Misperceptions of Social Rejection: A Closer Examination of Inter-rater Discrepancies

Previous research suggests that discrepant ratings of symptoms, behaviors, and competencies may have important implications for childhood adjustment. Consequently, several theoretical explanations regarding the meanings and implications of inter-rater discrepancies have been proposed. The current study examined several largely unexplored issues regarding the statistical and conceptual properties of discrepant ratings; these include heterogeneity of agreement, shared method variance, and direction-specific effects. In a sample of 384 seventh and eighth-grade adolescents, it was found that each of the issues is integral in the interpretation of significant relationships between discrepant ratings of social rejection and childhood adjustment variables. More specifically, results demonstrated that the influence of shared method variance appears to be largely related to heterogeneity of agreement, and that accounting for shared method variance can increase, decrease, or even change the direction of the relationship between discrepancy scores and other variables. Also, results showed that the magnitude of discrepancies can be unrelated, equally related, or differentially related to the adjustment variables for the different forms of bias. Altogether, the findings of this study have implications for the theoretical conceptualization and statistical analysis of inter-rater discrepancy scores.

Gene flow dynamics in Baboons - The influence of social systems

Die Beziehung zwischen Genen und Verhalten ist in der Evolutionsbiologie von besonderem Interesse. Bestimmte Verhaltensweisen können die genetische Struktur natürlicher Populationen gestalten, dadurch deren genetische Diversität verändern und so ihr evolutives Schicksal beeinflussen. Abwanderung aus der Geburtsgruppe ist eine dieser Verhaltensweisen. Sie beeinflusst Genfluss, dessen Ausmaß die genetische Struktur von Populationen bestimmt. Paviane (Gattung Papio) sind ein besonders interessantes Forschungssystem um die Beziehung zwischen Verhalten und populationsgenetischer Struktur zu untersuchen. Die Evolution der Paviane wurde sowohl von historischem als auch gegenwärtigem Genfluss geprägt. Innerhalb dieser Gattung treten sowohl die überwiegende Abwanderung von Männchen als auch die überwiegende Abwanderung von Weibchen auf. Zudem wurde ihre gegenwärtige Verbreitung maßgeblich von Populationsausbreitung und –rückzug beeinflusst und es tritt häufig Genfluss zwischen verschiedenen Arten auf. In meiner Doktorarbeit untersuchte ich, wie verschiedene Abwanderungsmuster den Genfluss bei Pavianen beeinflussen. Damit hoffe ich zu einem besseren Ver-ständnis der Wechselbeziehung zwischen Verhaltensökologie und Genetik in natürlichen Populationen beizutragen. Ich fokussierte mich darauf, wie Unterschiede in den Sozialsystemen unterschiedlicher Pavianarten deren genetische Struktur beeinflussen. Die beobachteten Muster nutzte ich, um auf das geschlechtsspezifische Abwanderungsmuster bei Guineapavianen zu schließen, eine der am wenigsten untersuchten Pavianarten. Zudem untersuchte ich, wie sowohl historischer als auch gegenwärtiger Genfluss die genetische Struktur der Guineapaviane formten und ob es möglich ist von der Populationsausbreitung der Paviane Rückschlüsse auf die menschliche Evolutionsgeschichte zu ziehen. Um diese Fragen zu beantworten nutzte ich einen populationsgenetischen Ansatz, basierend auf im gesamten Verbreitungsgebiet gesammelten Kotproben, deren exakter geographischer Ursprung bekannt war. Ich analysierte sowohl autosomale Mikrosatelliten als auch Sequenzen der mitochondrialen Hypervariablen Region I. Meine Ergebnisse zeigen, dass die genetische Struktur der Guineapaviane am besten durch die überwiegende Abwanderung von Weibchen erklärt werden kann, sowohl in einem lokalen als auch im globalen Kontext. Weiblicher Genfluss führt zu einer hohen Diversität innerhalb von Populationen sowie einem Fehlen von genetisch-geographischer Struktur in mitochondrialer DNA. Nukleäre DNA hingegen zeigt eine starke globale geographische Struktur und Männchen sind im Vergleich zu Weibchen durch eine stärkere lokale Struktur gekennzeichnet. Dies entspricht den Vorhersagen für ein System, in welchem hauptsächlich Weibchen abwandern und Männchen in ihrer Geburtsgruppe verbleiben. Insgesamt scheint lokal begrenzte Abwanderung den wirksamen Genfluss auf eine Distanz unter 200 km zu beschränken, was zu einem starken Isolation-durch-Distanz Effekt und genetisch differenzierten Populationen führt. Anzeichen für Populationsausbreitung, die graduelle Struktur genetischer Variation, und mögliche Hinweise auf das “Allele-surfing” Phänomen, deuten auf eine historische westwärts gerichtete Ausbreitung von Guineapavianen hin. Introgressive Hybridisierung mit benachbarten Anubispavianen könnte genetische Muster im Bereich der Kontaktzone erklären, muss aber im Detail noch untersucht werden. Zusätzlich konnte ich zeigen, dass Mantelpaviane vermutlich im gleichen Zeitraum des Späten Pleistozäns von Afrika nach Arabien wanderten, wie Hypothesen für den modernen Menschen vorschlagen. Meine Studie ist die erste umfassende Analyse der genetischen Populationsstruktur der Guineapaviane und liefert Belege für die überwiegende Abwanderung von Weibchen in dieser Art. Dies untersützt die Ansicht, dass das Sozialsystem der Guineapaviane einige vergleichbare Merkmale zum System der Mantelpaviane aufweist und deutet somit darauf hin, dass während der Evolution dieser beiden Arten besondere evolutionäre Drücke gewirkt haben, die sie von allen anderen Pavianarten abgrenzen. In Kombination mit dem starken Einfluss von Populationsausbreitungen auf ihre Verbreitung und genetische Diversität, bekräftigt meine Arbeit Paviane als interssanten analogen Modellorganismus, der helfen kann, die Prozesse die während der Evolution des Menschen maßgeblich waren, aufzuklären.

