Meanwhile, for the consolidation of Brazilian exportation of meat in natura and its derivatives it is necessary that the products have food safety. Among the microorganisms used as indicators of food security, Salmonella has a special attention because it causes severe diseases to animals and humans. This study was developed with the objective to isolate Salmonella during the cattle slaughter in a state inspection slaughterhouse located in the city of Pelotas, Rio Grande do Sul. A hundred and fifty samples were collected from three different points of slaughter: leather of the animal after bleeding (P1), the carcass after evisceration (P2) and cecum contents (P3). The isolates were tested for their antimicrobial susceptibility in vitro using amoxicillin, ampicillin and ciprofloxacin. As results, two (1,33%) strains were isolated from bovines and classified as Salmonella. No strain was isolated from buffalos. The two isolates were susceptible to amoxicillin and ciprofloxacin. Besides the investigation of the presence of Salmonella in the cattle flowchart, a review about Salmonella pathogenicity islands (SPI) was performed. According to the results, we conclude that there is the risk of incorporating the bacteria at the slaughter time by the ruminants, contaminating the carcasses. The implementation and maintenance of means to control the quality in the market facilities are important to prevent or reduce the health risk to employees and consumers. / O Brasil possui um dos maiores rebanhos de ruminantes do mundo e destaca-se como um grande exportador de carne. Entretanto, para que a exportação brasileira de carne in natura e seus derivados seja consolidada é importante que o produto tenha segurança alimentar. Entre os micro-organismos usados como indicadores de segurança alimentar, Salmonella se destaca por causar muitas doenças em animais e humanos. Este estudo foi realizado com o objetivo de isolar Salmonella durante o abate de ruminantes em um frigorífico sob inspeção estadual, localizado na cidade de Pelotas, Rio Grande do Sul, que utiliza medidas para o controle de qualidade. Cento e cinquenta amostras foram coletadas de três pontos diferentes do abate: couro do animal após a sangria (P1), carcaça após a evisceração (P2) e conteúdo cecal (P3). Os isolados foram testados quanto à suscetibilidade antimicrobiana in vitro usando amoxicilina, ampicilina e ciprofloxacina. Como resultados, duas (1,33%) cepas foram isoladas de bovinos e classificadas como Salmonella. Nenhuma cepa foi isolada de búfalos. Os dois isolados apresentaram suscetibilidade à amoxicilina e à ciprofloxacina. Além da investigação da presença de Salmonella no fluxograma de ruminantes, foi realizada uma revisão sobre ilhas de patogenicidade em Salmonella (SPI). De acordo com os resultados, conclui-se que existe o risco da bactéria ser incorporada na linha de abate pelos ruminantes e, dessa forma, provocar a contaminação das carcaças. A implementação e a manutenção de medidas para o controle de qualidade nos estabelecimentos são importantes para prevenir ou reduzir o risco à saúde dos funcionários e consumidores.

Using remotely-sensed habitat data to model space use and disease transmission risk between wild and domestic herbivores in the African savanna

The interface between protected and communal lands presents certain challenges for wildlife conservation and the sustainability of local livelihoods. This is a particular case in South Africa, where foot-and-mouth disease (FMD), mainly carried by African buffalo (Syncerus caffer) is transmitted to cattle despite a fence surrounding the protected areas.The ultimate objective of this thesis was to improve knowledge of FMD transmission risk by analyzing behavioral patterns of African buffalo and cattle near the Kruger National Park, and by modelling at fine spatial scale the seasonal risk of contact between them. Since vegetation is considered as a primary bottom-up regulator of grazers distribution, I developed fine-scale seasonal mapping of vegetation. With that purpose, I explored the utility of WorldView-2 (WV-2) sensor, comparing object- (OBIA) and pixel-based image classification methods, and various traditional and advanced classification algorithms. All tested methods produced relatively high accuracy results (>77%), however OBIA with random forest and support vector machines performed significantly better, particularly for wet season imagery (93%).In order to investigate the buffalo and cattle seasonal home ranges and resource utilization distributions I combined the telemetry data with fine-scale maps on forage (vegetation components, and forage quality and quantity). I found that buffalo behaved more like bulk feeders at the scale of home ranges but were more selective within their home range, preferring quality forage over quantity. In contrast, cattle selected forage with higher quantity and quality during the dry season but behaved like bulk grazers in the wet season.Based on the resource utilization models, I generated seasonal cost (resistance) surfaces of buffalo and cattle movement through the landscape considering various scenarios. These surfaces were used to predict buffalo and cattle dispersal routes by applying a cumulative resistant kernels method. The final seasonal contact risks maps were developed by intersecting the cumulative resistant kernels layers of both species and by averaging all scenarios. The maps revealed important seasonal differences in the contact risk, with higher risk in the dry season and hotspots along a main river and the weakest parts of the fence. Results of this study can guide local decision makers in the allocation of resources for FMD mitigation efforts and provide guidelines to minimize overgrazing. / Doctorat en Sciences / info:eu-repo/semantics/nonPublished

