Enterococcus isolados de alimentos: caracterização molecular e perfil de resistência a antimicrobianos

Os enterococos são patógenos com considerável capacidade de expressar resistência a vários antimicrobianos e com abundante representatividade em alimentos. Este estudo teve por finalidade realizar o isolamento de cepas do gênero Enterococcus em alimentos, representados por carne de frango e carne suína (crua e processada), analisar o perfil de resistência antimicrobiana através do método de disco-difusão e identificar através de técnicas moleculares as espécies dos isolados e os genes codificadores de resistência para vancomicina e tetraciclina. Foram analisadas 36 amostras, totalizando 54 cepas isoladas pertencentes ao gênero Enterococcus. A espécie detectada em maior ocorrência nos isolados de carne crua de frango (40%) e carne crua suína (29%) foi E. faecalis e nos isolados de produtos cárneos processados (100%) E. faecium. E. casseliflavus/E. flavencens teve seu isolamento apenas em produto processado. A espécie E. gallinarum não foi confirmada entre as amostras de alimentos analisadas. As espécies E. faecalis e E. faecium isoladas a partir de carne crua de frango foram significativamente mais resistentes que as isoladas das demais amostras, apresentando altos percentuais de resistência para: estreptomicina, ciprofloxacina, norfloxacina, eritromicina, vancomicina e tetraciclina. Isolados de carne crua de frango e suína apresentaram gene de resistência para tetraciclina e vancomicina. Portanto, sugere-se que, alimentos de origem animal possam desempenhar um papel importante na disseminação e transferência de enterococos resistentes e/ou genes de resistência aos seres humanos. / Enterococci are pathogens with considerable ability to express resistance to multiple antimicrobials and abundant representation in foods. This study aimed to carry out the isolation of strains of the genus Enterococcus in food, represented by chicken and pork (raw and processed), to analyze the profile of antimicrobial resistance by the disk diffusion method and by molecular techniques to identify species isolates and genes encoding resistance to vancomycin and tetracycline. 36 samples, totaling 54 strains belonging to the genus Enterococcus were analyzed. The species detected in most occurred in isolates from raw chicken meat (40 %) and raw pork (29%) was isolated in E. faecalis and processed meat products (100%) E. faecium. E. casseliflavus / E. flavencens had its isolation only in the processed product. The species E. gallinarum was confirmed between food samples surveyed. E. faecalis and E. faecium isolates from raw chicken meat isolated were significantly more resistant than the other samples, showing high percentage of resistance to: streptomycin, ciprofloxacin, norfloxacin, erythromycin, vancomycin and tetracycline. The isolated raw poultry and swine gene showed resistance to tetracycline and vancomycin. Therefore, it is suggested that food of animal origin may play an important role in the dissemination and transfer resistant enterococci and / or resistance genes to humans.

