A Clorose Variegada de Citrus (CVC) é uma doença que vem atingindo grandes plantações de laranjas no Brasil e outros países como Estados Unidos, França e Espanha. O principal efeito da doença é o surgimento de manchas amarelas nas folhas que progridem para toda a extensão levando-a à necrose. Os frutos são pequenos, apresentam casca dura, sendo impróprios para o consumo. O agente causador da CVC é a bactéria Xylella fastidiosa que é limitada ao xilema. A bactéria é transmitida por insetos conhecidos como \"cigarrinhas\" que se alimentam na seiva do xilema. Esta bactéria apresenta em seu genoma um Operon contendo 9 genes (B,C,D,E,F,H,J,K,M) responsáveis pela biossíntese do exopolissacarídeo denominado goma fastidiana, que pode estar envolvido na sua patogenicidade. No presente trabalho, realizou-se estudos com as enzimas GumD (uma enzima glicosiltransferase I que faz a primeira adição de glicose-l-fosfato ao lipídeo prenol) e GumC (que provavelmente está envolvida na etapa de polimerização e/ou secreção do polissacarídeo formado através da membrana da bactéria), com o objetivo de contribuir para melhor entendimento da via biossintética. O gene gumD, que codifica a enzima GumD, foi clonado nos vetores de expressão pMAL¬c2x e pKK223-3. A proteína GumD foi purificada através das cromatografias de troca aniônica e filtração a gel. O peso molecular e o pI da enzima foram determinados usando respectivamente a técnica de filtração a gel e o sistema Fast de eletroforese. A enzima foi caracterizada quanto ao seu enovelamento através da técnica de Dicroísmo Circular, apresentando um espectro característico de estrutura secundária enovelada, composta predominantemente por a-hélices. O gene gumC, que codifica para a proteína GumC, foi clonado nos vetores de expressão pMAL-c2x (no qual a enzima é expressa em fusão com a proteína MBP - Maltose Binding Protein) e pET29a (a proteína é expressa sem fusão). A proteína em fusão GumC-MBP foi parcialmente pura através da coluna de amilose e foi caracterizada através da técnica de Imunoblotting. A enzima GumC expressa no vetor pET29a está em fase de purificação. Anticorpos anti-GumC-MBP foram produzidos em camundongos e serão utilizados como uma forma de caracterizar a proteína GumC expressa sem fusão. Estudos estruturais destas enzimas poderão trazer informações fundamentais para o conhecimento da via biossintética, assim como para o desenvolvimento de inibidores específicos / The Citrus Variegated Chlorosis (CVC) is a serious disease of orange trees in countries like Brazil, USA, France and Spain. Typical disease symptoms include conspicuous variegations with chlorotic areas on the upper side and small necrotic lesions on the lower side of the leaves. The affected fruits are smaller, hardened and without commercial value. Citrus variegated chlorosis is caused by Xylella fastidiosa, which is a xylem-limited bacterium. X fastidiosa is transmitted by specific sharpshooter leafhoppers when the insect feeds on the xylem sap. X fastidiosa has a nine-gene operon (B, C, D, E, F, H, J, K, and M genes) responsible for the biosynthesis of exopolysaccharides, denoted fastidian gum, which can be involved in its pathogenicity. GumD glycosyltransferase enzyme adds the first sugar, glycose-l-phosphate, to the prenol lipid. GumC enzyme probably is involved in the polymerization and/or exportation of the fastidian gum through the membrane of the bacterium. Studies were done on the GumD and GumC enzymes with the aim of getting a better understanding of the exopolysaccharide biosynthetic pathway. The gumD gene was cloned into the pMAL-c2x and pKK223-3 expression vectors, and GumD protein was purified through ion exchange and size exclusion chromatography. Then GumD molecular mass and pl were determined using, respectively, size exclusion chromatography and electrophoresis. In addition, according to circular dichroism spectroscopy, GumD prevalent secondary structure is u-helix. On the other hand, the gumC gene was cloned in two expression vectors: pMAL-c2x (the protein is expressed in fusion with Maltose Binding Protein ¬MBP) and pET29a (the protein is expressed without fusion in our strategy). GumC¬MBP, the protein in fusion, was partially purified using an amylose column and the fusion between GumC and MBP was confirmed with imunoblotting technique. Purification trials of GumC enzyme expressed in pET29a are in course. Anti-GumC¬MBP antibodies were already produced in mice and they will be used to characterize the GumC protein expression without fusion. Structural studies of GumD and GumC enzymes will provide information about the fastidian gum biosynthetic pathway, as well as to the development of specific inhibitors