Enhancing public participation on the ward committee system: the case of Buffalo City Metropolitan Municipality

This study investigated the role of ward committees in enhancing service delivery through public participation with reference to the Buffalo City Metropolitan Municipality in the Province of the Eastern Cape in South Africa. This study provided a motivation for the study to be undertaken, problem statement, objectives of the study, hypothesis, research design and methodology, ethical considerations, delimitation of the study and the overview of chapters. The Constitution of the Republic of South African of 1996 provides for the creation of three spheres of government, namely, the national sphere, the provincial sphere and the local government sphere. These spheres are interdependent from each other. Local government is one of the spheres of government created to bring government closer to the people, as well as giving communities a sense of involvement in the political and governance processes that control their daily lives. The need for effective democratic local government as a vehicle for development and national integration is imperative (Reddy, 1996:3). Municipalities are established in terms of section 12 of the Municipal Structures Act 117 of 1998, wherein Members of Executive Councils (MEC‟s) may establish a municipal area which the Demarcation Board demarcates in the province in terms of the Municipal Demarcation Act 27 of 1998.

Enhancing the accountability of ward councillors through public participation in Buffalo City Metropolitan Municipality

In terms of section 152(1)(a) of the Constitution of the Republic of South Africa, 1996 local government should provide a democratic and accountable service to local communities. Therefore, elected representatives and municipal officials must be accountable for their actions. This study investigates the influence of public participation in enhancing the accountability of ward councillors in the Buffalo City Metropolitan Municipality (BCMM). For the purpose of the study, the hypothesis was formulated that accountability of ward councillors is hampered by improper public participation mechanisms in Buffalo City Metropolitan Municipality (BCMM). In order to achieve the goal of this study and to test the hypothesis, it was essential to review literature on the accountability of ward councillors and public participation in local government. Primary data was obtained by interviewing ward committees and ward councillors from Buffalo City Metropolitan Municipality. Interviews were conducted with ward councillors and ward committees in wards 13 and 14 of BCMM. The major findings of the study, inter alia, are that ward councillors give feedback to their constituencies. Ward committees are instrumental in encouraging members of the public to attending these meetings. The most popular forum for public participation is the Independent Development Planning (IDP). It was established that approachability, accessibility and visibility of both ward councillors and ward committees in the community enhance public participation. The study concludes with recommendations that will assist the BCMM to enhance the accountability of ward councillors through public participation.

Evaluation of rural sanitation in Buffalo City Metropolitan Municipality

This research paper is a qualitative evaluation of rural resident’s views in Buffalo City Metropolitan Municipality regarding the municipalities (BCM) performance in sanitation in rural areas. The study was conducted using interviews and some participant observation as a strategy for data collection. The data collected was analyzed using the basic needs theoretical framework. The study found that sanitation is the least talked about subject because of the stigma attached to it. The study found that Buffalo City Municipality initiatives in sanitation have little impact or has not yet transformed the lives of rural people. It has been found that the municipality is responding slowly in sanitation, and this situation impacts negatively on the lives of rural people as they are exposed to health hazards and diseases. The study questions the sustainability of toilets built in rural areas as many toilets have no roofs and door. It has been found that sanitation is still a challenge in the Buffalo City Municipality as some people still relieve themselves in the bushes. Some people even defecate into plastic bags and then throw the bags as far as they can. The study found that the environment is deeply affected by the lack of sanitation as both air and water are contaminated by waste disposal caused by poor sanitation. The general feeling of the residents of Buffalo City Municipality with regard to sanitation service delivery is that they were not happy at all. The residents are not happy with the state of sanitation services rendered by the municipality. The study calls upon the municipality to speedily address rural sanitation to avoid service delivery protest. The research calls upon organizations and companies to get sanitation issues as part of their function. The research made a number of recommendations in an effort to help municipalities accelerate the delivery of sanitation in rural areas. It is hoped that the recommendations would help the Buffalo City Municipality in ensuring that communities are equipped and empowered to meaningfully participate in making policies, regarding their development. The recommendations will contribute towards encouraging community participation in the decision making within their municipalities. It is the aim of the study for other municipalities to use Buffalo City Municipality example as a benchmark for best practices. In conclusion, the rural areas of Buffalo City Metropolitan Municipality need more serious commitment from the municipality regarding sanitation and other basic services.

