Low cost, more efficient, and less toxic synthetic routes to conjugated polymers

As key components of flexible organic electronics, the synthesis of polythiophenes via less toxic and more cost-effective routes is demanded. An efficient synthetic route for the production of thienyl-containing homopolymers and copolymers has been developed. The synthetic approach consists of: i) the synthesis in high yield and high purity of thienyl borane monomers protected with N-methyliminodiacetic acid (MIDA) via C-H electrophilic borylation. This reaction uses a combination of inexpensive reagents BCl3, AlCl3, and 2,6-dichloropyridine (Cl2Py) for the regioselective electrophilic aromatic substitution of thiophenes, followed by addition of a second aprotic amine pre-esterification to reduce the Brønsted acidity of the reaction mixture. In situ esterification provided the desired thienyl MIDA boronateester monomers in one-pot at room temperature. ii) Subsequent Suzuki-Miyaura polymerisation of the synthesised monomers to produce well defined thienyl containing pie-conjugated polymers in high molecular weight and high yields. Key reaction parameters for successful Suzuki-Miyaura polymerisation of thienyl-derived MIDA boronate esters under mild temperatures (i.e., 55 °C in THF) were found: a) an optimal monomer:H2O:base ratio, which enables controlled hydrolysis of the BMIDA moiety into its corresponding boronic acid at appropriate rates for high fidelity polymerisation. b) Nature of the base, where K3PO4 provided the best results for production of homopolymers (e.g., rr-P3HT), or KOH which gave excellent results for the formation of copolymers across a range of electronically different comonomers (e.g., pCPDT-BT). Thus, it is demonstrated that the approach is a general strategy for the highly efficient production of thienyl containing pie-conjugated regioregular, regiosymmetric and Donor-Acceptor polymers.

Příprava a studium vlastností polymerů na bázi bis(terpyridyl)fluorenů / Preparation and characterization of bis(terpyridyl)fluorene based polymers

The diploma thesis presents the preparation of ,-bisterpyridinylfluorenes (unimers) containing fluorene unit in the central block either directly attached to terpyridine (tpy) end groups or via 1,4-phenylene or thiophene-2,5-diyl linker. Synthesized unimers were subsequently used for the preparation of metallo-supramolecular polymers with Zn2+ and Fe2+ ions. The prepared unimers were characterized by NMR, IR and HR-MS, their optical properties were investigated using absorption and emission spectroscopy and cyclic voltammetry. Molecular weight distribution for the prepared Fe-polymers was determined by GPC. The number-average degree of polymerization equal to 7 for an equimolar ratio between unimer and Fe2+ in a diluted solution was determined for polymers composed of unimer with thiophene-2,5- diyl linker. By using the viscometric measurements the ongoing constitutional dynamics in solutions of metallo-supramolecular polymers was proven. The overstoichiometric amount of metal ions in solution resulted in equilibrium shortening of the polymer chains together with end-capping of terminal tpy ligands by Mt2+ ions. Unimer with thiophene-2,5-diyl linker exhibited unique properties involving significantly red-shifted absorption and emission maxima, as well as the different behavior during assembling...

Le myricanol est un [7,0]-métacyclophane naturel qui appartient à la famille des diarylheptanoïdes cycliques et qui possède des propriétés structurales et biologiques intéressantes (activité anti-Alzheimer ou anticancéreuse). L’objectif de ce travail de thèse est la préparation ambitieuse en voie racémique puis atropostéréosélective du myricanol, rendue délicate de part la tension de cycle existante. Pour se faire, deux nouvelles approches rétrosynthétiques ont été considérées. Une première route racémique a été envisagée et a permis de préparer le macrocycle par le biais d’une réaction de métathèse croisée suivie d’une réaction de Suzuki-Miyaura domino intramoléculaire avec un rendement de 2.55% en 11 étapes. Une autre voie racémique a également été explorée où la préparation du noyau biarylique du myricanol a été envisagé par une réaction de couplage intermoléculaire métallo-catalysée suivie d’une réaction de métathèse cyclisante. Des intermédiaires avancés ont également été préparés. / The myricanol, a natural [7.0]-meta-cyclophane which belongs to the family of strained and cyclic diarylheptanoids, possess an interesting structure and attractive biologically activities (anti Alzheimer and anti cancer properties). Actually only two synthesis of racemic (+/-)-myricanol have been reported in the literature. The goal of this research was to prepare this strained cyclophane in a racemic and then in an atropostereoselective route taking into account the challenging ring closure. Thus a linear diarylheptanoid was prepared using an efficient cross-metathesis reaction followed by an intramolecular Suzuki-Miyaura domino reaction giving rise to the desired cyclophane with 2.55% overall yield in 11 steps. On the other side, the biaryl core of myricanol was envisaged by an intermolecular metallo-catalysed coupling reaction between already highly functionalized fragments, followed by a ring closure metathesis. Two advanced intermediates were already attempted.