The analysis of survival data with a possible cure fraction has attracted much interest in the last two decades. Various models and estimating methods have been proposed for such data and they have been applied in many fields, especially in cancer clinical trials. In the thesis, we consider some new general cure models, which include existing survival models as their special cases. We also consider a nonparametric estimation of cure rate. The estimator is proved consistent and asymptotically normal. Adverse effects of psychotherapy : Outcomes of a combined Internet treatment for Social Anxiety Disorder.

Social Anxiety Disorder (SAD) is a mental disorder with high prevalence but low treatment accessibility. A way to facilitate care to these patients is through Internet based treatment. As is the case of most psychological treatments, much has been studied about positive effects but there is a gap regarding adverse effects. This study seeks to fill this gap in the case of an Internet delivered treatment. From a total of 127 participants who took part in a treatment that combined attention biased modification (ABM) and Internet based cognitive behavior therapy (iCBT), 21 (16.5%) presented adverse effects. Most of the adverse effects were detected after participants had received iCBT (13.4%), and these were mostly related to deterioration of symptoms (5.5%), negative wellbeing (3.9%) and emergence of new symptoms (1.6%). Perceived side effects after attention training were less common (4.7%). A thematic analysis showed also that the techniques used in treatment, the lack of time to complete treatment and becoming aware with the impairments of the disorder and its consequences could influence the emergence of adverse effects.

