Molekulární fylogeneze a genetická diverzita nejbližších příbuzných rodu Pisum

During the process of domestication and selection reduction of genetic diversity of cultivated crops occurred. Currently the main interest of breeders is to transfer some of crop wild relatives’ genes to cultivated crops. Those genes of interest are related e.g. with desease and pest resistance or drought resistance. Hybridization of wild and cultivated species is prevented by reproductive isolation. The frequent phonomenon in hybrids is called nuclear-cytoplasmic incompatibility, which is manifested by reduced fertility, sterility or lethality. In this phenomenon occurs conflict between nuclear-encoded genes with genes encoded in the organellar genomes. The identified accD candidate gene responsible for nuclear-cytoplasmic incompatibility in pea is highly variable due to insertions and deletions. High variability of the accD gene was also confirmed in the genera Lathyrus and Vicia. Variability was observed in gene sequence lenghts caused by presence of indels and single nucleotide polymorphisms. In comparative analysis with other regions of cpDNA commonly used in phylogenetics, the region of the accD gene had the highest value of parsimonially informative sites. The phylogeny derived from the region of the accD gene corresponds to the phylogeny based on combined chloroplast markers, therefore the accD gene may be suitable for this type of analysis.

Effet de la salinité pendant la croissance de plants de riz sur la biosynthèse de la 2-acétyl-1-pyrroline (2AP), composé aromatique clé des variétés de riz aromatique,le rôle de la 1-pyrroline dans la biosynthèse de la 2AP ainsi que l'origine du groupe acétyle ont été étudiés. La variété aromatique de Camargue (France), Aychade, a été cultivée soit dans un milieu liquide soit dans des pots en serre. Dans le milieu de culture liquide, une augmentation significative de la teneur en 2AP dans les feuilles a été observée quelle que soit la concentration en NaCl (de 1,7 à 170 mM). Une corrélation positive a été mise en évidence entre la teneur en sel et en 2AP. A de plus fortes concentrations en NaCl, il a également pu être montré qu'il y avait une relation entre la concentration en proline, un précurseur aminé majeur de la 2AP, et la teneur en 2AP. Inversement, les changements en acide -aminobutyrique (GABA), un des métabolites compétitif de la biosynthèse de la 2AP, n'ont pas été corrélés avec ceux de la 2AP. Dans les expérimentations menées en serre, la variété Aychade a été cultivée dans une solution saline à 30 mM, correspondant à une conductivité électrique de 3800 ± 400 S.cm-1, valeur considérée comme suffisante pour induire un stress salin sur le plant de riz. Cette solution saline a été appliquée à des dates et pour des durées différentes en fonction du cycle de développement de la plante. A la phase végétative, une relation positive a été observée dans les feuilles entre le traitement salin et la teneur en 2AP ainsi qu'en proline. Il est intéressant de noter que tous les grains provenant des plants traités avec la solution saline contenaient des teneurs nettement plus élevées en 2AP par rapport au contrôle. La synthèse de la 2AP la plus importante s'est produite lorsque les plants ont été soumis à un traitement salin pendant l'ensemble des phases végétative et reproductive. Cependant, dans le dernier cas, le rendement en grain était beaucoup plus faible. En supplémentant des cals de riz avec de la 1-pyrroline, il a été démontré que cette imine cyclique était un facteur limitant pour la biosynthèse de la 2AP non seulement dans la variété aromatique, mais également dans la non aromatique. Les essais conduits avec le glucose et les acides gras marqués 13C ont montré que ces molécules pourraient être précurseurs du groupe acétyle de la molécule de 2AP. / The effect of salinity during rice plant growth on the biosynthesis of 2-acetyl-1-pyrroline (2AP), characteristic flavour compound of fragrant rice cultivars was studied. Additionally role of 1-pyrroline in 2AP biosynthesis together with source of acetyl group were investigated. Aychade, a fragrant rice was cultivated either in a liquid medium adapted for seedlings or in pots in a greenhouse. A significant increase in 2AP levels in the leaves was observed whatever the concentration of NaCl (1.7 to 170 mM NaCl) in the culture medium. A positive correlation was found between the level of salt and that of 2AP. There was a relationship between the concentration of proline, a major amino precursor of 2AP and 2AP level at highest concentrations of NaCl. Conversely the changes in -aminobutyric acid (GABA), a competitive metabolite in 2AP biosynthesis were not correlated with those of 2AP. In greenhouse experiments, Aychade cultivar was grown under one level of salt solution, 30 mM that corresponds to EC of 3800 ±400 S.cm-1, value considered sufficient to induce salt stress in rice. Timing and duration of application of salt solution varied according to the growth stages. At vegetative phase a positive relation was observed between salt treatment and 2AP as well as proline levels in the leaves. Interestingly grains from all salt treated plants contained significantly higher levels of 2AP than those from control. The highest synthesis occurred when plants were subjected to salt treatment during whole vegetative and reproductive phases. However in the latter case crop yield decreased significantly. Through supplementation of rice callus with 1-pyrroline it was demonstrated that this cyclic imine was limiting factor for the 2AP biosynthesis not only in a fragrant variety but also in a non fragrant variety. Experiments with 13C labelled glucose and fatty acids showed that glucose and fatty acids could be precursors of acetyl group of 2AP.

