SK Channel Modulators as Drug Candidates and Pharmacological Tools

The small- and intermediate-conductance Ca2+ activated K + (SK/IK) channels play a fundamental role in the regulation of neurons in the central nervous system. In animal models, SK/IK channel positive modulators have been shown to be effective in reducing the symptoms of neurological diseases such as ataxia. Ataxia is a lethal neurological rare disease characterized by lack of balance and incoordination of muscle movements, often as a result of cerebellar or spinocerebellar neurodegeneration. SK/IK channel modulators have been developed over the past few decades. Currently available modulators are often weak in potency. Lack of knowledge about the binding site for the compounds is the main reason hindering the development of more potent and effective therapeutics targeting SK channels. Dr. Zhang and his colleagues recently discovered the binding pocket for these positive modulators of SK/IK channels. This pocket is located at the interface between the channel and calmodulin. Dr. Zhang and his colleagues performed screening of a large number of compounds in silico, to find those fitting into the binding pocket. I performed electrophysiological recordings to evaluate the efficacy and the potency of these modulators on SK2 channels. We discovered a correlation between the total binding energy values calculated from the structures and the potencies determined from electrophysiological recording.

A comparative membrane surface analysis between two human hepatocarcinoma cell lines ( SK-HEP-1 and Hep G2 cells ) using Atomic Force Microscope

Atomic force microscopy (AFM) can be used to acquire high-resolution topographical images of surfaces, but has the additional capability of detecting the local nanometer scale mechanical properties. For these reasons, it becomes a standard research tool in the surface science recently. In this paper, we used AFM to measure the several properties of two different human hepatocellular carcinoma cell lines, Hep G2 ( known as well differentiated and more highly carcinomatous hepatoma cell lines ) and SK-HEP-1 ( known as poorly differentiated and more lightly carcinomatous hepatoma cell lines ) cells fixed on the glass substrate, which including the surface morphology and the relationship between the cantilever deflections and loading forces ( force curve ). Considered the heterogeneous characteristics of the cell surface, the preferred experimental method is to make pixel-by-pixel force curves in a designated area ( force map ) , both adhesion forces and elasticity associated with different locations on the cell surfaces will be obtained. Finally, we use Hertzian model to calculate Young's modulus of Hep G2 and SK-HEP-1 respectively. Based on these results, we can understand the surface properties of two human hepatocarcinoma cell lines with different differentiated stage. The results showed the difference of the morphology, height, cell migration, degree of cell aggregation, roughness, elasticity, adhesive force of two cells. SK-HEP-1 cell has the wide distance of the folds, better cell migration, homogeneous properties of elasticity. It can be assumed that the SK-HEP-1 cells have a dense network structure of actin filaments under the cell membrane like branches (branched networks); Hep G2 cell has the narrow distance of the folds, poor cell migration, heterogeneous properties of elasticity. It can be assumed that the Hep G2 cells have the individual actin filaments and cross-linked network structure of actin filaments under the cell membrane. The above results can be speculated that the elastic properties of the membrane surface will be influenced of actin filaments.

TSP - Infrastructure for the Traveling Salesperson Problem

The traveling salesperson or salesman problem (TSP) is a well known and important combinatorial optimization problem. The goal is to find the shortest tour that visits each city in a given list exactly once and then returns to the starting city. Despite this simple problem statement, solving the TSP is difficult since it belongs to the class of NP-complete problems. The importance of the TSP arises besides from its theoretical appeal from the variety of its applications. In addition to vehicle routing, many other applications, e.g., computer wiring, cutting wallpaper, job sequencing or several data visualization techniques, require the solution of a TSP. In this paper we introduce the R package TSP which provides a basic infrastructure for handling and solving the traveling salesperson problem. The package features S3 classes for specifying a TSP and its (possibly optimal) solution as well as several heuristics to find good solutions. In addition, it provides an interface to Concorde, one of the best exact TSP solvers currently available. The purpose of my Master's Thesis is to determine the value of the SK Slavia Praha a.s. as of 30.06.2013. I focus on comparing differences between valuation of classical manufaturing company and valuation of sport club in theoretical part. I describe suitable methods for valuation of company and apparatus, that I chose to valuation of club of course in theoretical part. I use financial analysis to analyse main indicators, which affect the value of company in practical part. I use yield and property method for valuation. Property method is more meaningful due to the fact that company does not have profit in recent years. Sports club in the Czech Republic are not traded on stock market unfortunately, so we can't compare result with anything. On the other hand Slavia changed owners recenty unfortunately, so estimate value could be based on the price which current owner bought it, but this value is only for comparison, relative to situation which club was at that time.