Insights Into The Mechanism Of Actions Of Luteinizing Hormone And Prostaglandin F2α In The Regulation Of Corpus Luteum Function Of Monoovulatory Species

Corpus luteum (CL), a transient endocrine structure formed from the ruptured ovarian follicle after ovulation, secretes progesterone (P4) that is essential for establishment and maintenance of pregnancy in mammals. The biosynthesis and secretion of P4 from CL depends, in general, on trophic hormones of the anterior pituitary gland and on hormones or factors originating from ovary, uterus, embryo and placenta. The structure and function of CL tissue is regulated by intricate interplay between two types of factors, namely, the luteotrophic factors, which stimulate CL growth and function, i.e., P4 secretion, and the luteolytic factors, which inhibit CL function and lead to luteal regression. In monoovulatory species such as higher primates and bovines, a striking diversity in the regulation of CL function exists not only between species, but also within the species during different stages of the luteal phase. In higher primates, unlike other species, one of the important characteristics of CL regulation is that, during non-fertile cycle, circulating LH appears to be the sole trophic factor responsible for maintenance of its function, and during fertile cycle, chorionic gonadotropin (CG), an LH analogue, originating from placenta maintains CL function. In higher primates, the role/involvement of luteolytic factors during luteolysis remains elusive. On the other hand, in the bovine species, the role/involvement of luteolytic factor, prostaglandin (PG) F2α during luteolysis is well established. It should be pointed out that in both the species, the mechanism of luteolysis is still poorly understood and the work presented in this thesis attempts to address these lacunae. Further, in bovines, studies have been carried out to examine potential trophic factor(s) responsible for the maintenance of CL function. Chapter I provides an extensive review of literature on CL structure and function with emphasis on factors that influence its growth, development, function and demise in primates and bovines. In Chapter II, employing bonnet monkey (Macaca radiata) as the representative animal model for higher primates, various studies have been conducted to examine the role of molecular modulators involved in regulation of CL function, particularly during spontaneous luteolysis. Although, it is well established that LH is essential for the maintenance of CL function in higher primates, the mechanism(s) responsible for the decline in serum P4 levels at the end of non-fertile cycles, without a concomitant change in circulating LH milieu, remains to be addressed. Several experiments have been conducted to examine the component(s) of luteotrophic (LH/CG) signaling that is/are modulated during luteolysis in the bonnet monkey CL. To understand the relative lack of responsiveness of CL to the circulating LH during the late luteal phase, LH/CG receptor (R) dynamics (expression of LH/CGR and its various transcript variants) was examined throughout the luteal phase and during different functional states of the monkey CL. The results indicated presence of LH/CGR mRNA, its transcript variants and functional LH/CGR protein in the monkey CL on day 1 of menses. Moreover, the functionality of receptors was tested by confirming the biological response of the CL to bolus administration of exogenous LH preparations, which eventually suggested factor(s) downstream of LH/CGR activation to account for the decline in CL function observed during non-fertile cycle. Studies have been conducted to identify molecular modulators that would selectively exploit intraluteal processes to regulate trophic signaling pathways that are critical to the control of luteal function. Immunoblot and qPCR analyses were carried out to examine presence and activation of Src family of kinases (SFKs) and cAMP-phosphodiesterases (PDEs) during various functional states of CL. The results revealed an increased activation of Src (phosphorylated at Tyr 416) during spontaneous and PGF2α/CET-induced luteolysis that may participate in the regulation of cAMP levels in part by increasing the cAMP-PDE activity observed during spontaneous luteolysis. This observation raised the question on the possible mechanism by which CG, an analog of pituitary LH, rescues CL function during early pregnancy. Thus, subsequent experiments involving LH/hCG administration in CET-treated animals as well as simulated early pregnancy animal model were conducted and the results revealed that, a bolus of LH/hCG decreased Src activation and cAMP-PDE activity accompanying a momentous increase in cAMP levels in both these models that further led to a concomitant increase in P4 secretion. Although the mechanisms of action of LH/CG involve modulation of a number of signaling pathways in the CL, by far, the results from various experiments suggested that it leads to activation of Src kinase and cAMP-PDE, thus causing inhibition of various elements of the primary signaling cascade- AC/cAMP/PKA/CREB during spontaneous luteolysis. One of the consequences of activation of Src kinase and cAMP-PDE was the regulation of expression of genes associated with steroidogenesis and it was observed that expression of SR-B1, a membrane receptor associated with trafficking of HDL-CE into the luteal cells, was lower in the regressed CL. The results taken together suggest that the decrease in responsiveness of CL to LH milieu during non-fertile cycles is not associated with changes in LH/CGR dynamics, but, is instead coupled to the activation of Src kinase and cAMP-PDE, inhibition of molecules downstream of LH signaling, and a decrease in the SR-B1 expression that regulates cholesterol economy of the luteal cell, and in turn, P4 secretion. The control of primate CL function appears to be dominated by the luteotrophic factors (LH/CG) over the luteolytic factors, since the process of luteal regression was overcome by administration of LH/CG. Further, in the primate CL, the molecular modulators of LH/CG signaling (Src kinase and PDE) are maintained in the repressed state by the luteotrophic factor LH/CG for maximum steroidogenic function. In contrast, in non-primate species, without invoking a role for the luteotrophic factor, essentially the synthesis and secretion of luteolytic factor, PGF2α, from the uterus is kept in check during pregnancy by the trophoblast derived IFN- and thus allowing CL to continue to function that is essential for maintenance of pregnancy. In the bovine species, the mechanism of PGF2α-induced luteolysis that involves a change in expression of genes associated with various processes of cellular function is poorly understood. Experiments were conducted utilizing buffalo cows (Bubalus bubalis) as a model system, to determine temporal changes in the global gene expression profile of the CL in