A policitemia vera (PV) é uma doença mieloproliferativa clonal que afeta o progenitor hematopoético multipotente causando o acúmulo de eritrócitos, leucócitos e plaquetas morfologicamente normais e seus precursores, na ausência de um estímulo definido. Alterações na regulação da apoptose parece ser uma das causas do acúmulo de células eritróides sem a necessidade de eritropoetina. A mutação JAK2 V617F foi descrita como um dos eventos envolvidos na fisiopatologia das Doenças Mieloproliferativas Crônicas, sendo relacionada a um pior prognóstico. Esta mutação resulta na ativação constitutiva da enzima Janus Kinase 2 e conseqüentemente das vias de sinalização associadas ao estímulo celular mediado por citocinas e fatores de crescimento. O objetivo deste estudo foi investigar as alterações da expressão de proteínas e genes envolvidos na regulação da apoptose em pacientes com PV sem tratamento. Para tanto foi quantificada a expressão de genes e proteínas da família Bcl-2 e da via extrínseca da apoptose em células precursoras e leucócitos desses pacientes e de indivíduos saudáveis. Os leucócitos do sangue periférico de 12 pacientes e 14 controles foram obtidos pelo método de Haes-esteril. As células precursoras CD34+ destes pacientes e de 19 controles foram separadas em coluna imunomagnética. A detecção da expressão gênica e protéica das moléculas anti- e pró-apoptóticas foi realizada pelas técnicas de PCR em tempo real e Western-blot, respectivamente. Foi avaliada ainda a resistência à morte celular induzida por agentes apoptogênicos dos linfócitos dos pacientes. As células CD34+ e maduras mostraram expressão aumentada dos genes a1, mcl-1, noxa, c-flip e ciap-2. A expressão dos genes bid , bim-EL , fas e faim estava elevada e a dos genes bcl-2, bcl-xL e bax diminuída, somente nas células CD34+. Nos leucócitos, observou-se ainda maior nível de expressão do gene bok e expressão diminuída dos genes bcl-2, bcl-xL, bad, bax, faim, dr4, dr5 e trail. Os genes bcl-w, bak, bik, fasL e ciap-1 não mostraram diferença significativa na expressão em células CD34+ e leucócitos de pacientes e controles (p>0,05). Houve significância estatística nas correlações entre: expressão de dr5 e mutação JAK2 V617F; bim-EL com concentração de hemoglobina e com hematócrito; bcl-w e número de plaquetas; a1, bad, bax nas células CD34+ e bad em leucócitos com aumento do baço. Os linfócitos dos pacientes foram mais resistentes a apoptose mediada por ACT D 1uM e 5uM, ARA-C 25uM, CHX 25uM, VP-16 e VM-26 25uM. Houve correlação entre a percentagem de alelos mutados para JAK2 V617F e o perfil de resistência aos indutores de apoptose dos linfócitos dos pacientes a ACT D 1 uM e 5uM e STS 5uM. Observou-se ainda correlação estatística entre: expressão de a1 com ACT D 5 uM, CHX 50uM e ARA-C 25uM; bad com CHX 25uM e teniposídeo 25uM; bcl-2 com ACT D 5uM , CHX 50uM, STS 1uM e 5uM e VP-16 a25uM ; bid com ACT D 5uM e VM26 25uM; bik com ACT D 5uM , STS 1uM e 5uM e VM-26 25uM, ARAC 25uM e VP16 25Um; bim-EL com CHX 25uM e teniposídeo 25uM; bok com ACT D 5uM STS 1uM e 5uM, ARAC 25uM e etoposídeo 25uM; ciap-1 com ACT D 1uM e 5uM, STS 1uM e VP16 25uM; dr4 com ACT D 5uM, ARAC 25uM, VP16 a 25uM; dr5 com ACT D 5uM, STS 1uM, ARAC 25uM e VP16 25uM; faim com ACT D 5uM; mcl-1 com ARAC 25uM e STS 1uM; trail com ACT D 5uM. As proteínas A1, BAD e c-FLIP tiveram o mesmo perfil de expressão dos seus respectivos genes, contudo os níveis de expressão protéica para BCL-xL estavam aumentados e para BIM-EL diminuídos, resultados discordantes com os obtidos nas expressões gênicas. Polycythemia vera (PV) is a clonal myeloproliferative disease arising in a multipotent haematopoietic progenitor cell causing the accumulation of erythrocytes, leukocytes and platelets morphologically normal and their precursors in absence of a definable stimulus. Apoptosis deregulation seems to be one of the causes of erythroid cells accumulation without the need for erythropoietin. The JAK2 V617F mutation has been described as one of the events involved in pathophysiology of chronics myeloproliferative diseases, being related to worse prognosis. This mutation results in constitutive activation of the enzyme Janus kinase 2 and consequently of signaling pathways associated with cell stimulation mediated by cytokines and growth factors. This study aimed to investigate changes in expression of proteins and genes involved in regulation of apoptosis in PV patients without treatment. Was quantified for both gene and proteins expression of the Bcl-2 family and the extrinsic pathway of apoptosis members in precursor cells and leukocytes of patients and healthy subjects. Peripheral blood leukocytes of 12 patients and 14 controls were obtained by the Haes-sterile method. CD34+ precursor cells of these patients and 19 controls were separated by immunomagnetic column. Anti- and pro-apoptotic gene and protein expression detection was performed by real-time PCR and Western-blot techniques respectively. It also assessed the resistance to cellular death induced by apoptogenics agents in PV lymphocytes. CD34+ and mature cells showed increased expression of a1, mcl-1, noxa, c-flip and ciap-2. Gene expression of bid, bim-EL , fas e faim was high and to genes bcl-2, bcl-xL and bax diminished, only in CD34+ cells. In leukocytes, there was even greater level of expression of bok and reduced of bcl-2, bcl-xL, bad, bax, faim, dr4, dr5 and trail. Not significant difference was found to bcl-w, bak, bik, fasL and ciap-1 expression in CD34+ cells and leukocytes of patients and controls (p> 0.05). There was statistical correlation between: dr5 expression and JAK2 V617F mutation; bim-EL with hemoglobin concentration and hematócrito; bcl-w and platelets counts; a1, bad, bax in CD34+ cells and bad in leukocytes with enlarged spleen. PV lymphocytes were more resistant to apoptosis mediated by ACT D 1uM and 5uM, ARA-C 25uM, CHX 25uM, VP-16 25uM and VM-26 25uM. There was correlation between JAK2 V617F allele burden and resistance profile to apoptosis inducers in PV lymphocytes to ACT D 1uM and 5uM and STS 5uM. There was also correlation between: a1 expression with ACT D 5 uM, CHX 50uM and ARA-C 25uM; bad with CHX 25uM and teniposide 25uM; bcl-2 with ACT D 5uM, CHX 50uM, STS 1uM and 5uM and VP -16 25uM; bid with ACT D 5uM and VM-26 25uM; bik with ACT D 5uM, STS 1uM and 5uM, VM-26 25uM, ARA-C 25uM and VP-16 25uM; bim-EL with CHX 25uM and teniposide 25uM ; bok with ACT D 5uM, STS 1uM and 5uM, ARA-C 25uM and etoposide 25uM; ciap-1 with ACT D 1uM and 5uM, STS 1uM and VP-16 25uM; dr4 with ACT D 5uM, ARA-C 25uM, VP-16 25uM; dr5 with ACT D 5uM, STS 1uM, ARA-C 25uM and VP-16 25uM; faim with ACT D 5uM, mcl-1 with ARA-C 25uM and STS 1uM; trail with ACT D 5uM. The A1, BAD and c-FLIP proteins have the same profile of expression of their genes, however the levels of protein expression in BCL-xL were increased and decreased for BIM-EL, discordant results in related with the one obtained by gene expressions. Taken together these results suggest that apoptotic machinery is deregulated, which could contribute, at least in part, with myeloaccumulation observed in the SP and BM of PV patients.

