In vitro absorption of valine, threonine and methionine by the small intestine of sheep

Duodenal, jejunal and ileal sections from sheep were used to study <i>in vitro</i> absorption of valine, threonine and methionine. Everted sacs were incubated for 45 minutes at 39 C under an atmosphere of 95% O₂ - 5% CO₂ in Krebs-Ringer bicarbonate buffer (mucosal and serosal fluid) containing 5 μmoles per ml of the test amino acid. The hydrolyzed tissue and mucosal and serosal fluids were analyzed for amino acid content. The duodenum absorbed smaller quantities of amino acids from the mucosal fluid than the jejunum and significantly (P<.01) less than the ileum. The release of amino acids from duodenal tissue was small for threonine and methionine and negative for valine. Accumulation of amino acids by duodenal tissue was not significantly different from the ileum, but this represented a larger part of the amount absorbed from the mucosal fluid. The duodenum appears to be of limited importance as a site of amino acid absorption. The jejunum readily absorbed valine and methionine, but absorbed only a small amount of threonine from the mucosal fluid. The release of amino acids into the serosal fluid by the jejunum was very slight in comparison to the ileum (P<.01). Tissue accumulation of amino acids was significantly less (P<.05) than in either the duodenum or ileum. While the jejunum generally absorbed large amounts of amino acids from the mucosal fluid, it did not release or accumulate amino acids in large amounts. The ileum maximized movement of all amino acids. It absorbed significantly more (P<.01) amino acids from the mucosal fluid and released significantly more (P<.01) amino acids into the serosal fluid. The accumulation of valine and threonine by ileal tissue was significantly greater (P<.05) than the jejunum. The ileum, therefore, appears to be the most active and efficient site of valine, threonine and methionine absorption in the sheep. / Master of Science

Exigências de valina para frangos de corte / Requirements of valine for broilers

O presente estudo teve como objetivo avaliar níveis ótimos de valina digestível. Foram conduzidos três ensaios com frangos de corte da linhagem Cobb nas fases inicial (1-14 dias), crescimento (14-28 dias) e terminação (28-42 dias), com ensaios realizados separadamente para cada fase, tendo como base o método dose-resposta. Para os ensaios de 1 a 14dias e de 14 a 28 dias, 672 amimais foram distribuídos em delineamento inteiramente ao acaso, em oito tratamentos (7 níveis de valina e 1 controle), com 7 repetições, sendo cada unidade experimental composta por 12 aves , porém no ensaio de 28 a 42 dias foram utilizados 560 frangos, igualmente distribuídos como nos ensaios anteriores em 8 tratamentos com 7 repetições com 10 aves cada. Foram formuladas dietas basais pela técnica da diluição, deficientes em valina, contendo níveis de energia, minerais e vitaminas conforme recomendações de Rostagno et al. (2005), para cada fase. As recomendações dos níveis de valina digestível para cada fase, obtidas no presente estudo, foram realizadas com base nas respostas de CA. As recomendações obtidas pelo método dose resposta são de 0,917; 0,905 e 0,783% de valina digestível para frangos de 1 a 14 dias, 14 a 28 e 28 a 42 dias de idade, respectivamente. / This study was to evaluate optimal levels of digestible valine. Three trials were conducted with broiler chickens of Cobb in the initial phase (1-14 days), growth (14-28 days) and termination (28-42 days), with tests performed separately for each phase, based on the method dose-response. For tests 1 to 14 days and 14 to 28 days, 672 animals it were distributed in a completely randomized in eight treatments (seven levels of valine and 1 control), with 7 replicates, each replicate consisting of 12 birds but the test of 28 to 42 days 560 chickens were used, equally distributed as in previous trials in 8 treatments with 7 replicates of 10 birds each. Basal diets were formulated by the dilution technique, deficient in valine, containing levels of energy, minerals and vitamins as recommended by Rostagno et al. (2005), for each phase. The recommended levels of digestible valine for each phase, obtained in this study were based on responses from CA. The recommendations obtained by the dose response are 0.917, 0.905 and 0.783% of digestible valine for broilers from 1-14 days, 14-28 and 28-42 days of age, respectively.

