Análise histológica e imunoistoquímica do tecido pulpar de ratos Wistar submetidos a procedimento clareador com duas concentrações de peróxido de hidrogênio

Foram utilizados dois protocolos de clareação dentária com peróxido de hidrogênio (H2O2) nos molares superiores de 40 ratos Wistar, formando os grupos Controle (gel placebo), BLUE (H2O2 a 20% 1x50min) e MAXX (H2O2 a 35%, 3x15min). Após 2 e 30 dias, os ratos foram eutanasiados e as maxilas processadas para avaliação histológica (H.E.) e imunoistoquímica. Foram realizados os testes estatísticos referentes a cada caso, considerando p<0,05. Aos dois dias, foi observada necrose no terço oclusal do grupo MAXX, e infiltrado inflamatório moderado no BLUE (p<0,05). Aos 30 dias, não houve infiltrado inflamatório nos grupos, e grande área da câmara pulpar foi ocupada por dentina terciária. A marcação para PCNA aos 2 dias, demonstrou maior nível de proliferação celular no terço médio do grupo BLUE (p<0,05) e no terço cervical do MAXX (p<0,05), com redução significativa aos 30 dias no terço cervical (p<0,05). Influência de uma substância antioxidante na união de materiais resinosos à dentina após a realização de clareamento /

Materiais e métodos: Foram testados 6 grupos. GI- Controle: somente restaurações de Resina Composta (RC); GII- Ascorbato de Sódio (AS) + RC; GIII- clareamento com peróxido de carbamida 10% (PC) + RC; GIV- PC + AS + RC; GV- clareamento com peróxido de hidrogênio 35% (PH) + RC; GVI- PH + AS + RC. Após os tratamentos, os dentes foram seccionados e descalcificados. Os cortes foram montados em lâminas de vidro e corados pelo método Brown & Brenn. A análise em microscopia óptica comum (Axiophot) em 400X, evidenciou os tags e camada híbrida formados. Resultados: Os dados foram tabulados, sendo obtidas as respectivas médias para comprimento dos tags e a espessura da camada híbrida: GI- 10 e 3.1μm, GII- 9.2 e 2.4 μm, GIII- 5.4 e 1.5 μm, GIV- 6.8 e 2.1 μm, GV- 5.7 e 1.6 μm, GVI- 7.1 e 2.1 μm. Conclusão: O clareamento dental com PC ou PH prejudica a formação de tags resinosos e camada híbrida. O uso do AS aumentou o comprimento destas estruturas / Abstract: This study analyzed the influence of 10% sodium ascorbate (SA) on tag and hybrid layer formation in dentin after bleaching. Materials and Methods: Six groups were tested: GI- Control: restoration; GII- Sodium Ascorbate (SA) + restoration; GIII- Bleaching with carbamide peroxide (CP) + restoration; GIV- bleaching with CP + SA+ restoration; GV- bleaching with 35% hydrogen peroxide (HP) + restoration; and GVI- HP + SA + restoration. After treatments, the teeth were sectioned and decalcified. The length of resin tags and thickness of the hybrid layer were analyzed using optical microscopy (Axiophot). Ação de enxaguatórios clareadores e sua associação ao clareamento caseiro com peróxido de carbamida /

Orientador: Caneppele, Taciana Marco Ferraz / Banca: Eduardo Bresciani / Banca: Carlos Martins Agra / Resumo: Objetivos: o objetivo deste estudo foi comparar a efetividade de enxaguatórios clareadores sobre dentes, clareados previamente ou não, expostos a corantes alimentícios. Material e Método: Cento e vinte espécimes de esmalte e dentina de 3 mm de diâmetro foram obtidos a partir de incisivos bovinos. Os espécimes foram manchados por 14 dias em caldo de manchamento. Após o manchamento, a leitura inicial da cor foi realizada, por meio do espectrofotômetro CM-2600d (Konica Minolta, Osaka, Japão). Sessenta espécimes (Subgrupos PC) foram submetidos ao clareamento com Peróxido de Carbamida a 10% por 14 dias. Após, foram subdividos em 4 subgrupos, com ciclos de manchamento (5 min) e enxaguatório (2 min) por 12 semanas, sendo PC-LI: Listerine Whitening Pré- escovação; PC-PL: Plax Whitening; PC-BP: Bromelina + Papaína; PC-AG: água deionizada. Os subgrupos LI, PL, BP e AG foram submetidos aos mesmos ciclos citados, porém não foram submetidos ao clareamento prévio. A leitura de cor dos espécimes foi feita após 4, 8 e 12 semanas de tratamento com os enxaguatórios. Os dados foram coletados e submetidos à Análise de variância (ANOVA) de medidas repetidas. A comparação entre os subgrupos para a avaliação das diferenças estatísticas foi realizada pelo Teste de Tukey. Todos os testes empregados admitiram 5% como nível de significância estatística. Os resultados mostraram que os valores de ΔL*, Δa*, Δb* e ΔE* confirmaram o clareamento para os subgrupos PC-LI, PC-PL, PC-BP e PC-AG, após o tratamento