A bactéria XvIella fastidiosa tem sido associada com doenças economicamente importantes como a clorose variegada de citros (CVC) em citros no Brasil e a doença de Pierce em videiras nos Estados Unidos (Hopkins, 1989 e Purcell et al., 1997). A importância econômica da indústria de citricultura no Brasil e o grande prejuízo causado pela CVC em pomares brasileiros têm resultado em um extensivo programa de pesquisa que começou com o sequenciamento do genoma completo da X Fastidiosa (Simpson., et ai 2000). A bactéria é transmitida por insetos conhecidos como \"cigarrinhas\" que se alimentam da seiva do xilema. O principal efeito da doença é o surgimento de manchas amarelas nas folhas que progridem para toda a extensão levando-a à necrose. Os frutos de plantas infectadas são pequenos (representando 1/3 do tamanho de um fruto normal), apresentam casca dura e são impróprios para o consumo. Esta bactéria apresenta em seu genoma um Operon contendo 9 genes (B,C,D,E,F,H,J,K,M) responsáveis pela biossíntese do exopolissacarídeo denominado goma fastidiana, que pode estar envolvido na sua patogenicidade. No presente trabalho temos como objetivo estudar as enzimas GUMC, que provavelmente está envolvida na etapa de polimerização e/ou secreção do polissacarídeo formado através da membrana da bactéria, e GUMK, uma glucuronosiltransferase ou glicosiltransferase IV, que adiciona uma molécula de ácido glicurônico à molécula da manose para a formação do tetrassacarídeo (manose-α-1,3- glucose-β-1,4-glucose-P-P-polyprenol). O gene gumC foi clonado no vetor de expressão pMALc-2x, a proteína de fusão GUMC-MBP foi expressa com uma massa molecular calculada de 98kDa e foi purificada através de coluna de afinidade. A proteína de fusão foi caracterizada pela reação com o anticorpo anti-MBP por imunoblotting. Através da técnica de Espalhamento Dinâmico de Luz (EDL), foram realizados estudos de agregação com a proteína de fusão GUMC-MBP. Ensaios de cristalização foram realizados e microcristais da proteína de fusão GUMC-MBP foram obtidos. Após a clivagem da proteína de fusão com a enzima Factor Xa, a enzima GUMC foi separada através da cromatografia de troca aniônica. A massa molecular da enzima foi determinada através da cromatografia de filtração em gel, superose 12, eluindo na forma monomérica com 53kDa. A proteína GUMC foi caracterizada pela reação com o anticorpo anti-GUMC-MBP (produzido em camundongos) através da técnica de imonublotting. A enzima também foi caracterizada através da técnica do dicroísmo circular (CD), apresentando um espectro característico de estrutura secundária enovelada, composta predominantemente por α-hélices. Foram realizadas 9 construções para a retirada das regiões transmembrânicas das regiões 5 e 3° do gene gumC (1 construção para o vetor de expressão pET29b e 8 para o vetor pMALc-2x). Em uma das construções, a proteína de fusão modificada GUMC-MBP 5C foi expressa no vetor de expressão pMALc-2x e foi purificada em coluna de afinidade. A proteína de fusão modificada GUMCMBP foi clivada com a enzima Factor Xa. Estudos de agregação molecular das proteínas de fusão GUMC-MBP e GUMC-MBP modificada C5 e das enzimas GUMC e GUMCC5 modificada poderão ser realizados futuramente. O gene gumK foi clonado no vetor pMAc-2x, a proteína de fusão GUMK-MBP foi expressa, com uma massa molecular calculada de 86kDa e foi purificada em coluna de afinidade. A proteína de fusão foi caracterizada por imunoblotting e medidas de dicroísmo circular foram realizadas. Após clivagem com o Factor Xa, a