Global concrete production is increasing, which brings some environmental burden. A certain solution is the use of secondary raw materials, which has been a long-standing practice in the construction industry. One of the most widespread secondary raw materials is power fly ash. However, in recent years trends have been directed towards maximizing their utility properties. The aim of this work was to study the possibilities of activation of power plant fly ash as an active admixture in concrete. The thesis is divided into two parts, the theoretical and the practical part. The theoretical part describes in detail the possibilities of power plant fly ash activation for use as an active admixture in concrete. The practical part of the thesis is focused on mechanical activation, where the knowledge of the theoretical part is verified on various fly ash.

This master´s thesis deals with the suppression of the growing of algae in cooling towers. Subject of the work is suggestion and testing surface modification of the existing mature concrete in order to suppress the growth of algae. In the frame surface modification, several commercially available and newly developed systems were tested. Testing systems are based on the basis of portland cement, geopolymers or formation of insoluble complex compounds containing metal elements (Zn, Cu). Experimental methods were applied in the real environment of cooling towers of Dukovany Nuclear power plant.

Ground granulated blast furnace slag is a by-product of the steel industry and is often used in combination with ordinary Portland cement as a binder in concrete. When concrete is exposed to high temperatures, physical and chemical transformations lead to significant loss of mechanical properties. This study aims to investigate the effect of high temperatures and rehydration on the mechanical properties, microstructure and phase composition of alkali activated slag. The results of the research could make an important contribution to decisions made concerning the reconstruction of structures affected by fire. In suitable cases it would be possible to regenerate parts of a structure instead of totally rebuilding it.

It is generally believed that the presence of male traits is regulated by androgens. This thesis is designed to reveal the influence of testosterone on male traits in the leopard gecko, Eublepharis macularius. The noninvasive skin application mixture of vegetable oil and testosterone was used to manipulate the hormonal status of females and castrated males. This method has proven to be an effective alternative to the widely used hormonal implants. Manipulation was successfully validated, and there was apparent progressive increase of male sexual behavior in both groups. In females, the onset lasted much longer time, but both groups were able to perform all components of males sexual behaviour. Furthermore, I found that, contrary to widespread view, male sexual behavior persisted several weeks after reduction of testosterone levels in both manipulated groups. This PhD thesis reports the development of novel C-H activation strategies and aqueous-phase Suzuki-Miyaura cross-coupling reactions for the synthesis of modified deazapurine nucleobases. The methodologies of chemo- and regioselective synthesis of highly functionalized deazapurines have been developed by using modern C-H activation chemistry. Various functional groups such as amino-, imido-, silyl- and phosphonyl- were introduced by C-H activation reactions. Amino deazapurine derivatives were synthesized by developed Pd/Cu-catalyzed direct C-H amination and C-H chloroamination of 6-substituted 7-deazapurines with N-chloro-N- alkyl-arylsulfonamides. C-H imidation reactions of pyrrolopirimidines were performed under ferrocene catalysis with N-succinimido- or N-phtalimidoperesters. In order to obtain silylated derivatives, Ir-catalyzed C-H silylations of phenyldeazapurines with alkyl silanes were designed. Highly interesting deazapurine phosphonates were prepared by using Mn-promoted C-H phosphonation method and were further transformed into the corresponding phosphonic acids. Title: Comparison of Special Breathing Exercises versus Specialized Physiotherapeutic Interventions for Static Apnea Objectives: The aim of this diploma thesis is to look deeper into the issue of improving performance in static apnea, one of the competitive disciplines of freediving and compare its training with 1) special breathing exercises for divers (Mana, 2018) and 2) physiotherapeutic intevrentions based on working with the diaphragm and deep stabilization system of the spine (Kolář, 2009). The aim was to find out which group achieved a more effective result and what is the overall diference between the training result of both groups in terms of posture quality and performance in static apnea. Methods: The experiment involved 14 healthy people, men and women, aged 19-23 years old. The experiment consisted of three parts: 1) Initial measurement of static apnea and examination of four tests to determine the quality of one's postural muscles, 2) four-week lasting training according to the division into groups (breathing exercises for divers or training based on physiotherapy intervention), 3) final measurement of static apnea and re-examination of posture tests. This work is devoted to describe the possibilities of mechanical activations of admixtures used in concrete. Then, the verification options of grind fly ash, both from brown and black coal, from the classical as well as fluidized combustion and examination their impact on physical and mechanical properties of concrete, especially on long-term development to old age 180 days. Further attention is paid to assessing the impact of the use of mechanically activated fly ash on hydration heat evolution and the associated volume changes in the age of 1 to 90 days. In this work are presented the results of basic technological tests on concretes using admixtures, especially fly ash.

Title: Evaluation of the effect of exercise according to the Propriofoot concept and the methodology of Sensorimotor Training by Janda and Vávrová on foot pressure distribution Objectives: In this thesis the following three goals were set: First, to find out if and how does the percentual foot pressure distribution between the left and right foot in standing change after a four-week long exercise according to either the methodology of Sensorimotor training by Janda and Vávrová or the Propriofoot concept. Second, if and how will the segmental foot load change. Third, if the prospective changes of foot load will agree or differ between the two groups. Methods: A pilot study for quantitative research using experimental scientific method was conducted, with two groups consisting of ten members each, aged thirty to fifty and diagnosed with flat foot. At the beginning, all probands underwent diagnostic measurement using the "Plantograf V10" device. Then they attended a four-week long individual therapy with an exercise plan created according to one of the two methods in consideration. Analýza pluripotentního programu genové exprese v časných embryích a embryonálních kmenových buňkách / Analysis of pluripotent gene expression program in early embryos and embryonic stem cells

Pluripotence je schopnost buňky diferencovat do jakéhokoliv buněčného typu. Formuje se během časného embryonálního vývoje u savců a její vznik je spojen s reprogramací genové exprese na globální úrovni. Proces přirozeného vzniku pluripotence není stále zcela pochopen. Pro získání nového pohledu na události, které vedou ke vzniku pluripotence u savců, studovali jsme změny v genové expresi během oocyt-zygotického přechodu u myši. V tomto modelovém systému, oplodněné vajíčko podstoupí reprogramaci, která vede k vytvoření pluripotentních blastomer. Tyto blastomery zakládají samotné embryo. Cílem mé diplomové práce bylo analyzovat aktivaci transkripce během časného vývoje a vyvinout metodu pro monitorování exprese genů v oocytech, časných embryích a embryonálních kmenových buňkách. Metoda využívá kvantitativní PCR a umožnuje změřit expresi až 48 vybraných genů, které slouží jako markery pro maternální degradaci, aktivaci pluripotentního programu a diferenciaci do zárodečných linií. Dále ukazujeme, že náš systém monitoruje dynamiku transkriptomu během oocyt-zygotického přechodu, a získané výsledky jsou srovnatelné s daty naměřenými pomocí jiných metod. Díky našemu bioinformatickému přístupu jsme navíc identifikovali nové oocyt-specifické a zygotické nekódující RNA. Klíčová slova: pluripotence,...