com peróxido de carbamida, e para os subgrupos PC-LI, PC-PL, LI e PL, após os ciclos manchamento-enxaguatório. Os subgrupos PC-BP e BP assemelharam-se aos subgrupos controle PC-AG e AG. Conclui-se, desse modo, que o enxaguatório bucal Listerine Whitening apresentou maior efeito clareador, seguido do enxaguatório Plax Whitening. Porém, nenhum desses enxaguatórios foi capaz de produzir clareamento semelhante ao evidenciado pelo.. / Abstract: Objectives: The aim of this study was to compare the effectiveness of whitening mouthrinses on teeth previously whitened or not, exposed to food dyes. Material and Methods: One hundred and twenty specimens of enamel and dentin of 3 mm in diameter were obtained from bovine incisors. The specimens were stained for 14 days in broth staining. After staining, the initial color reading was performed by a spectrophotometer CM-2600d (Konica Minolta, Osaka, Japan). Sixty specimens (Subgroups CP) were submitted to whitening (Carbamide Peroxide 10%) for 14 days. Then they were subdivided into 3 subgroups, and were submitted to cycles of staining (5 min) and mouthrinse (2 min) for 12 weeks, with CP-LI: Listerine Whitening Pré- Escovação; CP-PL: Plax Whitening; CP-BP: Bromelain + Papain; CP-DW: deionized water. LI, LP, BP and DW subgroups were submitted to the same cited cycles, but without bleaching before. The color measurements were performed after 4, 8 and 12 weeks of treatment with mouthrinses. Data were collected and subjected to analysis of variance (ANOVA) for repeated measures. The comparison between the subgroups for the evaluation of statistical differences was performed by Tukey test. All tests admitted statistical significance level of 5%. The results showed that the values ΔL*, Δa*, Δb* and ΔE* confirmed bleaching to PC-LI, PC-PL, PC-BP and PC-DW subgroups, after treatment with carbamide peroxide and to CP-LI, CP-PL, LI and PL subgroups, after the staining-mouthrinse cycles. CP-BP and BP subgroups were similar to CP-DW and DW. We conclude, therefore, that the mouthrinse Listerine Whitening presented the highest bleaching effect, followed by mouthrinse Plax Whitening. Both of them were able to maintain the bleaching effect of CP after 12 weeks of dye-rinse cycles. Orientador: Andréa Abi Rached Dantas / Resumo: Após o clareamento dental, radicais livres permanecem na estrutura dental interferindo negativamente na adesão, além disso também geram redução da resistência dental à fratura. Sugere-se que substâncias antioxidantes podem ser capazes de reverter tais efeitos. Assim, esse estudo buscou avaliar os efeitos da solução de alfatocoferol em diferentes concentrações sobre a resistência à fratura, formação de camada híbrida e resistência de união adesiva de dentes tratados endodonticamente e clareados. Foram realizados 3 experimentos. Experimento 1: 60 incisivos bovinos receberam tratamento endodôntico e foram distribuídos em 6 grupos (n=10): G1 - coroas apenas restauradas; G2 - coroas clareadas e restauradas; G3 - coroas clareadas e aplicação de solução de ascorbato de sódio 10%; G4 ao G6: coroas clareadas e aplicação das soluções de alfatocoferol nas concentrações de 15%, 20% e 25%, respectivamente. Os espécimes foram submetidos ao teste de resistência à fratura em máquina de ensaios mecânicos (EMIC). Experimento 2: 60 incisivos bovinos foram tratados endodonticamente, distribuídos e tratados, de acordo com o grupo, de forma idêntica ao experimento 1. Os espécimes foram seccionados na junção cemento-esmalte e no sentido vestíbulo-lingual, lixados e polidos, passaram por processo de desproteinização com ácido clorídrico 18% e hipoclorito de sódio 5%. To minimize these undesirable effects, the use of antioxidants to neutralize free radicals before the final restoration is recommended. This study evaluated the effects of alphatocopherol solution at different concentrations on the fracture resistance, hybrid layer and shear bond strength of endodontically treated teeth subjected to tooth bleaching. Three experiments were done. Experiment 1: 60 bovine incisors were endodontically treated and then divided into 6 groups (n=10): G1 - the teeth immediately restored with composite resin; G2 - the teeth were bleached and immediately restored; G3 - the teeth received treatment similar to that used for G2, but with 10% sodium ascorbate gel applied after the bleaching protocol; G4 to G6: the teeth received treatment similar to that used for G3, rather than sodium ascorbate, it was applied alphatocopherol at concentrations of 15%, 20% and 25%, respectively. Ação do fluor na microdureza do esmalte humano submetido a dois tipos de agentes clareadores /