A Novel Method for the Synthesis of Indolo[2,1-a]isoquinolines

MSc dissertation School of Chemistry Faculty of Science 0004984F / Many azapolycyclic aromatic ring systems, whether they are naturally occurring or synthetically made, display important biological activities. One important class of naturally occurring azapolycyclic aromatic ring systems are the dibenzopyrrocoline alkaloids, which contain an indole ring fused to an isoquinoline moiety, where they share a common nitrogen. The basic skeleton of these alkaloids is the indolo[2,1-a]isoquinoline nucleus. Both the dibenzopyrrocoline alkaloids and the indolo[2,1-a]isoquinolines have been found to inhibit tubulin polymerization and thus possess antitumour and antileukemic activities. In our laboratories, a variety of indolo[2,1-a]isoquinolines, for example 5,12- dimethyl-6-phenylindolo[2,1-a]isoquinoline, have been synthesized using the Suzuki-Miyaura cross coupling reaction and reaction conditions for the formation of aromatic rings (KOBut in DMF and a light source – developed in our laboratories) as key steps. In this dissertation we discuss the synthesis of (±)-5,6-dihydro-6-phenylindolo[2,1-a]isoquinolin-5-ol and ethyl indolo[2,1- a]isoquinoline-6-carboxylate using these reaction conditions as our key steps. The syntheses commenced with the N-protection of isatin with a benzyl and an ethyl acetate group to afford 1-benzylindoline-2,3-dione and ethyl 2-(2,3- dioxoindolin-1-yl)acetate respectively. The next step was the synthesis of the brominated compounds 1-benzyl-2-bromo-1H-indole and ethyl 2-(2-bromo- 1H-indol-1-yl)acetate by means of a functional group interconversion of the oxygen in the 3-position to two chlorine atoms, followed by hydrodehalogenation, using zinc in AcOH, and then bromination, using POBr3 in CH2Cl2. Having obtained the brominated compounds we went on and coupled them with 2-formylphenylboronic acid using the Suzuki-Miyaura cross coupling reaction to obtain the coupled products 2-(1-benzyl-1H-indol-2- yl)benzaldehyde and ethyl 2-(2-(2-formylphenyl)-1H-indol-1-yl)acetate in 92 and 77% yield, respectively. Aromatisation of ethyl 2-(2-(2-formylphenyl)-1Hindol- 1-yl)acetate to ethyl indolo[2,1-a]isoquinoline-6-carboxylate occurred smoothly in 2 minutes using 10 mol % KOBut in DMF at room temperature. Using the same reaction conditions on 2-(1-benzyl-1H-indol-2- yl)benzaldehyde to form 6-phenylindolo-[2,1-a]isoquino-line resulted in (±)- 5,6-dihydro-6-phenylindolo[2,1-a]iso-quinolin-5-ol being obtained in 75% yield (7:3 ratio of anti:syn). An attempt to dehydrate this compound using p-TSA in CH2Cl2 in the presence of molecular sieves was not successful. Time constraints prevented any further attempts at dehydrating (±)-5,6-dihydro-6- phenylindolo[2,1-a]iso-quinolin-5-ol. In conclusion, we managed to synthesize (±)-5,6-dihydro-6-phenylindolo[2,1- a]isoquinolin-5-ol and ethyl indolo[2,1-a]isoquinoline-6-carboxylate using the Suzuki-Miyaura cross coupling reaction and specific reaction conditions, using the base KOtBu, for the formation of aromatic rings, both as key steps.

