THE SPATIALITY IN STORYTELLING

Theatre has always been played a irreplaceable role in people’s lives, even nowadays where people have multiple choices for entertainment. Some theater architecture has also become the symbol of the city, such as Paris Opéra and Sydney Opera House. By taking a close look at various case studies, one will understand how the theatre architecture corresponds with their city representing its history, culture and visions for the future. The development of my thesis is based on the integration of the ‘space’ of storytelling and the space of design. Will the quality of space bring out the memories that have been stored with in it? How the space provide with most flexibility for its users? Explorations in acoustic and lighting design as well as the spectators’ experience in the space will be discussed. My attention is to create a design that will heighten perception and arouse the audience’s sensory experience even before the start of the show. The relationship between nature and the architecture is also one of my attentions. Questions arise such as how to use a multidisciplinary approach to investigate the intersection of architecture and landscape. Should the architecture become a new definition towards the context or an expression that abstract from the landscape? This thesis explores the relationship between the performer and audience, and how the architecture space can contribute to the theater experience. The design for this project offers one solution for the whole site serving as a place for performing arts as well as a public space and a destination for the city of Boston.

Analysis and Improvement of Cross-dock Operations in Less-than-Truckload Freight Transportaion Industry

The less-than-truckload (LTL) transportation industry is highly competitive with low profit margins. Carriers in this industry strive to reduce costs and improve customer service to remain profitable. LTL carriers rely on a network of hubs and service centers to transfer freight. A hub is typically a cross docking terminal in which shipments from inbound trailers are unloaded and reassigned and consolidated onto outbound trailers going to the correct destinations. Freight handling in a hub is labor intensive, and workers must quickly transfer freight during a short time period to support customer service levels. Reducing shipment transfer time in hubs offers the opportunity to reduce labor costs, improve customer service, and increase competitive advantages for carriers. This research focuses on improving the efficiency of hub operations in order to decrease the handling costs and increase service levels for LTL carriers. Specifically, the following two decision problems are investigated: (1) assigning trailers to dock doors to minimize the total time required to transfer shipments from inbound trailers to destination trailers and (2) sequencing unloading and loading of freight to minimize the time required by dock employees. The trailer-to-door assignment problem is modeled as a Quadratic Assignment Problem (QAP). Both semi-permanent and dynamic layouts for the trailer-to-door assignment problem are evaluated. Improvement based heuristics, including pair-wise exchange, simulated annealing, and genetic algorithms, are explored to solve the trailer-to-door assignment problem. The freight sequencing problem is modeled as a Rural Postman Problem (RPP). A Balance and Connect Algorithm (BCA) and an Assign First and Route Second Algorithm (AFRSA) are investigated and compared to Balanced Trailer-at-a-Time (BTAAT), Balanced Trailer-at-a-Time with Offloading (BTAATWO), and Nearest Neighbor (NN). The heuristics are evaluated using data from two LTL carriers. For these data sets, both the total travel distance and the transfer time of hub operations are reduced using a dynamic layout with efficient freight sequencing approaches, such as the Balance and Connect Algorithm (BCA), the Balanced Trailer-at-a-Time with Offloading (BTAATWO), and the Nearest Neighbor (NN). Specifically, with a dynamic layout, the BCA reduces travel distance by 10% to 27% over BTAAT and reduces the transfer time by 17% to 68% over BTAAT. A simulation study is also conducted for hub operations in a dynamic and stochastic environment. The solutions from the door assignment and freight sequencing approaches are evaluated in a simulation model to determine their effectiveness in this environment. The simulation results further demonstrate that the performance measures of hub operations are improved using a dynamic layout and efficient freight sequencing approaches. The main contributions of this research are the integer programming models developed for the freight sequencing problem (FSP), based on the Rural Postman Problem (RPP). This is the first known application of the RPP for the FSP. Efficient heuristics are developed for the FSP for a single worker and for multiple workers. These heuristics are analyzed and compared to previous research using industry data. / Ph. D.

Caractérisation structurale et biophysique de Elmo1 et des interactions avec son partenaire