Orientador: Clóvis Pagani / Banca: Camillo Anauate Netto / Banca: Wagner de Oliveira / Resumo: 0 objetivo deste estudo foi avaliar a capacidade do flúor de induzir a remineralização do esmalte humano submetido às técnicas de clareamento caseiro e profissional. Sessenta dentes foram divididos em cinco grupos: G1 - controle; G2- peróxido de carbamida a 10%; G3 - peróxido de carbamida a 10% e flúor a 0,05%; G4 - peróxido de hidrogênio a 35%; G5 - peróxido de hidrogênio a 35% e flúor a 0,05%. A microdureza Vickers de todos os corpos-de-prova foi mensurada antes e após o tratamento. Posteriormente foram realizadas análises em Espectrômetro de Absorção Atômica (EAA) e em Microscopia Eletrônica de Varredura (MEV). Os dados foram submetidos à análise estatística de variância e testes de Dunnett e Tukey. Os resultados evidenciaram redução significante da microdureza do esmalte clareado e o peróxido de carbamida promoveu redução significativamente maior do que o peróxido de hidrogênio. Dentes expostos ao flúor apresentaram menor redução da microdureza, sendo que essa diferença não foi significante nos dentes clareados com peróxido de carbamida e significante nos dentes clareados com peróxido de hidrogênio. Por meio da análise em EAA, observou-se que os grupos que não foram expostos ao flúor perderam mais cálcio do que os seus correspondentes que foram remineralizados. Ao MEV apresentaram-se alterações na morfologia da supertfcie do esmalte nos dentes clareados, sendo que estas não estavam distribuídas uniformemente em toda a superfície. Concluiu-se que a fluorterapia é um método adequado para minimizar a redução da microdureza após o clareamento caseiro ou de consultório / Abstract: The aim of this study was to evaluate the ability of fluoride to induce remineralizatian of the human dental enamel subjected to nightguard vital bleaching and professional bleaching. Sixty teeth were divided into five groups: control; 10% peroxide carbamide; 10% peroxide carbamide and 0.05% fluoride; 35% hydrogen peroxide; 35% hydrogen peroxide and 0.05% fluoride. The Vickers microhardness of all specimens was measured before and after treatment. Complementary analysis comprised Atomic Absorption Spectrometry (AAE) and Scanning Electron Microscopy (SEM). Data were subjected to statistical analysis of variance (ANOVA), Dunnett's and Tukey's tests. The results revealed that there is significant reduction of enamel microhardness when subjected to the bleaching agents, compared to the control group. Carbamide peroxide promoted reduction of microhardness statistically bigger than hydrogen peroxide. The specimens that were exposed to fluoride showed a statistically significant smaller reduction of microhardness, compared with those not treated with fluoride. This difference was non-significant in teeth treated with carbamide peroxide and significant in teeth treated with hydrogen peroxide. Analysis by AAE .displayed that the groups not exposed to fluoride lost more calcium than their correspondents with remineralization. The SEM findings revealed morphologic changes in the enamel surface of bleached teeth, though these were not uniform through the entire surface. Os fatores em estudo foram: técnicas em 5 níveis: controle, microabrasão (Opalustre – Ultradent Products Inc. Utah, USA), clareamento dental (Opalescence Boost PF 38 % - Ultradent Prodcuts Inc. Utah, USA) e a associação dos dois procedimentos (imediata e tardia); assim como o tempo de manutenção dos espécimes in situ: 0, 1, 3, 7 dias. As variáveis de resposta foram microdureza e a rugosidade superficial. Foram utilizados 50 dentes bovinos hígidos, seccionados em discos de esmalte/dentina com 5,7mm de diâmetro. Os espécimes selecionados foram aleatoriamente divididos em 5 grupos de estudo (n=10): Grupo Controle + 0, 1, 3, 7 dias in situ; GI Clareamento + 0, 1, 3, 7 dias in situ; GII Microabrasão + 0, 1, 3, 7 dias in situ; GIII Microabrasão + Clareamento + 0, 1, 3, 7 dias in situ e GIV Microabrasão + 0, 1, 3, 7 dias in situ + Clareamento. Os resultados foram submetidos à análise de variância de dois fatores (ANOVA) para medidas repetidas, seguidas pelo teste de Tukey a 5%. Foi observado que a rugosidade superficial do esmalte dental não foi alterada pela realização do clareamento