O sistema olfatório desempenha um papel relevante na exploração do ambiente e no reconhecimento social e sexual de mamíferos. Por meio deste sistema os animais podem reconhecer, detectar e discriminar uma grande quantidade de odorantes de estruturas químicas variadas, sinais químicos no ambiente essenciais para a sobrevivência e os ferormônios, que desencadeiam comportamentos sociais e reprodutivos. Algumas doenças e certos tipos de injúrias fisiológicas podem provocar a morte destas células, o que pode levar a perda da sensibilidade olfatória, embora já se saiba que este epitélio apresenta grupos de neurônios capazes de regeneração. A partir deste contexto, a terapia celular acaba sendo uma alternativa para o tratamento de patologias as quais acometem o sistema olfatório, como por exemplo, a anosmia que pode causar problemas graves, desde acidentes com gás ou comida estragada até depressão e distúrbios alimentares, causadas pela perda do paladar. Objetivou-se com este trabalho avaliar a recuperação da função olfatória de ratos anósmicos, bulbectomizados e submetidos a terapia celular com células-tronco provenientes do epitélio olfatório de ratos wistar. Para tanto foram utilizados 21 ratos machos Wistar de sessenta dias de idade, onde três foram utilizados para obtenção das células tronco do epitélio olfatório, dois para o controle cirúrgico, e restante foram divididos em 4 grupos: GI, GII, GIII e GIV os quais foram transplantados após 3, 7, 14 e 21 dias após a bulbectomia, respectivamente. A técnica cirúrgica foi realizada com incisão de pele, tecido subcutâneo e periósteo, seguida de abertura em janela de formato ovalado e remoção dos bulbos olfatórios mediante aspiração. Para a comprovação da anosmia após a cirurgia, os ratos foram submetidos ao teste comportamental do \"odor de gato\", e os do grupo controle após o período experimental foram sacrificados e a área encefálica da lesão causada pela cirurgia foi coletada onde foram realizadas análises histopatológicas. Os animais do GI, GII, GIII e GIV após 3, 7, 14 e 21 dias após bulbectomia foram anestesiados e receberam células tronco (1x106) do EOR no mesmo local da realização da bulbectomia, e posteriormente foram submetidos ao teste comportamental do \"odor de gato\". Transcorrido o período experimental, foram eutanasiados e os fragmentos de encéfalo foram coletados para análise histopatológica e imunohistoquímica. Os resultados evidenciam que realização da intervenção cirúrgica demonstrou remoção parcial do BO, com destruição da conexão nervosa entre os bulbos olfatórios e o epitélio olfatório. Ainda, a partir do teste comportamental do \"odor de gato\", e pela análise histopatológica das lesões causadas pela cirurgia, que evidenciou extensa área de necrose, com presença de hemossiderina e astrogliose reativa, constatou-se que a técnica empregada para promoção da bulbectomia foi eficaz para promoção da anosmia. A partir da análise comportamental, dos animais submetidos a terapia celular, os animais do GII e GIII apresentaram modificações no comportamento olfativo, com comportamento olfativo positivo ao \"odor do gato\", aversão comportamento defensivo, enquanto 100% dos animais do GI e GIV não apresentaram nenhuma modificação no comportamento olfativo. As análises por imunohistoquímica evidenciaram marcação positiva para o GFP, o que indica a presença das células tronco transduzidas com eGFP nos locais das lesões e ainda a expressão positiva do GFAP que evidencia a presença de astrogliose reativa com presença de cicatriz glial nos locais das lesões. / The olfactory system plays an important role in the exploration of the environment and in social and sexual behavior in mammals. Disturbances of the olfactory system such as observed in anosmia has also been related to accidents caused by gas leak and intoxications by food poisoning, in addition to eating disorders due to relation of the olfactory system with the taste. Through the olfactory system, animals recognize, detect and discriminate a large amount of odorants in a variety of chemical products, including pheromones, as well as in the environment which may guarantee their survival. Some diseases and injuries cause death of cells from the olfactory system leading to decrease or loss of the smell sensitivity. It is known that the olfactory epithelium has cells capable of regenerating neurons. Therefore, the utilization of these cells in cell therapy represents an alternative for the treatment of the olfactory system disorders. The objective of this study was to evaluate the regenaration of olfactory function of bulbectomized rats that were subjected to cell therapy with cells isolated from the olfactory epithelium. Twenty one male Wistar rats, sixty days of age were included in this study. Three rats were used for isolation of the olfactory epithelium cells, two for the surgical control and the remaining were divided into 4 groups: GI, GII, GIII and GIV corresponding to groups where cells were transplanted after 3, 7, 14 and 21 days after bulbectomy, respectively. The surgical technique was performed with skin, subcutaneous and periosteum incisions, followed by craniectomy and the removal of olfactory bulbs upon aspiration. For proof of anosmia, the rats were subjected to behavioral testing know as \"cat odor\". The animals of the control group were sacrificed and brain, with the lesion area, collected and processed for histopathological analysis. The animals of the experimental groups (GI, GII, GIII and GIV) were anesthetized and received heterologous cells (1x106) from olfactory epithelium, thorugh the same venue of the bulbectomy. These animals subsequently underwent behavioral \"cat odor\" testing. After 3, 7, 14 and 21 days of the cell injection, the rats were euthanized and the animals brains were collected for histopathological and immunohistochemical analyses. The results show that the surgical procedure promoted partial removal of olfactory bulbs, with destruction of the neural connection between the olfactory bulb and the olfactory epithelium. The behavioral \"cat odor\" test and the histopathological exams of the lesions, which revealed a large area of necrosis with presence of hemosiderin and reactive astrogliosis, demonstrated that the bulbectomy technique used to promote anosmia was effective. The behavioral test showed that the animals from GII and GIII presented changes in olfactory sensitivity, with positive \"cat odor\" aversion and defensive reaction. This test did not show change in GI and GIV groups. Immunohistochemistry analysis was positive for GFP, suggesting the presence of eGFP transduced cells at the sites of injury. In addition, the expression of GFAP positive cells demonstrated the presence of reactive astrogliosis with glial scar at the sites of injury.

