Investigations of Novel NxSy Technetium and Rhenium Chelants

This thesis describes the synthesis and complexation of amino acid based chelants for technetium and rhenium. All of the chelates were of the NxSy type. An initial N2S2 diamido dithiol peptide of the type mercaptoacetic acid-serine-cysteine was synthesized and coordinated with both rhenium and technetium. The complexes were analysed by NMR spectroscopy and found to retain the benzyl moieties. Syn and anti diastereoemers were produced with one conformation preferred over the other. This was not true of another chelant RP294 supplied by Resolution Pharmaceuticals. The N2N1'S1 chelant consisted of dimethyl-glycine-serine-cysteineglycine and was a novel variation of the N2S2 tripeptide used initially. The coordination of this N3S chelant to technetium resulted in the formation of equal 1:1 quantities of diastereomers. Analysis was done by NMR spectroscopy and electrospray mass spectrometry. The two diastereomers were found to slowly interconvert and a mechanism for the interconversion proposed. The radiopharmaceutical RP128 (dimethylglycineserine-cysteine-glycine-threonine-lysine-proline-proline-arginine), of which the RP294 is the chelating portion, was coordinated to technetium. Analysis of the bifunctional chelant and its coordination complex by NMR spectroscopy demonstrated that the targeting portion of the molecule was unaffected by the technetium. Two diastereomers of the TcO(RP128) complex were found, as seen for the chelating portion of the molecule, TcO(RP294). / Thesis / Master of Science (MS)

Quantitative Conjugate Imaging of Iodine-123 and Technetium-99m Labeled Brain Agents in the Basal Ganglia

In the research reported in this dissertation, the concept of classic conjugate imaging, a non-tomographic nuclear medicine technique, is modified such that activity of a radiopharmaceutical distribution in the striata can be estimated. A mathematical model is developed that extended the application of classic conjugate imaging to estimation of two distinct and aligned activity distributions. Error analysis of the mathematical model is performed to characterize the accuracy of the model and to benchmark the limitations of the model. Phantom experiments are performed to demonstrate the practical application of the model and to evaluate its accuracy. A Monte Carlo simulation model of conjugate imaging of activity uptake in the striata of a primate is developed to evaluate the accuracy of the modified conjugate imaging technique as applied in the use of a dedicate conjugate imaging system. In addition, the simulation model is used to determine and characterize the shielding design of the small field of view gamma cameras comprising the dedicated conjugate imaging system. The application of scatter correction is investigated to address the downscatter of high-energy photon emissions into the photopeak window and the inclusion of scattered primary photons in the photopeak window. In this dissertation, it is shown that the modified conjugate imaging technique developed can be used to estimate accurately activity uptake in each of two distinct and aligned activity distributions. The accuracy of the technique is shown to be comparable to that of clinical quantitative SPECT. The modified conjugate imaging technique used with the dedicated conjugate imaging system may, therefore, be a viable quantitative nuclear medicine technique for activity estimation of radiopharmaceutical uptake in the striata of Parkinsonian and schizophrenic patients. The portability and low cost relative to SPECT systems make a dedicated conjugate imaging system advantageous for clinics with Parkinsonian and schizophrenic patients, who are unable to travel due to physical or mental limitation.

Neste trabalho foram estudadas as formas de coordenação de piridinonas, ditiocarbazatos e N-(N\'\',N\'\'-dialquilaminotiocarbonil)-N\'-(2-hidroxifenil)benzoamidinas aos centros metálicos Re (V) e Tc (V). Neste sentido, os complexos triclorobis(trifenilfosfina)oxorrênio(V), [ReOCl3(PPh3)2]57, tribromobis(trifenilfosfina)oxorrênio(V), [ReOBr3(PPh3)2]57, e tetraclorooxorenato(V) de tetrabutilamônio, (NBu4)[ReOCl4]58, foram sintetizados e utilizados como precursores. O precursor tetraclorooxotecnetato(V) de tetrabutilamônio, (NBu4)[TcOCl4], foi sintetizado e disponibilizado pelo grupo de Prof. Dr. Ulrich Abram, da Universidade Livre de Berlin, durante o período de trabalho realizado na Alemanha. <br /> Foram utilizadas as seguintes técnicas para a caracterização dos complexos sintetizados: espectroscopia de absorção na região do infravermelho, análise elementar (C, H, N e S), espectroscopia de ressonância magnética nuclear (31P, 1H e no caso de um dos complexos, também 13C), espectrometria de massas (MS-ESI) e determinação de estruturas cristalinas e moleculares, quando possível, por difração de raios X em monocristal. Por se tratar de um elemento radioativo, a avaliação do percentual de tecnécio em seus complexos foi determinada por meio da técnica de cintilação líquida. <br /> Deste o início deste trabalho foram sintetizados e caracterizados por diversas técnicas 14 novos complexos de rênio e 1 de tecnécio, todos inéditos. Seis ditiocarbazatos, sendo 5 inéditos, também foram sintetizados e caracterizados. / This work evaluated the coordination mode of pyridinones, dithiocarbazates and N-(N\'\',N\'\'-Diethylaminothiocarbonyl)-N\'-(2-hydroxyphenyl)benzamidine, to the metal centers Re (V) and Tc (V). For this purpose, the compounds trichlorobis(triphenylphosphine)oxorhenium(V), [ReOCl3(PPh3)2]43, tribromobis(triphenylphosphine)oxorhenium(V), [ReOBr3(PPh3)2]43, and tetra-n-butyl)ammonium tetrachlorooxorhenate(V), (NBu4)[ReOCl4]44, were synthesized and used as starting materials. The precursor tetra-n-butylammonium tetrachlorooxotechnetate(V), (NBu4)[TcOCl4] was synthesized and released by the Prof. Dr. Ulrich Abram\'s group, at the Freie Universität Berlin, during my research time in Germany. <br /> The complexes were characterized by infrared absorption spectroscopy, elemental analyses (CHNS), mass spectrometry with electrospray ionization (MSESI), nuclear magnetic resonance spectroscopy (31P, 1H and, in one case, also 13C) and had their crystalline and molecular structures, when possible, properties determined by X-ray diffraction on single crystals. Due to its radioactive element, the 99Tc values were determined by standard liquid scintillation counting. <br /> Since the beginning of this work were synthesized and characterized by various techniques 14 new rhenium and 1 technetium complexes, all of then unknown in the literature. All the complexes presented here were obtained in good or excellent yields. Six new dithiocarbazates were obtained and characterized, of which 5 are unknown.