Development of a thermometric sensor for fructosyl valine and fructose using molecularly imprinted polymers as a recognition element

Nature has always served as a model for mimicking and inspiration to humans in their efforts to improve their life. Researchers have been inspired by nature to produce biomimetic materials with molecular recognition properties by design rather than evolution. Molecular imprinting is one way to prepare such materials. Such smart materials with new functionalities are at the forefront of the development of a relevant number of ongoing and perspective applications ranging from consumer to space industry. Molecularly imprinted polymers were developed by mimicking the natural enzymes or antibodies that serve as host for binding target molecules. These imprints were used as a recognition element to substitute natural biomolecules in biosensors. The concept behind molecular imprinting is to mold a material (with the desired chemical properties) around individual molecules. Upon removal of the molecular templates, one is left with regions in the molded material that fit the shape of the template molecules. Thus, molecular imprinting results in materials that can selectively bind to molecules of interest. Imprinted materials resulted in applications ranging from chemical separation to bioanalytics. In this work attempts were made particularly in the development of molecularly imprinted polymer based thermometric sensors. The main effort was focused towards the development of an covalently imprinted polymer that would be able to selectively bind fructosyl valine (Fru-Val), the N-terminal constituent of hemoglobin A1c ß-chains. Taking into account the known advantages of imprinted polymers, e.g. robustness, thermal and chemical stability, imprinted materials were successfully used as a recognition element in the sensor. One of the serious problems associated with the development of MIP sensors and which lies in the absence of a generic procedure for the transformation of the polymer-template binding event into a detectable signal has been addressed by developing the "thermometric" approach. In general the developed approach gives a new insight on MIP/Analyte interactions. / In dem Bestreben, ihr eigenes Leben zu verbessern, haben die Menschen stets die Natur nachgeahmt und sich von ihr inspirieren lassen. Die Natur hat Forscher zur Erzeugung smarter biomimetischer Stoffe mit molekularen Erkennungseigenschaften nach dem Vorbild der Evolution inspiriert. Eine der Methoden zur Herstellung solcher Substanzen ist das molekulare Prägen. Smarte Materialien mit neuen Eigenschaften stehen an der Spitze der Entwicklung potentieller Anwendungen vom Verbraucher bis hin zur Raumfahrtindustrie. Durch Nachahmung von natürlichen Enzymen oder Antikörpern wurden molekular geprägte Polymere (MIPs) entwickelt, die der Bindung von Zielmolekülen dienen. Diese geprägten Polymere (imprints) wurden anstelle von Biomolekülen als Erkennungselemente in Biosensoren eingesetzt. Das Konzept, das dem molekularen Prägen zugrunde liegt, besteht in der Formung eines Polymers (mit den entsprechenden chemischen Eigenschaften) um einzelne Zielmoleküle herum. Nach Entfernen dieser molekularen Template bleiben Abdrücke im Polymer übrig, die der Form der Templatmoleküle entsprechen. Mit Hilfe des molekularen Prägens kann man also Stoffe herstellen, die sich selektiv an bestimmte Moleküle binden können. Geprägte Polymere finden breite Anwendung, etwa in chemischen Aufreinigungsprozessen und der Bioanalytik. Hauptanliegen der vorliegenden Arbeit war es, thermometrische Sensoren auf der Basis molekular geprägter Polymere zu entwickeln. Die Anstrengungen richteten sich vor allem auf die Entwicklung eines kovalent geprägten Polymers, das in der Lage ist, selektiv Fruktosyl-Valin (Fru-Val), den N-terminalen Bereich von Hämoglobin A1c, zu binden. Aufgrund der bekannten Vorzüge geprägter Polymere – z. B. Robustheit und thermische und chemische Stabilität – wurden geprägte Polymere erfolgreich als Erkennungselement im Sensor angewendet. Eine der größten Herausforderungen bei der Entwicklung von MIP-Sensoren, das Fehlen eines generischen Verfahrens zur Umwandlung der Bindungsreaktion in ein nachweisbares Signal, wurde mit der Entwicklung der thermometrischen Methode in Angriff genommen. Diese Methode führt allgemein zu neuen Einsichten in die Interaktionen zwischen MIP und Analyt.

Suplementação de aminoácidos de cadeia ramificada em dietas com redução protéica para frangos de corte / Branched chain amino acids supplementation in diets with protein reduction for broilers chickens

