Úloha fosforylace proteinů v progamické fázi vývoje samčího gametofytu tabáku / The role of protein phosphorylation during progamic phase of tobacco male gametophyte development

Fíla, Jan January 2016 (has links)
v angličtině (English abstract) Tobacco male gametophyte has a strongly dehydrated cytoplasm and represents a metabolically inactive stage. Upon cytoplasm rehydration, pollen grain becomes metabolically active and after the activation is finished, the pollen tube growth through a selected pollen aperture starts. The rehydration together with metabolic activation are accompanied by the regulation of translation and post-translational modifications (mainly phosphorylation) of the existing proteins. In this Ph.D. thesis, there were identified phosphopeptides from tobacco (Nicotiana tabacum) mature pollen, pollen activated in vitro 5 min and pollen activated in vitro 30 min. The total proteins from the above male gametophyte stages were extracted. The protein extract was trypsinized and the acquired peptide mixture was enriched by MOAC (metal oxide/hydroxide affinity chromatography) with titanium dioxide matrix. The enriched fraction was subjected to liquid chromatography coupled with tandem mass spectrometry (LC- MS/MS). Totally, there were identified 471 phosphopeptides, carrying 432 exactly localized phosphorylation sites. The acquired peptide identifications were mapped to 301 phosphoproteins that were placed into 13 functional categories, dominant of which were transcription, protein synthesis,...

Vliv facilitačních technik na kognitivní funkce u roztroušené sklerózy mozkomíšní / The effect of facilitation physiotherapy on cognitive functions in multiple sclerosis patients

Kořínková, Markéta January 2012 (has links)
Cognitive function disorders have an enormous impact on the life quality of a patient, his or her family and close people. It influences the patient's job, social interaction and independence. This diploma thesis focuses on the possibility of improving cognitive functions concerning multiple sclerosis with the help of facilitation physiotherapy. Twelve disordered volunteers participated a two-month physiotherapeutic programme (motor programs activating therapy), for 1 hour, twice per week. Before the beginning and after the end of the therapeutic programme an examination by a clinic test PASAT and the monitoring of functional magnetic resonance was done. The results of functional magnetic resonance were further compared with the results of group of healthy volunteers. Concerning clinic test, the results of the disordered patients have improved after the therapy. In functional magnetic resonance statistics the earlier start and change of a curve progress of brain activation was discovered, which portrays improving speed of processing information at multiple sclerosis disordered.

Aktivace kolimačního systému lineárního urychlovače fotonovým zářením a její vliv na dávky obdržené personálem. / Activation of collimating system of linear accelerator through photon radiation and its impact on the doses received by the staff.

KUBÍKOVÁ, Adéla January 2017 (has links)
The diploma thesis on the topic "Activation of Collimating System of Linear Accelerator through Photon Radiation and its Impact on the Doses Received by the Staff" is divided into two parts, a theoretical and a practical part. The introductory theoretical part is devoted to the basic information about ionizing radiation. What are the types of ionizing radiation, its characteristics, properties and sources of ionizing radiation. Furthermore, there are mentioned direct and indirect interactions of ionizing radiation, a brief description of charged particle accelerators, collimator systems and radioisotope irradiation devices. Another part deals with radiation protection, basic legislation, methods and principles of radiation protection, personal dosimetry and medical supervision of radiation workers. Literary sources, available internet links and valid legal regulations were used for the work. In the practical part, a number of measurements were carried out to measure and to analyse the doses of secondary radiation of the linear accelerator collimator system for different photon beam energies, depending on time, distance from source, position of radiologic assistant around the table when working with the patient, as well as sizes of irradiated field. The aim of the work was to find out how large the doses of secondary radiation are, although measurable, but their values are not sufficient to be detected by personal dosimeters of radiological assistants. Confirm the hypothesis that the doses from the secondary radiation of the linear accelerator collimator system are so low that the dose power does not exceed 0.031 mGy/hr. The results obtained from various measurements are processed into transparent tables and graphically displayed. Based on the results of the measurements, the hypotheses were confirmed.

