Avaliação do processo de reparo ósseo após enxerto ósseo autógeno tratado com tetraciclina hidroclorídrica : estudo histomorfológico em tíbia de coelhos /

Foram realizadas análises histológicas e submetidas a tratamento estatístico após histometria. Os resultados mostraram que não houve, estatisticamente, diferenças significantes entre o grupo controle onde não houve nenhuma implantação, e o grupo tratado utilizando a tetraciclina. O processo de reparo ocorreu de maneira similar em ambos os grupos. Não houve reabsorção completa dos fragmentos ósseos implantados, permanecendo até o período estudado de 30 dias pós-operatórios. / Abstract: The aim of the present study was to evaluate the bone repair process in experimental bone cavities which were realized in rabbit tibias when particles of homogenous bone graft were implanted and treated with tetracycline. Histolological analysis were carried out and undergone a statistical treatment after HISTOMETRIA. The results showed that the hydrochloric tetracycline didnþt interfered on the bone graft performance, which was used on the treatment of experimental bone defects. There wasnþt histological evidence of the decalcifier action of tetracycline; it could be the medicine exposion time of the bone graft. Evaluating the statistical results; it was observed that the tetracycline didnþt interfered on bone repair process, moreover its presence doesnþt cause delay or any significant interference that changes the healing process. / Mestre

Amostras foram obtidas das superfícies radiculares de dentes extraídos de humanos, sendo submetidos à instrumentação rotatória e raspagem e aplainamento radicular previamente a aplicação do agente condicionante. O cloridrato de tetraciclina foi aplicado por contato ou por fricção com bolinhas de algodão trocadas a cada 30 segundos, em diferentes concentrações: 50, 125, 250 e 500 mg/ml e em diferentes intervalos: 1, 2 e 3 minutos. O grupo controle consistiu de aplicação de soro fisiológico nos mesmos modos e tempos dos grupos testes. Concluímos que: -as concentrações de 125 e 250 mg/ml de tetraciclina foram as que apresentaram os melhores resultados na remoção da "smear layer"; - a aplicação por fricção mostrou ser mais eficiente na remoção da "smear layer"; - os resultados não foram tempo-dependentes. Quando verificou-se as variáveis conjugadas, a concentração de 50 mg/ml sob fricção por 1 minuto e a concentração de 125 mg/ml sob fricção por 1 minuto foram as que apresentaram os melhores resultados. / Abstract: The goal of this study is to assess the capacity of tetracycline HCl in removing -in vitro, in different concentrations, during different intervals and according to different modes of application-radicular smear layer resulting from scaling and planing root surfaces of human extracted teeth, which had been, previously, submitted to rotary instrumentation. Cotton pellets, soaked in a solution of tetracycline HCl, in concentrations of 50, 125, 250, or 500mg/ml, were either gently "placed" or "rubbed" on the prepared root surfaces for either one, two or three minutes. Pellets were changed every 30 seconds. The control group consisted of applying physiological serum during the same intervals and according to the same modes of application of those used for the testing-groups. As a conclusion, the strongestly recommended concentrations of tetracycline for removing radicular smear layer were the ones of 125 and 250 mg/ml; that rubbing root surfaces proved to be the most efficient on the removal of radicular smear layer; and that the results reached in the present study were not time-dependent. When all assessment criteria were simultaneously applied, the best results derived not only from rubbing the prepared surfaces in a concentration of 50 mg/ml of tetracycline HCl, during one minute, but also from rubbing them in a concentration of 125 mg/ml of tetracycline HCl, during one minute. / Mestre

