Living in a predation matrix : Studies on fish and their prey in a Baltic Sea coastal area

Ahlbeck, Ida January 2012 (has links)
This thesis was written within the framework of a biomanipulation project where young-of-the-year (YOY) pikeperch (Sander lucioperca) were stocked to a Baltic Sea bay to improve water quality through a top-down trophic cascade. The aim of my doctorial studies was however focused on a broader ecological question, namely predation (the main driving force in a biomanipulation). Hence, this thesis consists of four papers where we study the interactions between predator and prey using fish and zooplankton and how these interactions can be measured. In paper I we evaluated the performance of different diet analysis methods by individual based modelling and found that when having a nutritional gain perspective, mass based methods described diets best. Paper II investigated how the explorative, foraging and anti-predator behaviour of the YOY pikeperch used for stocking were affected by their rearing environment (pond vs. tank rearing). The more complex and varied environment in the semi-natural ponds seemed to promote a more flexible and active behaviour, better equipping young fish for survival in the wild. For paper III we studied the diel vertical migration in the six copepodite stages of the zooplankton Acartia spp. and Eurytemora affinis in relation to fish biomass, phytoplankton abundance and temperature. Both species migrated and in addition showed increased migration range with size within species, indicating evasion from visual predators. Paper IV addressed the movement of littoral Eurasian perch (Perca fluviatilis) via stable isotope signatures (13C and 15N) and body condition. We found clear indications of sedentarity and intra-habitat dietary differences. Interactions between predators and prey are complex and affected by both physiological and environmental characteristics as well as behavioural traits. The results in this thesis suggest that different species and even different life stages pursue different strategies to survive. / At the time of doctoral defense, the following papers were unpublished and had a status as follows: Paper 1: Submitted.  Paper 2: In press.  Paper 4: Submitted.

Expression analysis of the 3p25.3-ptelomere genes in epithelial ovarian cancer

Rossiny, Vanessa Delphine. January 2008 (has links)
Microarray expression analysis was carried out to identify genes with a role in epithelial ovarian cancer (EOC). The U133A Affymetrix GeneChipRTM was used to determine the expression patterns of the 3p25.3-ptel genes represented on the microarray in 14 primary cultures of normal ovarian surface epithelial (NOSE) samples, 25 frozen malignant ovarian tumor samples and four EOC cell lines. Seven genes with differential expression patterns in the tumor samples compared to the NOSE samples were identified as candidates for further analysis, starting with ARPC4, SRGAP3 and ATP2B2. Although none of the candidates had been previously studied in ovarian cancer, several had either family or pathway members that had. Expression patterns seemed unaffected by either tumor histopathological subtype or the allelic imbalances observed with loss of heterozygosity (LOH) analysis. The absence of association with genomic context suggested that differential expression was the result of transcriptional regulation rather than direct targeting.

A study to control the sensitivity of primers for burbot (Lota lota), salmon (Salmo salar) and brown trout (Salmo trutta)

Borgiel, Björn January 2018 (has links)
eDNA is a fast and popular method to collect information about species presence in the environment. eDNA is DNA that is collected from environmental samples, such as water, from DNA that is expelled from organisms interacting with their environment. eDNA is an effective way to find species with small populations and alien species. There are two ways to analyze eDNA, with high-throughput DNA sequencing methods and DNA metabarcoding or use of species-specific primers and PCR. In this study, we focus on the latter, designing species-specific primers for Burbot, Brown trout, and Atlantic Salmon, testing their validity in detecting eDNA of these species with functional PCR. We also evaluated eDNA collection methods, testing different scenarios in aquarium tanks with different number of dead and alive fishes. Primers and experimental setup such as use of different temperatures of the PCR reaction used in this study didn’t result in a functional PCR as determined by electrophoresis gel. There are some problems with the design of the PCR methods for eDNA since the purpose is to design methods that can identify certain species. However, future development of eDNA methods will probably include sequencing and not detection of PCR product sizes. eDNA methods will complete traditional trapping methods like net and electrofishing, but not replace them.

Évaluation de la capacité à innover : une approche par auto évaluation et suivi supporté par des analyses multicritères dynamiques / Evaluation of Innovation capabilities : A self assessment approach and monitoring supported by multi-criteria dynamic analysis

Galvez Manriquez, Daniel 09 February 2015 (has links)
Des scénarii sont simulés afin de tester les résultats de la mise en œuvre des différentes stratégies / One of the most important challenges in the current industrial policy of developed and emerging countries is to support innovation in SMEs, issue which has been approach as a problem only dependent on the availability of resources. Innovation capability does not have an appropriate framework of evaluation, mainly because of its complexity and abstract nature. Without proper indicators, it is not possible to establish objectives and identify the efforts and resources necessary for its improvement. In response to this, the ERPI laboratory has developed an evaluation approach of innovation capability, taking into account internal practices to promote innovation. A field of knowledge has been developed in this direction allowing for the proposal of a novel metrology, the Potential Innovation index (PII), based on multi-criteria analysis methods for studying the dynamics within companies. In this thesis, we propose an optimization model that replicates the evaluation procedure of innovation capability to generate an action plan, i.e., the model returns concrete points in which companies can work to improve its innovation system. The model considers factors such as the particular situation of the company, the economic efforts associated with the proposed strategy, the goals of the entrepreneur, etc. Three optimization strategies are proposed: access to a higher category with a minimum of effort, maximize resource efficiency and maximize innovation performance with a defined budget. Scenarios are simulated to test the results of the implementation of the different strategies

In this research, propagation phenomena of acoustic waves in one-dimensional and two-dimensional cavities is investigated and a methodology, based on energy methods, is proposed to predict acoustic response of these structures, at mid and high frequencies. These methods, originated from Energy Flow Analysis, use as primary variables the energy density and the energy flow. Two methodologies are used to solve approximated energy differential equations derived from EFA: Energy Finite Element Method (EFEM) and Energy Spectral Element Method (ESEM). The Spectral Element Method (SEM), is used to solve the wave propagation differential equations of acoustic in one-dimensional cavities, moreover this method is applied to validate the proposed methodology. Different acoustics cavities were investigated, such as simple waveguide, waveguide with geometric discontinuity and waveguide with discontinuity of the average properties. To this purpose, coupling relationships that represent those discontinuities are developed. A simulated example involving a closed rectangular acoustic cavity is analyzed with ESEM and the results are compared with those obtained by EFA, the Energy Finite Element Method (EFEM) and the acoustic analytical solution (SAA) of forced Helmholtz equation for rectangular two-dimensional closed cavities. An experimental investigation of acoustic response of a closed two-dimensional acoustic cavity was performed to verify the results of ESEM. The simulated models results, calculated in terms of energy variables, to analyze cavity, showed a good agreement with the experimental result / Doutorado / Mecanica dos Sólidos e Projeto Mecanico / Doutor em Engenharia Mecânica