The Genetic Structure and Dispersal Patterns of the Nigeria-Cameroon Chimpanzee (Pan troglodytes ellioti)

The goal of this study was to examine several aspects of the population genetics and population biology of the Nigeria-Cameroon chimpanzee at seven sampling locations in the south of Taraba State, Nigeria. Three of the sampling locations are within GGNP and two are situated just outside the southern boundary of GGNP. The final two sampling locations are found within Ngel Nyaki forest reserve, at each of the two forest fragments inside the reserve. Ngel Nyaki forest reserve was the focus of the study and the principal goal was to 16 determine if the community of chimpanzees at Ngel Nyaki forest reserve has become isolated from the chimpanzees at GGNP using microsatellite loci extracted from non-invasive sources of DNA. In Chapter two, the methods used to extract and amplify the DNA and the protocols used to confirm the genotypes are outlined. Chapter three examines the population structure of the chimpanzees among the regions sampled in this study, particularly addressing the question as to whether the chimpanzees at Ngel Nyaki forest reserve are isolated from the chimpanzees at GGNP. Chapter four investigates patterns of sex biased dispersal in the Nigeria-Cameroon chimpanzee. In Chapter five, population viability analysis is used to determine the fate of the chimpanzees at Ngel Nyaki forest reserve under a range of management scenarios. Chapter six summarizes the conclusions of the study and presents a conservation strategy to ensure the viability of the population of chimpanzees at Ngel Nyaki forest reserve.

Nucleotide Substitution Patterns in Vertebrate Genomes

The rates and patterns at which nucleotide substitutions occur vary significantly across the genome sequence of vertebrates. A prominent example is the difference in the rate of evolution of functional sequences versus nonfunctional (neutrally evolving) sequences, which is explained by the influence of natural selection on functional sequences. However, even within neutrally evolving sequences there is striking variation in the rates and patterns of nucleotide substitutions. Unraveling the underlying processes that induce this variation is necessary to understand the basic principles of variation in neutral substitution profiles, which in turn is crucial for the identification of regions in the genome where natural selection acts. This research question builds the main focus of the present thesis. I have studied the causes and consequences of variation in different patterns of nucleotide substitutions. In particular, I have investigated substitutional strand asymmetries in mammalian genes and could show that they result from the asymmetric nature of DNA replication and transcription. Comparative analysis of substitutional asymmetries then suggested that the organization of DNA replication and the level of transcription are conserved among mammals. Further, I have examined the variation in CpG mutation rate among human genes and could show that beside DNA methylation also GC content plays a decisive role in CpG mutability. In addition, I have studied the signatures of GC-biased gene conversion and its impact on the evolution of the GC isochore structure in chicken. By comparison of the results in chicken to previous results in human I found evidence that karyotype stability is critical for the evolution of GC isochores. Finally, beside the empirical studies, I have performed theoretical investigations of substitution rates in functional sequences. More precisely, I have explored the temporal dynamics of estimates of the ratio of non-synonymous to synonymous substitution rates dN/dS in a phylogentic-population genetic framework.

Meiotic Recombination in Human and Dog : Targets, Consequences and Implications for Genome Evolution

Understanding the mechanism of recombination has important implications for genome evolution and genomic variability. The work presented in this thesis studies the properties of recombination by investigating the effects it has on genome evolution in humans and dogs. Using alignments of human genes with chimpanzee and macaque orthologues we studied substitution patterns along the human lineage and scanned for evidence of positive selection. The properties mirror the situation in human non-coding sequences with the fixation bias ‘GC-biased gene conversion’ (gBGC) as a driving force in the most rapidly evolving regions. By assigning candidate genes to distinct classes of evolutionary forces we quantified the extent of those genes affected by gBGC to 20%. This suggests that human-specific characters can be prompted by the fixation bias of gBGC, which can be mistaken for selection. The gene PRDM9 controls recombination in most mammals, but is lacking in dogs. Using whole-genome alignments of dog with related species we examined the effects of PRDM9 inactivation. Additionally, we analyzed genomic variation in the genomes of several dog breeds. We identified that non-allelic homologous recombination (NAHR) via sequence identity, often GC-rich, creates structural variants of genomic regions. We show that these regions, which are also found in dog recombination hotspots, are a subset of unmethylated CpG-islands (CGIs). We inferred that CGIs have experienced a drastic increase in biased substitution rates, concurrent with a shift of recombination to target these regions. This enables recurrent episodes of gBGC to shape their distribution. The work presented in this thesis demonstrates the importance of meiotic recombination on patterns of molecular evolution and genomic variability in humans and dogs. Bioinformatic analyses identified mechanisms that regulate genome composition. gBGC is presented as an alternative to positive selection and is revealed as a major factor affecting allele configuration and the emergence of accelerated evolution on the human lineage. Characterization of recombination-induced sequence patterns highlights the potential of non-methylation and establishes unmethylated CGIs as targets of meiotic recombination in dogs. These observations describe recombination as an interesting process in genome evolution and provide further insights into the mechanisms of genomic variability.

