Geographic Provenancing of Unprocessed Cotton Using Elemental Analysis and Stable Isotope Ratios

Cotton is the most abundant natural fiber in the world. Many countries are involved in the growing, importation, exportation and production of this commodity. Paper documentation claiming geographic origin is the current method employed at U.S. ports for identifying cotton sources and enforcing tariffs. Because customs documentation can be easily falsified, it is necessary to develop a robust method for authenticating or refuting the source of the cotton commodities. This work presents, for the first time, a comprehensive approach to the chemical characterization of unprocessed cotton in order to provide an independent tool to establish geographic origin. Elemental and stable isotope ratio analysis of unprocessed cotton provides a means to increase the ability to distinguish cotton in addition to any physical and morphological examinations that could be, and are currently performed. Elemental analysis has been conducted using LA-ICP-MS, LA-ICP-OES and LIBS in order to offer a direct comparison of the analytical performance of each technique and determine the utility of each technique for this purpose. Multivariate predictive modeling approaches are used to determine the potential of elemental and stable isotopic information to aide in the geographic provenancing of unprocessed cotton of both domestic and foreign origin. These approaches assess the stability of the profiles to temporal and spatial variation to determine the feasibility of this application. This dissertation also evaluates plasma conditions and ablation processes so as to improve the quality of analytical measurements made using atomic emission spectroscopy techniques. These interactions, in LIBS particularly, are assessed to determine any potential simplification of the instrumental design and method development phases. This is accomplished through the analysis of several matrices representing different physical substrates to determine the potential of adopting universal LIBS parameters for 532 nm and 1064 nm LIBS for some important operating parameters. A novel approach to evaluate both ablation processes and plasma conditions using a single measurement was developed and utilized to determine the “useful ablation efficiency” for different materials. The work presented here demonstrates the potential for an a priori prediction of some probable laser parameters important in analytical LIBS measurement.

Social status and diet. Reconstruction of diet of individuals buried in some early medieval chamber graves from Poland by carbon and nitrogen stable isotopes analysis

Yes / The study presents results of the investigations of diet based on carbon and nitrogen stable isotope ratios (δ13C and δ15N) of the bone collagen of individuals buried in medieval elite chamber graves from the territory of the state of the first Piasts, Poland (the second half of the 10th and the first half of the 11th century). The aim of the research was to determine the diet of individuals buried in such funerary structures, to compare this with commoners buried in ordinary graves, and investigate any sex-related patterns. Rib bone samples were taken from individuals buried in chamber graves at Bodzia, Dziekanowice, Pień and Sowinki. Results indicate that the elite male diet was based on C3 plants with possible contribution of some C4 plants (millet) and substantial consumption of animal proteins including fish. The bone collagen δ13C and δ15N of male chamber burials suggested consumption of higher trophic level foodstuffs (meat and fish) whilst the female diet, and that of the juveniles, was similar to the commoners in the rest of the population.

Evaluating the Role of Seagrass Beds as a Food Resource in Port Everglades, Florida

Seagrass habitats are highly productive ecosystems that support marine food webs and provide essential habitat for a variety of species. Seagrass coverages are declining in abundance worldwide. For southeastern Florida in particular, one of the main causes of the decline is disturbance from dredging and removal of substrate. Seagrass beds at three locations in the vicinity of Port Everglades, Florida were assessed for their trophic contribution to the marine organisms in the area. Seagrasses, algae, invertebrates, and vertebrates from the beds were identified and analyzed for stable carbon and nitrogen isotope ratios to determine their contribution as a food source. Significant differences were found in both δ13C and δ15N between both seagrass species and among the three sites. The δ13C of Johnson’s seagrass Halophila johnsonii ranged from -16.28 to -11.27‰ while shoal grass Halodule wrightii ranged from -15.78 to -13.36‰. The δ15N for H. wrightii were more constrained than those of H. johnsonii, 4.69 to 7.08‰ versus 0.80 to 7.86‰, respectively. Neither seagrass species appeared to be a dominant food source for marine organisms at all three study sites. However, the δ13C and δ15N of both seagrass species, Halophila johnsonii and Halodule wrightii, did fall in the fractionation range of potential consumers, -28.78 to -17.11‰ and 1.96 to 12.63‰, indicating that these animals could be ingesting pieces of seagrass while grazing on epiphytes and other primary producers in the area. Epiphytes found on the seagrass blades appeared to be a greater trophic contributor to local organisms. So while the seagrass species in question may not have been major contributors to the diet of many of the local consumers, the seagrasses nontheless played a vital role as habitat for the epiphytes that did serve as a trophic resource in these communities.