Efeitos, percepção de fármacos e comunicação química em peixes / Effects, perception of drugs and chemical communication in fish

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES / The presence of drugs in aquatic environments has been studied for decades and their actions have as consequence changes of several physiological mechanisms. It is known that there is a complex chemical communication between the species that inhabit this environment, but the perception of intra and interspecific communication substances is unknown. The studies developed aim to evaluate if the fish are able to identify different stress situations and drugs, analyzing the neuroendocrine, behavioral and osmoregulatory effects. Seven studies were elaborated: 1) acute stress test and osmoregulatory evaluation with fluoxetine; 2) test of the action of fluoxetine in different responses of stressors (physical or chemical); 3) individual perception test to different conspecific situations; 4) testing preference at different concentrations of drugs and detection by olfaction; 5) experimental anosmia test in type-anxiety behavior; 6) experimental anosmia test, by ZnSO4, in behavioral and physiological responses; 7) test of the effects of acute stress on social behavior and "anxiety" in jundias (Rhamdia quelen). Based on the results it was verified that the acute exposure to fluoxetine is able to inhibit osmoregulatory changes caused by stress. Fluoxetine attenuates the cortisol response to physical stressor stimulus, but not to chemical stressor stimulus. Zebrafish can perceive and trigger aversive behaviors when in contact with conditioned waters of physical, chemical, and food stress (acute fasting). Zebrafish is attracted to psychotropic drugs such as diazepam, fluoxetine, risperidone and buspirone, which are probably detected by olfaction. Temporary experimental anosmia (by lidocaine and ZnSO4) modulates anxiety behaviors in adult zebrafish. Jundia can be used for behavioral studies of "anxiety" and social interaction. In general, these results contribute to a better understanding of the effects and perceptions of pharmacological action and chemical communication in fish. / A presença de fármacos em ambientes aquáticos tem sido estudada há décadas e suas ações têm como consequências alterações de diversos mecanismos fisiológicos. Sabe-se que existe uma complexa comunicação química entre as espécies que habitam esse ambiente, mas a percepção de substâncias das comunicações intra e interespecíficas é desconhecida. Os estudos desenvolvidos objetivam avaliar se os peixes são capazes de identificar diferentes situações estressoras e fármacos, analisando os efeitos neuroendócrinos, comportamentais e osmorregulatórios. Foram elaborados sete estudos: 1) teste de estresse agudo e avaliação osmorregulatória com fluoxetina; 2) teste da ação da fluoxetina em diferentes respostas de estressores (físicos ou químicos); 3) teste de percepção individual a diferentes situações de coespecíficos; 4) teste de preferência a diferentes concentrações de fármacos e detecção pela via olfatória; 5) teste de anosmia experimental em comportamento tipo-ansiedade; 6) teste de anosmia experimental, por ZnSO4, em respostas comportamentais e fisiológicas; 7) teste dos efeitos do estresse agudo sobre comportamento social e "ansiedade" em jundiás (Rhamdia quelen). Com base nos resultados, verificou se que a exposição aguda de fluoxetina é capaz de inibir as alterações osmorregulatórias causadas pelo estresse. A fluoxetina atenua a resposta de cortisol a estímulo estressor físico, mas não a estímulo estressor químico. O zebrafish pode perceber e desencadear comportamentos aversivos quando em contato com águas condicionadas de estresse físico, químico e alimentar (jejum agudo). O zebrafish apresenta atração por psicofármacos como diazepam, fluoxetina, risperidona e buspirona, os quais provavelmente são detectados pela via olfatória. A anosmia experimental temporária (por lidocaína e ZnSO4) modula comportamentos de ansiedade em zebrafish adulto. O jundiá pode ser utilizado para estudos comportamentais de "ansiedade" e interação social. De modo geral, esses resultados contribuem para um melhor entendimento dos efeitos e percepções da ação farmacológica e de comunicação química em peixes.

