The Ph.D. thesis deals with one of the most important characteristics of hardened concrete; i.e. the modulus of elasticity. The thesis aims to describe and evaluate the important factors that affect the final value of concrete modulus of elasticity, especially those that can be influenced during construction. Another aim is to find ways to determine the static modulus of elasticity of concrete by non-destructive means (primarily ultrasonic pulse method, resonance method and use of electronic rebound hammers), thus to determine the possibility of creating calibration curve for NDT determination of the modulus of elasticity.

Dynamický dekodér pro rozpoznávání řeči / Dynamic Decoder for Speech Recognition

The result of this work is a fully working and significantly optimized implementation of a dynamic decoder. This decoder is based on dynamic recognition network generation and decoding by a modified version of the Token Passing algorithm. The implemented solution provides very similar results to the original static decoder from BSCORE (API of Phonexia company). Compared to BSCORE this implementation offers significant reduction of memory usage. This makes use of more complex language models possible. It also facilitates integration the speech recognition to some mobile devices or dynamic adding of new words to the system.

This thesis deals with inspection and constructionally technical research of reinforced concrete prefabricated construction from the second half of 20th century. In theoretical part, the research of literature, documents and valid standards is performed, with a focus on production and construction of prefabricated elements and also on the metodology of research. In practical part, the detection of reinforcement of individual elements of the construction and concrete properties is performed. The result of this work is the assessment of the state of the construction, static calculation of the ceiling panel and idea proposal for eventual reconstruction.

Charakterizace adsorbčních vlastností probiotických bakterií / Characterization of adsorption properties of probiotic bacteria

The aim of the diploma thesis is to characterize the adsorption properties of probiotic bacteria. Characterization was performed using of designed and optimized spectrophotometric method of determination of adhesion of probiotic bacteria to mucin as the main viscoelastic component of mucus. In order to provide a more detailed description of the adhesion interactions involved in the adhesion of probiotic bacteria to mucin, the viability of selected probiotic bacterial strains Lactobacillus rhamnosus CCM 1825, Lactobacillus plantarum CCM 7039, Lactobacillus delbrueckii subsp. bulgaricus CCM 7190, Lactobacillus acidophilus CCM 4833, Lactobacillus casei CCM 4798, Bifidobacterium breve CCM 7825, Bacillus coagulans CCM 2658 and a potential probiotic strain of Lactobacillus zeae CCM 7069. Moreover, bacterial adhesion to carbohydrates was also determined for all the bacteria tested. The last proposed and optimized technique was dynamic and electrophoretic light scattering characterizing surface properties such as -potential, average size distribution and isoelectric point. From this method, information was obtained on aggregation of bacterial cells and on electrostatic interactions. The combination of these methods was used as a complex tool to characterize adhesion of the tested bacterial cultures as a very specific, sensitive and key parameter of a successful probiotic microorganism that is influence by the multiple effects.

Dynamický částicový systém jako účinný nástroj pro statistické vzorkování / A dynamical particle system as a driver for optimal statistical sampling

The presented doctoral thesis aims at development a new efficient tool for optimization of uniformity of point samples. One of use-cases of these point sets is the usage as optimized sets of integration points in statistical analyses of computer models using Monte Carlo type integration. It is well known that the pursuit of uniformly distributed sets of integration points is the only possible way of decreasing the error of estimation of an integral over an unknown function. The tasks of the work concern a survey of currently used criteria for evaluation and/or optimization of uniformity of point sets. A critical evaluation of their properties is presented, leading to suggestions towards improvements in spatial and statistical uniformity of resulting samples. A refined variant of the general formulation of the phi optimization criterion has been derived by incorporating the periodically repeated design domain along with a scale-independent behavior of the criterion. Based on a notion of a physical analogy between a set of sampling points and a dynamical system of mutually repelling particles, a hyper-dimensional N-body system has been selected to be the driver of the developed optimization tool. Because the simulation of such a dynamical system is known to be a computationally intensive task, an efficient solution using the massively parallel GPGPU platform Nvidia CUDA has been developed. An intensive study of properties of this complex architecture turned out as necessary to fully exploit the possible solution speedup.

The diploma thesis deals with the determination of the influence of size, shape and type of test specimen on values of modulus of elasticity of light-weight concrete. A lot of different specimens were prepared from two concrete’s mixtures for the experiment. Tests for measurement of static modulus of elasticity and dynamic modulus of elasticity by ultrasonic impulse velocity method were made. The results were assessed and summarized in tabular and graphical form.

The Diploma thesis is focus on polymer modified bitumen for warm mix asphalt. The theoretical part of the thesis describes additives which influence the properties of bitumen for their use and reduction of production and service temperatures. Further, the theoretical part describes the conclusions from the researches. This part also describes empirical and functional tests of bituminous binders. The practical part presents the results of individual tests. Tests were performed on samples of non-aged, aged by RTFOT and RTFOT + PAV methods. The results of the work and the assessment of the suitability of bituminous binders are summarized at the end of the thesis.

This diploma thesis deals with the preparation and characterization of stealth liposomes and their combination with trimethylchitosan (TMC). This complex could find application in the field of inhalation administration. Stealth liposomes were prepared from neutral phophatidylcholine, negatively charged fosfatidic acid and polyethyleneglycol bounded to phosphatidylethanolamine. We have managed to prepare stealth liposomes with suitable properties that should guarantee passive targeting without evocation an immune response, despite the content of the negative component. We also found a suitable method of preparation for stealth liposome–TMC complex, where the change of size and zeta potential confirmed the non–covalent bound between two components despite the content of the polyethyleneglycol.

Generace kovových nanočástic v nízkoteplotním plazmatu v kapalině / Generation of metallic nanoparticles by non-thermal plasma in liquids

This thesis focuses on the process of nanoparticle generation using new source of nonthermal plasma combining corona and pin-hole discharge in liquids. The theoretical part is focused on generation of metallic nanoparticles using various types of plasma discharge, the properties of metallic nanoparticles, their preparation by other methods and methods of characterization of nanoparticles. The experimental part deals with the preparation of copper, silver and gold nanoparticles from solutions of their precursors. The influence of experimental conditions, such as the influence of voltage polarity, effect of precursor concentration, effect of added electrolyte or reducing agent were investigated. All samples were analyzed by UV-VIS spectroscopy. Dynamic light scattering was used to determine the sice of nanoparticles. To confirm the presence of nanoparticles, samples were analyzed using scanning microscope with and energy dispersion spectrometer for elemental analysis.
