The development of Deoxyribonucleic Acid (DNA) based methods for the identification and authentication of medicinal plant material

Herbal medicines are growing in popularity in the Western world and are becoming more stringently regulated under new EU legislation. Within the arena of herbal medicines, St. John’s Wort (SJW), Hypericum perforatum, is a top ten best seller with clinical evidence to support its use as an anti-depressant. A fundamental requirement of the new legislation is to prove the identity of the plant material in question. This is currently achieved via morphological and chemical methods, neither of which are ideal. A wide range of DNA based methods have been applied to this arena, standardisation is required to realise the potential of DNA based techniques. The DNA barcoding initiative aims to produce sequence data for all plant species, capable of species identification. The proposal is to use these data to design fast and effective DNA based methods of identification. For assay design, the putative barcode region nrITS was selected as a platform. Three assays were designed; • A PCR assay designed to hyper variable sequences within a barcode region. This assay is capable of distinguishing SJW from other closely related species. • A quantitative qPCR assay designed to measure total DNA and specific SJW DNA within a mixed sample. • A multiplex PCR incorporating fluorescently labelled primers, allowing amplicon detection by capillary electrophoresis. This assay identifies four separate Hypericum species, including SJW, with a mixed sample in one reaction. The suitability of the nrITS and three other barcode regions is assessed based on sequence data generated for 32 vouchered samples of different Hypericum species, and a Lithuanian sample set of 22 and 16 H. perforatum and H. maculatum samples respectively. The matK is currently unusable, the rbcL highly conserved, trnH-psbA problematically variable and the nrITS proved to be ideal for assay design.

Neste estudo realizamos uma prova de conceito através da combinação de três técnicas com a finalidade de monitorar a mosturação e fermentação da cerveja durante o processo de fabricação. O princípio deste trabalho é baseado em uma análise espectral, utilizando um equipamento de absorção na região do infravermelho médio por transformada de Fourier (FTIR - Fourier Transform Infrared) de amostras coletadas durante a fabricação da cerveja. Combinado com técnicas de processamento de Análise de Componentes Principais e Redes Neurais Artificiais é possível quantificar a concentração dos principais carboidratos e etanol presentes nestas amostras. Estas medidas físicas e químicas irão permitir a redução de erros durante a produção de cerveja além de optimizar as reações enzimáticas intrínsecas de suas principais etapas de análise. As técnicas ópticas de absorção, juntamente com o processamento neural, apresentam grandes vantagens, principalmente devido ao fato de serem facilmente adaptáveis aos equipamentos industriais, fornecendo respostas em curtos intervalos de tempo com alta sensibilidade e especificidade. / This study is fundamentally a proof of concept. By the combination of three techniques, our aim is to develop a new method of monitoring beer wort production and fermentation during brewing. The principle is based on spectral analyses, using Fourier Transform Infrared (FTIR) spectroscopy to collect absorption data from beer wort samples. This data is refined by the application of a statistical method, Principal Component Analysis (PCA), to reduce the number of variables. A computational method, Artificial Neural Network (ANN), enables quantification of carbohydrates and ethanol concentrations. Such physical-chemical measurements are expected to allow both reduction of mistakes during beer processing and optimization of enzymatic reactions, enhancing brewing processes. Optical absorption techniques associated with Artificial Neural Network present great advantages, mainly because the first ones are more easily inserted in industries than the latter ones, since they enable assessing the process status at short intervals, with high sensibility ; specificity.

Atua hidrolisando ligações alfa-1,4 do amido, mais precisamente nas cadeias lineares da amilose e amilopectina, liberando carboidratos de cadeias menores (maltose, glicose e maltotriose), necessários para processo de fermentação pela levedura cervejeira. Essa quebra ocorre basicamente para disponibilizar açúcares que posteriormente servirão para formação de álcool na cerveja. A atividade enzimática é inicialmente dependente da solubilização das enzimas, como exemplo na mosturação onde os grãos moídos são imersos em água. Existem outros fatores como temperatura, tempo e pH que garantem máxima eficiência enzimática, o que de certa forma é de grande importância para cervejaria na busca de um rendimento otimizado durante a brassagem. Este trabalho propôs um estudo da beta amilase em diferentes períodos de atuação sobre o amido presente no mosto cervejeiro avaliando qual o melhor período de atuação enzimática. Foi utilizada uma única temperatura, diferentes tempos de atuação, que vão de zero até 60 minutos, intercalados em cinco minutos. Todas as amostras foram fermentadas e analisadas individualmente, buscando identificar qual o melhor período de atuação desta enzima e como os mostos produzidos em diferentes períodos de tempo sofreram fermentação, de acordo com a graduação alcoólica, teor alcoólico e extrato real. / Beta amylase is an enzyme of the malt that has the property of breaking down starch into fermentable sugars. Connections acts hydrolyzing alpha -1 ,4 starch, more precisely in the linear chains of amylose and amylopectin, lower releasing carbohydrate chains ( maltose, maltotriose and glucose ) necessary to process the fermentation by the brewing yeast. This breakdown occurs primarily to provide sugars that subsequently serve to formation of alcohol in beer . The enzymatic activity is initially dependent on the solubilization of enzymes , such as the mashing where ground beans are immersed in water . There are other factors such as temperature , time and pH to ensure maximum enzyme efficiency, which is of great importance for brewery in search of a performance optimized during mashing somehow . This paper proposed a study of beta amylase in different periods of activity on the starch present in the beer wort evaluating the best period of enzymatic activity . A single temperature , different operating times , ranging from zero to 60 minutes , interspersed in five minutes was used . All samples were fermented individually analyzed in order to identify the best period of activity of this enzyme and the musts produced in different periods of time have undergone fermentation , according to alcohol content , alcohol content , real extract and apparent extract.