enzima GUMK foi purificada através da cromatografia de troca catiônica. Ensaios de cristalização foram realizados onde microcristais e cristais em forma de agulha foram obtidos. Futuramente, estudos estruturais e funcionais destas enzimas poderão trazer informações sobre a patogenicidade da X fastidiosa, assim como possibilitar o desenvolvimento de inibidores específicos / The bacterium Xylella fastidiosa has been associated with diseases of economically importante crop as the citrus variegated chiorosis (CVC) in citrus in Brazil and the Pierce\'s disease in grapevines in the United States (Hopkins. 1989 and Purcell et al., 1997). Tvpical disease svmptoms include conspicuous variegations. with chlorotic amas on the upper side and small necrotic lessions on the lower side of the leaves. The affected fruits are smaller. often no more than one-third of the diameter of healthy fruits, hardened and without commercial value. X fastidiosa has a nine-gene operou (B, C. D. E. F, H. J. K. and M genes) responsible for the biosvnthesis of exopolysaccharides, denoted fastidian gum, that mas be linked directly to the pathogenecin - of the microorganism. In this work our main aim is to study the GLIM(\' enzyme, that may be responsible for gum polymerization or secretion through the membrane of X fástidiosa and GUMK enzyme, a glucuronosvltransferase or glycosyltransferase IV. that adds a glucuronic acid to tetrasaccharide manose-α-1,3- glucose-β-1,4-glucose-P-P-po1visoprenyl. The gume gene was cloned into the pMAL-c2x expression vector, and GIIMC-MBP fusion protein was expressed with estimate molecular mass 98kDa. was purified through affinin - column and was characterized through the reaction of antiMBP antibodv by imunoblotting. Molecular aggregation studies of GII.MC-MBP fusion protein were measured by Dynamic Light-Scattering (DLS). Crystallization screens were made and microcrvstals were obtained. The GUMC-MBP fusion protein were cleavaged with Factor Xa and the GIM\' enzyme purified through ion exchange chromatography. The GUMC molecular mass were determined using size exclusion chromatography, superosel2. Anti-GUMC-MBP antibodies were already produced in mice and were used to characterize the GUMC protein. by imunoblotting technique. In addition, according to circular dichroism spectroscopy, GUMC protein prevalent secondary structure is α-hefix. Nine constructions for the gtmiC gene. without 5 - and 3° transmembrane regions (one for cloning in pET29b and 8 for cloning in pMALc-2x) were carried out. The modified fusion protein GLIMC-MBP 5C were purified through affmin - resin and cleavaged with the Factor Xa enzvme. In the future. aggregation states of GIIMC-MBP and modifiecl GUMC -MBP C5 fusion proteins. and GII.MC and modificai GUMC C5 enzymes could be measured by DLS and compared. The gumK gene. cloned into the pMALc-2x expression. vector. expressed a GIIMK-MBP fusion protein soluble with molecular mass 86kDa. This fusion protein was purified through affmin - resin. The GIIMK-MBP fusion protein was characterized for imunoblotting and CD measurements. The GUMK-MBP fusion protein was cleavaged with Factor Xa enzvme and the GUMK enzyme was purified through cationic Exchange chromatography. Crvstallization screens were made and micmcrystals and crystals in needle form were obtained. Future enzvmes characterization three-dimensional structure will allow - further structural and functional studies on the pathogenicity of X. fastidiosa. as well as to development of specific inhibitors