Esta tese foi realizada objetivando comparar 4 programas alimentares, em dietas à base de milho e farelo de soja, formuladas com ou sem restrição de proteína bruta (PB), suplementadas ou não com L-Val e L-Ile, e usando diferentes níveis de lisina digestível (dig.). Os programas alimentares (PRG) foram: PRG 1, PB limitada a um mínimo (22,4, 21,1, 19,8, 18,4% para as fases pré-inicial, inicial, crescimento e final, respectivamente, com relações de AA:Lis definidos apenas para Met+Cis (0,72) e Tre (0,65); PRG 2, a PB não foi restrita e as relações estendidas para Val (0,77) e Ile (0,67); PRG 3, mesmas restrições do PRG 2 e suplementadas com L-Val; PRG 4, mesmas restrições do PRG 3, e suplementadas com L-Ile. Dois experimentos foram realizados, um com 1.800 e outro com 4.800 pintos machos Cobb x Cobb 500 de 1 dia de idade. No primeiro experimento, as rações foram formuladas usando os 4 PRG e níveis de Lis dig. de 1,324%, 1,217%, 1,095% e 1,006% ou 5% maiores, para cada fase, totalizando 8 tratamentos e 9 repetições cada. Para o segundo experimento, os 4 PRG foram utilizados até 1-21 d, totalizando 4 tratamentos com 48 repetições. E de 22 a 42 dias, cada PRG foi subdividido nos 4 PRG, passando cada PRG da fase anterior a receber todos os 4 PRG para as fases de crescimento e final, totalizando 16 tratamentos com 12 repetições cada. Não houve interação entre os tratamentos em ambos os experimentos, com exceção para o ganho de peso (GP) e conversão alimentar (CA) de 36 a 43d, no primeiro experimento, onde aves alimentadas com PRG 2 demonstraram melhorias no GP e CA quando alimentadas com um aumento de 5% no nível de Lis. Também, o GP e a CA acumulados aos 35 e 43d foram melhores quando as aves foram alimentadas com o PRG 2, sem diferença estatística para o PRG 3 e 4. Um aumento em 5% no nível de Lis dig. resultou na melhoria da CA acumulativa aos 43d. A gordura abdominal, como porcentagem da carcaça eviscerada aos 43d, foi maior para as aves alimentadas com a PRG 1. No segundo experimento, as aves alimentadas com PRG 2 tiveram melhor CA de 22 a 42d e de 1 a 42d, mas sem diferença estatística para os PRG 3 e 4. As aves alimentadas com PGR 1 de 1 a 42d apresentou o menor GP e o pior CA sem diferenças entre os PGR 2 e 3, e os PGR 3 e 4, respectivamente. Os dados do presente estudo demonstram que a suplementação de dietas para frangos de corte com fontes sintéticas dos cinco primeiros AA limitantes permitiram resultados de desempenho semelhantes a uma dieta com PB restrita a um valor mínimo. / This thesis was carried out to compare four corn-soy feeding programs formulated with or without crude protein (CP) restrictions supplemented with or without L-Val and L-Ile, and using different digestible (dig.) Lys levels. For the second experiment, the 4 PRG were used from 1 to 21 d, totaling 4 treatments with 48 replicates. From 22 to 42d, each PRG was subdivided in 4 PRG having the same rational as it was done to 21d, where each of the 4 PRG began receiving all 4 PRG for grower and finisher phases, totaling 16 treatments with 12 replicates each. No interaction was found between treatments in both experiments, with one exception for body weight gain (BWG) and feed conversion ratio (FCR) from 36 to 43d, in the first experiment, with birds fed PRG 2 demonstrating improvements in BWG and FCR when fed the 5% increasing in dig. Lys. And also, cumulative BWG and FCR at 35 and 43d and in each individual feeding phases showed broilers from PRG 2 having the best BWG and FCR; however, mean separations using Tukey showed no difference from birds fed PRG 3 and 4. Feeding a dietary program with 5% increase in dig. Lys resulted in improvement in cumulative FCR at 43d. Abdominal fat, as a percentage of the eviscerated carcass at 43d, was highest for birds fed the PRG 1. For the second experiment, birds fed with PRG 2 led to the best FCR from 22 to 42d and from 1 to 42d, but without statistical difference from PRG 3 and 4. Birds fed PGR 1 from 1 to 42d showed the lowest BWG and the highest FCR without statistical differences from PGR 2 and 3 and PGR 3 and 4, respectively. Data from the current study demonstrate that the supplementation of broiler diets with crystalline sources of the first five limiting AA allowed performance results similar to those produced when CP is restricted to a minimum.