Les protéines eucaryotes Elmo (Engulfment and Cell Motility) forment une famille de régulateurs conservés qui jouent un rôle central dans les processus biologiques reposant sur le remodelage du cytosquelette d'actine comme la phagocytose et la migration cellulaire et régulé par les GTPases de la famille Rho. Les protéines Elmo régulent la fonction des protéines Dock (Downstream of Crk), qui sont des Facteurs d'Echange de Guanine (GEF) atypiques pour les GTPases Rac1 et Cdc42. Le mécanisme de régulation connu à ce jour, repose sur l'interaction entre les 200 résidus C-terminaux d'Elmo et les 180 premiers résidus N-terminaux de Dock. Cependant, le rôle précis des différents domaines et motifs identifiés dans ces régions n'est pas encore défini. En effet, les données fonctionnelles, biochimiques et structurales rapportées à ce jour semblent contradictoires quant à la contribution de l'extrémité C-terminale d'Elmo qui comprend un motif polyproline et le domaine SH3 N-terminal de Dock. Nous avons donc étudié la contribution de l'extrémité C-terminale de Elmo1 à l'interaction entre Elmo1 et le domaine SH3 de Dock1 en utilisant notamment la résonance plasmonique de surface. Nos données démontrent la capacité du domaine SH3 de Dock1 à interagir avec Elmo1, indépendamment de l'extrémité C-terminale contenant le motif polyproline. Toutefois, la présence de cette région conduit à une augmentation significative du temps de demi-vie du complexe Elmo1/Dock1. En parallèle, des expériences de diffusion des rayons X aux petits angles nous ont permis de déterminer des enveloppes tridimensionnelles de la protéine Elmo1. Ces données nous permettent ainsi de proposer un premier modèle à basse résolution dans lequel nous localisons les parties N et C-terminales de Elmo1. De façon surprenante cette étude semble indiquer un changement de conformation de la région N-terminale de Elmo1 ainsi qu'une interaction possible de cette même région avec le domaine SH3 de Dock1.

Tem sido demonstrado que na hipertensão arterial ocorre aumento de ouabaína plasmática, um inibidor da Na+K+-ATPase. Em 1998, Ferrari et al. desenvolveram uma molécula denominada de rostafuroxina, a qual é capaz de antagonizar os efeitos da ouabaína por deslocar a ligação desse glicosídeo com a Na+K+-ATPase. Dentro desse contexto, parece razoável sugerir que um anti-hipertensivo capaz de antagonizar os efeitos da ouabaína possa representar uma nova ferramenta farmacológica para o tratamento da hipertensão arterial. Baseado em tais premissas, o presente estudo avaliou o papel da ouabaína por meio do tratamento com rostafuroxina sobre: a pressão arterial, a reatividade vascular em artérias de resistência e a atividade simpática de ratos DOCA-sal. Os resultados demonstraram que os animais DOCA-sal tratados com rostafuroxina apresentaram redução da pressão arterial sistólica e da hiperatividade simpática e melhora na função vascular. Assim, sugere-se que a ouabaína seja um possível alvo para o tratamento da hipertensão arterial dependente de volume. / It has been shown that some types of hypertension have increased plasma levels of ouabain, a factor inhibitor of Na+K+-ATPase. In 1998, Ferrari et al. developed a molecule called rostafuroxin that antagonizes the effects of ouabain. In this context, it seems reasonable to suggest that an antihypertensive capable of antagonizing the effects of ouabain might be a new and specific pharmacological tool for the treatment of hypertension. In so doing, the present study aimed to evaluate the role of endogenous ouabain by treatment with rostafuroxin on blood pressure, vascular reactivity in resistance arteries and sympathetic activity of DOCA-salt rats. Our data have shown that the treatment with rostafuroxin decreased the systolic blood pressure and sympathetic activity and improve the vascular function of the DOCA-salt rats. Thus, it is suggested that ouabain is a putative target for the treatment of volume-dependent hypertension.

O objetivo deste trabalho é apresentar as formas adquiridas pela cultura do trabalho portuário avulso após o processo de modernização, ocorrido em 1993, ano de promulgação da Lei nº 8.630, que rege sobre a reforma portuária brasileira. O caso estudado é o Porto de Santos. O estudo levanta a hipótese que com a regulamentação da atividade sindical e a vinculação do trabalho portuário avulso ao sindicato (closed shop), fato ocorrido nos anos 1930, esta cultura passa a basear-se na figura da entidade sindical. O processo de regulação econômica e reforma do estado brasileiro, iniciado no Governo Fernando Collor de Mello, acaba com o closed shop e vincula o trabalho portuário a um órgão gestor de mão-de-obra. A lei aparece como marco, pois determina o fim do sistema de closed shop. Para análise da formação desta cultura utiliza-se a noção de experiência e o conceito de consciência de classe de E. P. Thompson e o conceito de habitus de Pierre Bourdieu. / The objective of this work is to present the forms acquired for the culture of the dock work after the process of modernization, occurrence in 1993, year of promulgation of the Law nº 8.630, that it prevails on the Brazilian port reform. The studied case is the Port of Santos. The study it raises the hypothesis that with the regulation of the syndical activity and the entailing of the dock work to the union (closed shop), fact occurred in years 1930, this culture starts to base in the figure of the labor union. The process of economic regulation and reform of the Brazilian state, initiate in the Government Fernando Collor de Mello, finishes with closed shop and ties the port work with a managing agency of man power. The law appears as landmark, therefore determines the end of the closed-shop system. For analysis of the formation of this culture it is used experience notion and the concept of class conscience of E.P. Thompson and the concept of habitus of Pierre Bourdieu.