This thesis examines the relationship between the Holy Spirit and the Word of God and attempts to describe the precise nature of this relationship. Thus, after the introduction, a survey of research and a methodological introduction, the study illuminates the background of the Pauline understanding of the relationship between word and Spirit by examining the same relationship in the Old Testament, in Early Judaism, in the Jesus tradition and in the wider Graeco-Roman world. It emerges that Paul - in analogy to the Old Testament, Early Judaism and the theology of Jesus - saw a close relationship between the Holy Spirit and God's word. Obviously for Paul the Holy Spirit as a divine person of the trinity is more than the word but in his - mainly soteriological - work the Spirit has bound himself to God's word. Paul makes it clear that the Spirit works with the word everything for the spiritual life. Zusammenfassung In vorliegender Arbeit wird die Frage nach dem Zusammenhang von Gottes Geist und dem Wort Gottes bei Paulus aufgegriffen und der Versuch unternommen, eine Antwort zu geben. Dabei wird - nach der Einleitung, dem Forschungsüberblick und einer methodischen Einführung -, als Hintergrund des paulinischen Verständnisses von Wort und Geist, das Alte Testament, Frühjudentum, die Jesusüberlieferung und der Hellenismus auf diesen Zusammenhang hin befragt. Dabei zeigt sich, dass der Apostel Paulus in Analogie zum Alten Testament, Frühjudentum und der Theologie Jesu, einen engen Zusammenhang zwischen Heiligem Geist und Gottes Wort lehrt (,,Wort-Theologie"). Der Geist Gottes ist für Paulus als göttliche Person der Trinität natürlich mehr als das Wort, doch in seinem - vor allem soteriologischen - Wirken, hat er sich an das Wort Gottes gebunden. Paulus macht deutlich, dass der Geist mit dem Wort alles zum geistlichen Leben wirkt: Glaube (Wiedergeburt), Innewohnung des Geistes, ewiges Leben, Führung, Erkenntnis, Kraft, Gaben u.v.a. / New Testament / (M.Th.(New Testament))

<p>This paper deals with prehistoric charred organic material, interpreted as bread, found in graves and in the underlying settlement at Kalvshälla, Järfälla parish in Uppland. The aim was to categorize the organic material morphologically and with the help of Fourier transformed Infrared spectroscopy (FT-IR) to see whether it was bread or not. Material from 13 graves (dated to Late Roman Iron Age until Viking Age) and 3 finds from the settlement have been analysed. Differences were noted both in shape, porosity and structure. Some of the material has been interpreted as cereal based foodstuff instead of bread due to morphological discrepancies. No clear differences between material from the graves and the settlement can be seen. One find from a posthole can be dated to late Bronze Age, which is unusual. The bread in the graves was intentionally given to both adult females and males maybe as a symbol of their roles as the good householders. Several finds of root tubers of dropwort (Filipendula vulgaris) were identified in female graves from Early Iron Age, but also in two male graves with bread from Vendel period. The FT-IR method does not give a clear answer to whether the organic material can be bread or not. The method requires careful sampling procedures and several samples from each concretion to get good results.</p>

Wir versuchen herauszufinden, ob das subjektive Maß der Cloze-Vorhersagbarkeit mit der Kombination objektiver Maße (semantische und n-gram-Maße) geschätzt werden kann, die auf den statistischen Eigenschaften von Textkorpora beruhen. Die semantischen Maße werden entweder durch Abfragen von Internet-Suchmaschinen oder durch die Anwendung der Latent Semantic Analysis gebildet, während die n-gram-Wortmaße allein auf den Ergebnissen von Internet-Suchmaschinen basieren. Weiterhin untersuchen wir die Rolle der Cloze-Vorhersagbarkeit in SWIFT, einem Modell der Blickkontrolle, und wägen ab, ob andere Parameter den der Vorhersagbarkeit ersetzen können. Unsere Ergebnisse legen nahe, dass ein computationales Modell, welches Vorhersagbarkeitswerte berechnet, nicht nur Maße beachten muss, die die Relatiertheit eines Wortes zum Kontext darstellen; das Vorhandensein eines Maßes bezüglich der Nicht-Relatiertheit ist von ebenso großer Bedeutung. Obwohl hier jedoch nur Relatiertheits-Maße zur Verfügung stehen, sollte SWIFT ebensogute Ergebnisse liefern, wenn wir Cloze-Vorhersagbarkeit mit unseren Maßen ersetzen. / We try to determine whether it is possible to approximate the subjective Cloze predictability measure with two types of objective measures, semantic and word n-gram measures, based on the statistical properties of text corpora. The semantic measures are constructed either by querying Internet search engines or by applying Latent Semantic Analysis, while the word n-gram measures solely depend on the results of Internet search engines. We also analyse the role of Cloze predictability in the SWIFT eye movement model, and evaluate whether other parameters might be able to take the place of predictability. Our results suggest that a computational model that generates predictability values not only needs to use measures that can determine the relatedness of a word to its context; the presence of measures that assert unrelatedness is just as important. In spite of the fact, however, that we only have similarity measures, we predict that SWIFT should perform just as well when we replace Cloze predictability with our measures.