response to PGF2α treatment. For this purpose, CL tissues were collected on day 11 of estrous cycle without treatment (designated as 0 h) and at 3, 6 and 18 h post PGF2α treatment for various analyses. Global changes in gene expression pattern in the CL were investigated employing Affymetrix GeneChip bovine genome array and the results are presented in Chapter III. The hybridization intensity values obtained by microarray analysis were subjected to R/Bioconductor tool. Following the application of highly stringent statistical filters to eliminate false positives, a set of differentially expressed genes were identified. The differentially expressed genes were further classified based on a fold change cut-off filter of ≥2, and the analysis revealed 127 genes to be differentially expressed within 3 h of PGF2α administration, of these 64 and 63 genes were up-regulated and down-regulated, respectively. Analysis of microarray data at 6 h post PGF2α administration revealed 774 genes to be differentially expressed, of which 544 genes were up-regulated, while 230 genes were down-regulated. The microarray analysis performed on CL tissues collected at 18 h post PGF2α administration showed that out of the total 939 differentially expressed genes, 571 genes were up-regulated, while 368 genes were down-regulated. Analysis of the ontology report for the biological processes category showed that initially in response to PGF2α administration, genes regulating steroidogenesis, cell survival and transcription were differentially regulated in the CL, but at later time points, differential expression of genes involved in apoptosis, PGF2α metabolism, tissue remodeling and angiogenesis was observed. Further, involvement of molecules downstream of LH/IGF-1 activation was investigated and the results obtained indicated that PGF2α interfered with the LH/IGF-1 signaling since the expression of LH/CGR, GHR and pAkt were down-regulated following PGF2αadministration. Furthermore, the functional luteolysis observed post PGF2αadministration appeared to be due to an interruption in cholesterol trafficking to inner mitochondrial membrane, since StAR expression was inhibited. The results obtained also demonstrated that the expression of AGTR1, VEGFR2 and R3 were down-regulated following PGF 2α administration. Further, the data obtained also suggested modulation of expression of pro- and anti-angiogenic factors upon PGF2α-treatment indicative of an involvement of other autocrine or paracrine factor(s) in the regression of bovine CL. This was an interesting finding as it suggests a novel and potential functional relationship between angiogenesis and the luteolytic response of CL to PGF2α administration. In bovines, despite extensive research being carried out to examine factors involved in the regulation of development and function of the CL, the trophic factor(s) required for maintenance of CL function, especially, P4 biosynthesis and secretion are not well characterized. It was hypothesized that the function of the CL during its finite lifespan must be responsive to LH as well as to various growth factors. Thus, experiments were conducted to examine the effects of increased LH and GH/IGF-I on the maintenance of CL function during mid luteal phase and post PGF2α administration and the results of these studies are presented in Chapter IV. To elucidate the role of LH as a trophic factor in the regulation of CL function, effects of increased endogenous LH through GnRH administration and exogenous hCG injections were examined. The results indicated an absence of noticeable effect of various hCG/GnRH treatments on circulating P4 levels. On the other hand, administration of GH resulted in increased serum IGF-1 and P4 levels. It was further observed that the administration of a combination of hCG and GH increased serum P4 levels better than treatment with GH alone. Further experiments were carried out to examine the complex reciprocal relationship between LH/GH and PGF2α on expression of genes involved in the regulation of luteal structure and function. In buffalo cows, administration of exogenous hCG and/or GH following inhibition of CL function by PGF2α administration did not prevent the PGF2α-induced decline in serum P4 levels, but PGF2-mediated decrease in expression of LH/CGR and GHR genes was prevented upon GH administration. However, the decrease in StAR expression was not restored by hCG and GH treatments, thereby indicating that PGF2 action was not prevented by hCG and/or GH treatments. Taken together, the results of studies carried out in buffalo cows employing various experimental model systems suggest essential role for LH and GH/IGF-1, however, these factors were unable to reverse PGF2α-induced luteolysis. Further, our crucial findings of the effects of increased endogenous LH and IGF-1, in addition to their relationship with luteolytic agents such as PGF2α will open new avenues for studying the mechanisms involved in the regulation of structural and functional properties of the buffalo CL. It is well known that a large number of buffalo cows experience loss of pregnancy and infertility due to inadequate luteal function and/or failure of timely insemination. Results from our studies suggest that the incorporation of PGF2α and hCG or GH/IGF-1 protocols in buffalo cows to be beneficial for improving their breeding efficiency as these protocols are likely to increase luteal function with defined luteolysis. To summarize, the results of studies described in the present thesis provide new insights into the physiological and molecular mechanisms involved in the regulation of CL function during luteolysis in the monoovulatory species. The results suggest that the maintenance of CL function appears to be dependent on both luteotrophic and luteolytic factors, but with a varied degree of dominance between the two species examined. Further, the results indicate that while the luteotrophic factors (LH/CG) dominate the CL regulation in primates, the regulation of CL function in bovines is dominated by the actions of luteolytic factor (PGF2α). In monoovulatory species, the luteotrophic and luteolytic factors following binding to their specific plasma membrane receptors on the luteal cells, would counteract each other and modulate activation of various downstream signaling molecules subsequently leading to regulation of gene expression and P4 secretion (Fig.5.1). Surprisingly, regulation of apoptotic gene expression and CL regression are still unclear. In bovines, PGF2α of uterine origin mediates changes in luteal gene expression and results in decreased P4 secretion, principally by reduction in StAR level. The present study suggests that during luteolysis PGF2α affects the genes regulated by LH, by interfering with LH (and perhaps IGF-1) signaling leading to alteration in the expression of genes crucial for CL structure and function. Trauma and recovery in Janet Frame’s fiction

