Construction of a Synthetic Human VL Phage Display Library and Isolation of Potential Neuropilin-1-specific VL Therapeutics from the Library

Antibody phage display technology mimics the natural immune system, and has been widely used for rapid isolation of single-domain antibodies (sdAbs) with various binding specificities and affinities in the micromolar to low nanomolar range. SdAbs are the variable regions of immunoglobulins (e.g., VH, VL, VHH) and serve as potential probes with therapeutic value. The small size, high solubility, high expression and stability, and high specificity and affinity for the cognate antigen, make sdAbs ideal in improving drug delivery and the overall therapeutic value of antibodies. The main objective of this thesis was to construct a large VL phage display library (~1010 diversity); analyze it via sequence analysis, and to subtractively pan the library for isolation of Neuropilin-1 (NRP1)-specific VLs. Neuropilin-1 (NRP1), a cell-surface receptor for both vascular endothelial growth factor (VEGF) and class 3 Semaphorins (Sema3A), contributes to neuron cell death through its interaction with Sema3A in stroke patients. Disruption of this NRP1-Sema3A interaction would allow for axonal outgrowth and neuron regeneration in the area of the brain affected by stroke. Construction of the synthetic phage antibody library utilized a single VL framework with selected positions in the complementarity-determining regions (CDRs) targeted for randomization in vitro using synthetic oligonucleotides that introduced sequence degeneracy. Specific VLs were then selected from the repertoire through subtractive panning against a cell line endogenously expressing NRP1 (PC12) as well as a negative cell line that does not express NRP1 (HEK293) with competitive elution carried out using a synthetic Sema3A-derived peptide. Fifteen VL clones were isolated, cloned in E. coli, expressed and purified, and of these, nine were determined to be non-aggregating by size exclusion chromatography. Further studies will determine the potential therapeutic use of these VL sdAbs as agents in recovery from stroke and neuron degeneration.

Vastus medialis oblique : vastus lateralis muscle imbalance in patellofemoral pain syndrome (PFPS) patients

BACKGROUND and AIMS. Patellofemoral pain syndrome (PFPS) is complex and challenging musculoskeletal disorder. Maltracking of the patella is considered to be one of the primary causative factors. Vastus Medialis Oblique (VMO) and Vastus Lateralis (VL) muscle imbalance in terms of EMG magnitude and timed onset is implicated in either initiating or perpetuating the patellofemoral pain (Cowan et al, 2002, Witvrouw et al, 1996). Many physiotherapeutic treatments are aimed at addressing this muscle imbalance despite a lack of evidence confirming or refuting it exists and it's association with pain and function. The ultimate aim of the study was therefore to establish if it is appropriate to continue treating muscle imbalance in patients with clinically defined PFPS. OBJECTIVES. The overall objectives of the study were to establish: 1. If VMO-VL muscle imbalance exists in PFPS patients and if so is it specific to this condition or does a similar VMO-VL muscle imbalance exists in a healthy population? 2. If muscle imbalance does exist is it related to clinical symptoms used as indications of pain syndrome in clinical practice? 3. Is muscle imbalance associated with lower limb muscle physiology i.e. lower limb and quadriceps muscle strength in both fresh and fatigued states. METHODS. The study employed a cross-sectional design. 63 patients with patellofemoral pain syndrome (PFPS) and 63 age/sex matched healthy subjects were recruited and VMO & VL normalised EMG RMS amplitude and time onset differences were assessed during functional and experimental tasks. Additionally, correlations with pain level, functional status, muscular flexibility and biomechanical characteristics of the lower limb were explored. RESULTS. The results revealed that the VMO-VL activation patterns are task specific and most significantly related to functional stepping down task at a fast speed of execution (p=0.000). This interesting link between the type of muscle contraction, the speed of execution and the recruitment pattern of the VMO-VL was also confirmed by the non-functional isokinetic eccentric contraction (p=0.000). Additionally, it is the timing of the VMO-VL activation rather the intensity that is important. Also, a correlation appears to exist between activation pattern and duration of symptoms and knee functional performance (p=0.03) but not with the level of pain. CONCLUSION. The findings of the study suggest that the VMO-VL muscle imbalance does exist in a clinically defined PFPS population. Unlike previous studies however, this thesis suggests that specificity of the functional activities and speed of execution have a significant role to play in the muscular performance and it could be argued that this translates to a role in PFPS. It would therefore seem appropriate to continue addressing and treating this complex and challenging issue with physiotherapeutic interventions but this may need to be targeted to interventions that are tailored to addressing issues in relation to stepping down and at fast speed.