Estudos bioanalíticos envolvendo a Xylella fastidiosa / Bioanalytical studies involving Xylella fastidiosa

Este trabalho apresenta estudos bioanalíticos envolvendo a bactéria Xylella fastidiosa. A X. fastidiosa é uma bactéria aeróbica, responsável pela doença Clorose Variegada dos Citros. Durante o projeto genoma foi possível mapear diversas proteínas, sendo uma das enzimas a hidroxinitrila liase (HNL). As indústrias de química fina, em especial a farmacêutica, vêm utilizando enzimas para produção de enantiômeros que visam à formação de novas drogas quirais com alto teor de pureza. As enzimas HNLs são proteínas presentes na superfamília α/β-hidrolases, da qual também fazem parte as lipases e esterases. As hidroxinitrilas são empregadas na síntese de compostos quirais, para melhores condições em biocatálise. HNLs são enzimas que catalisam a formação reversível de cianoidrinas utilizando ácido cianídrico (HCN) e aldeídos ou cetonas. Por esta razão, foi analisado o potencial de biocatálise enantiosseletiva da XfHNL referente ao substrato (R,S)-ibuprofeno e a síntese do éster racêmico, α-metil benzil acetato, e também a síntese de cianoidrinas catalisadas pela XfHNL. Pelas três reações estudadas neste trabalho foi possível observar que XfHNL não possui enantiosseletividade em dois dos substratos testados. Também foi estudado neste trabalho a expressão proteica nos meio de cultura líquidos XDM2, XDM4 e XDM5 até então não estudados por eletroforese OFFGEL, uma nova plataforma semi-preparativa para fracionamento de proteínas. Foi realizado um estudo preliminar desse meios, para avaliar a expressão proteica, e também o meio de cultura BCYE para visualizar possíveis fatores sinal difusível (DSF) por espectrometria de massas e seu comportamento em eletroforese capilar. Por fim, foi fabricado um microdispositivo de microfluídico feito em poliéster-toner (PT) para biomimetizar o xilema para o estudo in vitro de X. fastitiosa e seu comportamento na colonização. Assim sendo, foi possível visualizar o crescimento, a formação de biofilme e a presença de goma xantana dentro do microchip. / This work involves bioanalytical studies of Xylella fastidiosa. The X. fastidiosa is an aerobic bacteria, responsible for the disease Citrus Variegated Chlorosis. During the genome project was possible to characterize several proteins, one of the enzymes hydroxynitrila lyase (HNL). Industries, especially pharmaceuticals, have been using enzymes for production of enantiomers aimed at the formation of new chiral drugs with high purity. Enzymes are proteins present in HNLs superfamily α / β-hydrolases, which are also part of lipases and esterases. The HNLs have been employed in the synthesis of chiral compounds for better conditions in biocatalysis. HNLs are enzymes that catalyze the reversible formation of cyanohydrins using hydrocyanic acid (HCN) and aldehydes or ketones. For this reason, we investigated the potential of enantioselective biocatalysis of XfHNL respect to substrate (R,S)-ibuprofen and synthesis of racemic ester, α-methyl benzyl acetate. Also, the synthesis of cyanohydrins catalyzed by XfHNL. By three reactions involved in this work, it was observed that there were not XfHNL enantioselectivity in 2 of the substrates studied. Also, it was studied protein expression in liquid culture medium XDM2, XDM4 and XDM5 hitherto studied by electrophoresis OFFGEL, a new platform for semi-preparative protein fractionation. We conducted a quick study and through this liquid medium, BCYE, to view possible DSFs by mass spectrometry and their behavior in capillary electrophoresis. And finally, it was produced a PT microdevice in order to mimic the xylem vessels to study of X. fastitiosa in vivo and its behavior. Accordingly, it is possible to display the growth, biofilm formation and the presence of xanthan gum in the microchip.