Focusing on four novels by Janet Frame in dialogue with texts by Freud, Zizek, Lacan, and Silverman, my project theorizes trauma as the basis for both an ethical and an interpretive practice. Frame's fiction develops a cultural psychology, showing how the factors of narcissistic fantasy and the incapacity to mourn contribute to physical and epistemic aggression committed along divides of ethnicity, gender, and linguistic mode of expression. Employing trauma as a figure for an absolute limit to what can be remembered or known, I suggest that reconciliation with whatever is inaccessible, lacking, or dead within an individual or collective self fosters a non-violent relation with others. I begin by querying the place of "catharsis" within hermeneutic literary interpretation, focusing on the construction of Frame within the New Zealand literary industry. With Erlene's adamantine silence at its centre, Scented Gardens for the Blind (1964) rejects the hermeneutic endeavour, exemplified by Patrick Evans' critical work on Frame, to make a text "speak" its secrets. My readings of Intensive Care (1910) and The Adaptable Man (1965) address inter-generational repetitions of violence as the consequences of the failure to recognise and work through the devastations of war. The masculine fantasy of totality driving the Human Delineation project in Intensive Care has a linguistic corollary in Colin Monk's pursuit of the Platonic ideality of algebra, set against Milly's "degraded" punning writing. In The Adaptable Man, the arrival of electricity ushers in a new perceptual regime that would obliterate any "shadow" of dialectical negativity or internal difference. The thesis ends with a swing toward conciliation and emotional growth. The homosexual relationship depicted in Daughter Buffalo (1972) offers a model of transference, defined as a transitional, productive form of repetition that opens Talbot to his ethnic and familial inheritance. The intra- and inter-population relatedness of bovine tuberculosis-infected and -uninfected African buffaloes (Syncerus caffer caffer) in the Kruger National Park

