Transgenerational Effects of Early Exposure to Soy Isoflavones on Reproductive Health and Bone Development in CD-1 Mice

Previous studies showed that early exposure to soy isoflavones resulted in improved bone mineral density (BMD) and bone quality that resulted in stronger bones in CD-1 mice. This study investigated whether the benefits to bone health are transferred to second generation (F2) females and if there are any adverse effects on reproductive health. First generation (F1) female CD-1 mice received subcutaneous injections of the isoflavones daidzein (DAI) and genistein (GEN) or corn oil from postnatal day (PND) 1 to 10 or 21. F1 and F2 treated-females experienced earlier pubertal onset and lengthened anogenital distance but only F1 had reduced fertility, histological abnormalities in the uterus and ovaries, and altered estrous cycling. F2 had higher BMD and stronger bones at 4 months of age. In conclusion, early life exposure to soy isoflavones compromise reproductive function but confer a transgenerational benefit to bone development in CD-1 mice.

Soy Isoflavone Supplementation Does Not Alter Distribution of Circulating Lymphocytes or Natural Killer Cell Activity in Postmenopausal Women

A growing body of evidence has demonstrated that soy isoflavone consumption may protect against the development of various chronic diseases. This defense could be linked to isoflavone-induced alterations in immune function. However, to date, no study has examined the effect of soy isoflavone supplementation on human immunity in vivo. Establishing whether isoflavones affect immunity in aging adults is particularly relevant since compromised immune function has been observed in this population. Therefore, the purpose of this double-blind, placebo-controlled, 4-wk intervention trial was to investigate whether supplementation with soy isoflavones influenced the distribution and/or function of specific lymphocytes in postmenopausal women. Healthy postmenopausal women (50-69 y), who were not using hormone replacement therapy, were randomly divided into 2 treatment groups. The experimental group (n=9) consumed two-50 mg soy isoflavone tablets/d for 4 wk, while the control group (n=9) received placebo tablets. Fasting blood samples were drawn at baseline and on d 28 to assess distribution of T-helper cells (CD3+CD4+), T-cytotoxic cells (CD3+CD8+), total T lymphocytes (CD3+), B lymphocytes (CD19+) and natural killer (NK) cells (CD16+CD56+) via flow cytometry. Cytotoxicity of NK cells was quantified based on lactate dehydrogenase release of lysed K562 cancer cells following co-culture with NK cells from subjects. Analysis of plasma isoflavone concentrations by HPLC demonstrated a significant increase (p<0.005) in plasma genistein concentration in the experimental group after 4 wk of supplementation. However, there was no alteration in lymphocyte distribution or NK cell activity in response to isoflavone supplementation, suggesting that short-term soy isoflavone supplementation does not alter these parameters of immunity in healthy postmenopausal women. / Master of Science