Une série de calixarènes portant une unité imidazolium ou deux, en conformation cône ont été synthétisés et caractérisés par différentes méthodes. Ces sels chargés positivement constituaient des précurseurs de carbènes N-hétérocycliques pour la synthèse de complexes de nickel correspondants. Les complexes Ni-NHC-calix[4]arènes obtenus ont été testés dans la catalyse de la réaction de couplage croisé de Suzuki-Miyaura. Des moyennes à bonnes conversions ont été obtenues selon l’espacement du centre métallique de la cavité calixarénique, ainsi que le nombre de centre métallique présents sur la même plateforme. / A series of new calix[4]arenes bearing one or two imidazoliums units in the lower rim were synthesized as precursors of N-heterocyclic carbenes for the synthesis of the corresponding nickel complexes. All products obtained were characterized by NMR spectroscopy, microanalysis and mass spectroscopy and was revealed to be in cone conformation. The application of Ni-NHC-calix[4]arene in Suzuki-Miyaura cross coupling showed moderate to good conversions. This is been depending on the length of the alkyl chain and the number of the catalytic centers grafted on the platform.

Synthèses, caractérisation et étude structurale de complexes de type carbènes N-hétérocycliques basés sur des Calix[4]arènes / Synthesis, Characterization and X-Ray Structures of N-heterocyclic Carbene Palladium Complexes based on Calix[4]arenes

Cette thèse apporte une contribution au développement de complexes carbènes N-hétérocycliques-palladium basés sur des calix[4]arènes: leur synthèse, leur caractérisation ainsi que l'évaluation de leur activité catalytique est presenté. Pour cela, un calix[4]arène mono-substitué a été préparé conformément aux procédures classiques, puis les précurseurs de nouveaux ligands NHC ont été obtenus. Subissant l'alkylation avec du n-butylbromide et puis une métallation subséquente avec du palladium et de la pyridine, quatorze nouveaux complexes ont été obtenus. Après les caractérisations complètes en solution et à l'état solide, l'évaluation de l'activité catalytique a été réalisée dans réactions couplage de Suzuki-Miyaura. De bonnes performances ont pu être observées permettant d'obtenir des taux de transformation de 99% avec des quantités de catalyseur descandant jusqu'à 0.08 mol%. L'étude de conformation ainsi que les résultats catalytiques en catalyse n'ont pas permis de mettre en évidence un effet supramoléculaire de la cavité macrocyclique envers le processus de couplage. Dans le but d'orienter le centre catalytique à l'intérieur de la cavité, des contraintes stériques ont été appliquées sur des nouveaux Pd-NHC-complexes. Ainsi des complexes dimeriques de palladium ont été synthétisés et entièrement caractérisés. Plusieurs approches pour construire des linkers intramoléculaires permettant de fixer la conformation ont été énvisagées mais n'ont pas permis à ce jour un gel conformationnel / This thesis will focus on development of N-heterocyclic carbene palladium complexes based on calix[4]arenes: from synthesis, characterization and structural study to evaluation of catalytic activity. A new series of calix[4]arene supported N-heterocyclic carbene palladium complexes was developed and fully characterized. A mono-substituted calix[4]arene was prepared through conventional procedures, following with the attachment of imidazolyl derivative groups to compose the precursors of novel NHCs ligands. Undergoing the alkylation with n-butylbromide and corresponding metallation with palladium and pyridine, original complexes were obtained. After a full characterization in solution and solid state, the evaluation of catalytic activity was undertaken through Suzuki-Miyaura cross-coupling reactions which revealed good performances. The conformational study as well as the catalytic results in catalysis did not allow putting in evidence a supramolecular effect of the macrocycle cavity towards the coupling process. With the aim of localize catalytic center inside of the cavity, steric constrain was involved in the form of new Pd-NHC-complexes. Dimeric Pd complexes were synthesized and fully characterized as well. Several approaches to construct intramolecular linkers for fixing the conformation were elaborated. The conception of ‘flexible steric bulky’ inspired the research on confined structure of calix[4]arene, such as encapsulation and capped configuration. Synthesis and Studies of Dendritic Poly (Ether Imine) Boronates and Cholesteryl-Functionalized Mesogens

