Using of PCR-DGGE Technique to Analyze the Microbial Diversity in Biofiltration System of Water Treatment PlantShiu, Chih-ping 23 August 2007 (has links)
This study investigated the microbiota in ten different drinking water treatment pools, particles in the Biological Activated Carbon Filtration (BACF) bed, and two mimic columns in the Cheng-Ching Lake Water Treatment Plant. Assimilable organic carbon (AOC) is one of the main nutrition sources for microbes to survive in tap water. Over growing microbes not only decrease the water quality, but also contaminate the water treatment system and distribution system. In this study, we used two molecular biology techniques, the polymerase chain reaction (PCR) and denaturing gradient gel electrophoresis (DGGE), to analyze the dynamic microbial communities and biodiversities in the drinking water cleaning system and the micorbiota that exist in the BAC and anthracite filtration pellets. The bacterial 16S rDNA sequences resulted from PCR-DGGE were compared with the data in the Ribosomal Database Project Bank to construct a phylogenetic tree which allowed us to understand the microbial communities and biodiversities in the drinking water treatment pools and the filtration pellets. The total bacterial count and PCR-DGGE profiles showed that the drinking water quality had been improved during the treating processes and most of the microbes in raw water were removed. The scanning electron microscopy clearly indicated the biofilms were developed on the pellet surface. From the mimic column studies, the PCR-DGGE profiles suggested that various microbial communities were present on different depth of the columns samples. In comparing the 16S rDNA sequences with Gene Bank, many are new category bacteria were found and most of them are unculturable. Most of these microbes belong to the beta-proteobacterium. Although many bacteria were located on the surface of the filtration pellet, the BAC and anthracite could still absorb AOC efficiently to enhance the bacteria growth. The over growing bacteria might release out and contaminate the drinking water. Therefore, we suggest that it is important to backwash the filter bed frequently in order to diminish microbes of the filtration pellet and avoid re-contaminate the drinking water.

Through comparative and qualitative research method, this dissertation first analyzes the individual values in political freedom and equality emphasized by modern Western individualism, will further discuss collective consciousness in the form of emotional and moral achievements as emphasized by traditional Chinese Confucianism as comparative reference basis and finally lists common grounds and differences between these two. This dissertation researches political thought and philosophy, mainly aiming at the political objectives of argument foundations, essential connotations and achievements in traditional Chinese political thought, illuminates collective sentiment thinking and will analyze and explain the reasons for a lack of Western individualism in traditional Chinese political thought, caused by the environmental conditions created by that thinking. The author hopes to be able to offer a comparison of nature and characteristics of Chinese and Western culture and thinking and a few answers and explanations regarding the problems the Chinese society is facing since it encountered and started to learn from Western democratic political systems more than 100 years ago.

Besides changing from hunting to farming technology has greatly improved the lives of local residents, a large number of Han Chinese Immigration has also developing the continental culture; trade demand from the Netherlands, Taiwan is seen at the world stage, also the first time for Taiwan to accept the baptism of different cultures; Prior to this, Japan desires Taiwan for a long time, but their moves and policies end up giving up invasive intentions. While Spain for the sake of trade and dissemination of cultural purposes to compete with the Netherlands, Taiwan has already evolved into East Asia as the most important geopolitical position.
The Zengs¡¦ developments in Taiwan, showing they play essential role in the history of Taiwan. As establishment of the first regime, it is absolutely a glorious event. Moreover, its implementation in the military, economics, political, internal affairs and diplomacy, until the present Taiwan still had far-reaching impact. The cultural monument and place names can be powerful proof.
Zeng and Qing¡¦s proposals can reflect the status quo in Taiwan and the failure of Zeng¡¦s regime can be a good lesson for leaders in Taiwan. If Taiwan can not get lessons from history, the tragedy will be inevitable. The ultimate solutions can be extracted from the lesson. Facing the rising power of China, the Republic of China undergo severe political environment, which is comparable to Zeng¡¦s condition. The focus of this study hope to provide different points of view to review the past and develop a future module.

The ritual context of morality books : a case-study of a Taiwanese spirit-writing cultClart, Philip Arthur 05 1900 (has links)
The present study focusses on the description and analysis of the
religious beliefs and practices of a central Taiwanese spirit-writing cult or
"phoenix hall" (luantang). A phoenix hall is a voluntary religious association of congregational character centring upon communication with the gods by means of the divinatory technique of "spirit-writing" (fuluan). While spirit-writing can be and is used as an oracle for the solving of
believers' personal problems, its more high-profile application is for the writing of so-called "morality books" (shanshu), i.e., books of religious instruction and moral exhortation. Spirit-writing cults are nowadays the most important sources of such works. Much attention has been given to morality books as mirrors of the social concerns of their times, but comparatively little work has been done on the groups that produce them and the meaning these
works have for them. An adequate understanding of the meanings and
functions of morality books, however, is impossible without some knowledge of the religious groups that produce them and the role played by morality books in their beliefs and practices. It is the objective of this thesis to provide a detailed description and analysis of one such group, the "Temple of the Martial Sage, Hall of Enlightened Orthodoxy" (Wumiao Mingzheng Tang), a phoenix hall in the city of Taizhong that was founded in 1976 and has played a significant role in the modern development of the shanshu genre through the active and varied publications programme of its publishing arm, the Phoenix Friend Magazine Society. The study utilizes data
extracted from the Hall's published writings as well as interview, observation,
and questionnaire data collected during an eight month period of field research in Taizhong. Part I provides a macrohistorical overview of the development of
spirit-writing cults on the Chinese mainland (chapter 1) and on Taiwan (chapter 2) since the nineteenth century, leading up to the case-example's microhistory (chapter 3). Part II is devoted to an account of the beliefs and practices of the Wumiao Mingzheng Tang, including descriptions and
analyses of its organization, deities, ritual activities, concepts of moral
cultivation, and of the body of morality book literature it has produced over
the years. The appendix contains samples of the cult's morality book and scriptural literature, as well as of various liturgical texts.

Chinese Idioms (Cheng-yu) for Second-Language Learners: Toward a Pedogogical ApproachStellard, Lisa 06 1900 (has links)
x, 56 p. / This thesis considers the importance of cheng-yu (Chinese language-specific, idiomatic lexical-syntactic units) in second-language acquisition at the intermediate and advanced levels and examines current research in order to formulate an instructional approach. Informed by linguistic and pedagogical research on idioms generally, the study aims to apply research relevant to Chinese language idioms specifically. Main topics to be considered include: the importance of cheng-yu in higher-level discourse including the social significance of cheng-yu use; the patterned structure of cheng-yu; idiom teaching methods and a review of idiom content and presentation in commonly-used Chinese language textbooks; and the significance of corpus data in cheng-yu instruction. Results confirm the importance of cheng-yu in discourse and recommend practicable, research-based classroom strategies.