Infant mortality and isotopic complexity: new approaches to stress, maternal health and weaning.

Objectives Studies of the carbon and nitrogen stable isotope ratios (δ13C and δ15N) of modern tissues with a fast turnover, such as hair and fingernails, have established the relationship between these values in mothers and their infants during breastfeeding and weaning. Using collagen from high-resolution dentine sections of teeth which form in the perinatal period we investigate the relationship between diet and physiology in this pivotal stage of life. Materials and Methods Childhood dentine collagen δ13C and δ15N profiles were produced from horizontal sections of permanent and deciduous teeth following the direction of development. These were from two 19th-century sites (n = 24) and a small number (n = 5) of prehistoric samples from Great Britain and Ireland. Results These high-resolution data exhibit marked differences between those who survived childhood and those who did not, the former varying little and the latter fluctuating widely. Discussion Breastfeeding and weaning behavior have a significant impact on the morbidity and mortality of infants and the adults they become. In the absence of documentary evidence, archaeological studies of bone collagen of adults and juveniles have been used to infer the prevalence and duration of breastfeeding. These interpretations rely on certain assumptions about the relationship between isotope ratios in the bone collagen of the adult females and the infants who have died. The data from this study suggest a more complex situation than previously proposed and the potential for a new approach to the study of maternal and infant health in past populations.

Identifying cohorts using isotope mass spectrometry: the potential of temporal resolution and dietary profiles

Yes / Archaeological skeletal material from most sites represents a cross-sectional, opportunistic sample of the burials. These are influenced by the proportion and area of the site which is excavated, the taphonomic conditions and survival of tissues. This may not be representative of the population, and in an attritional cemetery may represent a long period of use, during which humans will have differing life-courses. Here we describe a commingled skeletal assemblage, the only human remains recovered from the historically significant medieval site of St Stephen’s Chapel, Palace of Westminster, London. Using carbon (δ13C) and nitrogen (δ15N) stable isotope ratios of bulk bone collagen and incremental dentine to investigate dietary life histories from 5 individuals, we combine the evidence with radiocarbon dating to assign them to two different temporal cohorts.

Nearshore habitat and land-use effects on trophic interactions and growth of largemouth bass and bluegill in Indiana's glacial lakes

Glacial lakes, such as those in the Midwest region of the United States, are ecologically and economically important, and they provide a wide range of ecosystem services, such as habitat for wildlife and fishes, flood control and recreational boating. Glacial lakes often support locally important sport fisheries, such as largemouth bass <i>Micropterus salmoides</i> and bluegill <i>Lepomis macrochirus</i>,<i> </i>which are partially dependent on suitable habitat within lakes. Nearshore vegetation is often removed by lakeshore landowners for perceived aesthetics and boat access, or by area managers as a form of indirect fisheries management and invasive species control. The connection between nearshore vegetation and fish population health, though widely studied is somewhat unclear. In the two research chapters of this thesis we attempted to further understanding of the environmental factors that influence vegetation abundance and distribution, how vegetation abundance and distribution influences fish population abundance and size structure, and how young-of-year (YOY) largemouth bass utilize habitats within the nearshore environment. In the first research chapter, we used structural equation modeling (SEM) and data collected by the Indiana Department of Natural Resources to quantify the complexity of relationships among catchment characteristics (e.g., catchment size), lake morphology, water quality, vegetation abundance and distribution, and fish population abundance and size structure. Across multiple lakes, lake productivity was more influential in explaining cross-lake variation of largemouth bass and bluegill proportional stock density (PSD) and largemouth bass catch per unit effort (CPUE) than vegetation. This may be a result of the feedback between phytoplankton production and rooted vegetation production. The models we constructed provide insights into the complexity of environmental variables that influence nearshore vegetation and fish populations. In the second research chapter we used stable isotopes (δ<sup>13</sup> Carbon, δ<sup>15</sup> Nitrogen, δ<sup>18</sup> Oxygen and δ<sup>2</sup> Hydrogen) to examine the consistency of habitat use and foraging of YOY largemouth bass within Indiana glacial lakes. We observed spatial variation in stable isotope ratios of YOY largemouth bass between habitat types and sites. Additionally, there were significant, positive relationships between δ<sup>13</sup>C of locally collected potential prey items and δ<sup>13</sup>C of YOY largemouth bass suggesting localized foraging patterns. Later in the summer, as young bass grew in size and likely switched to piscivory, we did not observe similar spatial variation in young bass stable isotopes or spatial relationships between prey and the young bass suggesting more homogeneous foraging patterns. Understanding the habitat use patterns of young bass may allow for more efficient and effective management of the nearshore environment. Overall, a greater consideration for the complexity of relationships between nearshore habitat and fish populations may facilitate more effective management.

