Polysaccharide fractions obtained from the red alga Gracilaria intermedia: a structural chemical study, evaluation of antioxidant activity and rheological characterization / FraÃÃes polissacarÃdicas obtidas a partir da alga vermelha gracilaria intermedia: um estudo quÃmico estrutural, avaliaÃÃo de atividade antioxidante e caracterizaÃÃo reolÃgica

CoordenaÃÃo de AperfeiÃoamento de Pessoal de NÃvel Superior / The seaweed Gracilaria are important sources of agar sulfated polysaccharides that are commonly used in the food, pharmaceutical and biotechnology industries. For characterization of the media, the kelp was subjected to analysis of their chemical composition, which indicated their percentages of moisture, ash, protein and lipids. Extractions in which extraction by enzymatic digestion native extract (a seaweed is not subjected to pretreatments and obtaining the polysaccharides is by their boiling) is: For evaluation of red seaweed polysaccharides Gracilaria intermedia fractions were obtained for the following extraction methods seaweed is pretreated with NaOH and KOH in concentrations of 1% and 5%. The extracted material was subjected to colorimetric test chemical composition (protein content determination and quantification of carbohydrates, sulfate and 3,6-anhydro-galactose). The material was further analyzed by infrared spectroscopy with Fourier transform and Nuclear Magnetic Resonance ( NMR). Antioxidant activity with each of the fractions obtained tests were performed. Antioxidants tests (total antioxidant capacity, chelation of ferrous ion) indicated that such polysaccharides have the ability to scavenge free radicals and significant dose-dependent manner. Due to these results, the polysaccharide obtained from seaweed Gracilaria show potential intermediate that they will not be used as a synthetic antioxidant compound. Experiments to evaluate the rheological behavior of aqueous solutions of polysaccharides under study were performed at 25  1 ÂC, using stationary and dynamic oscillatory measurements, which indicated that the material obtained should be classified as pseudoplastic and highlighted the skills of such polymer solutions in acting as thickeners and gelling agents. / Algas marinhas do gÃnero Gracilaria sÃo importantes fontes de Ãgar, polissacarÃdeos sulfatados que sÃo comumente usados nas indÃstrias alimentÃcia, farmacÃutica e biotecnolÃgica. Para caracterizaÃÃo do material de trabalho, a alga marinha foi submetida à anÃlise de sua composiÃÃo centesimal, a qual indicou seus percentuais de umidade, cinzas, proteÃnas, lipÃdios e carboidratos totais. Para avaliaÃÃo dos polissacarÃdeos da alga vermelha Gracilaria intermedia foram obtidas fraÃÃes pelas seguintes metodologias de extraÃÃo: 1. extraÃÃo por meio de digestÃo enzimÃtica, 2. extraÃÃo nativa (a alga nÃo à submetida à prÃ-tratamentos e a obtenÃÃo dos polissacarÃdeos ocorre pela sua fervura) e 3. extraÃÃes nas quais a alga à prÃ-tratada com NaOH e KOH nas concentraÃÃes de 1% e 5%. Os materiais extraÃdos foram submetidos a testes colorimÃtricos de composiÃÃo quÃmica (determinaÃÃo do teor de proteÃnas e quantificaÃÃo de carboidratos, sulfato e 3,6 anidrogalactose). Os materiais foram ainda analisados por espectroscopia de infravermelho com transformada de Fourier e RessonÃncia MagnÃtica Nuclear (RMN). Foram realizados testes de atividade antioxidante com cada uma das fraÃÃes obtidas. Os testes antioxidantes realizados (capacidade antioxidante total e quelaÃÃo do Ãon ferroso) indicaram que tais polissacarÃdeos possuem capacidade de sequestrar radicais livres de maneira significativa e dose-dependente. Devido a estes resultados, os polissacarÃdeos obtidos a partir da alga marinha Gracilaria intermedia mostram potencial de virem a ser utilizados como um composto antioxidante nÃo sintÃtico. Experimentos para avaliar o comportamento reolÃgico de soluÃÃes aquosas dos polissacarÃdeos em estudo foram realizados a 25  1 ÂC, usando medidas estacionÃrias e dinÃmicas oscilatÃrias, que indicaram que os materiais obtidos devem ser classificados como pseudoplÃsticos e apontaram as habilidades de tais soluÃÃes polimÃricas em atuar como agentes espessantes e gelificantes.
