Evaluation of some pseudomonas species isolated from Hogsback forest reserve for the production of antibacterial compoundsMkono, Yonela Pelokazi January 2017 (has links)
Pseudomonas species are Gram-negative bacteria most abundant in soil and water bodies, with the capacity to thrive in varied environments. They are largely associated with resistant pathogenic bacteria linked to human and plant diseases. Species such as Pseudomonas aeruginosa have been particularly targeted as case studies due to the extremity to which they pose a threat to human health. With more focus directed at using these species for biocontrol and bioremediation purposes, their role in bioactive compound production may be equally important. As the crisis on antimicrobial resistance still persists, the need for effective antimicrobial compounds is ever more urgent and solutions may possibly still be dormant in bacterial species whose potential has not been fully investigated. On a bid to source out potential antimicrobial compound producers, soil samples were collected from Hogback forest reserve in the province of the Eastern Cape, South Africa. For bacterial screening, M1 and R2A agar were used and the cultures grown at 37˚C for a period of seven days. After the presumed Pseudomonas species were identified, antimicrobial production was determined by submerged fermentation method using nutrient broth as media of choice. Active isolates were further studied to determine the optimum conditions which best facilitate for antimicrobial compound production, with parameters such as temperature (25˚C – 40˚C) and pH (4 – 9) considered. The role plasmids play in antimicrobial compound production was also investigated. Each isolate was grown in fermentation media containing Sodium dodecyl sulphate and Ethedium Bromide, at varying concentrations, to facilitate for plasmid curing. With each sample, distinct colonies were identified with varying pigmentations most dominant being a cream colour. The identity of the isolated strains was achieved through sequencing of 16S rDNA. Phylogenetic analysis showed that isolate A16 had 80 percent homology with Pseudomonas plecoglossicida strain P4 and share a close ancestor with isolates Y52 and Y81, also isolate Y89 showed a 90 percent homology with Pseudomonas sp. Co-11a. With the exception of isolate A16, the isolates which were active against Gram-negative bacteria lost activity as the screening processes continued. When looking at temperature variations, isolates Y81 and A16 were highly active with maximum activity observed at 35˚C while Y89 performed best at 25˚C and Y52 showed constant activity across all studied temperatures. The plasmids in all isolates were found to be 48.5 kb in size with the exception of isolate Y89 which was 20 kb. The plasmids were cured at concentrations of (1 mg/ml; 5 mg/ml; 7 mg/ml; 10 mg/ml; 11 mg/ml) SDS and (125 μg/ml; 6.5 μg/ml; 5μg/ml) EtBr. The curing process also showed changes in both the antimicrobial activity of the isolates as well as their physical characteristics. The isolates are the first reported Pseudomonas species from Hogsback forest reserve with the ability to produce antimicrobial compounds which are active against Gram-positive and Gram-negative bacteria. These mesophilic bacteria also show that plasmids do not pay any role in the production of antimicrobial compounds and that the biosynthesis genes are highly likely to be chromosomal borne meaning that the production cannot be linked to horizontal transfer of genes. Therefore, these isolated Pseudomonas species provide a potential reservoir of antimicrobial compounds which may play an important role in the antimicrobial resistance phenomenon.

The study explored professional nurses' experience of working in a remote rural hospital in the Eastern Cape Province. A qualitative phenomenological design was used and the study was conducted in a remote rural hospital in region `D'. Eight professional nurses were selected non-randomly from a population of professional nurses who had been working in the hospital for more than one year. Purposive sampling was used to select the participants and semi-structured phenomenological interviews were conducted to collect data. Colaizzi's eight-step method was used for data analysis. The themes that emerged from the data analysis were shortage of human and material resources, poor access, communication problems and lack of safety and insecurity. The study found that the professional nurses at the remote rural hospital experience many obstacles to quality service delivery. / Health Studies / M.A. (Health Studies)