A Clorose Variegada de Citrus (CVC) é uma doença que vem atingindo grandes plantações de laranjas no Brasil e outros países como Estados Unidos, França e Espanha. O principal efeito da doença é o surgimento de manchas amarelas nas folhas que progridem para toda a extensão levando-a à necrose. Os frutos são pequenos, apresentam casca dura, sendo impróprios para o consumo. O agente causador da CVC é a bactéria Xylella fastidiosa que é limitada ao xilema. A bactéria é transmitida por insetos conhecidos como \"cigarrinhas\" que se alimentam na seiva do xilema. Esta bactéria apresenta em seu genoma um Operon contendo 9 genes (B,C,D,E,F,H,J,K,M) responsáveis pela biossíntese do exopolissacarídeo denominado goma fastidiana, que pode estar envolvido na sua patogenicidade. No presente trabalho, realizou-se estudos com as enzimas GumD (uma enzima glicosiltransferase I que faz a primeira adição de glicose-l-fosfato ao lipídeo prenol) e GumC (que provavelmente está envolvida na etapa de polimerização e/ou secreção do polissacarídeo formado através da membrana da bactéria), com o objetivo de contribuir para melhor entendimento da via biossintética. O gene gumD, que codifica a enzima GumD, foi clonado nos vetores de expressão pMAL¬c2x e pKK223-3. A proteína GumD foi purificada através das cromatografias de troca aniônica e filtração a gel. O peso molecular e o pI da enzima foram determinados usando respectivamente a técnica de filtração a gel e o sistema Fast de eletroforese. A enzima foi caracterizada quanto ao seu enovelamento através da técnica de Dicroísmo Circular, apresentando um espectro característico de estrutura secundária enovelada, composta predominantemente por a-hélices. O gene gumC, que codifica para a proteína GumC, foi clonado nos vetores de expressão pMAL-c2x (no qual a enzima é expressa em fusão com a proteína MBP - Maltose Binding Protein) e pET29a (a proteína é expressa sem fusão). A proteína em fusão GumC-MBP foi parcialmente pura através da coluna de amilose e foi caracterizada através da técnica de Imunoblotting. A enzima GumC expressa no vetor pET29a está em fase de purificação. Anticorpos anti-GumC-MBP foram produzidos em camundongos e serão utilizados como uma forma de caracterizar a proteína GumC expressa sem fusão. Estudos estruturais destas enzimas poderão trazer informações fundamentais para o conhecimento da via biossintética, assim como para o desenvolvimento de inibidores específicos / The Citrus Variegated Chlorosis (CVC) is a serious disease of orange trees in countries like Brazil, USA, France and Spain. Typical disease symptoms include conspicuous variegations with chlorotic areas on the upper side and small necrotic lesions on the lower side of the leaves. The affected fruits are smaller, hardened and without commercial value. Citrus variegated chlorosis is caused by Xylella fastidiosa, which is a xylem-limited bacterium. X fastidiosa is transmitted by specific sharpshooter leafhoppers when the insect feeds on the xylem sap. X fastidiosa has a nine-gene operon (B, C, D, E, F, H, J, K, and M genes) responsible for the biosynthesis of exopolysaccharides, denoted fastidian gum, which can be involved in its pathogenicity. GumD glycosyltransferase enzyme adds the first sugar, glycose-l-phosphate, to the prenol lipid. GumC enzyme probably is involved in the polymerization and/or exportation of the fastidian gum through the membrane of the bacterium. Studies were done on the GumD and GumC enzymes with the aim of getting a better understanding of the exopolysaccharide biosynthetic pathway. The gumD gene was cloned into the pMAL-c2x and pKK223-3 expression vectors, and GumD protein was purified through ion exchange and size exclusion chromatography. Then GumD molecular mass and pl were determined using, respectively, size exclusion chromatography and electrophoresis. In addition, according to circular dichroism spectroscopy, GumD prevalent secondary structure is u-helix. On the other hand, the gumC gene was cloned in two expression vectors: pMAL-c2x (the protein is expressed in fusion with Maltose Binding Protein ¬MBP) and pET29a (the protein is expressed without fusion in our strategy). GumC¬MBP, the protein in fusion, was partially purified using an amylose column and the fusion between GumC and MBP was confirmed with imunoblotting technique. Purification trials of GumC enzyme expressed in pET29a are in course. Anti-GumC¬MBP antibodies were already produced in mice and they will be used to characterize the GumC protein expression without fusion. Structural studies of GumD and GumC enzymes will provide information about the fastidian gum biosynthetic pathway, as well as to the development of specific inhibitors