Late gestation lysine and energy effects in sows and dose-responses to tryptophan and valine in finishing pigs

Dritz / The overall goal of this dissertation was to evaluate the effects of different amino acid (AA) levels on performance of pigs under commercial conditions. To reach this objective, a total of 12 experiments were conducted. For the sow research, 1,102 highly prolific sows were used to determine the effects of AA and energy intake during late gestation on piglet birth weight and reproductive performance of sows. Weight gain depended on the energy and AA intake levels while sows fed increased amount of energy had increased stillborn rate; however, there was no statistical differences due to energy intake in stillborn rate of gilts. The modest increase in individual piglet birth weight is due to energy rather than AA intake during late gestation. Pre-weaning mortality was reduced in piglets suckling from sows fed high AA diets during late gestation while subsequent reproductive performance was not affected. With recent advances in statistical computing capability, linear and non-linear mixed models were refined to estimate the AA ratio dose-response relationships. Then, 4 experiments using 2,420 wean-to-finish pigs were conducted to validate the methods for estimating the standardized ileal digestible (SID) AA to lysine (AA:Lys) ratio requirement. Subsequently, 7 experiments using 7,562 pigs were conducted to estimate the SID tryptophan (Trp) to Lys and Valine (Val) to Lys ratio requirements of wean-to-finish pigs. In 11- to 20-kg pigs, optimum SID Trp:Lys ranged from 16.6% for maximum mean G:F to 21.2% for ADG. In 30- to 125-kg pigs, optimum SID Trp:Lys ratio ranged from 16.9% for maximum mean G:F to 23.5% for ADG. However, 18% SID Trp:Lys captured 96 and 100% of the maximum mean ADG and G:F for finishing pigs, respectively. In 25- to 45-kg pigs, optimum SID Val:Lys ratio ranged from 72.3% for maximum mean G:F to 74.4% for ADG with 99% of the maximum mean ADG and G:F at approximately 69% and 65% SID Val:Lys ratio, respectively. In conclusion, optimum SID Trp:Lys and Val:Lys were consistently higher for ADG than G:F. This finding is critical for conducting economic evaluations and reference tables such as NRC (2012) should consider presenting requirement values for different response criteria.

Os dados do presente estudo demonstram que a suplementação de dietas para frangos de corte com fontes sintéticas dos cinco primeiros AA limitantes permitiram resultados de desempenho semelhantes a uma dieta com PB restrita a um valor mínimo. / This thesis was carried out to compare four corn-soy feeding programs formulated with or without crude protein (CP) restrictions supplemented with or without L-Val and L-Ile, and using different digestible (dig.) Lys levels. Feeding programs (PRG) were: PRG 1, CP was restricted to a minimum (22.4, 21.1, 19.8, 18.4% for pre starter, starter, grower and finisher phases, respectively) with AA to Lys ratios set only for TSAA (0.72) and Thr (0.65); PRG 2, CP was not restricted while minimum ratios were also extended to Val (0.77) and Ile (0.67); PRG 3, restrictions were as in PRG 2 but with L-Val added; PRG 4, restrictions were as in PRG 3, but with L-Ile added. For this two experiments were conducted, one using 1,800 and the other using 4,800 oneday- old Cobb 500 male broiler chicks. For the first experiment, feeds had formulated using the 4 PRG and dig. Lys as 1.324%, 1.217%, 1.095% and 1.006% or 5% higher, for each phase, totaling 8 treatments with 9 replicates each. For the second experiment, the 4 PRG were used from 1 to 21 d, totaling 4 treatments with 48 replicates. From 22 to 42d, each PRG was subdivided in 4 PRG having the same rational as it was done to 21d, where each of the 4 PRG began receiving all 4 PRG for grower and finisher phases, totaling 16 treatments with 12 replicates each. No interaction was found between treatments in both experiments, with one exception for body weight gain (BWG) and feed conversion ratio (FCR) from 36 to 43d, in the first experiment, with birds fed PRG 2 demonstrating improvements in BWG and FCR when fed the 5% increasing in dig. Lys. And also, cumulative BWG and FCR at 35 and 43d and in each individual feeding phases showed broilers from PRG 2 having the best BWG and FCR; however, mean separations using Tukey showed no difference from birds fed PRG 3 and 4. Feeding a dietary program with 5% increase in dig. Lys resulted in improvement in cumulative FCR at 43d. Abdominal fat, as a percentage of the eviscerated carcass at 43d, was highest for birds fed the PRG 1. For the second experiment, birds fed with PRG 2 led to the best FCR from 22 to 42d and from 1 to 42d, but without statistical difference from PRG 3 and 4. Birds fed PGR 1 from 1 to 42d showed the lowest BWG and the highest FCR without statistical differences from PGR 2 and 3 and PGR 3 and 4, respectively. Data from the current study demonstrate that the supplementation of broiler diets with crystalline sources of the first five limiting AA allowed performance results similar to those produced when CP is restricted to a minimum.