dental com/sem período in situ, mas foi significativamente influenciada pela realização da microabrasão associada ou não com o clareamento dental, com/sem período in situ. Já a microdureza do esmalte dental, em uma comparação entre grupos, sofreu redução significativa nos grupos onde a microabrasão do esmalte foi realizada, independente da associação com o clareamento dental; sendo que após o período in situ observou-se o reestabelecimento da microdureza... / The aim of this in situ study was to evaluate if the association of microabrasion enamel with dental bleaching causes damage to microabrasioned enamel properties, through the surface roughness, hardness of dental enamel and scanning electron microscopy tests. The factors under study were: procedure on 5 levels: control, microabrasion (Opalustre - Ultradent Products Inc. Utah, USA), dental bleaching (Opalescence Boost PF 38 % - Ultradent Prodcuts Inc. Utah, USA) and the association of the two procedures (immediate and late), as well as the maintenance time of the specimens in situ: 0, 1, 3, 7 days. The response variables were microhardness and surface roughness. Fifty healthy bovine teeth were used, sectioned into discs of enamel/dentin with 5.7 mm diameter. The selected specimens were randomly divided into 5 groups (n=10): control group + 0 , 1 , 3 , 7 days in situ; GI dental bleaching + 0, 1, 3, 7 days in situ; GII Microabrasion + 0, 1, 3, 7 days in situ; GIII Microabrasion + dental bleaching + 0, 1, 3, 7 days in situ and GIV Microabrasion + 0, 1, 3, 7 days in situ + dental bleaching. The results were subjected to two way repeated measures ANOVA for followed by the Tukey test at 5 %. It was observed that the enamel roughness surface was not altered by dental bleaching, with/without in situ period, but was significantly influenced by microabrasion, associated or not with dental bleaching, with/without in situ period; whereas the enamel microhardness, in a comparison between groups, was significantly decreased in the groups where enamel microabrasion was performed, regardless of association with dental bleaching; after the in situ period, there was satisfactory reestablishment of enamel microhardness. Influência da tiouréia no efeito clareador do peróxido de hidrogênio /

Para tal, foram misturados dentro de um frasco Becker uma associação de cromógenos (vinho tinto, tabaco e café), peróxido de hidrogênio em quantidade suficiente para que se obtivesse uma concentração final de 35%, solução de hidróxido de sódio para o ajuste de pH e concentrações variadas de tiouréia como catalisador químico. Foram testadas cinco diferentes concentrações de tiouréia (0,05%, 0,125%, 0,25%, 0,375% e 0,425%), sendo pH inicial de todas as associações ajustado para 7. Um grupo controle sem catalisador também foi avaliado. Após a mistura, a cor inicial das soluções e a concentração de peróxido foram avaliados empregando, respectivamente, espectrofotômetro colorimétrico e um titulador automático. As leituras foram repetidas após 30 minutos. A mudança na variação de cor (∆E) e o percentual de queda na concentração de peróxido após a reação foram analisados estatisticamente, assim como a correlação entre as duas variáveis foi avaliada. Os dados foram analisados utilizando os testes de ANOVA a um fator e Tukey. Diferenças significavas foram observadas para a variação de cor (p=0,0001) e para a concentração de H2O2 (p=0,0001). (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: The aim of this study was to evaluate thiourea (T) effectiveness as a chemical catalyst on hydrogen peroxide bleaching efficiency. For that, an association of chromogens (red wine, tobacco and coffee) were mixed in a Becker bottle with hydrogen peroxide in sufficient amount to obtain a final concentration of 35%, sodium hydroxide solution for pH adjustment and a range of different concentrations of thiourea as a chemical catalyst. Five different concentrations of thiourea were tested (0.05%, 0.125%, 0.25%, 0.375% and 0.425%), with the pH adjusted to 7. A control group without a catalyst was also evaluated. After mixing, the baseline color of the solutions and the peroxide concentration were evaluated using a colorimetric spectrophotometer and an automatic titrator. The readings were repeated after 30 minutes. The color change (ΔE) and the percentage of peroxide concentration reduction after the reaction were statistically analyzed, as well as the correlation between the two variables.