The African buffalo (Syncerus caffer) is a member of one of Africa’s most well known tourist attractions and unique grouping of mammals – the ‘big five’. Historical records indicate that during the 19th century approximately 3 million African buffaloes inhabited almost the whole of sub-Saharan Africa. Several factors such as disease, habitat fragmentation, over-hunting and drought reduced the buffalo population to approximately 400 000 by 1990. The African buffalo is host to a variety of sub-acute diseases, such as bovine tuberculosis (BTB), foot-and-mouth disease (FMD) and corridor disease (CD). Disease is an important factor which influenced African buffalo populations throughout the continent and more specifically the Kruger National Park (KNP) and is largely responsible for the fact that buffaloes are restricted to enclosed areas with strict regulations imposed on their movement. The social organization of animals influences the distribution and spread of a disease - especially in the case of the African buffalo in the KNP. The emergence of BTB in the largest conservation area in South Africa (the KNP), threatens wild and domestic animals and humans who are in close proximity to the Park. The potential economic losses associated with this disease are excessive. The results presented in this thesis provide baseline information into the genetic status of sampled African buffaloes in the KNP, genetic relatedness between sampled individuals as well as BTB associations between sampled African buffaloes in the KNP, based on a limited dataset of 181 animals. Twelve microsatellite markers were used to evaluate 181 samples which were collected from 39 locations dispersed throughout the KNP. Specific population genetic parameters revealed information based on the intra and inter - relationships at the ‘per population’ level as well as at the ‘per prevalence group’ level. Evidence indicates a medium to high level of genetic diversity, a low to medium level of inbreeding (inbreeding coefficient (Fis) for each group ranges between 0.143 and 0.147) and a relatively high level of migration for buffaloes associated with each prevalence group. Pairwise relatedness estimates were determined between individuals, to reveal their level of relatedness (unrelated, full siblings, parent-offspring or half siblings), based on Queller and Goodnight’s (1989) coefficient of relatedness. Relatedness was determined on different levels, intra and interpopulation level, BTB infected and BTB uninfected group level as well as prevalence group levels. Evaluation of data based on these different levels and between different groups, painted an overall picture of the disease condition and genetic relatedness within and between sampled BTB infected and BTB uninfected buffaloes. Evidence indicated that the greater majority of our sampled African buffaloes (BTB infected or uninfected), were genetically unrelated (in terms of sibling and parent-offspring relationships), irrespective of their disease status. Johannes Gelbke (1846 Radeberg – 1903 Buffalo/ USA) „Unter den vielen tüchtigen Musikern, welche die Stadt Buffalo beherbergt, ist Herr Johannes Gelbke sicherlich der allertüchtigsten einer.“ So beginnt in dem Buch „Die Geschichte der Deutschen in Buffalo und Erie County, N.Y., mit Biografien und Illustrationen hervorragender Deutsch-Amerikaner, welche zur Entwickelung der Stadt Buffalo beigetragen haben“ der Beitrag über Johannes Gelbke. Geboren und aufgewachsen in Radeberg, erhielt er seine musikalische Ausbildung am „Gymnasium zum Heiligen Kreuz Dresden“ (Kreuzschule), seine ersten Kompositionen entstanden dort. Er wird Musiklehrer in Leipzig und wirkt als Sänger, Chorleiter und Gesangsvereins-Dirigent, ab 1869 auch in Wurzen. 1882 komponierte er sein sog. Heimatlied: „Horch, die alten Eichen rauschen“ (Text: Emil Schimpke). Im November 1882 wanderte Gelbke nach Buffalo / USA aus, wo er bis zu seinem Tode 1903 als Dirigent, Komponist und Chorleiter gewaltiger Chöre arbeitete. Nach Deutschland ist er nie zurückgekehrt.