The growing demand for governments and organisations to demonstrate principles of accountability, transparency and participatory decision making, which are the cornerstones of good governance, has fostered renewed efforts to transform the public service to ultimately produce tangible results and meet the needs of the beneficiaries of service delivery. Effective performance management practised across the entire organization in an integrated, iterative and sustained way that can help public service organizations overcome the challenges and deliver more with less. This study focuses on the assessment of monitoring and evaluation as the control measure to enhance organisational performance with the reference to the Eastern Cape Provincial Treasury and assess critical role in supporting performance management and also provides methodological options to support performance management. Monitoring and Evaluation (M&E) are management activities that are necessary to ensure the achievement of policy goals in the form of concrete results. The study revealed that the current M and E system is not effective in that there are no clearly defined standards of what a quarterly progress report entail, especially the portfolio of evidence and because the main problem stems from the definition of what constitutes departmental performance. In determining what needs to be done, the effectiveness suffers since we opt for mainly indicators that are “safe”, that the department are sure they can achieve and not necessarily those that will enhance organizational performance. The study revealed that each programme manager should monitor and evaluate his/her processes on a continuous basis without having to wait for M&E unit. M&E unit will then provide an objective evaluation of the process already in place and constant engagement between M&E unit and line management, feedback sessions between staff and M&E unit are essential about the achievement of targets as outlined in the APP and Operational Plan. The study further revealed that there should be continuous meetings to discuss how directorates should sufficiently report on their quarterly targets to avoid inadequate reported information and the department must go step by step in performing activities towards achieving a set vision and on way to the vision, not forgetting to take away risks that may take us to another direction that was not intended as per our vision. The study thus recommends that, inter alia, the department must introduce strong mechanisms to deal with non-implementation of corrective measures regarding under performance and the poor quality of information provided in performance reports related to the performance indicators in the Annual Performance Plans.The introduction of an early warning sign system to warn all programmes when the performance of their planned indicators are not be achieved. The Top Management should continue to use the performance management information as the tool to improve the organisational performance by linking and aligning individual, team and organisational objectives and results, it also provides a means to recognise and reward good performance and to manage under-performance. However the Top Management should be able to conduct in-depth analysis into underperformance to detect whether the underperformance is related to the bad crafting of the indicators or if it relates to the work not done. The department should invest money on training of officials in M&E because it plays a critical role supporting performance management at various levels, in that it contributes to a thinking that is results oriented and also provides methodological options to support performance management. This means that all officials must be capacitated to manage M&E systems in the department that will ensure the production of accurate, objective and reliable information. The M&E unit in the department must facilitate such skills development and provide reasonable technical support where required. The training could be designed to instill a deeper understanding and knowledge of the concepts of M&E and their correct application in order to avoid misunderstandings and misinterpretations which could hinder the achievement of the desired results.

The approaches of traditional healers in the treatment of HIV/ AIDS: the case of Chris Hani District Municipality, Cala, Tsengiwe, Eastern cape, South AfricaMati, Similo January 2017 (has links)
In South Africa, just like in any other country within the African continent, traditional healing remains an integral part of many communities and this is not just restricted to the rural communities only, as is sometimes assumed. The main aim of this research was to explore the approaches of traditional healers in the treatment of HIV/AIDS in the Chris Hani District Municipality, Eastern Cape. The following research objectives were followed regarding the approaches of traditional healers in the treatment of HIV/AIDS: (i) to assess how traditional healers and people living with HIV/AIDS in Tsengiwe village understand HIV/AIDS, (ii) to investigate the reasons people living with HIV/AIDS consult traditional healers in Tsengiwe village, (iii) to assess Tsengiwe village traditional healers’ treatment strategies for HIV/AIDS, (iv) to establish how traditional healers view their role in the treatment of HIV/AIDS in Tsengiwe village. A qualitative research design was utilized, using in-depth interviews with traditional healers and focus group discussions with caregivers and people living with HIV/AIDS respectively. A type of non-probability sampling known as purposive sampling was used. A total of sixteen (16) participants were interviewed. The findings in this research revealed the following themes: (i) HIV/AIDS is incurable and it is understood by symptoms, (ii) belief system entrenched in traditional healing, (iii) cleansing rituals and traditional medicinal remedies and, (iv) strengthening relations between stakeholders. While traditional healers expressed a willingness to work with biomedical professionals in the management of HIV/AIDS, caregivers and people living with HIV/AIDS preferred going to clinics and hospitals for treatment. Furthermore, people living with HIV/AIDS in this research never admitted to consulting traditional healers for their ailments, only saying that they choose to self-medicate.

