebrafish and mammals. The expression level for each of the three zebrafish HSFs was determined in adult tissues and in developing embryos by quantitative reverse transcription polymerase chain reaction (qPCR) analysis. Expression of both hsf4 transcripts was observed predominantly in the eye but only observed in developing embryonic tissue at 60 hours post fertilization or later. This, together with the lack of an observable phenotype following MO knockdown of hsf4, suggests that HSF4 likely has a role in later stages of lens development. Additionally, hsf1 and hsf2 expression were detected in all tissues and in all stages of development as well as being present as maternal transcripts in zebrafish eggs. The results presented in this thesis demonstrate that while zebrafish HSFs share some similarity with HSF proteins from other species, they also have structural characteristics and expression patterns unique to the zebrafish.