Sports spectators is free entertainment for millions of people around the world. Here I will focus on football spectators who have in their heart the club SK Slavia Praha, who is one of the oldest football club in our country and have a fan base. As the name suggests, I will be interested in the phenomenon of a football fan, which differs from the viewer, among other things, that he identifies with his club. The main aspect of this work will be a comparison of two historical sections, the period before and after the fall of the communist regime in 1989 and Police supervision is connected. I will also be interested in the relationship to the biggest rival within AC Sparta Praha, or the possibility of club artifacts, which are now available in a wealth of, but always be. Even fans have their asociations. Aside from the fact that they are associated fan support such as an expert and tribune, which have great influence as a club support and at the end of the work focusing on their relationship to the club, why they chose Slavia and what it means for them. For this work I chose the method of qualitative research, which oral history and field research, which I found due to the nature of the work appropriate. 1 This MA thesis starts to explore a new exciting part of socialization theory, by applying the research into socialization in the management of the Czech football club SK Slavia Praha. This is done trough the evaluation of the end of the year 2007, when SK Slavia Praha for the first and last time played the group stage of UEFA Champions League. This MA thesis tries to prove that there was indeed a transfer of norms and values from UEFA to SK Slavia Praha. Next to this, this MA thesis also tries to prove that the previous socialization is not anymore apparent in the club, since the contacts with UEFA (and ECA) have since then lost because of the lack of international matches played by SK Slavia Praha. This research is done through interviews with the employees of the club, both the ones working in the club in 2007, and the ones who are employed in 2014. Molekulare Charakterisierung und Expressionsanalyse spannungsabhängiger und kalziumsensitiver Kaliumkanäle aus dem ZNS der Regenbogenforelle (Oncorhynchus mykiss)

Kaliumkanäle ermöglichen es, den geladenen Kaliumionen selektiv die hydrophobe Lipidphase von Zellmembranen zu passieren. Sie operieren in ihren Funktionen als Antagonisten von Natrium- und Kalziumkanälen, um die elektrische Erregbarkeit von Zellen zu kontrollieren. Im ZNS der Vertebraten existiert eine Vielzahl verschiedener Kaliumkanäle, welche die zeitliche Struktur von Aktionspotentialen festlegen. Im Rahmen der vorliegenden Dissertation wurden bisher unbekannte Kanäle aus den Familien der spannungsgesteuerten Kv1-/Kv3- und der kalziumsensitiven SK-/BK-Kaliumkanäle aus dem ZNS der Regenbogenforelle kloniert. Diese wurden durch heterologe Expression in Insekten- und Säugerzellen einer biophysikalischen und pharmakologischen Charakterisierung zugeführt. Gegen fünf der bekannten Kanäle wurden spezifische Antikörper hergestellt. Mit Hilfe immunhistochemischen und PCR-Techniken konnte die gewebetypische und entwicklungsspezifische Expression der Kaliumkanäle untersucht werden. Mit den in dieser Arbeit präsentierten Daten konnte die Grundlage für eine differentielle Betrachtung der funktionellen Bedeutung und des Expressionsmusters der entsprechenden Kaliumkanäle im nativen Kontext gelegt werden.