On the implementation effectiveness and efficiency of ecological interventions in operational contexts : the case of Working for WaterMcConnachie, Matthew Morgan January 2013 (has links)
There is little understanding of the implementation efficiency and effectiveness of restoring plant invaded landscapes within operational contexts. South Africa's Working for Water (WfW) programme is arguably the most ambitious alien plant control programme in the world, yet little is known about its cost-effectiveness and the challenges it faces in linking poverty and environmental objectives. My first aim was to assess the cost-effectiveness of invasive plant removal, and the factors that underpin its effectiveness over large spatial and temporal scales. The second aim was to compare the accuracy of evidence-based findings with managers' experience-based beliefs, and to assess whether managers are willing to change their beliefs after being exposed to it. The third aim was to assess the costs and benefits of removal versus removal followed by active native re-vegetation. My final aim is to assess the challenges and lessons learnt by managers linking ecological restoration with poverty alleviation objectives, specifically within the public works model. My study area was focused primarily on two WfW river catchment projects in the western region of the Eastern Cape province. I adopted an interdisciplinary approach drawing from a range of methods such as observational studies, statistical modelling and interviews with managers. The key findings were that control efforts in the two catchment projects are largely inadequate owing to many sites being re-invaded and not enough resources being allocated to the catchments. It would take between 54 and 695 years to clear the respective catchments. In terms of cost-effectiveness, my results exceeded previous estimates by 1.5 to 8.6 times for each catchment project. After being exposed to the evidence-based findings, the managers did not change their beliefs when it came to forecasting the future effectiveness. I found that active native re-vegetation after removal of invasive plants is very costly and that priority should be given to understanding the effectiveness of the removal treatments on native species recovery. The managers cited significant challenges in effectively and efficiently meeting the programmes dual objectives. Based on a broader review of the public works literature I recommend WfW re-examine the type of public works they currently use.

The contribution of agriculture involved livestock and crop production. Wages and government grants were other livelihood sectors that were looked into. This study was important in determining the change in livelihood strategies in the last decade and to quantify the NTFPs used at the two sites. It was carried out in Guquka and Koloni, both part of the central Eastern Cape. Information on direct use value of the NTFPs used, the quantities and local price; crop production outputs and inputs and the costs. Data were collected via a questionnaire.

The contribution of agriculture to rural development in Mbashe MunicipalityMalusi, Phaphama Paul January 2017 (has links)
Despite the decreasing contribution of agriculture to Gross Domestic Product (GDP) in South Africa, agricultural activities are critical in the development of rural households. These activities are an important route through which these rural communities can reduce poverty. This is mainly because agriculture has been the main source of income and employment in rural areas in South Africa and other less developed countries (LDC). However, it has not generated sufficient income to sustain rural households. Rural areas are characterised by factors such as low socio-economic conditions, low standards of living, lack of access to markets and inadequate infrastructure that does not create an enabling environment for private sector involvement. The private sector is not actively involved or engaged in rural development. Rural development involves improvement in the economic security of people in rural areas. It is poor households that look for employment in agriculture. The poor are portrayed in terms of qualifications, employment, access to resources, basic health services, skills possessed, self- sustaining activities, and income and expenditure patterns. Many individuals in rural areas are pushed into non-agricultural activities due to lack of opportunities in agricultural activities. Lack of opportunities in agriculture can be caused by subsistence agriculture in small holdings of land and lack of appropriate skills sets for certain agricultural activities. Ultimately, it is poor individuals who increase their dependence on agriculture. The primary objective of this research was to obtain a better understanding of the contribution of agriculture in rural development in the Mbhashe Local Municipality (MLM). Furthermore, the necessary conditions required to sustain livelihoods in the rural areas were identified. This was mainly done in an attempt to improve the quality of life in the rural areas. The study was based on three objectives. The first objective was to identify the constraints to agricultural activities in the Mbashe Local Municipality; the second was to investigate the plight of rural people in the Mbhashe Local Municipality. Lastly, the third objective was to investigate the contribution of agriculture in rural development in the study area. The main finding with regards to the first objective is that facors such as lack of infrastructure, low socio-economic conditions, lack of appropriate skills and lack of opportunities in agriculture are some of the constraints to agricultural activities. Investments in basic infrastructure need to be significant so that funds can flow into the rural areas to be invested in livelihood activities in order to create more employment opportunities. With regard to the second objective, the main finding is that rural people need to be empowered through increased knowledge and skills. This involves improving literacy and skills training, because the outcome of this training is that they are able to stand on their own feet and provide for their families. The main finding with regard to the third objective is that rural areas, by nature, are not easy to service but have indigenous knowledge that can be used in agriculture. However, the government must support them with the necessary infrastructure. Agriculture has been a major source of income and employment in the Mbashe Municipality.