Clinical manifestations and anthropometric profiles of visceral leishmaniasis in selected centres in Ethiopia

Visceral leishmaniasis is a severe systemic illness and early case management is important for the avoidance complications and control of the disease. Improving health workers’ knowledge on leishmaniasis is essential in improving the control programme. A quantitative, retrospective study of patient records and descriptive, explorative study of health care professionals’ knowledge on leishmaniasis were conducted. Data was collected from patient records (n=299) using a structured audit tool and from health care professionals (n=55) by means of a structured questionnaire. The study findings highlight that the commonest clinical manifestations of visceral leishmaniasis are fever and splenomegaly. Severe malnutrition and HIV co-infection contribute to mortality. The findings indicate the need for training to improve health care professionals’ awareness of visceral leishmaniasis. Leishmaniasis disease surveillance and support by the regional and district heath offices should be improved / Health Studies / M.A. Public Health (MPH)

The focus of this work is to verify the possibility of very rapid collection of accurate data that can be easily analyzed and used for planning and business purposes. For data collection was chosen method of averaging the fullest and data analysis was used new software LČRTax, ULT and set of polynomials. The data found standing crops were analyzed with data after felling. In this work were used electronic devices allowing for a very accurate data. Digitech Professional caliper and Vertex Laser Altimeter. The results are confronted with at the end of the current economic situation.

Development and simulation of hard real-time switched-ethernet avionics data network

The computer and microelectronics technologies are developing very quickly nowadays. In the mean time, the modern integrated avionics systems are burgeoning unceasingly. The modern integrated modular architecture more and more requires the low-latency and reliable communication databus with the high bandwidth. The traditional avionics databus technology, such as ARINC429, can not provide enough high speed and size for data communication, and it is a problem to achieve transmission mission successfully between the advanced avionic devices with the sufficient bandwidth. AFDX(Avionics Full Duplex Switched Ethernet) is a good solution for this problem, which is the high-speed full duplex switched avionic databus, on the basis of the Ethernet technology. AFDX can not only avoid Ethernet conflicts and collisions, but also increase transmission rate with a lower weigh of the databus. AFDX is now adopted by A380,B787 aircraft successfully. The avionics data must be delivered punctualy and reliablely, so it is very essential to validate the real-time performance of AFDX during the design process. The simulation is a good method to acquire the network performance, but it only happends in some given set of scenarios, and it is impossible to consider every case. So a sophisticatd network performance method for the worst-case scenario with the pessimistic upper bound requires to be deduced. The avionic design engineers have launched many researches in the AFDX simulation and methods study. That is the goal that this thesis is aimming for. The development of this project can been planned in the following two steps. In the first step, a communication platform plans to be implemented to simulate the AFDX network in two versions – the RTAI realtime framework and Linux user space framework. Ultimately, these frameworks need to be integrated into net-ASS, which is an integrated simulation and assessment platform in the cranfield’s lab.The second step deduces an effective method to evaluate network performance, including three bounds(delay,backlog and output flow), based on the NC. It is called Network Calculus. It is an internet theory keeping the network system in determistic way. It is also used in communication queue management. This mathematics method is planed to be verified with simulation results from the AFDX simuation communication platform, in order to assure its validity and applicability. All in all, the project aims to assess the performance of different network topologies in different avionic architectures, through the simulation and the mathematical assessment. The technologies used in this thesis benefit to find problems and faults in the beginning stage of the avionics architecture design in the industrial project, especially, in terms of guarantee the lossless service in avionics databus.