Peruvian Pinnipeds as Archivists of ENSO Effects

Environmental fluctuations in the eastern Pacific Ocean are reflected in the tissues of some of its most vulnerable apex predators, the Peruvian fur seal (PFS) Arctocephalus australis ssp.and the South American sea lion (SASL) Otaria byronia.These large pinnipeds live in sympatry along the Pacific coastline of South America and forage within the neritic waters over the continental shelf. The coastal waters off Peru are a region of great environmental fluctuations due to periodic, oscillating El Niño- La Niña- Southern Oscillation (ENSO) events, which result in ecosystem-wide food web changes. Pinniped vibrissae (whiskers) are continuously growing keratinous tissues and reflect the incorporation of prey from the region. Fine resolution sampling along their length provided trophic information on a weekly to monthly time scale over several years. Vibrissae were obtained from 2010-2016 from female (n=47), male (n=17) and newborn pup (n=6) fur seals and male (n=6) sea lions collected during pinniped rookery health assessments. Stable isotope ratios (δ13C and δ15N) in vibrissae infer temporal primary production and dietary variations in individuals. ENSO conditions were dictated by the sea surface temperature anomaly (SSTA) recordings from the Niño 1+2 Index region over 12 years. Fluctuations in vibrissae δ15N were correlated to varying SSTA readings (p15N (p15N signatures (p13C revealed minor fluctuations ranging from -18.13 to -13.17‰over the 12-year period, δ13C did reveal a significant depletion and enrichment oscillation during specific points in time, such as the 2014-2016 El Niño event, which ranged from -15.09 to -13.83‰(p=0.040). Stable isotope signatures varied between genders, but not by species. Female fur seal stable isotope signatures were significantly more depleted in both δ13C and δ15N than males (p13C and δ15N were inversely correlated to each other from 2014 to 2016; this was during the strongest El Nino-Southern Oscillation event on record (p=0.002). As δ13Csignatures became more enriched, δ15N signatures depleted from 2014 until 2016. This suggests that when ENSO warm phase conditions occur, the environments resources change in historic foraging grounds, possibly forcing pinnipeds to travel farther distances offshoreto forage or altering what they feed on throughout these stressful conditions.This study serves to help understand of the impact of oceanographic influences on these pinnipeds.ENSO conditions influence the trophic dynamics and resultant survival adaptations in both of these vulnerable Peruvian marine mammals.

Weaned Upon A Time : Studies of the Infant Diet in Prehistory

This thesis is concerned with how prehistoric infants were fed in different physical and cultural environments, and in particular what impact the economic, social, and epidemiological changes associated with the development of agriculture had on infant feeding practices. In order to examine these effects, stable isotope ratio analysis has been used to assess the duration of breastfeeding and weaning in a variety of prehistoric contexts. The first study is of Pitted Ware Culture hunter-gatherers at the site of Ajvide on Gotland, Sweden. Breastfeeding usually continued for at least two years, but there was some variation in supplementary foods, which is attributed to seasonal variations in resource availability. The second study analysed a number of Neolithic and early Bronze Age sites from south-east Poland. Breastfeeding duration varied both within and between sites and ranged from six months to five years. The third study found that the infant feeding practices of two Iron Age populations on Öland, Sweden, were very varied, and infants may have been fed differently depending on their social status. The fourth study is of the childhood diet in the Únětice Culture of south-west Poland. Individual diets changed little during the lifetime, suggesting that eventual adult identity was determined early in life. A small number of infants in the study were found to have breastfed for differing lengths of time. The final paper considers the health consequences of introducing animal milks into the infant diet in a prehistoric context, and finds that their availability is unlikely to have made it possible to safely wean infants earlier. Comparison of the results from the four stable isotope studies to those of other published studies reveals that the modal age at the end of weaning was slightly lower in agricultural communities than hunter-gatherer communities, but the range of ages was similar. Weaning prior to the age of eighteen months was rare before the post-medieval period.