O objetivo deste estudo in vitro foi avaliar, por meio de microscopia eletrônica de varredura, a remoção da smear layer e a exposição da matriz colágena da dentina produzidas pela aplicação de cloridrato de tetraciclina (TTC-HCl). O cemento radicular das amostras foi removido com fresas diamantadas, processo seguido de raspagem e alisamento radicular com cureta. As 450 amostras foram divididas em 10 grupos: controle (soro fisiológico) e os que compreendiam a aplicação de TTCHCl nas concentrações: 10mg/ml, 25mg/ml, 50mg/ml, 75mg/ml, 100mg/ml, 125mg/ml, 150mg/ml, 200mg/ml e 250mg/ml. Todos os grupos receberam essa aplicação de diferentes formas (passiva, pincel e fricção) e em diferentes tempos (1, 2 e 3 minutos). A avaliação foi feita por um examinador treinado, calibrado e que não conhecia o grupo ao qual as amostras pertenciam, utilizando o índice de Sampaio (1999) modificado para este estudo. Os dados foram analisados pelos testes de Kruskal- Wallis e de Dunn. As concentrações de 50mg/mL e 75mg/mL aplicadas por fricção e por pincelamento foram as mais efetivas na remoção de smear layer e na exposição de colágeno. O modo de aplicação passivo mostrou-se inferior aos demais (p=0,0001) com relação aos mesmos parâmetros, assim como o tempo de aplicação de 1 minuto também foi inferior em relação aos demais (p=0,002). Concluiu-se que as concentrações de 50mg/mL e 75mg/mL aplicadas por pincelamento ou fricção durante 2 ou 3 minutos foram as mais efetivas. / Abstract: Smear layer removal and collagen fibers exposure may improve periodontal treatment and regeneration. This in vitro study assessed smear layer removal and collagen fibers exposure after application of tetracycline hydrochloride (TTC-HCl) on root surfaces by means of scanning electron microscopy. Root cementum was removed with diamond burs followed by scaling and root planning. Four hundred and fifty samples were divided into 10 groups: control (application of saline) and application of TTC-HCl 10mg/ml, 25mg/ml, 50mg/ml, 75mg/ml, 100mg/ml, 125mg/ml, 150mg/ml, 200mg/ml and 250mg/ml. The TTCHCl application was performed in all groups in 3 different ways (passive, brushing and burnishing) and in 3 different times (1, 2 and 3 minutes). A previously trained, calibrated and blind examiner evaluated the photomicrographs using the Sampaio index (1999) modified for this study. Statistical analysis was performed by Kruskal-Wallis and Dunn tests. The 50mg/mL and 75mg/mL concentrations applied by burnishing or brushing were the most effectives in smear layer removal and collagen fibers exposure. Both, the passive mode of application (p=0.0001) and 1-minute time of application (p=0,002) were the less effectives. In conclusion, the 50mg/mL and 75mg/mL concentrations applied by brushing or burnishing during 2 or 3 minutes were the most effectives. / Doutor