O hipoestrogenismo decorrente da menopausa acelera o envelhecimento da pele. Apesar de possuir efeitos positivos na pele, dois grandes estudos apontaram para os efeitos adversos da TH (terapêutica hormonal) estroprogestiva. Os fitoestrógenos são utilizados como terapia alternativa para mulheres com contraindicação à TH clássica. Dentre os fitoestrógenos, as isoflavonas daidzeína (D) e genisteína (G) são encontradas em pequenas quantidades na soja e possuem capacidade de promover efeitos benéficos na pele quando administradas por via oral e subcutânea. Entretanto, nenhum estudo investigou se estas isoflavonas podem aumentar o colágeno da pele quando administradas por via tópica, e se este efeito está relacionado com os receptores de estrógeno. Desta forma, o objetivo deste trabalho foi desenvolver formulações cosméticas contendo extrato de soja biotransformado pelo fungo Aspergillus awamori e analisar o potencial estrogênico deste extrato na produção de colágeno. Para contemplar nosso objetivo, foram produzidos quatro diferentes tipos de extrato de soja biotransformado, e após definir qual o extrato com maior concentração das isoflavonas (D e G), este extrato (ESBE) foi submetido a testes de citotoxicidade em células L929 (mouse fibroblast cell line - clone 929 da cepa L) e HDFa (Human Dermal Fibroblast, adult). Na segunda etapa foram desenvolvidas formulações cosméticas do tipo gel, creme e gel-creme incorporadas com ESBE, as quais foram utilizadas na realização de testes de estabilidade preliminar e em ensaio de permeação e retenção cutânea. Finalmente, foi realizado teste de eficácia para avaliar a produção de colágeno após tratamento com ESBE em cultura de fibroblastos humanos primários (HDFa). Os resultados demonstraram que a biotransformação é um processo eficaz para aumentar a concentração de D e G; o ESBE não demonstrou citotoxicidade em células L929 e HDFa; as formulações incorporadas com ESBE demonstraram possuir estabilidade preliminar; a formulação do tipo gel demonstrou ser a melhor opção para incorporação do ESBE visto que facilitou a retenção das isoflavonas na pele sem permitir a permeação de compostos para o líquido receptor; e finalmente, o ESBE é capaz de estimular a produção de colágeno em fibroblastos primários nas concentrações de 59 e 1,33?g/mL. Concluímos que o ESBE estimula a produção de colágeno, principalmente na concentração de 1,33?g/mL. Phytoestrogens are used as an alternative therapy for women with contraindications to classical HRT. The isoflavones daidzein and genistein are phytoestrogens found in small amounts in soybean grains and with ability to promote beneficial effects on the skin when administered orally and subcutaneously. However, no studies have investigated whether these isoflavones may increase skin collagen content when administered topically, and if this effect is associated with the estrogen receptors. Thus, the aim of this study was to develop cosmetic formulations containing soybean extract biotransformed by the fungus Aspergillus awamori and analyze the estrogenic potential of this extract on the production of collagen. To achieve our purpose, four different types of biotransformed soybean extract were produced, and after define which extract has the highest concentration of isoflavones (D and G), this extract (ESBE) was subjected to cytotoxicity tests in L929 (mouse fibroblast cell line - strain L) and HDFa cells (Human Dermal Fibroblasts , adult) . In the second stage cosmetic formulations type gel, cream and gel-cream incorporated with ESBE were developed. These formulations were used in preliminary stability tests and assays to analyze permeation and retention of compounds on the skin. Finally, efficacy test evaluated the production of collagen after treatment of primary human fibroblasts with ESBE. The results showed that the biotransformation is an effective method for increasing the concentration of D and G; ESBE did not show cytotoxicity in L929 and HDFa cells; the cosmetic formulations incorporated with ESBE shown to have preliminary stability; gel-type formulation proved to be the best option for incorporation of ESBE because it was able to facilitate the retention of the isoflavones in the skin without allowing the permeation of compounds into the liquid receiver; And finally, the ESBE was able to stimulate collagen production in primary fibroblasts at concentrations of 59 and 1,33 ?g / mL. We conclude that the ESBE can stimulate collagen production, mainly in the concentration of 1,33?g / mL. We suggest that the mechanism responsible for this effect may involve genistein binding to estrogen receptors, however, other mechanisms of action of soy isoflavones, as protection against oxidative stress, induction of expression of antioxidant enzymes and the presence of protease inhibitors proteins (BBI and STI) must be taken into account.

The Combined Effects Of Genistein And Daidzein On Adipocyte Differentiation

Dietary soy isoflavones have been shown to ameliorate insulin resistance and Type 2 diabetes. However, many in vitro studies used supra-physiological concentrations of individual isoflavones that make it difficult to interpret the results as potential mechanisms in vivo. Since the insulin-sensitizing effects of thiazolidinediones, anti-diabetic drugs, have been shown to be mediated through activation of peroxisome proliferators-activated receptor gamma (PPARγ), the key transcription factor for adipocyte differentiation, we examined the effects of the two main soy isoflavones genistein and daidzein either as individual compound or combined on adipocyte differentiation and PPARγ expression, as well as whether the Wnt/β-catenin signaling pathway is the underlying molecular mechanism. In 3T3-L1 cells, genistein and daidzein significantly enhanced adipocyte differentiation. Similarly the expression of PPARγ increased particularly at 20 µmol/L. The stimulatory effect is greater when the two isoflavones are combined, indicating a synergistic effect. Genistein and daidzein also increased the relative abundance of insulin-responsive glucose transporter 4 (GLUT4) mRNA with a greater effect when combined. Wnt10b expression was not affected by soy isoflavones treatments, while Wnt5b expression was only increased by the combination of genistein and daidzein. Our results suggest, that the combination of soy isoflavones has a greater effect in increasing the newly formation of adipocytes that are highly insulin-sensitive via an increase in PPARγ expression as well as increasing the expression of GLUT4. However, genistein and daidzein actions on Wnt signaling remain unclear. These data further support the epidemiological findings for the beneficial effect of soy consumption on insulin sensitivity.