Cellules solaires avec absorbeur II - VI nanostructuré : matériaux et propriétés / Extremely thin absorber “eta” solar cells with nanostructured II-VI absorber : materials and properties

L’objectif de ce travail est d’élaborer des méthodes peu chères pour produire desmatériaux semi-conducteurs pouvant entrer dans la fabrication de cellules solaires de type"eta" (extremely thin absorber). Ces cellules sont constituées d’une couche extrêmement fined’un adsorbeur inorganique dont la bande interdite est situé entre 1.1 et 1.8 eV placée entredeux nanostructures transparentes l’une de type n et l’autre de type p et dont les bandesinterdites doivent être supérieurs à 3.3 eV. Une couche compacte et des nanofils de ZnO ontété préparés en mode galvanostatique. Les dimensions des nanofils ont été contrôlées à l’aidede la couche compacte et de la densité du courant appliqué. La photosensibilisation desnanofils par des couches uniformes de CdS, CdSe et CdTe préparée par la méthode SILAR(Successive Ionic Layer Adsorption and Reaction) a été étudiée. Les propriétés de cescouches ont été améliorées par recuit et traitement chimique. En ce qui concerne les finescoquilles de CdTe deux autres méthodes de sensibilisation ont été également étudiées : la CSS(Close Spaced Sublimation) et les QDs (Quantum Dots). La première méthode conduit à unfaible recouvrement alors que la seconde produit un matériau mal défini optiquement. Leshétérostructures formées sur les nanofils ont été complétées par une couche de CuSCN, unsemi-conducteur de type p, préparée par trois méthodes différentes. L’influence de lamorphologie de ces couches sur les propriétés des cellules eta a été étudiée. Les filmspréparés par électrodéposition et SILAR sont plus rugueux que ceux obtenus parimprégnation et leur conductivité est moins bonne. Les hétérostructures (avec CdS et CdSecomme absorbeurs) ont été testées dans une cellule photoélectrochimique et les rendementsobtenus (jusque 2%) montrent une amélioration certaine des propriétés de ces matériauxpréparée par SILAR-modifiée ainsi que des interfaces ZnO/absorbeur. La qualité desmatériaux obtenus par SILAR montre qu’aujourd’hui on peut s’attendre à une Renaissance decette technique. / The development of semiconducting materials for the extremely thin absorber (eta)solar cell using cheap and scalable methods was the main objective of this work. The eta-solarcell is composed of all inorganic materials consisting of an extremely thin layer of absorbingmaterial (1.1 <Eg< 1.8 eV) sandwiched between nanostructured transparent electron and holeconductors (Eg ≥ 3.3 eV). Compact and defect free ZnO thin film and nanowires (NWs) wereprepared galvanostatically. The ZnO nanowire dimensions were controlled with the ZnO seedlayer or the applied current density. The photosensitization of the ZnO nanowires withconformal layers of CdS, CdSe and CdTe prepared by Successive Ionic Layer Adsorption andReaction (SILAR) was studied. The improvement of the absorber structural and opticalproperties by annealing and chemical treatment was achieved. The Close Spaced Sublimation(CSS) and Quantum Dot (QD) sensitization were also used for CdTe thin shell deposition,while the first method produced low coverage, the second resulted in better coverage but withnot optimal optical features. The ZnO NW/absorber heterostructure was completed with ahole conducting CuSCN layer. The influence of the CuSCN layer (prepared by three methods)morphology on the eta-solar cell performance is discussed. Electrodeposited and SILARprepared films exhibited rougher surfaces than that by the Impregnation technique (whichaffects the electrical conductivity). The ZnO/absorber core/shell heterostructures were alsotested in a photoelectrochemical cell. The recorded efficiencies (up to 2 %) for the case ofCdS and CdSe photosensitizers demonstrated an improvement of the ZnO/absorber interfacesand the material quality achieved by the modified-SILAR technique. These results let us toconsider that today a Renascence of the SILAR method is happening.