Estudos fitoquímicos e avaliação do potencial anti-helmíntico da opuntia ficus-indica /

Esta planta apresenta propriedades medicinais interessantes. O. ficus tem um bom potencial nutritivo é fácil cultivare muito utilizada na alimentação de pequenos ruminantes especialmente em zonas áridas do nordeste do Brasil. Embora esta planta seja usada como alimento para ovinos, a literatura não contém quaisquer relatórios sobre as suas propriedades anti-helmínticas. Portanto, este trabalho teve como objetivo avaliar a atividade ovicida e larvicida da O. ficus por meio dos extratos hidroalcoólicos de cladódios e frutas em ensaios de eclosão dos ovos (EHA), desenvolvimento larval (LDA) e migração larval (LMIA) usando ovos e larvas de nematoides gastrointestinais de ovinos naturalmente infectados. Para o EHA foi observada uma inibição da eclodibilidade de ovos maior do que 90% para ambos os brutos extratos dos cladódios e frutos na concentração de 50 mg/mL ou mais elevada (p <0,05). Ambos os extractos mostraram uma resposta dose dependente com o mesmo valor de IC 50 de 9,9 mg/mL. No LDA, o extrato hidroalcoólico bruto dos cladódios mostrou 98,7% de inibição do desenvolvimento das larvas a 12,5 mg/ml, enquanto o extrato hidroalcoólico bruto dos frutos apresentou 100% de atividade a 50 mg/ml, respectivamente, com um IC 50 de 3,4 mg/ml e 3,7 mg/ml (p <0,05). No LMIA, os resultados foram maiores do que 65% de inibição da migração das larvas na a parir da concentração de 1,56 mg/mL com IC 50 de 0,35 mg/mL e 0,76 mg/mL, respectivamente para os extrato brutos dos frutos e cladódios. O fracionamento dos extratos hidroalcoólicos brutos dos frutas e cladódios forneceu quatro fracções. Entre os extratos fraccionados, os extratos aquoso residual e em acetato de etilo dos cladódios, na concentração mais baixa testada (1,56 mg / ml) resultou em 100% de inibição do... / Abstract: Opuntia fícus-indica (O. ficus), which is native to Mexico and is known as "palma forrageira" in Brazil, is a species belonging to the family Cactaceae. This plant displays interesting medicinal properties. O. ficus display good nutritional potential is easy to cultivate and is used as feed for small ruminants feed especially in arid areas of northeastern Brazil. Although this plant serves as food for sheep, the literature does not contain any reports on its anthelmintic properties. This paper reports on the ovicidal and larvicidal activity of O. ficus from hydroalcoholic extracts of cladodes and fruits through egg hatch assay (EHA), the larval development assay (LDA), and the larval migration assay (LMIA) using eggs and larvae of gastrointestinal nematodes of naturally infected sheep. For EHA was observed an egg hatch inhibition greater than 90% for both the crude extracts of cladodes and fruits when using a concentration of 50 mg/mL or higher (p < 0.05). Both extracts showed a dose-dependent response with the same value to LD 50 of 9.9 mg/mL. In the LDA, crude hydroalcoholic extract of cladodes showed 98.7 % of inhibition of larval development at 12.5 mg/ mL while the crude hydroalcoholic extract of fruits showed 100% at 50 mg/mL, respectively with an LD 50 of 3.4 mg/mL and 3.7 mg/mL (p < 0.05). In the LMIA, crude extracts showed results greater than 65% inhibition of larval migration at 1.56 mg/mL revealing LD 50 of 0.35 mg/mL and 0.76 mg/mL for the extract of fruit and cladodes, respectively. Fractionation of the crude hydroalcoholic extracts of fruits and cladodes yielded four fractions. BOTTLED FANTASIES: COLLEGE STUDENTS' INTERPRETATIONS OF ALCOHOL ADVERTISING AND ITS EFFECTS