O presente estudo teve como objetivo avaliar níveis ótimos de valina digestível. Foram conduzidos três ensaios com frangos de corte da linhagem Cobb nas fases inicial (1-14 dias), crescimento (14-28 dias) e terminação (28-42 dias), com ensaios realizados separadamente para cada fase, tendo como base o método dose-resposta. Para os ensaios de 1 a 14dias e de 14 a 28 dias, 672 amimais foram distribuídos em delineamento inteiramente ao acaso, em oito tratamentos (7 níveis de valina e 1 controle), com 7 repetições, sendo cada unidade experimental composta por 12 aves , porém no ensaio de 28 a 42 dias foram utilizados 560 frangos, igualmente distribuídos como nos ensaios anteriores em 8 tratamentos com 7 repetições com 10 aves cada. Foram formuladas dietas basais pela técnica da diluição, deficientes em valina, contendo níveis de energia, minerais e vitaminas conforme recomendações de Rostagno et al. (2005), para cada fase. As recomendações dos níveis de valina digestível para cada fase, obtidas no presente estudo, foram realizadas com base nas respostas de CA. As recomendações obtidas pelo método dose resposta são de 0,917; 0,905 e 0,783% de valina digestível para frangos de 1 a 14 dias, 14 a 28 e 28 a 42 dias de idade, respectivamente. / This study was to evaluate optimal levels of digestible valine. Three trials were conducted with broiler chickens of Cobb in the initial phase (1-14 days), growth (14-28 days) and termination (28-42 days), with tests performed separately for each phase, based on the method dose-response. For tests 1 to 14 days and 14 to 28 days, 672 animals it were distributed in a completely randomized in eight treatments (seven levels of valine and 1 control), with 7 replicates, each replicate consisting of 12 birds but the test of 28 to 42 days 560 chickens were used, equally distributed as in previous trials in 8 treatments with 7 replicates of 10 birds each. Basal diets were formulated by the dilution technique, deficient in valine, containing levels of energy, minerals and vitamins as recommended by Rostagno et al. (2005), for each phase. The recommended levels of digestible valine for each phase, obtained in this study were based on responses from CA. The recommendations obtained by the dose response are 0.917, 0.905 and 0.783% of digestible valine for broilers from 1-14 days, 14-28 and 28-42 days of age, respectively.

The DNA sequence and transcriptional analyses of Drosophila melanogaster transfer RNA valine genes

The nucleotide sequence of the single Drosophila meianogaster tRNA gene contained in the recombinant plasmid, pDtl20R was determined by the Maxam and Gilbert method. This plasmid hybridizes to the 90 BC site on the Val Drosophila polytene chromosomes, a minor site of tRNA4 hybridization. The Val nucleotide sequence of the tRNA4 gene present in pDtl20R differs at four Val positions from the sequence expected from that of tRNA4 . The four differences occur at nucleotides 16, 29, 41 and 57 in the coding region. Comparison of the DNA sequence of pDtl20R to that of the plasmid pDt92R, which also hybridizes to the 90 BC site, indicates that the Drosophila fragments contained in these two plasmids are either alleles or repeats. The implications of these findings are discussed. An in vitro transcription system was developed from a Drosophila Schneider II cell line. This homologous cell-free extract support specific and accurate transcription of various Drosophila tRNA Val genes. The major product of transcription is a tRNA precursor which is processed to a tRNA sized species. Transfer RNA valine genes originating from different sites on the Drosophila chromosomes are transcribed at different rates. Comparison of the sequences in the internal promoter regions of the various genes indicates that the few differences within the coding regions may not be responsible for the observed difference in the rates of transcription. This conclusion is substantiated by studies with hybrid genes constructed during the course of this work. Preliminary evidence indicates that the Val tRNA gene which is transcribed at the highest rate may be preceded in its 5'-flanking region by a positively modulating sequence. Val The precursor RNAs directed by various tRNA genes are also processed at different rates. Transcription and processing experiments with hybrid genes suggest that nucleotide changes within the coding region, which do not affect the rate of transcription, influence the rate of processing. Time course and competition experiments demonstrate that at least two kinetic steps are required for the formation of a stable transcription complex. Studies with an in vitro constructed mutant missing in nucleotides 51-61 in the tRNA coding region suggests that this deleted region (which is highly conserved in eukaryotic tRNAs) may be involved in the primary interaction required for tRNA gene transcription. / Science, Faculty of / Microbiology and Immunology, Department of / Graduate