As mulheres na fase reprodutiva apresentam menor incidência de doenças cardiovasculares (DCV), em relação aos homens de mesma idade. Porém, essa vantagem desaparece na pós-menopausa, sugerindo que os hormônios sexuais femininos exercem algum efeito cardioprotetor. Um dos mecanismos propostos para explicar essa proteção é o fato dos estrógenos promoverem a produção de importantes fatores vasoativos pelo endotélio vascular, entre eles o óxido nítrico e a prostaglandina I2. Com a diminuição da quantidade de estrógenos circulante, as mulheres na pós-menopausa, estão mais suscetíveis à disfunção endotelial e a doenças cardiovasculares. Estudos têm demonstrado que a terapia de reposição hormonal (TRH) utilizada por mulheres na pós-menopausa combate os sintomas deste período, melhora o quadro de disfunção endotelial e o perfil lipídico e aumenta a síntese de fatores vasoativos, que auxiliam na prevenção da DCV. Entretanto a TRH vem sendo questionada por grandes estudos como o WHI (Writing Group for the Women\'s Health Initiative Investigators) e o HERS (Heart and Estrogen/Progestin Replacement Study), que mostraram um risco aumentado de desenvolvimento de câncer de mama e endometrial em mulheres fazendo uso da TRH. Objetivos: estudar os efeitos dos tratamentos com ISO e VIB isolados e combinados no tecido ósseo de ratas OVX. Métodos: 50 ratas Wistar com 6 meses de idade foram SHAM operadas ou OVX e após três meses, divididas em 5 grupos: SHAM; OVX; OVX tratadas diariamente por gavage com 200mg/kg de ISO por 90 dias consecutivos (OVX-ISO); OVX tratadas com VIB, 20 min./dia, 30 Hz, 0.6g, 5 dias por semana por 90 dias (OVX-VIB) e OVX tratadas com ISO e VIB (OVX-ISO+VIB). Após o tratamento, os fêmures e tíbias foram coletados e seus comprimento e espessura foram medidos. Os fêmures foram processados para histomorfometria e aos métodos histoquímicos, bioquímico e do TUNEL. As tíbias foram congeladas e destinadas para testes físicos e biomecânicos. Resultados: Os tratamentos não exerceram efeitos significantes no peso corporal e nas medidas de comprimento e espessura, exceto na espessura das tíbias dos grupos OVX-ISO e OVX-ISO+VIB que foi significantemente maior comparado ao grupo SHAM. Os testes físicos mostraram que o peso imerso, a porcentagem de material orgânico e a densidade óssea foram maiores nos grupos SHAM, OVX-VIB e OVX-ISO+VIB quando comparados aos outros grupos, enquanto que o volume ósseo, a densidade mineral óssea, porc entagem de material mineral e o peso das cinzas foram maiores nos grupos OVX e OVX-ISO do que nos outros grupos e não houve diferenças significantes nos parâmetros biomecânicos analisados entre os grupos. Nos fêmures, os tratamentos não exerceram efeitos significantes no volume ósseo trabecular, porém, o tratamento combinado exerceu efeitos positivos no osso cortical. Os tratamentos tiveram efeitos positivos em relação à quantidade e na organização das fibras colágenas e esse efeito foi mais expressivo no grupo OVX-ISO+VIB. O método do TUNEL não detectou diferenças nas células TUNEL-positivas entre os grupos. Conclusão: Esses resultados demonstram que o tratamento combinado (ISO+VIB) exerce maior efeito nos constituintes do tecido ósseo de ratas ooforectomizadas osteopênicas, quando comparado aos tratamentos isolados. / Introduction: It has been shown that both soy isoflavones (ISO) and low intensity high frequency mechanical vibration (VIB) have protective effects in bone tissue of ovariectomized (OVX) rats. Objective: We investigated whether combined treatments with ISO and VIB would exhibit cooperative effects in bone tissue of OVX rats. Methods: Fifty female Wistar rats at the age of 6 months were either sham-operated or OVX. After operation, the rats were divided in five groups: SHAM; OVX; OVX treated daily by gavage with 200mg/kg/body weight of ISO for 90 days (OVX-ISO); OVX treated with VIB, 20 min./day, at 60 Hz, 0.5g, 5 days/ week for 90 days (OVX-VIB) and OVX treated with ISO and VIB (OVX-ISO+VIB). After treatment, the femurs and tibias were collected and its length and thickness were measured. The femurs were then processed for histomorphometry, histochemistry and biochemical assay. The tibias were frozen for subsequent physical and mechanical tests. Results: Treatments did not have significant effects on body weight and in the bone measurements, except in the tibial thickness of the OVX-ISO and OVX-ISO+VIB groups that was significant higher than the SHAM group. Physical tests showed that the immersed weight, content of organic material and bone density were higher in the SHAM, OVX-VIB and OVX-ISO+VIB groups than the other groups, while the bone volume, bone mineral density, mineral material and the ash weight were higher in the OVX and OVX-ISO groups than the other groups and there were no significant differences in the mechanical tests among the groups. Treatment did not have significant effects in the trabecular bone volume, but it was not detected cortical bone loss in the group treated with the combined therapy. The treatments showed positive effects in the content of glycosaminoglycans, as well as in the organization of collagen fibers and these effects was higher in the OVX-ISO+VIB group. The TUNEL-positive cell profile was similar among the groups. Conclusion: These results demonstrate that the combined treatment (ISO+VIB) have better effects in the bone tissue constituents of ovariectomized osteopenic rats, when compared with the treatments isolated. / TEDE / BV UNIFESP: Teses e dissertações