Die eingehende Analyse ausgewählter Aspekte der Wort-Ton-Gestaltung in Gustav Mahlers Vokalsinfonik ist Schwerpunkt der vorliegenden Untersuchung. Prämisse ist Mahlers Ausarbeitung ideenprogrammatischer Intentionen, wobei er sich die besondere Stärke der Musik, geistige Sachverhalte in gewisser Weise präziser als Sprache (die hier ins Symbolhafte ausweichen muss) verdeutlichen zu können, offensichtlich bewusst zunutze gemacht hatte. Die Differenziertheit der musikalischen Textinterpretation, zugleich Fokus der Analysen, weist deutliche graduelle Unterschiede auf. Sie reicht vom ,Vertonungscharakter‘ bis hin zur innigsten Anverwandlung von Musik und Textaussage bzw. -semantik. Wesentlich ist das oft proportionale Verhältnis von Grad der Freiheit, die sich Mahler hinsichtlich der Beziehung der Musik zur formalen Topographie des Textes nimmt, zu Intensität und Vielschichtigkeit der musikalischen Textexegese. Je stärker sich ihre musikalische Umsetzung in Richtung ‚Semantik-Ausarbeitung‘ bewegt, verliert die äußerliche Sprache an Materialität, wobei das im Lied von der Erde zutage tretende Amalgam von sprachlicher Semantik und Musik gesondert zu nennen ist. Letztendlich wird die überwiegend eher unbestimmte Textsemantik durch Mahlers Musik denotiert und in zuvor im Bereich der Vokalsinfonik nicht erreichter Intensität zum Klingen gebracht. / Focal point of this thesis is the detailed analysis of selected aspects of the relationship between music and text in Gustav Mahler’s vocal-instru-mental symphonies. Mahler’s elaboration of programmatic intentions functions as background. The composer obviously used the specific quality of music which is able to make spiritual or immaterial facts clearer than language (which here often has to switch over to symbolism). The variety of the text interpretation by Mahler’s music (on which the analyses are focused) shows distinct differences: from a relatively simple setting to music-character to an intense amalgam of music and semantics of the words. An important result of the analyses is the approximate proportionality between independency of Mahler’s music from the text’s formal parameters and intensity of the musical exegesis of the words. The more the music tends to elaborate the semantics, the more decreases the materiality of language. In this context the semantics-music-amalgam in the final movement of Das Lied von der Erde represents an unique example. Ultimately Mahler’s music supplies the denotation of the text semantics. This intermixture shows previously – in the history of development of vocal symphonic tradition – not heard intensity.

Untersuchungen zum Sprachentwicklungsverlauf von Kindern mit Lippen-, Kiefer-, Gaumenspalten im Alter von 0-1;6 Jahren

Bereits in frühen Phasen der Sprachentwicklung sind bei Kindern mit Spaltbildungen im Vergleich zu Kindern ohne Spaltbildungen Abweichungen beobachtet worden. In der Fachliteratur sind allerdings meist nur undifferenzierte Aussagen über den frühen strukturellen Verlauf zu finden. Daher wurde eine Untersuchung an 100 Kindern mit Spaltbildungen im Alter von 0-1;6 Jahren durchgeführt. Untersuchungsgegenstand waren die kindlichen Vokalisationen in der 1.Lallphase, der 2.Lallphase (prälinguistische Phase) und der Phase des Sprechbeginns (frühe linguistische Phase). Die Analyse umfaßte phonetische Parameter, den aktiven Wortschatz sowie die Silben- und Wortstrukturen. Die Auswertung ergab hinsichtlich des temporalen Verlaufs beim überwiegenden Teil der Probanden keine Unterschiede, hinsichtlich des strukturellen Verlaufs jedoch deutliche, typisch-spezifische Unterschiede im Vergleich zu altersgleichen Kindern ohne Spaltbildungen. / Even at the earliest stages, differences in speech development can be observed between children with cleft palate and those without cleft palate. One finds, however, only undifferentiated statements in the scientific literature on the early structural and linguistic phases. We therefore undertook an investigation of 100 children with cleft palate between the ages of 0 and 1;6 years. The subjects of the investigation were the children s vocalizations during the first and second babbling phases (prelinguistic phase) and during the onset of speech (early linguistic phase). The analysis included phonetic parameters and active vocabulary, as well as syllable and word structures. Our results showed no differences among the majority of the cildren, in terms of temporal development, and only typical-specific differences in structural development, in comparison with children of the same age without cleft palate.
