SHEAR RHEOMETRY PROTOCOLS TO ADVANCE THE DEVELOPMENT OF MICROSTRUCTURED FLUIDS

<p></p><p>This doctoral dissertation takes the reader through a journey where applied shear rheology and flow-velocimetry are used to understand the mesoscopic factors that control the flow behavior of three microstructured fluids. Three individual protocols that measure relative physical and mechanical properties of the flow are developed. Each protocol aims to advance the particular transformation of novel soft materials into a commercial product converging in the demonstration of the real the chemical, physical and thermodynamical factors that could potentially drive their successful transformation. </p> <p> </p> <p>First, this dissertation introduces the use of rotational and oscillatory shear rheometry to quantify the solvent evaporation effect on the flow behavior of polymer solutions used to fabricate isoporous asymmetric membranes. Three different A-B-C triblock copolymer were evaluated: polyisoprene-<i>b</i>-polystyrene-<i>b</i>-poly(4-vinylpyridine) (ISV); polyisoprene-<i>b</i>-polystyrene-<i>b</i>-poly(<i>N</i>,<i>N</i>-dimethylacrylamide) (ISD); and polyisoprene-<i>b</i>-polystyrene-<i>b</i>-poly(<i>tert</i>-butyl methacrylate) (ISB). The resulting evaporation-induced microstructure showed a solution viscosity and film viscoelasticity strongly dependent on the chemical structure of the triblock copolymer molecules. </p> <p> </p> <p>Furthermore, basic shear rheometry, flow birefringence, and advanced flow-velocimetry are used to deconvolute the flow-microstructure relationships of concentrated surfactant solutions. Sodium laureth sulfate in water (SLE<sub>1</sub>S) was used to replicate spherical, worm-like, and hexagonally packed micelles and lamellar structures. Interesting findings demonstrated that regular features of flow curves, such as power-law shear thinning behavior, resulted from a wide variety of experimental artifacts that appeared when measuring microstructured fluids with shear rheometry.</p> <p> </p> <p>Finally, the successful integration of shear rheometry to calculate essential parameters to be used in a cost-effective visualization technique (still in development) used to calculate the dissolution time of polymers is addressed. The use of oscillatory rheometry successfully quantify the viscoelastic response of polyvinyl alcohol (PVA) solutions and identify formulations changes such as additive addition. The flow behavior of PVA solutions was correlated to dissolution behavior proving that the developed protocol has a high potential as a first screening tool.</p><br><p></p>

Chimie dynamique: reconnaissance de nucléobases par des récepteurs synthétiques et isomérie cis-trans d'hydrazones acylées.Ce travail traite du développement des systèmes moléculaires qui peuvent s'adapter à l'addition de substances qui agissent comme un gabarit. Cette approche permet d'isoler une espèce majeure à partir d'un mélange de composés par le biais de la chimie combinatoire dynamique (CCD). La première partie de ma thèse de doctorat inclus l'utilisation d'un ADN simple brin (ADNsb) comme un gabarit pour le transfert d'information par auto-assemblage de récepteurs sans avoir besoin d'enzyme. De nouveaux récepteurs de l'adénine et de la guanine (pinces A et G) solubles dans l'eau ont été conçues dans ce but. Une approche utilisant la résonance magnétique nucléaire (RMN) a été utilisée pour déterminer l'affinité de liaison comme preuve d'une reconnaissance spécifique et efficace. Une évaluation dans l'eau par dichroïsme circulaire (CD) et mesure de la température de fusion par UV (Tm) a été réalisée. Cela a permis de tester respectivement la capacité d'auto-assemblage entre les pinces et un modèle ADNsb, et la force du processus de coopérativité. La deuxième partie de ce travail est axée sur le tri spontanné de motifs pyridine acylhydrazone et sur les configurations intéressantes qu'ils adoptent. Nous avons étudié la synthèse d'une série de motifs pyridine acylhydrazone: dimère, trimères et pentamères. Des études RMN ont permis d'évaluer les changements dans l'équilibre configurationnel cis / trans de ces systèmes dynamiques. Les études ont montré que l'équilibre attendu est biaise la cis acylhydrazone pyridine isomère a été observée par diffraction des rayons X. / Dynamic chemistry: nucleobase recognition by synthetic receptors and cis-trans acylhydrazone isomerism. This work deals with the development of molecular systems which can adapt upon the addition of substances that act as templates. This approach enables one major species to be identified from a mixture of compounds through the use of dynamic combinatorial chemistry (DCC). The first part of my PhD included the use of a single stranded DNA (ssDNA) as a template for information transfer via the self-assembly of receptors without the need for enzymes. New water soluble adenine and guanine receptors (A and G clamps) were designed and synthesised for this purpose. Nuclear magnetic resonance (NMR) titration studies were carried out to calculate the binding affinity and as a proof of specific and efficient recognition. An assessment in water via circular dichroism (CD) and UV temperature melting (Tm) studies was carried out. This tested the ability for self-assembly between the clamps and a ssDNA template and the strength of the cooperative process respectively. The second part of my PhD focused on the self-sorting of acylhydrazone pyridine motifs and the interesting configurations they adopt. The feasibility to synthesise these acylhydrazone pyridine motifs (dimer, trimers and pentamers) was investigated. X-ray and NMR studies showed that the equilibrium was found to be biased in an unusual way, and the cis acylhydrazone pyridine isomer was observed.