Degradação de tetraciclina utilizando o processo foto-fenton /

Dessa forma, neste trabalho foi avaliada uma alternativa para a degradação do antibiótico tetraciclina (TC): a utilização do processo foto-Fenton. O monitoramento da concentração de TC foi feito por análises de Cromatografia Líquida de Alta Eficiência (CLAE) e a remoção da carga orgânica foi avaliada por determinações de Carbono Orgânico Total (COT). Dois sistemas de fotodegradação foram empregados, um utilizando radiação artificial e outro solar. Foi avaliada a influência de alguns fatores na fotodegradação de TC (24,0 mg L-1) como a utilização de ferrioxalato de potássio (FeOx) e Fe(NO3)3 que foram testados como fonte de ferro e apresentaram resultados diferentes. Sob radiação artificial, Fe(NO3)3 apresentou os melhores resultados pois a total oxidação do composto alvo foi atingida em apenas um minuto de tratamento, enquanto que na presença de FeOx a total oxidação ocorreu em 10 minutos. Quando a concentração de Fe(NO3)3 foi variada de 0,200 mol L-1 para 0,100 mol L-1 foi verificado um decréscimo na remoção de COT de 74% para 47%. Já sob radiação solar, os melhores resultados foram obtidos com FeOx, pois em 30 segundos de tratamento a TC foi totalmente oxidada. Outro parâmetro avaliado foi a influência da concentração de H2O2 e a maior concentração estudada (20 mmol L-1) prejudicou a fotodegradação, enquanto que a menor concentração testada (3,00 mmol L-1) manteve boa remoção de COT (76%) e de oxidação de TC (100%). A degradação de TC foi também avaliada em matrizes como água superficial e efluente de estação de tratamento de esgoto. Nessas matrizes, FeOx sob radiação solar manteve bons resultados para a oxidação de TC, com o qual foram alcançados percentuais de 85 e 74% de remoção de COT na água superficial e efluente de ETE, respectivamente. / Abstract: Pharmaceutical residues are being found frequently in aquatic environments what may cause environmental problems. Therefore, in this work an alternative for the degradation of the tetracycline (TC) antibiotic was evaluated: the use of the photo-Fenton process. The monitoring of the TC concentration was made by High Performance Liquid Chromatography (HPLC) analysis and the removal of the organic load was evaluated by Total Organic Carbon determination (TOC). Two systems of photodegradation were employed, one using artificial radiation and other using solar radiation. The influence of some factors in the photodegradation of TC (24,0 mg L-1) was evaluated such as the use of potassium ferrioxalate (FeOx) and Fe(NO3)3 that were tested as iron source and presented different results. Under artificial radiation, Fe(NO3)3 presented the best results since total oxidation of the target compound was achieved in only one minute of treatment, while in presence of FeOx the total oxidation occurred in 10 minutes. When the concentration of Fe(NO3)3 was varied from 0,200 mol L-1 to 0,100 mol L-1 a decrease in the TOC removal from 74% to 47% was verified. Regarding solar radiation, best results were obtained with FeOx, since in 30 seconds of treatment, the TC was totally oxidized. Other parameter evaluated was the influence of the H2O2 concentration. The highest concentration studied (20 mmol L-1) hindered the photodegradation, while the lowest concentration (3,00 mmol L-1) tested provided good TOC removal (76%) and TC oxidation (100%). The degradation of TC was also evaluated in matrices such as surface water and effluent of sewage treatment plant. FeOx under solar radiation maintained good results for the TC oxidation with which percentages of 85% and 74% of TOC removal were achieved, respectively. / Mestre

Efeito do condicionamento radicular na reparação periodontal de defeitos tipo deiscência : Estudo experimental em cães /

Defeitos tipo deiscência com 5 × 3 mm foram criados cirurgicamente na face vestibular das raízes mesiais dos primeiros molares, segundos e terceiros pré-molares superiores de sete cães. Assim, em cada animal foram criados seis defeitos que foram posteriormente submetidos ao acúmulo de placa por 45 dias. Após este período, foi realizado o exame clínico (baseline) dos dentes envolvidos no experimento e, em seguida, um retalho de espessura total foi rebatido. As superfícies radiculares foram raspadas e os defeitos receberam um dos seguintes tratamentos: I) irrigação com solução salina estéril (controle); II) ácido cítrico (25%, 3 min); III) EDTA (24%, 3 min); IV) tetraciclina hidroclorada (50 mg/ml, 3 min); V) ácido cítrico (3 min) + tetraciclina hidroclorada (3 min); VI) tetraciclina hidroclorada (3 min) + ácido cítrico (3 min). Após três meses de cicatrização, foi realizado o exame clínico final e os animas foram sacrificados para obtenção dos blocos contendo dente e tecidos periodontais adjacentes para análise histológica. Clinicamente, foi verificado que houve uma redução significativa da profundidade de sondagem e um significativo ganho de inserção clínica em todos os grupos após o tratamento dos defeitos (p < 0.05), porém sem diferença estatisticamente significante entre os grupos experimentais (p > 0.05). Em relação aos parâmetros histológicos avaliados, também não foi constatado uma diferença significativa entre os grupos (p > 0.05). (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: The purpose of this study was to clinically and histologically evaluate the effect of root surface conditioning with citric acid (CA), tetracycline hydrochloride (TTC HCl), and ethylenediaminetetraacetic acid (EDTA) on the periodontal repair of dehiscence-type defects in dogs. Dehiscence defects measuring 5 × 3 mm were surgically created on the buccal aspect of mesial roots of the maxillary first molars and second and third pre-molars of seven mongrel dogs. For each animal, a total of six defects were created and exposed to plaque accumulation for 45 days. After this period, clinical evaluation (baseline) of all teeth was performed and a full thickness buccal flap raised. The root surfaces were scaled and the defects were randomly assigned to one of the following treatments: I) irrigation with sterile saline solution; II) citric acid (25%, 3 min); III) EDTA (24%, 3 min); IV) tetracycline hydrochloride (50 mg/ml, 3 min); V) citric acid (3 min) and tetracycline hydrochloride (3 min); VI) tetracycline hydrochloride (3 min) and citric acid (3 min). After 3 months of healing, the clinical parameters (final) were evaluated and the animals were sacrificed to obtain block sections of the teeth and surrounding tissues for histological analyses. Clinically, all treatments yielded significant improvements in terms of probing depth decrease and attachment clinical level gain compared to baseline values (p < 0.05), however without significant differences among the experimental groups (p > 0.05). Epithelial apical migration as well as newly formed cementum and alveolar bone were similar in all treatment groups. No statistically significant differences were found among the groups in relation to the histological parameters evaluated (p > 0.05). Efeitos do condicionamento radicular com diferentes agentes para a adesão de plasma rico em plaquetas e de células sanguíneas : estudo in vitro /