Subtle Differences in Brain Architecture in Patients with Congenital Anosmia

People suffering from congenital anosmia show normal brain architecture although they do not have functional sense of smell. Some studies in this regard point to the changes in secondary olfactory cortex, orbitofrontal cortex (OFC), in terms of gray matter volume increase. However, diffusion tensor imaging has not been explored so far. We included 13 congenital anosmia subjects together with 15 controls and looked into various diffusion parameters like FA. Increased FA in bilateral OFC confirms the earlier studies reporting increased gray matter thickness. However, it is quite difficult to interpret FA in terms of gray matter volume. Increased FA has been seen with recovery after traumatic brain injury. Such changes in OFC point to the plastic nature of the brain.

Understanding subjective measures of olfaction and cognition : A study on the occurrence of subjective olfactory and/or cognitive decline and their effect on future behavioral performance

Dementia is a growing burden for society, and it is of interest to discover it at an early stage. Both subjective cognitive decline (SCD) and subjective olfactory decline (SOD) has been associated with future cognitive decline and dementia. However, subjective measures have often been criticized and are still not fully understood. I aimed to examinate the frequency of SCD and SOD and whether they are likely to measure different things and what their longitudinal effects are. The baseline sample (N=784, 35-90 years, 51% female) were split into reported SCD, SOD, combined subjective olfactory and cognitive decline, and controls. Between-subjects and within-subjects statistical tests were conducted on a subset of participants (N=307, 45 to 90 years, 52% female) comparing SCD and SOD and their olfactory ability, cognitive performance, and demographics. In the baseline sample, a total of 21.1% reported a SOD whereas only 9.9% reported a SCD, only 2.7% reported both. SOD individuals had an emerged olfactory decline at follow up, their olfactory performance was associated with performance in several cognitive tests, this was not the case for the SCD individuals. The SOD and the SCD groups differ from each other, and they appear to be rather independent from each other. They might be complementary in understanding the aging brain.