Synthesis and Studies of Dendritic Poly(Ether Imine) Boronates and Cholesteryl-Functionalized Mesogens SYNOPSIS Dendrimers are hyperbranched synthetic macromolecules having branches-upon-branches structures, high molecular weights, globular shapes and monodispersities. Dendrimers possess a large number of modifiable functional groups at their peripheries. Initial efforts were largely concerned with the synthesis, design and development of new dendrimers. Exploring the chemical, biological and material applicability of these macromolecules are relevant to current interests, as a result of the unique structural features of dendrimers. Incorporation of transition metals and organic moieties at the peripheries of the dendrimers was studied to determine their efficacies in catalysis. Evolution of dendritic effects was observed in few instances, that were non-linear in nature. On the other hand, dendritic peripheries were also utilized to study mesogenic properties in liquid crystals. Chapter 1 of the Thesis gives an overview of the types of dendrimers, its structural features and their application in catalysis and as liquid crystalline materials. Chapter 2 describes the synthesis of a new type of poly(ethyl ether imine) dendrimer, having nitrogen as a branching unit, ethylene moiety as the spacer and an oxygen as the connecting linker. Synthesis, characterization, and studies of the photophysical properties of these dendrimers are described in this chapter. The molecular structure of second generation dendrimer is shown in Figure 1. Synthesis of this dendrimer was initiated using 2,2'-oxy-bis(ethan-1¬amine) as the core. The reaction sequence of two alternate nucleophilic substitutions and two alternate reductions, involving ethyl bromoacetate and bromoacetonitrile as monomers was employed in the synthesis of the dendrimer. The formation of dendrimers having ether linkage and tertiary amines as branching unit was established by spectroscopies and mass spectrometry. A number of functional groups, such as, acid, alcohol, amine, ester and nitrile are present at the peripheries of each generation the dendrimers that open up the possibilities for further studies. Carboxylic acid terminated poly(ethyl ether imine) dendrimers are substituted iminodiacetic acids, belonging to the class of polyaminocarboxylic acid. Methyl iminodiacetic acid boronates with NB coordination have emerged as an excellent substitute for unstable boronic acids. Upon increasing the steric bulk on the nitrogen moiety, the hydrolytic stability of the boronates to a base-catalyzed hydrolysis is increased. Combining the structure of carboxylic acid terminated dendrimer and the stability of the dendritic boronates, such dendritic iminodiacetic acids were reacted with arylboronic acids to prepare bis-and tetrakis-boronates (Figure 2). Kinetic hydrolytic studies of boronates were conducted to assess the stabilities of the newly synthesized dendritic boronates. From the studies it was observed that the tetrakis-boronate was ~20 times more stable in comparison with dimeric and monomeric boronates (Figure 3). Subsequent to synthesis and hydrolytic stability studies, C-C bond-forming Suzuki-Miyaura cross-coupling reactions were conducted. A comparison of the reactivities among monomeric, dimeric and tetrameric arylboronates in C-C bond-forming reactions showed a higher reactivity of monomeric and dimeric boronates, than the tetrameric aryl boronate to construct ter-and tetra-aryl in one-pot iterative manner (Figure 4). Chapter 3 of this Thesis describes the synthesis and characterization of dendritic boronates and studies of their hydrolytic stability in Suzuki-Miyaura cross¬coupling reactions to construct ter-and tetraaryls. Step-wise iterative synthesis of dendrimer allows a uniform branching throughout the structure. The first and second generation poly(ether imine) dendrimer series, having hydroxyl groups at their peripheries were chosen for further modification. A versatile mesogenic group, namely, cholesterol was covalently attached at the peripheries of the dendrimers with succinic moiety as linker, so as to install 4 and 8 cholesteryl moieties at the peripheries of the dendrimers (Figure 5), that were characterized by H, C NMR spectroscopies and elemental analysis, so as to confirm their structural homogeneities. Figure 5. Molecular structures of the first and second generation dendritic mesogens. Subsequent to synthesis and characterization, liquid crystalline properties of all the dendritic mesogens was assessed through differential scanning calorimetry (DSC), polarizing optical microscopy (POM) and X-ray diffraction (XRD) studies. In POM study, broken fan or leaf like texture revealed the lamellar arrangement, whereas homeotropic appearance of texture on surfactant (cetyltrimethylammonium bromide) coated substrate indicated the lamellar nature of G1-Et-(OCS)4, G1-Pr-(OCS)4 and G2-Pr-(OCS)8 (Figure 6). From DSC studies, the change in enthalpy was found to increase with increase in generation and change in enthalpy per mesogenic unit was found to be ~ -1 1-2 kJ mol, which indicated the mesophase arrangement to be lamellar. Decrease in the length of spacer dendritic backbone and increase in the generation increased the isotropization temperature of the dendritic liquid crystals. Variable temperature XRD studies were undertaken to characterize the mesophase property. Two sharp peaks in small angle region and a diffused halo in wide angle region in XRD pattern of the material suggested the smectic A (SmA) liquid crystalline arrangement of G1-Et-(OCS)4, G1-Pr-(OCS)4 and G2-Pr-(OCS)8 (Figure 7). The second generation ethyl-linker dendrimer G2-Et-(OCS)8 exhibited a layered structure with a superimposed in-plane modulation (SmÃ), the length of which corresponded to a rectangular column width. Chapter 4 describes the synthesis, characterization and studies of mesophase property and fluorescence property of cholesterol functionalized homologous pairs of the PETIM dendritic liquid crystals. Peripheral functionalization of the dendrimers provides an easy access to dendritic liquid crystalline materials. The covalent functionalization was extended further with the dendrimers for both the series, so as to have 2, 4, 8 and 16 cholesteryl groups at the peripheries of 0, 1, 2 and 3 generation dendrimers, respectively, having succinic amide and phthalic ester functionalities for 1, 2 and 3 generation dendrimers with 4, 8 and 16 cholesteryl groups. Subsequent to synthesis and characterization, mesophase property was studied through POM, DSC and XRD techniques. In POM study, a birefringent texture was observed in heating and cooling cycles. Leaflet, broken fan or bâtonnet like texture suggested the layered arrangement of the molecules (Figure 9). In DSC studiues, it was observed that the amide-linked dendrimers showed higher glass transition and isotropization temperatures than that of ester-linked dendrimers within the same generation irrespective of the back-bone of the dendrimer. Succinic moiety linked dendrimers showed lower glass transition temperature than that of phthalic moiety linked dendrimers and consequently, larger mesophase range. The change in enthalpy for isotropization was found to increase with increase in generation, whereas change in -1 enthalpy per mesogenic unit was 1-2 kJ mol, indicative of a layered arrangement in the mesophase. Appearance of two sharp peaks in small angle region and a wide halo in wide angle region in XRD pattern supported lamellar mesophase property of the material (Figure 10). On decreasing the temperature, increase in the layer thickness also suggested the smectic A arrangement of the molecules except third generation phthalate derivative G3-Pr-(OCP)16, which showed rectangular columnar mesophase. For all the dendromesogens, the layer thickness increased with the increase in generation. Upon protonation, the first generation dendrimer showed a change in mesophase from simple smectic A to modulated smectic A with decrease in layer thickness. The change in liquid crystal property of the dendromesogens from lamellar to columnar mesophase by changing the linker of the mesogen is unknown so far in the dendrimer liquid crystals. Chapter 5 gives details of synthesis, characterization and mesophase property study of ester-and amide-linked dendritic liquid crystals. Overall, the Thesis establishes a synthetic methodology for the synthesis of a new homologous series of poly(ether imine) dendrimers with ethyl spacer; synthesis of dendritic boronates and their studies in cross-coupling reactions through in-situ slow release of boronic acid; hydrolytic stability study showed higher stability of dendritic boronates which was used in one-pot iterative cross-coupling reactions to construct ter-and tetra-aryls. decrease in linker length in dendrimer backbone modified the thermal, as well as, mesophase behavior of the dendritic liquid crystals; change in the linker functionality from ester to amide changed the thermal behavior of dendritic liquid crystals; a switching of mesophase property from lamellar to columnar was observed by changing the rigidity of the linker from succinate to phthalate without changing the linker length.