O objetivo desta pesquisa foi o desenvolvimento, avaliação da estabilidade e da eficácia de formulações cosméticas contendo acetil hexapeptídeo-3 (AHP) ou acetil tetrapeptídeo-5 (ATP) ou mistura de di e tripeptídeos (DTP). Para tal, foram desenvolvidas 6 formulações à base hidroxietilcelulose (HEC), metilfenil polissiloxano (MFP) e álcool batílico e lecitina de soja (ABL) (F1), ou de HEC, MFP, ésteres de polietilenoglicol com cadeia alquílica com 12 a 20 carbonos (EP) (F2), ou de polímero de acrilatos (PA), MFP e ABL (F3) ou de PA, MFP e EP (F4), ou de HEC e MFP (F5) ou de PA e MFP (F6), contendo ou não (veículo) 10% de AHP, ou 10% de ATP ou 4% de DTP, as quais foram submetidas a um estudo de estabilidade física por determinação da reologia e a uma avaliação sensorial. Para o estudo de eficácia dos peptídeos objeto de estudo as formulações foram aplicadas nos antebraços, na face, na região dos olhos e da boca de 60 voluntárias, sendo realizadas medidas do conteúdo aquoso do estrato córneo, da perda de água transepidérmica (TEWL), do micro-relevo cutâneo e das propriedades mecânicas da pele (anisotropia), antes (basal), após 2 horas da aplicação única (efeitos imediatos) e após 15 e 30 dias de uso contínuo (efeitos a longo prazo) das formulações. Os dados foram analisados estatisticamente pelo teste paramétrico análise de variância. Nos testes de estabilidade física, as formulações de nos 1, 2 e 5 foram consideradas as mais estáveis e as formulações de nº 5 e 6 foram as que apresentaram melhor sensorial, sendo, portanto, selecionada a formulação de nº 5 como veículo para os estudos de eficácia. Na avaliação dos efeitos imediatos na pele, observou-se que as formulações estudadas proporcionaram um aumento significativo no conteúdo aquoso do estrato córneo dos antebraços, enquanto que, somente as formulações acrescidas dos peptídeos provocaram um aumento significativo neste conteúdo na face. Em relação ao micro-relevo cutâneo, as formulações contendo ou não os peptídeos provocaram uma redução do parâmetro Ser (aspereza da pele) e Rt (rugosidade) nos antebraços e a formulação contendo DTP provocou um efeito imediato significativo no parâmetro Sesm (textura da pele). Na avaliação dos efeitos a longo prazo, as formulações ocasionaram uma diminuição significativa na TEWL e as acrescidas dos peptídeos objetos de estudo proporcionaram um aumento significativo no conteúdo aquoso do estrato córneo na região dos olhos, enquanto que nos antebraços e na região da boca todas as formulações acrescidas ou não dos peptídeos ocasionaram um aumento nesse parâmetro. Em relação ao micro-relevo cutâneo, a aplicação das formulações contendo os peptídeos, na região da boca, provocou um aumento significativo nos valores de Sesm. A aplicação das formulações contendo os peptídeos objeto de estudo, nos antebraços ocasionou uma diminuição significativa da anisotropia da pele. A formulação nº 5 contendo os peptídeos objeto de estudo, apresentou efeitos imediatos na hidratação, aspereza, rugosidade e textura da pele e, efeitos a longo prazo, na hidratação, textura e anisotropia da pele, mostrando a importância do emprego destes peptídeos em produtos cosméticos com finalidades hidratantes e antienvelhecimento. / The aim of this study was the development, evaluation of the stability and the efficacy of cosmetic formulations containing acetyl hexapeptide-3 (AHP), or acetyl tetrapeptide-5 (ATP) or mixture of di- and peptides (DTP). For this purpose the 6 following formulations (F) were developed: F1- containing hydroxyethyl cellulose (HEC), methylphenyl polysiloxane (MPPS), batyl alcohol, and lecithin (BAL); F2- HEC, MPPS and C12-C20 PEG-8 alkyl ester (AE); F3- acrylate copolymer (AC), MPPS and BAL; F4- AC, MPPS and AE; F5- HEC and MPPS and F6- AC and MPPS, which were supplemented or not (vehicle) with 10% of AHP or 10% of ATP or 4% of DTP and submitted to the evaluation of physical stability by the determination of the rheologic behavior and also to the sensorial analysis. For clinical efficacy studies, formulations were applied to the volar forearm and to the face skin (eye corner and cheek region), of 60 female volunteers. Skin conditions in terms of skin moisture, transepidermal water loss (TEWL), skin mechanical properties (anisotropy) and skin microrelief, before and after 2 hours (immediate effects) and after 15 and 30-day period of daily application (long term effects). Results obtained were statistically analyzed using the ANOVA parametric test. In the rheological study, the formulations number 1, 2, and 5 were considered more stable than the others. Moreover, the formulations number 5 and 6 showed the best sensorial attributes, and for this reason the formulation number 5 was selected for the efficacy study. The evaluation of immediate effects of formulations showed that the application of the formulations studied in the volar forearm increased the water content of the stratum corneum, while only the formulations containing the peptides provoked an increased in this content, when face skin was analyzed. When the volar forearm skin microrelief was analyzed, the application of all the formulations studied provoked a significant decrease in skin roughness (Ser) and depth of roughness (Rt), furthermore only the formulation supplemented with the DTP showed a significant increase in the skin smoothness (Sesm). In the evaluation of long term effects, all formulations provoked a significant reduction in TEWL values and the formulations containing the peptides studied provoked a significant increase in the water content of the stratum corneum of eye corner; when the volar forearms and the cheek region were analyzed, all the formulations containing or not the peptides studied, provoked an increase in this parameter. When the skin microrelief was analyzed, only the formulation supplemented with AHP or ATP or DTP, provoked a significant increase in the skin smoothness (Sesm), when applied in the cheek region. The application of formulations containing the peptides studied in the volar forearms provoked a significant decrease in the skin anisotropy. The formulation nº 5 (F5) containing the peptides studied, showed immediate effects on skin hydration, roughness and smoothness, and also long-term effects on skin hydration, smoothness and mechanical properties, which demonstrated the importance of the use of these peptides in cosmetic formulations with hydration and anti-aging purposes.