The degree of phosphorus saturation of agricultural soils in Brazil and Germany: New approaches for risk assessment of diffuse phosphorus losses and soil phosphorus management

Diffuse Phosphor (P)-Austräge aus der Landwirtschaft tragen zur Gewässereutrophierung bei. Der Phosphorsättigungsgrad (DPS) ist ein etablierter Parameter, um das P-Austragsrisiko aus Böden zu erfassen. Ein bodentypunabhängiger Ansatz, der die Abschätzung des DPS durch eine einfache Standardmethode wasserlöslichen P (WSP) ermöglicht (WSP-DPS-Ansatz), wurde an europäischen Böden entwickelt. In der Dissertation wurde dieser Ansatz erstmalig: i) an tropischen Böden getestet und ii) dazu verwendet P-Austragsrisiken von Boden-P-Monitoringdaten und von landwirtschaftlichen Institutionen empfohlenen P-Gehalten abzuleiten. Neben dem DPS wurde der Einfluss der in Brasilien gängigen anorganischen Oberflächendüngung auf das Austragsrisiko mittels Laboranalysen und Feldstudien erfasst. Die Bodentypunabhängigkeit des WSP-DPS-Ansatzes wurde für Böden Brasiliens bestätigt. Infrarotspektroskopische Analysen lieferten eine Erklärung für relativ niedrige gelöste P-Konzentrationen im Oberflächenabfluss von Oxisols. Pedotransferfunktionen zwischen WSP und Methoden, die in Brasilien und Deutschland zur Abschätzung pflanzenverfügbaren P verwendet werden, ermöglichten die Berechnung von DPS-Werten aus Monitoringdaten. Erste DPS-Karten zeigten relative geringe Austragsrisiken für das Untersuchungsgebiet in Brasilien und hohe Risiken für Deutschland, die teilweise durch unterschiedliche empfohlene Boden-P-Gehalte erklärbar waren. Um mit einer einfachen und kosteneffizienten Methode sowohl die landwirtschaftliche Produktion als auch den Gewässerschutz zu berücksichtigen, wurden die Wasser- und CaCl2-Methode zur Abschätzung von pflanzenverfügbarem P mit dem WSP-DPS-Ansatz kombiniert. Dieser Ansatz könnte helfen die Herausforderungen zu lösen mit denen die Menschheit in den nächsten Jahrzehnten bezüglich P in der Landwirtschaft konfrontiert sein wird: Einer effizienten Nutzung der limitierten Ressource P und dem Schutz der Gewässer vor diffusen P-Einträgen. / Diffuse phosphorus (P) losses from agriculture contribute to the eutrophication of surface waters. The degree of P saturation (DPS) is an established parameter for assessing the risk of P loss from agricultural soils. A soil type-independent approach for estimating the DPS by a simple standard method of water-soluble phosphorus (WSP; the WSP-DPS approach) was developed on European soils. In the thesis, the WSP-DPS approach was for the first time: i) tested on tropical soils and ii) used to derive P loss risks from soil P monitoring data and from recommended soil P levels by agricultural institutions. In addition to DPS, laboratory analyses and field studies were combined to assess the risk of P loss associated with the superficial application of inorganic fertilizer, which is commonly used in Brazil. The soil type-independency of the WSP-DPS approach was confirmed for soils of Brazil. Infrared spectroscopic analyses provided an explanation for the relatively low dissolved P concentrations in the surface runoff of Oxisols. Pedotransfer functions were determined between WSP and methods used to estimate plant-available P in Brazil and Germany and allowed for the transformation of soil P monitoring data into DPS values. The first DPS maps revealed relatively low P loss risks for the investigation area in Brazil and high risks for Germany. This difference was partly explainable by the recommended soil P levels in the two countries. To consider both agricultural production and the protection of surface waters in soil P management with a simple and cost-effective method, the soil test methods of using water and CaCl2 to estimate plant-available P and the WSP-DPS approach were combined. This approach could help to solve the challenges humanity faces regarding P in agriculture in the coming decades: An efficient use of the limited resource P and the protection of surface waters from diffuse P losses.