Le sens du goût chez l'aveugle congénital

Thèse réalisée en collaboration avec le Département de neurosciences et pharmacologie de l'Université de Copenhague, Danemark. / La vision est cruciale dans la recherche et l’identification de nourriture. Non seulement elle déclenche le réflexe céphalique de la digestion mais, combinée à l’expérience alimentaire, elle aide à raffiner nos prévisions par rapport aux aliments. En un simple clin d’œil, la vue renseigne sur la disponibilité, l’identité, la comestibilité, les saveurs, les textures et les contenus nutritionnel, calorique et toxique des aliments qui nous entourent. Étant donnée l’importance de la vue dans l’expérience gustative, il est judicieux de se poser la question suivante : Qu’arrive-t-il au goût en absence de vision? Cette thèse répond à cette question par l’étude de cette modalité chez l’aveugle de naissance grâce aux techniques de psychophysique et d’imagerie cérébrale. De plus, les conséquences gustatives de la cécité sont comparées à celles suivant la perte d’un autre sens important dans l’appréciation des aliments, soit l’odorat (anosmie). Les résultats comportementaux démontrent premièrement que l’absence de vision depuis la naissance abaisse la sensibilité gustative, reflétée par des seuils élevés de détection et d’identification des cinq goûts de base (sucré, salé, acide, amer, umami). Deuxièmement, bien que les aveugles congénitaux aient plus de facilité à identifier les odeurs comestibles par leurs narines (voie olfactive orthonasale), ceux-ci perdent leur avantage par rapport aux voyants quand ils doivent identifier ces stimuli placés sur la langue (voie olfactive rétronasale). Les résultats d’imagerie indiquent en outre que les aveugles congénitaux activent moins leur cortex gustatif primaire (insula/opercule) et leur hypothalamus par rapport aux voyants durant une tâche gustative. De plus, l’absence d’activation dans le cortex (« visuel ») occipital chez l’aveugle pointe vers le manque de plasticité intermodale en gustation. Chez les anosmiques congénitaux d’autre part, non seulement l’absence d’odorat diminue l’habileté à reconnaître les goûts mais elle abaisse également la force du signal dans les aires olfactives (ex : cortex orbitofrontal médial) durant une tâche gustative. Les résultats chez l’aveugle contrastent grandement avec les études antérieures soulignant l’amélioration de leurs sens extéroceptifs tels que l’audition, l’olfaction (orthonasale) et le toucher qui font tous intervenir la plasticité intermodale. Par ailleurs, les données chez l’anosmique concordent avec ceux de la littérature indiquant une diminution similaire de la chémosensation trigéminale, laquelle est également associée à un affaiblissement du circuit neural des saveurs. Ceci suggère que le sens du goût ne soit pas utile aux handicapés visuels pour percevoir l’environnement extérieur et ainsi compenser leur perte de vision. De plus, bien que l’odorat participe à l’appréciation de la nourriture en bouche, sa perte n’entraîne pas de compensation sensorielle chez l’anosmique. Prises ensemble, ces données indiquent différents mécanismes d’adaptation suivant la cécité et l’anosmie. Elles soutiennent également le point de vue selon lequel la perception unifiée de goûts et de saveurs inclut non seulement les sens chimiques et le toucher mais également la vision. Considérant l’importance du goût et de l’alimentation dans la qualité de vie, ces résultats encouragent la société tout comme les professionnels de la réadaptation à faciliter l’accès à la nourriture ainsi qu’à l’enseignement culinaire chez les handicapés sensoriels. / Vision is crucial for seeking and identifying food. Not only does it trigger the cephalic digestion reflex but, when combined with the experience of eating, it helps to refine expectations about foods. In a single eye blink, sight informs us about the availability, identity, palatability, flavours, textures as well as nutritional, caloric and toxic contents of foods surrounding us. Given the importance of sight in the gustatory experience, one may therefore ask the following question: What happens to gustation without vision? This thesis answers this question by studying this modality in congenitally blind subjects using psychophysical and brain imaging techniques. Additionally, the gustatory consequences of blindness are compared to those following the loss of another important modality involved in the appreciation of food, i.e. the sense of smell (anosmia). Behavioural results first show that the absence of vision from birth reduces the gustatory sensitivity, as reflected by higher detection and identification thresholds of the five basic tastes (sweet, salty, acid, bitter, umami). Second, although congenitally blind subjects are better at identifying palatable odorant stimuli through their nostrils (orthonasal olfactory route), they lose this advantage over sighted people when identifying these stimuli placed on their tongue (retronasal olfactory route). Neuroimaging results also reveal that congenitally blind subjects activate the primary gustatory cortex (insula/operculum) and the hypothalamus less compared to blindfolded sighted participants. Moreover, the absence of occipital (“visual”) cortex activity in the blind points towards the lack of crossmodal plasticity in gustation. In congenitally anosmics, on the other hand, not only does the absence of smell lower the ability to recognize tastes but it also lowers the strength of the signal in olfactory areas (e.g. medial orbitofrontal cortex) during a gustatory task. The results in the blind greatly contrast with previous studies highlighting the enhancement of their exteroceptive senses such as audition, (orthonasal) olfaction and touch, all of which involve crossmodal plasticity. Moreover, data in the anosmic group are consistent with previous literature describing similar decrease of trigeminal chemosensation that is also associated with a weakening of the flavour neural network. This suggests that the sense of taste is not useful to the visually impaired to perceive their exterior environment and compensate for their lack of vision. Furthermore, although olfaction contributes to the appreciation of foods in the mouth, the lack of this modality does not drive sensory compensation in anosmic subjects. Taken together, these data indicate different adaptation mechanisms following blindness and anosmia. They also support the view according to which the unified perception of tastes and flavours includes not only the chemical senses (taste, smell and trigeminal chemosensation) and touch but also vision. Given the importance of taste and eating experience in quality of life, these results encourage society as well as rehabilitation professionals to facilitate access to foods and culinary lessons in sensory deprived subjects.

Idiopathic Phantosmia: Outcome and Clinical Significance

Background/Aim: Little is known about the clinical significance of phantosmia. The literature on phantosmia indicates that this symptom has a wide range of differential diagnoses. However, most cases of phantosmia remain of unknown origin. Our goal was to follow up patients with idiopathic phantosmia, with special regard to improvement rates and possible severe health conditions preceded by phantosmia of unknown origin. Methods: Forty-four patients with idiopathic phantosmia which had consulted our Ear-Nose-Throat Smell and Taste Clinic over the last 10 years were contacted by telephone and underwent a structured medical interview. Results: None of the patients had eveloped any severe health condition or Parkinson’s disease. More than 5 years after the occurrence of hantosmia, more than 50% of the patients experienced disappearance (31.8%) or improvement (25%). In the remaining cases, phantosmia did not change (38.7%) or became worse ( ! 5%). Conclusion: The main findings of the present study were that idiopathic phantosmia improves or disappears in almost two thirds of the patients after more than 5 years, and that idiopathic phantosmia seems to be more likely a harmless symptom rather than a reliable predictor of early Parkinson’s disease or other severe diseases. / Dieser Beitrag ist mit Zustimmung des Rechteinhabers aufgrund einer (DFG-geförderten) Allianz- bzw.
