Development of new methods for the asymmetric formation of C-N bonds / Développement de nouvelles méthodes de formation asymétriques de la liaison C-N

Au cours de ce travail de nouvelles méthodes pour la formation de liaison C-N ont été développées. Dans la première partie de cette thèse une application de catalyse métal-ligand bifonctionnelle pour la réaction énantiosélective aza-Michael est démontrée. Dans la deuxième partie nous présentons le travail sur les cyclisations, en utilisant des alcaloïdes du quinquina facilement disponibles, comme catalyseurs des plus prometteurs, fournissant des β-amino-acides d’indoline avec jusqu'à 98% ee. Parmi eux, l’hydroquinidine ressort du lot comme étant le catalyseur donnant le meilleur excès énatiomérique. La troisième partie est liée à l'élaboration d'un nouveau processus intermoléculaires de diamination de styrènes, diènes et triènes, utilisant des bis-sulfonylimides comme source d'azote, en combinaison avec le diacétate de iodosobenzène, qui fournit une approche intéressante et efficace de diamines vicinales biologiquement et chimiquement important. La réaction peut être effectuée à température ambiante sans avoir besoin de protection par atmosphère inerte. / The concept of metal-ligand bifunctionality was successfully applied for an enantioselective aza-Michael reaction by employing well-defined ruthenium amido complexes. The catalyst was optimised and the corresponding chiral indoline β-amino acid derivatives were obtained with high enantioselectivities. Next, a straightforward enantioselective bifunctional organocatalytic approach was also developed. Employing hydroquinidine as catalyst the corresponding cyclic products were obtained in excellent enantioselectivities and quantitative yields. These compounds can be selectively deprotected and applied to peptide synthesis. Finally, we have developed unprecedented diamination reactions of styrenes, butadienes and hexatrienes employing easily accessible hypervalent iodine(III) reagents under robust reaction conditions. The first examples of the metal-free 1,2-diamination of butadienes were demonstrated and this oxidation methodology was further extended to the highly attractive 1,4 installation of two nitrogen atoms within a single step.

Adições de aza-Michael em diazocetonas α,β-insaturadas e reações de inserção em ilídeos β-cetosulfoxônios / α,β-unsaturated diazoketones in Aza-Michael additions and β-keto sulfoxonium ylides in insertion reactions

O trabalho de tese é dividido em dois capítulos e visou explorar a química das diazocetonas α,β-insaturadas bem como dos ilídeos β-cetosulfoxônios. No primeiro capítulo é apresentado o emprego das diazocetonas α,β-insaturadas como novos aceptores em reações de aza-Michael. A adição conjugada de aminas primárias e secundárias foi explorada, assim como o uso de aminas quirais para avaliar a versão assimétrica da reação. Além da formação desses adutos, também foram investigadas estratégias para a construção de heterocíclicos nitrogenados a partir destes ou via protocolos \"one-pot\" (partindo das diazocetonas α,β-insaturadas) levando à formação de 2- e 3-pirrolidinonas. Por fim, foi avaliada a aplicação da metodologia desenvolvida na síntese do produto natural Barmumicina. O segundo capítulo se dispôs a investigar o emprego dos ilídeos β-cetosulfoxônios como substitutos de compostos diazocarbonílicos em reações de inserção. Num primeiro momento, avaliaram-se reações de inserção S-H intermolecular, visando a construção do fragmento β-cetotioéter. Alguns estudos competitivos e mecanísticos, bem como estratégias assimétricas também compõem o escopo. Num segundo momento, os ilídeos foram empregados em reações de inserção N-H intramolecular, no qual, a partir de sulfoxônios derivados de aminoácidos, vislumbrou-se a formação de 3-azetidinonas. / This thesis is divided into two chapters which are related to the chemistry of α,β-unsaturated diazoketones and β-ketosulfoxonium ylides. The first chapter presents the utility of α,β-unsaturated diazoketones as new aza-Michael acceptors. Conjugate addition of primary and secondary amines was explored, as well as the use of chiral amines to evaluate the asymmetric version of the reaction. In addition to the formation of these adducts, it was also investigated some strategies for the synthesis of 2- and 3-pyrrolidinones via the \"one-pot\" protocols (starting directly from the α,β-unsaturated diazoketones). Finally, the synthesis of the natural product Barmumicyn was evaluated from this methodology. The second chapter aimed to investigate the use of β-ketosulfoxonium ylides as diazocarbonyl compounds substitutes in insertion reactions. At first, the intermolecular S-H insertion reaction was studied aiming the construction of the β-ketothioether fragment. Some competitive and mechanistic studies, as well as asymmetric versions are also part of the scope. Secondly, some ylides were also employed in intramolecular N-H insertion reactions (from sulfoxonium amino acid derivatives) aiming the formation of 3-azetidinones.

Synthèse et réactivité d'énamides, de la diversité moléculaire à la synthèse de molécules bioactives et/ou naturelles / Synthesis and reactivity of enamides, toward the molecular diversity and the synthesis of bioactive and/or natural compounds

La nécessité grandissante de disposer d’une large librairie de diverses petites molécules pour le screening biologique constitue une puissante force motrice pour les chimistes organiciens et requiert en amont le développement de méthodologies rapides et efficaces. Dans ce cadre, nous nous sommes plus particulièrement intéressés à la fonctionnalisation d’énamides qui représentent des blocs moléculaires intéressants permettant d’introduire des fonctionnalités aminées dans des systèmes variés. Notre objectif a été de synthétiser des petites bibliothèques de molécules azotées à partir de substrats communs tout en mettant en oeuvre les différentes stratégies de la synthèse orientée vers la diversité et en s’attachant à respecter les règles suivantes : économie d’atomes, processus catalysés, synthèses rapides en peu d’étapes et contrôle de la stéréoselectivité. Dans un premier temps, nous avons principalement synthétisé divers énamides, nous permettant par la suite de développer des méthodologies innovantes et d’accéder à des « structures privilégiées » ou des fragments clés présents dans des produits naturels ou dans des substances potentiellement biologiquement actives en mettant en jeu des processus variés telles que des réactions d’aza-Michael, d’oxyamidation ou en cascade et la chimie du palladium avec de la CH insertion, des dioxoazoborocanes ou encore l’utilisation de l’auxiliaire chiral SAMP. / The continuing demand to synthesize new and original collections of small molecules for the biological screening is an attractive subject for organic chemists and requires upstream the development of fast and easy synthetic methods. In this context, we decided to focus particularly on the functionalization of enamides which represent valuable building blocks in order to introduce nitrogen based functionality into various organic systems. Our objective was to synthesize new nitrogen containing compound libraries starting from common substrates by applying Diversity-Oriented Synthesis strategy and following these rules: atom economy, catalyzed reactions, fast synthesis in few steps and control of stereoselectivity. Firstly we mainly synthesized enamides. Thereafter, we developped efficient methodologies giving access to motifs frequently found in “privileged structures” or key scaffolds present in natural products or potential bioactive compounds thanks to various processes like aza-Michael, oxyamidation or cascade reactions, palladium chemistry with CH activation, dioxoazoborocanes or chiral auxiliary SAMP.