Local economic development and poverty alleviation in Nkonkobe Local MunicipalityTsomo, Zixolele January 2016 (has links)
The purpose of this study was to investigate the impact of Local Economic Development (LED) in alleviating poverty in Nkonkobe Local Municipality in the Eastern Cape Province, a municipality that is predominantly rural in nature and also stricken by poverty. The research objectives were; to explore the implementation of LED strategies, to assess the degree to which various spheres of government work collaboratively to enhance efficiency and effectiveness in development planning; and to evaluate the extent to which job opportunities have been created as a result of LED practices in the municipality. To achieve these objectives, a qualitative research approach was utilised. The sample was selected using a purposive sampling technique whereby individuals who are directly involved in integrated development planning and LED planning were targeted. Those who met this criterion were considered suitable for providing relevant information regarding the implementation of LED in Nkonkobe Local Municipality. Based on the literature review presented in chapter 2, LED can be perceived as an integrated, multidisciplinary approach that seeks to alleviate poverty through pro-poor economic growth by promoting economic development initiatives in municipalities. Moreover, the legislative framework presented in the same chapter reveals that the South African government has employed various strategies in the implementation of LED in local communities. The study showed that the Nkonkobe Local Municipality LED strategy has limited impact on poverty alleviation due to a myriad of factors which reinforce and interact with each other, thereby limiting development and thus trapping residents in poverty. The findings suggest that there is significant room for a paradigm shift from predominantly pro-growth LED to pro-poor LED, and the need to adopt a comprehensive LED strategy that seeks to include both pro-growth and pro-poor strategies.

Exploring the management-union relationship in an Eastern Cape public sector departmentMkalipi, Nosivatho Getrude January 2017 (has links)
This study explored the relationship between management and the union in an Eastern Cape Public Sector Department. The study employed both qualitative and quantitative research tools to collect information from the respondents, who gave a view on their experiences of what the relationship is, and how it could be improved. The sample consisted of members of management and union executives, across employment levels. Both statistical and thematic analysis were used to analyse data from questionnaires and semi-structured interviews. The literature review defined the concept of trade unionism, management and trade union relationship in the workplace, and different frames of reference of the labour relations system. From the literature review it is noted that management and union relationships are more of a formal arrangement in the workplace; as such government, has developed tools to assist in this regard. Although that is not the case in the Department it is noted that the informal arrangement in the relationship between management and union is ‘working’ to some extent. Quality and sustainability of that, however, is questionable. The findings indicate that most members of management acknowledge the existence of the relationship between management and the union, but they are dissatisfied, as most members are not part of the engagements with the union. They are also of the view that it is in favour of the union. Unions on the other hand, view the existing relationship as working in favour of one union instead of the union collective. The union which finds favour is satisfied, and the other is not. It is recommended that the management and union engagement be formalized in order for the Department to work with unions in a more structured, professional manner, which would be inclusive of other members of management who feel left out. Not only that, but management would also be able to engage with the union as a collective, and not as an individual union.

The transition period post-apartheid in South Africa since 1994 brought a number of changes in policy making process and the initiated programs by the government in all level with the aim of addressing inequality and poverty reduction strategies. In South Africa prior 1994 division in terms of location access to resources and information was meant for the certain group and participation in the Community development matters. The aim of this research study is to seek and find out the impact of the government policy projects, programme (strategies) in the community to poverty reduction and try to identify the reasons for the failure of other project and to what extend is the capacity building in community members in terms of skill development before the implementation of projects for sustainability and viability of those projects. Also is to explore the community participation or involvement in policy making process such as the formulation of Integrated Development Plan .The study attempts to answer community involvement into their own development, their role and impact to fight poverty and other socio-economic challenges that are faced by the community. The research design for the study could be classified as the mixed design which will include qualitative and quantitative approaches. A sample of 30 participants will be drawn from the community unemployed respondents, project employees, those searching for employment, 5 municipality officials from Blue Crane Route, Ward Councillor and projects leaders. On my findings community participation on community development issues must be strengthen for the sake of development .The issue of centralisation of offices for availability of resources to same of the communities makes it difficult to access resources. Lack of skills and training acquired in the job market for artisans, technical and management skills.