This dissertation deals with the impact of alcohol advertising, which associates drinking alcohol with many desires and values psychologically appealing to young people. As traditional research on alcohol advertising's impact focused on a direct link between exposure and alcohol consumption, this study breaks the link into several mediating stages. Under the theoretical framework of the Message Interpretation Process (MIP) model, this study involves how college students interpret alcohol advertising, how their desire, wishful thinking and expectancies toward drinking may account for both the impact of exposure to alcohol advertising and reasons for drinking. Specifically, the purpose of the study is to (1) assess the MIP model and (2) explore the impact of anti-alcohol messages on college students' interpretation of alcohol advertising. An experiment was conducted with 94 college students. In the experiment, participants were grouped into three treatment groups: the first was exposed to logic-based anti-alcohol messages followed by alcohol advertisements, the second was exposed to emotion-based anti-alcohol messages followed by alcohol advertisements; and the third group was exposed to alcohol advertising only, as the control group. Results showed support for the MIP model in general. As posited by the model, realism and desirability are both related to identification with characters seen in alcohol advertising, which in turn is related to expectancies toward drinking, which is in turn related to intention to drink. The only two exceptions were the lack of significance for the social norm-identification link and relationships with negative expectancies. Compared with exposure, realism and desirability were better predictors for alcohol advertising's impact. The results also showed impact of logic-based anti-alcohol messages on identification and expectancies as well as impact of emotion-based anti-alcohol messages on desirability and identification. There was no significant difference between the two anti-alcohol groups. This study provides insights on how viewers draw from alcohol advertising to make sense of their own lives. Implications for anti-alcohol campaign are provided. Because negative expectancies toward drinking did not influence the intention to drink, the association between negative outcomes and drinking might not very well. Another association between positive outcomes and non-drinking may work better. Recommendations for future research as well as limitations of this study are discussed.

The logical anti-psychologism of Frege and Husserl

Frege and Husserl are both recognized for their significant contributions to the overthrowing of logical psychologism, at least in its 19th century forms. Between Frege's profound impact on modern logic that extended the influence of his anti-psychologism and Husserl's extensive attempts at the refutation of logical psychologism in the Prolegomena to Logical Investigations, these arguments are generally understood as successful. This paper attempts to account for the development of these two anti-psychologistic conceptions of logical objects and for some of the basic differences between them. It identifies some problems that are common to strongly anti-psychologistic conceptions of logic and compares the extent to which Frege's and Husserl's views are open to these problems. Accordingly, this paper is divided into two parts. Part I develops a conception of the problems of logical psychologism as they are distinctively understood by each philosopher, out of the explicit arguments and criticisms made against the view in the texts. This conception is in each case informed by the overall historical trajectories of each philosopher's philosophical development. Part II examines the two views in light of common problems of anti-psychologism.