Este trabalho apresenta estudos bioanalíticos envolvendo a bactéria Xylella fastidiosa. A X. fastidiosa é uma bactéria aeróbica, responsável pela doença Clorose Variegada dos Citros. Durante o projeto genoma foi possível mapear diversas proteínas, sendo uma das enzimas a hidroxinitrila liase (HNL). As indústrias de química fina, em especial a farmacêutica, vêm utilizando enzimas para produção de enantiômeros que visam à formação de novas drogas quirais com alto teor de pureza. As enzimas HNLs são proteínas presentes na superfamília α/β-hidrolases, da qual também fazem parte as lipases e esterases. As hidroxinitrilas são empregadas na síntese de compostos quirais, para melhores condições em biocatálise. HNLs são enzimas que catalisam a formação reversível de cianoidrinas utilizando ácido cianídrico (HCN) e aldeídos ou cetonas. Por esta razão, foi analisado o potencial de biocatálise enantiosseletiva da XfHNL referente ao substrato (R,S)-ibuprofeno e a síntese do éster racêmico, α-metil benzil acetato, e também a síntese de cianoidrinas catalisadas pela XfHNL. Pelas três reações estudadas neste trabalho foi possível observar que XfHNL não possui enantiosseletividade em dois dos substratos testados. Também foi estudado neste trabalho a expressão proteica nos meio de cultura líquidos XDM2, XDM4 e XDM5 até então não estudados por eletroforese OFFGEL, uma nova plataforma semi-preparativa para fracionamento de proteínas. Foi realizado um estudo preliminar desse meios, para avaliar a expressão proteica, e também o meio de cultura BCYE para visualizar possíveis fatores sinal difusível (DSF) por espectrometria de massas e seu comportamento em eletroforese capilar. Por fim, foi fabricado um microdispositivo de microfluídico feito em poliéster-toner (PT) para biomimetizar o xilema para o estudo in vitro de X. fastitiosa e seu comportamento na colonização. Assim sendo, foi possível visualizar o crescimento, a formação de biofilme e a presença de goma xantana dentro do microchip. / This work involves bioanalytical studies of Xylella fastidiosa. The X. fastidiosa is an aerobic bacteria, responsible for the disease Citrus Variegated Chlorosis. During the genome project was possible to characterize several proteins, one of the enzymes hydroxynitrila lyase (HNL). Industries, especially pharmaceuticals, have been using enzymes for production of enantiomers aimed at the formation of new chiral drugs with high purity. Enzymes are proteins present in HNLs superfamily α / β-hydrolases, which are also part of lipases and esterases. The HNLs have been employed in the synthesis of chiral compounds for better conditions in biocatalysis. HNLs are enzymes that catalyze the reversible formation of cyanohydrins using hydrocyanic acid (HCN) and aldehydes or ketones. For this reason, we investigated the potential of enantioselective biocatalysis of XfHNL respect to substrate (R,S)-ibuprofen and synthesis of racemic ester, α-methyl benzyl acetate. Also, the synthesis of cyanohydrins catalyzed by XfHNL. By three reactions involved in this work, it was observed that there were not XfHNL enantioselectivity in 2 of the substrates studied. Also, it was studied protein expression in liquid culture medium XDM2, XDM4 and XDM5 hitherto studied by electrophoresis OFFGEL, a new platform for semi-preparative protein fractionation. We conducted a quick study and through this liquid medium, BCYE, to view possible DSFs by mass spectrometry and their behavior in capillary electrophoresis. And finally, it was produced a PT microdevice in order to mimic the xylem vessels to study of X. fastitiosa in vivo and its behavior. Accordingly, it is possible to display the growth, biofilm formation and the presence of xanthan gum in the microchip.

Differentiation and Analysis of Xylella fastidiosa Subspecies fastidiosa Cultures Isolated from a Single Texas Vineyard using Simple Sequence Repeat Markers

Xylella fastidiosa subspecies fastidiosa is the causative agent of Pierce’s disease of grape and has caused significant crop stress and loss in vineyards throughout Texas. While multiple techniques are available to identify subspecies of X. fastidiosa, only simple sequence repeat markers can be used for the differentiation of isolates within individual subspecies. In this research, SSR markers were utilized to demonstrate the diversity of subsp. fastidiosa isolates from within a single vineyard. The distributions of strains defined within subsp. fastidiosa were also compared to epidemiological data to clarify any relationships. Initial results from isolation attempts indicate disease severity to have the largest impact on the success of isolation attempts with 7% of samples rated as ‘Healthy’ and 83% of samples rated as ‘Advanced’ producing successful isolations. A conventional PCR protocol employing 5 SSR markers was used to generate banding profiles for 97 isolates collected from 7 grape varieties planted in 5 blocks throughout a single Texas vineyard. SPSS statistical program was used to execute a hierarchical cluster analysis to produce a dendrogram which grouped isolates into 3 strain groups with 7% or 15% dissimilarity. Of the 3 epidemiological factors analyzed, the distribution of strains showed significant dependence on grape variety while having no dependence on disease severity or location within the vineyard.

