The versatile Generic Nanosatellite Bus (GNB) technology fused with the Canadian Advanced Nanospace eXperiment (CanX) missions is pioneered by the Space Flight Laboratory (SFL) at University of Toronto Institute for Aerospace Studies. Microspace philosophy has been employed at SFL to develop low cost nanosatellites with emerging technologies for education and research. This thesis provides an insight to the space systems engineering experience acquired by the author being a master’s student at SFL. This thesis describes the design, construction and testing of the VHF beacon transmitter for GNB. The umbilical Electrical Ground Support Equipment (EGSE) designed and built by the author is presented in this thesis. The assembly, integration and testing of the SFL ground station for the CanX missions has been explored. The on-orbit spacecraft operations for CanX-2 and NTS, from the launch campaign to the commissioning phase to nominal operations along with the anomalies faced and the contingency operations carried out by the author to date have been explained in detail in this thesis.

Validation of the North American Ice Service Iceberg Drift Model

Icebergs calved from high-latitude glaciers and ice shelves pose a threat to vessels and offshore infrastructure at a time when Arctic shipping and resource exploration are increasing. Knowledge of the location of potential ice hazards is therefore critical to ensure safe and efficient operations in this remote region. The Canadian Ice Service (CIS; Environment and Climate Change Canada) provides information to stakeholders on the observed and predicted distribution of icebergs in Canadian waters by combining iceberg observations with forecasts from the North American Ice Service (NAIS) iceberg drift model. The NAIS model estimates the forces acting on an iceberg to predict its future position and velocity. It is widely used for the east coast of Canada but largely unproven in the Arctic and suffers from insufficient validation due to a paucity of reliable in-situ observations of iceberg drift. This study represents the first comprehensive validation of the NAIS iceberg drift model for the Canadian Arctic. A total of 133 hindcast simulations for the period 2009-2019 were performed against in-situ drift observations of 44 icebergs. These data, collated in an iceberg beacon database (compiled by Carleton University and CIS), includes observations collected by Cryologgers; novel iceberg tracking beacons designed as part of this study. Quantified comparisons of the distance error between observed and modelled drift tracks indicate that the NAIS model produces realistic simulations of iceberg drift in Baffin Bay. Root mean square error after 24-hours of simulated drift ranged from 18-22 km and increased at a daily rate of 11-13 km, which is typical of previous model verification and validation studies. Improved model performance was observed for longer (>250 m) and deeper-keeled (>100 m) icebergs, which appears to counteract the model’s tendency to overestimate drift by reducing the influence of stronger surface ocean currents acting on the iceberg. Ocean current direction, wind direction, and iceberg keel geometry were identified by a sensitivity analysis as the model parameters and environmental driving forces that have the greatest influence on modelled iceberg drift. These results emphasize the need for accurate environmental information and underscore the importance of properly representing the physical characteristics of icebergs in drift models.

Design and Implementation of a Practical Aircraft Position and Reporting Identification Beacon (PRIB)

A transponder is a device that is used for tracking aircraft by mean of a secondary radar system, but it can be turned off deliberately, and it is an expensive item for small aircraft. These weaknesses have fatal consequences, as was shown with the terrorist attack on September 11 th , 2001, where four commercial aircraft under the control of international terrorists were used as missiles against the United Stated of America, killing thousands of people. These factors have shown a need for the development of an efficient aircraft tracking system, which does not rely on transponders. To this end a new tracking aircraft system is proposed, which will be referred to as the Positioning and Reporting Identification Beacon (PRIB) system. Due to size, mass, power, and financial constraints, the design must be small, light, power efficient, and cost-effective. The PRIB will acquire the aircraft's position from a dedicated GPS receiver and then transmit this information to a base station at a different location using a radio link. This thesis presents the design of a PRIB unit in light of the system constraints. In addition to the hardware design, the software needed by the unit to control and communicate with the ground stations is presented. The performance of the PRIB is analyzed and ways in which a PRIB could be manufactured using commercial off-the-shelf parts is discussed. / Master of Science

Excimer-Monomer Switching Molecular Beacon: The Study on Synthetic Cryptosporidum DNA Detection, Thermodynamics, and Magnesium Effects

Cryptosporidium parvum is a deadly waterborne protozoan parasite that invades the gastrointestinal tract of humans and causes severe to life-threatening gastro enteric disease. Due to the ubiquitous nature of Cryptosporidium parvum in the world’s water, it is necessary to determine the source of an outbreak. Rapid detection and identification of various genotypes of Cryptosporidium are a valuable goal in determining the source of the pathogen in a human epidemic. Exploitation of gene sequences specific to species is a powerful tool detecting pathogens. Molecular beacons are one of these tools for high selectivity and specificity detection of DNA and RNA. Molecular beacon is a single strand of DNA that forms a stem and loop structure, where the stem holds the DNA together and the loop detects a target sequence. This molecular beacon detection is determined by the changes of fluorescent emissions of fluorescent dye linked to the ends of the stem. In this thesis work, a new and novel molecular beacon was designed to detect the specific sequences from the heat shock protein gene of Cryptosporidium parvum that infects humans. This probe is synthesized by the conjugation of pyrene molecules to both ends of the stem which leads to a unique feature of pyrene excimer-monomer switching molecular beacon upon the hybridization of the loop sequence with the target DNA sequence. This thesis systematically investigates the physical binding (e.g., quantum yield) and thermodynamic properties, including enthalpy, entropy, and free energy of this excimer-monomer switching molecular beacon in the presences of complimentary, mismatched, and damaged DNA, respectively, in the three phases: phase one is the molecular beacon in the stem and loop structure, phase two is the molecular beacon hybridized to its target DNA, and phase three is the molecular beacon in a random coil. The effect of magnesium concentration on the binding and thermodynamic properties was also investigated. Finally, as a comparison, a conventional fluorescence resonance energy transfer-based molecular beacon with a fluorophore at the 5’ end and quencher at the 3’ end was used to assess selectivity and sensitivity in detection of DNA-DNA hybridization.

Analysis of beacon triangulation in random graphs

Our research focusses on the problem of finding nearby peers in the Internet. We focus on one particular approach, Beacon Triangulation that is widely used to solve the peer-finding problem. Beacon Triangulation is based on relative distances of nodes to some special nodes called beacons. The scheme gives an error when a new node that wishes to join the network has the same relative distance to two or more nodes. One of the reasons for the error is that two or more nodes have the same distance vectors. As a part of our research work, we derive the conditions to ensure the uniqueness of distance vectors in any network given the shortest path distribution of nodes in that network. We verify our analytical results for G(n, p) graphs and the Internet. We also derive other conditions under which the error in the Beacon Triangulation scheme reduces to zero. We compare the Beacon Triangulation scheme to another well-known distance estimation scheme known as Global Network Positioning (GNP).