Mechanismy vývoje převodního systému srdečního u obratlovců / Mechanisms of conduction system development in vertebrates

Šaňková, Barbora January 2016 (has links)
Group of specialized cells that form cardiac conduction system is responsible for generation and coordinated propagation of the electrical impulse in the heart. Changes in its development can be connected with arrhythmias; therefore, a good level of knowledge is necessary and relevant for basic science and clinical practice. For correct development of the conduction system are important genes coding gap junctions proteins, ion channels, transcription factors and other molecules involved in signaling cascades (endothelin, neuregulin). Development of conduction system is determined in addition to genetic factors also by epigenetics and environmental factors. This thesis with its individual papers on which it is based is addressing different aspects of conduction system development, which appears to be a complex process. Another feature which is linking all papers together, is the methodological approach enabling us to study function of the conduction system - optical mapping. In the first publication we studied by the means of in vitro organ culture the impact of work load without interfering hemodynamics on the conduction system maturation in the chick embryonic heart. The phenotype observed during experiments was developmental regression of conduction system maturation together with changes in...

Binární alkalicky aktivované kompozitní materiály s cihelným prachem / Binary alkali-activated composite materials with brick powder

Mizerová, Cecílie January 2018 (has links)
The thesis is focused on the application of brick powder in production of binary alkali-activated binders with metakaolin, slag and fly ash. The theoretical part summarizes recent fields of use of the ceramic waste in construction and binder industry and prerequisite properties of the brick powder for alkaline activation. Brick powder based geopolymers made in the experimental work were tested for their mechanical properties, porosity and microstructure, flow properties of the fresh geopolymers were evaluated by rheometric measurements. In accordance with the results it can be concluded that brick powder could be a suitable precursor for blended binders with metakaolin and slag, these samples exhibited good mechanical properties and microstructure characteristics. The combination with fly ash was less applicable due to a rapid setting, hardening retarder used in these binders caused significant deterioration of the mechanical properties of the material.

Stanovení možností zvyšování vazného potenciálu el. popílků pro výrobu cementových kompozitů / Determining the possibilities of increasing binding potencial of fly ash for the usage in cement composites

Jančaříková, Denisa Unknown Date (has links)
The deliberate use of fly ash in the production of concrete has been used for decades, but recent trends tend to maximise its utility properties. These are different types of activations from chemical through thermal to mechanical. An alkaline agent is added to chemical activation which, by etching the glass structure of the ash grain, promotes its reactivity. Mechanical activation is mainly focused on granulometry and particle size distribution curve. In this thesis three mechanical activation methods are compared: separation the ash into fractions by grain size, mixing these fractions to achieve the ideal particle size distribution curve and grinding. Four types of ash were collected from the Tušimice power plant – ash from individual electrostatic separators (I, II and III) and SESYP ash which represents the total volume of production. Ash from individual separators differ not only in the particle size but also in the chemical composition and reactivity. Better properties generally have smaller ash particles. This is used by mechanical activation by sorting. When mixing the individual ash fractions, the aim is to achieve an optimal grading curve of either the ash itself or the fine particle set in the concrete. In this work, ash is only used to calculate the mixing ratio and the grading curve according to Funk was selected as optimal. The last compared option of activation was grinding ash. Grinding was carried out in an industrial mill for 15, 30, 45, 60 and 75 minutes. The efficiency of the modified ash was monitored for cement paste in terms of rheology and for concrete in terms of consistency, strength and durability. Based on the results we can say that mechanical activation by sorting is suitable for special purposes, but it’s necessary to realise that coarser ash fractions remain unused. When mixing the fractions to ensure matrix density, the efficiency was shown mainly on the durability characteristics. Grinding cause positive effect.

Vliv velikosti částic odpadního skla na vlastnosti alkalicky aktivovaných aluminosilikátových kompozitů / Effect of Grain Size of Waste Glass on Properties of Alkali Activated Aluminosilicate Composites

Novák, Václav January 2019 (has links)
The diploma thesis is focused on the use of the waste glass with different fineness on alkali - activated composites, mainly based on slag and fly ash. The theoretical part is focused on materials that are most used for alkaline activation - slag, fly ash and their composites with waste glass. The theoretical part also deals with the alkaline activation of composites from these materials and the factors that influences the microstructure and properties of these composites. In the experimental part were prepared composites from slag and fly ash with a waste glass as substitute. These composites then have been examined on mechanical properties and microstructure, also how different fineness of glass influences these properties. Then it will be decided whether it is economically advantageous grinding waste glass to finer fractions