Biochemical and structural characterization of the ATP-dependent maturation factor of acetyl-CoA synthase

Acetyl-CoA Synthase (ACS) katalysiert die Reaktion eines Methylkations, Kohlenstoffmonoxid und CoA zu Acetyl-CoA. Das aktive Zentrum von ACS ist ein Ni,Ni-[4Fe4S]-Cluster (A-cluster), in dem zwei Nickel-Ionen mit einem kubanen [4Fe4S]-Cluster verbrückt sind. An der Biosynthese von komplexen Metallclustern sind in der Regel mehrere akzessorische Proteine, auch Maturationsfaktoren genannt, beteiligt. Die Biosynthese des A-Clusters wurde bisher noch nicht genauer untersucht und es war nicht bekannt welche Proteine die Biosynthese des A-Clusters katalysieren. In dieser Arbeit wurde das Protein AcsF als Maturationsfaktor der ACS identifiziert und seine biochemischen und strukturellen Eigenschaften wurden charakterisiert. AcsF und apoACS aus Carboxydothermus hydrogenoformans bilden einen stabilen Komplex, der zwei Nickel-Ionen binden kann. ApoACS hingegen kann unter den gleichen Bedingungen im Durchschnitt nur weniger als ein Nickel-Ion binden. Der Ni-ACS-AcsF Komplex, an dem zwei Nickel-Ionen gebunden sind, ist katalytisch jedoch nicht aktiv. Erst durch Zugabe von Mg-ATP kann die inaktive Spezies in eine aktive Form überführt werden. AcsF-Proteine gehören zur gleichen Protein-Familie wie CooC-Proteine, die Maturationsfaktoren der Kohlenstoffmonoxid Dehydrogenase. Ein Sequenzähnlichkeitsnetzwerk konnte zeigen, dass AcsF- und CooC-Proteine jeweils eine eigene Untergruppe in dieser Familie bilden. Die AcsF-Proteine von C. hydrogenoformans und Archaeoglobus fulgidus wurden kristallisiert und deren Kristallstrukturen gelöst. Durch einen Vergleich der Strukturen von AcsF mit den Strukturen von zwei CooC-Proteinen konnte aufgedeckt werden, dass die größten strukturellen Unterschiede zwischen AcsF- und CooC-Proteinen zwischem dem Switch I Motif und dem CXC Motif zu finden sind. / Acetyl-CoA synthase (ACS) catalyzes the reaction of a methyl cation, carbon monoxide and CoA to acetyl-CoA. The active site of ACS is a Ni,Ni-[4Fe4S] cluster (A-cluster), in which two nickel ions are bridged to a cubane-type [4Fe4S] cluster. Usually, several accessory proteins are involved in the biosynthesis of such complex metal clusters. However, the biosynthesis of the A-cluster had not yet been investigated and it was not known which accessory proteins take part in its assembly. In this work, the protein AcsF was identified as a maturation factor of ACS, and its biochemical and structural properties were characterized. AcsF and apoACS from Carboxydothermus hydrogenoformans form a stabile complex, that can bind two nickel ions. ApoACS alone, on the other hand, binds on average only less than one nickel ion under the same conditions. The Ni-ACS-AcsF complex, that contains two nickel ions, is not active, but the addition of Mg-ATP converts the inactive species into an active form. AcsF proteins belong to the same protein family as CooC proteins, the maturation factors of carbon monoxide dehydrogenase. A sequence similarity network showed that AcsF and CooC proteins each form their own subgroup within this family. The AcsF proteins from C. hydrogenoformans and Archaeobglobus fulgidus were crystallized and their crystal structures were solved. A comparison of the crystal structures of AcsF proteins with the structures of two CooC proteins revealed that the main structural differences between AcsF and CooC proteins can be found between the switch I motif and the CXC motif.