Diferentes substâncias têm sido empregadas para remover esta camada e expor fibras colágenas da superfície dental. A adesão de elementos sangüíneos a superfícies radiculares desmineralizadas e a estabilização do colágeno pelas fibras colágenas são de extrema importância no sucesso da cirurgia periodontal. O objetivo deste estudo foi avaliar os diferentes padrões de adsorção e adesão de plasma rico em plaquetas (PRP) e de PRP + células sangüíneas a superfícies radiculares quimicamente condicionadas. Oitenta dentes foram raspados, eqüitativamente divididos em 5 grupos: irrigação com soro fisiológico (controle), aplicação de solução de ácido cítrico a 25%, gel de EDTA a 24%, solução de cloridrato de tetraciclina a 50mg/mL e solução de citrato de sódio a 30%. Metade das superfícies condicionadas foi exposta ao PRP e a outra metade ao PRP e sangue fresco para avaliação com microscopia eletrônica de varredura. Não houve diferenças estatisticamente significantes entre os grupos quando aplicou-se o PRP seguido de sangue. O EDTA e o ácido cítrico mostraram-se mais efetivos na remoção de smear layer, porém o ácido cítrico foi o único agente que apresentou adesão de PRP nas superfícies radiculares. Dessa forma, o emprego do PRP sobre a superfície radicular pareceu favorecer a adsorção e adesão de células sangüíneas e a estabilização da rede de fibrina. / Abstract: Smear layer removal and collagen fibers exposure may improve periodontal treatment and regeneration. Different substances have been used to remove it and to expose collagen fibers from tooth surface. Blood elements adhesion to demineralized roots and clot stabilization by collagen fibers are extremely important for the success of periodontal surgery. The aim of this study was to evaluate the different patterns of platelet - rich plasma (PRP) and PRP + blood cells adsorption and adhesion to root surfaces chemically conditioned. Eighty teeth were planed and equitably divided into five groups: irrigation with saline solution (control), application of a 25% citric acid solution, 24% EDTA gel, 50mg/mL tetracycline hydrochloride and 30% sodium citrate solution. Half of the conditioned surface was exposed to PRP and another half to the PRP and fresh blood and prepared for scanning electron microscopy. Planed root surfaces and conditioned with EDTA and citric acid were more effective on smear layer removal, but citric acid was the only agent that showed blood cells adhesion to root surface. This way, PRP employments on root surface probably improve blood element adsorption and adhesion to root surface and fibrin network stabilization. / Doutor