Trends, cycles and institutions : -Job polarization and the business cycle in Europe

This thesis studies the cyclical aspect of job polarization in Europe. Contributions include offering a comparison to the findings of previous research on the United States, and extending the analysis by introducing labor market institutions. The analysis is done in two parts, first showing that the observed link between job polarization and jobless recoveries in the US is observed in Europe, but not across all countries and business cycles. In Scandinavia, the process of job polarization appears smoother than the spurts observed in the US. The second part involves regression analyses of the relationship between labor market institutions, the business cycle and occupational employment. The results indicate that stricter labor market institutions are less robustly associated with Routine employment than other occupational groups and that Routine employment is more sensitive to the business cycle than other types of employment. Further, rigid labor market institutions may prevent some of the Routine decline associated with economic downturns, while not necessarily affecting the long run employment. Limitations of the analysis regards rough estimates of the key variables, number of observations and the lack of identification associated with cross-country analyses.

Investigation of 1,900 Individual Field Aged Photovoltaic Modules for Potential Induced Degradation (PID) in a Positive Biased Power Plant

abstract: Photovoltaic (PV) modules undergo performance degradation depending on climatic conditions, applications, and system configurations. The performance degradation prediction of PV modules is primarily based on Accelerated Life Testing (ALT) procedures. In order to further strengthen the ALT process, additional investigation of the power degradation of field aged PV modules in various configurations is required. A detailed investigation of 1,900 field aged (12-18 years) PV modules deployed in a power plant application was conducted for this study. Analysis was based on the current-voltage (I-V) measurement of all the 1,900 modules individually. I-V curve data of individual modules formed the basis for calculating the performance degradation of the modules. The percentage performance degradation and rates of degradation were compared to an earlier study done at the same plant. The current research was primarily focused on identifying the extent of potential induced degradation (PID) of individual modules with reference to the negative ground potential. To investigate this, the arrangement and connection of the individual modules/strings was examined in detail. The study also examined the extent of underperformance of every series string due to performance mismatch of individual modules in that string. Random Walk With Absorbing Barriers Modeled by Telegraph Equation With Absorbing Boundaries

Organisms have movements that are usually modeled by particles’ random walks. Under some mathematical technical assumptions the movements are described by diffusion equations. However, empirical data often show that the movements are not simple random walks. Instead, they are correlated random walks and are described by telegraph equations. This thesis considers telegraph equations with and without bias corresponding to correlated random walks with and without bias. Analytical solutions to the equations with absorbing boundary conditions and their mean passage times are obtained. Numerical simulations of the corresponding correlated random walks are also performed. The simulation results show that the solutions are approximated very well by the corresponding correlated random walks and the mean first passage times are highly consistent with those from simulations on the corresponding random walks. This suggests that telegraph equations can be a good model for organisms with the movement pattern of correlated random walks. Furthermore, utilizing the consistency of mean first passage times, we can estimate the parameters of telegraph equations through the mean first passage time, which can be estimated through experimental observation. This provides biologists an easy way to obtain parameter values. Finally, this thesis analyzes the velocity distribution and correlations of movement steps of amoebas, leaving fitting the movement data to telegraph equations as future work.