The development of new tools for field and laboratory diagnosis of Pierces Disease

Pierce’s Disease (PD), caused by Xylella fastidiosa, is a devastating bacterial disease of grapevines. One of the few control options is roguing. Roguing depends on precise diagnosis of PD in vines. These experiments were conducted to improve available diagnostic protocols and enhance levels of disease control. Plots were selected from four different Texas vineyards with a total of four different varieties (Blanc duBois, Cabernet Sauvignon, Chardonnay, and Merlot). An infrared thermometer was used to take temperature measurements of the vines. Samples were taken of each of these vines at the same time and were tested for X. fastidiosa by culturing, Enzyme-Linked ImmunoSorbent Assay (ELISA), and Quantitative Real-Time Polymerase Chain Reaction (QRT-PCR). ELISA found an increase in plant temperature in samples that tested positive for X. fastidiosa, but QRT-PCR did not. An infrared thermometer could be used to detect asymptomatic vines, but there are several variables to consider such as grape variety and vineyard location. Grape varieties differed significantly in mean temperatures, as did vineyard locations. PD does not seem to have a pattern in which it spreads, although this could be because of the high level of disease incidence in the chosen vineyards. Both the ELISA and QRT-PCR tests have their own pros and cons for X. fastidiosa detection. ELISA takes approximately 6 hours and can be inaccurate in detecting X. fastidiosa. QRT-PCR takes 2-3 hours and is a much more sensitive test. A combination of techniques (PrepMan Ultra® and nucleic acid precipitation) can be used to clean QRTPCR samples when they have degraded and are being affected by inhibitors.

Investigations on the diagnosis, colonization, and epidemiology of grapevines with Pierce's disease

Pierce’s disease (PD) of grapevines, caused by Xylella fastidiosa, is devastating Texas vineyards. Two rapid diagnostic techniques, real-time polymerase chain reaction (PCR) and enzyme-linked immunosorbent assay (ELISA), were compared on the basis of cost, reliability, and their ability to quantify X. fastidiosa in diseased tissues. A high correlation was found between the two techniques for measuring bacterial titer in vitro. A similar relationship was not detected when applying the methods to diseased tissue. There was a 75% similarity between the techniques when used to diagnose PD in artificially infected grapevines. Where the two methods differed, real-time PCR was more successful in identifying plants known to be infected with the bacterium. In uninoculated grapevines, the two techniques were similar, where the positive rates were 7% and 4% for ELISA and real-time PCR respectively. In a second study, 3 grape cultivars, ‘Cynthiana’, ‘Cabernet Sauvignon’, and ‘Chardonnay’, were inoculated with 2 isolates of X. fastidiosa to measure disease development and colonization by the pathogen. The bacteria colonized similar distances from the inoculation point over a 25 week period in all three cultivars. Real-time PCR and ELISA absorbance values suggest that the concentrations of bacteria ranged between 104 and 106 cells/ml in a 1.27 cm section of grapevine cane. Concentrations of bacteria didn’t vary based on distance from the inoculation point. Marginal leaf-scorch symptoms were seen on ‘Cabernet Sauvignon’ and ‘Chardonnay’ grapevines 9 weeks post-inoculation. Leaf-scorch symptoms were not observed on ‘Cynthiana’. The vigor of all inoculated grapevines was reduced compared to negative control grapevines the season after initial infection. In a third study, a Texas vineyard planted in Viognier grapevines was surveyed for PD symptoms on 3 separate dates. In October 2003, 45/50 rows had significant aggregation of symptomatic grapevines according to Ordinary Runs Analysis. Aggregation of symptomatic grapevines was found down the row more often than across the row. The rapid rate of disease progress and mortality rate of vines in this vineyard suggest that vine-to-vine spread is occurring and that Viognier vines are highly susceptibly to PD.

Determining the threat of Pierce's disease to Virginia vineyards

Pierce's disease (PD) is a vascular disease of grapevines caused by <i>Xylella fastidiosa</i> (Wells et al.) (<i>Xf</i>) which is transmitted by xylophagous insect vectors. PD infection in Virginia vineyards was thought to be isolated to southeastern portions of the state as there have been no reports of vine loss in western Virginia and cold winter temperatures experienced there limit the effects of the bacterium from year to year. Upward trends in winter temperatures have raised PD concern in the mid-Atlantic. My risk assessment study found PD symptomatic vines beyond the modeled boundary for infection, confirmed <i>Xf</i>-positive with DAS-ELISA. Yellow sticky traps were used to survey Virginia vineyards throughout the 2006 and 2007 growing seasons to identify sharpshooter (Cicadellinae) species in six growing regions. <i>Graphocephala versuta </i>(Say) and <i>Oncometopia orbona</i> (Fabricius) (Hemiptera: Cicadellidae) were trapped in the greatest abundance and were both present in every region surveyed.