Distintos controles tecnológicos, metodologias de dosagem e características dos materiais, são alguns dos fatores que fazem com que o concreto tenha peculiaridades relacionadas à região do planeta onde o mesmo é produzido.O presente trabalho aplicou diversos métodos de controle e aceitação de estruturas de concreto aos resultados práticos oriundos do concreto lançado na execução do dique seco do Estaleiro Rio Grande em construção na cidade de Rio Grande - RS. Esta obra é uma importante fonte de informações sobre a resistência e o comportamento do concreto visto que, na mesma, foi empregado desde o início um rigoroso controle de qualidade; gerando dados raramente disponíveis em obras portuárias brasileiras. Foram utilizados como base para este trabalho dois traços de concreto; um com adição de fibras e fck= 35 MPa e outro com adição de sílica ativa e fck= 40 MPa. Foram realizadas comparações entre as estimativas de resistência das normas brasileira, européia e norte-americana de concreto. Realizadas análises estatísticas de modelos teóricos de distribuição de probabilidade destas resistências. Além disso, foram estabelecidas correlações entre as resistências à tração e à compressão do concreto. O incremento de resistência do concreto ao longo do tempo também foi estudado, tanto para compressão como para a tração. As análises apresentadas permitem atestar o atendimento dos concretos utilizados na execução do dique seco quanto aos requisitos estabelecidos pelas normas brasileiras de concreto. / Due to the variability in the results of the concrete acceptance tests, especially in the resistance tests, it is fundamental to use a large number of sample data to establish the assessment parameters and to calibrate the standard codes. Different technological controls, mixing methodologies and material characteristics are some of the factors that cause the concrete having peculiarities proper of the region where it is produced. The present work has applied different methodsof control and acceptance of concrete structures to the test results of the concreteused in the construction of the Estaleiro Rio Grande dry-dock, in the city of Rio Grande – RS. This construction site is an important source of information about the concrete resistance and behavior, because it was adopted, since the beginning of the construction, a rigorous quality control. This quality control produced a number oftest results that is seldom available in the Brazilian harbors construction sites. In this work two different concrete mixes were studied: first a fiber reinforced concrete (FRC) with fck = 35 MPa, and a second one with silica fume addition with fck = 40 MPa. The resistance assessment formulations of the Brazilian, European and North American standard codes were compared. A statistical analysis of best the probability distribution models for the resistance test results was made. Besides, it was determined a correlation between the concrete tensile and compressive resistances. The increasing of the concreteresistance with time was also studied to compression as well as to tension. The analyses performed have showed that both concrete mixes used in the construction of this dry-dock have attained the quality standards required by the Brazilian standard codes for concrete.

Utbyggnad av Gröna Lund : Ett förslag till utformning av Skeppsholmsviken 6 / Extension of Gröna Lund : A proposal for formation of Skeppsholmsviken 6

Syfte: Syftet med arbetet är att undersöka och gestalta utbyggnaden av Gröna Lund inom fastigheten Skeppsholmsviken 6 med hänsyn till funktion, upplevelse, kulturhistoriska värden, hållbarhet och allmänhetens intressen. Metod: Intervjuer genomfördes med personer som har varit inblandade i det pågående planarbetet och en enkätundersökning har delats på sociala medier för att ta reda på allmänhetens synpunkter och önskemål kring utbyggnaden. Utöver detta har detaljplaner, rapporter och ritningar från det pågående planarbetet utgjort del av faktainsamlingen inför arbetet. Resultat: Arbetet resulterade i fem byggnader placerade längs vattenlinjen mot Saltsjön och ett allmänt gångstråk i form av en brygga som ska kunna användas som passage förbi Skeppsholmsviken 6. Slutsats: Resultatet visar att en utbyggnad av Gröna Lund har stark kulturhistorisk förankring och att platsen i utbyggt utseende kommer kunna nyttjas bättre av både allmänheten och Gröna Lunds besökare. Utbyggnaden kommer även medföra positiva ekologiska, sociala och ekonomiska konsekvenser som minskad biltrafik, ökad trygghet, fler jobbmöjligheter och stärkt turistnäring. Det är möjligt att göra utbyggnaden ekologiskt hållbar genom att använda trä som byggnadsmaterial, grundlägga bryggan på pålar och höja bryggdäcket en bit över vattnet för att få ett större ljusinsläpp. / Aim: The aim of the thesis is to investigate and design the extension of Gröna Lund within the property of Skeppsholmsviken 6 in regards to the factors that affected the formation. Methods: Interviews were conducted with people involved in the current project. A survey was administered on social media to explore public opinion and requisition tied to the extension work. Furthermore, an audit of detailed plans, reports and drawings was carried through. Results: The constructions resulted in five buildings placed along the coastline facing Saltsjön and a public board walk in terms of a dock which should be accessible for passage through Skeppsholmsviken 6. Conclusions: The results show that the extension of Gröna Lund has a strong cultural and historical attachment and will benefit the public and visitors of Gröna Lund. Additionally, the extension will have positive ecological, social and economic consequences such as increased safety, tourism, job opportunities and reduced traffic. The extension can be made ecologically sustainable by using wood as a building material, build it on piles and raising the dock a bit over the water surface to get a larger light transmission.
