In-situ analýza složení záporné elektrody olověných sekundárních článků s využití rentgenového difraktometru / Using of X-ray for distribution analysis of electrodes for secondary cell application

This diploma thesis is focused on in-situ analysis of the composition of the negative electrodes of second secondary cells with using X-ray radiation. The first part is dedicated to the theory of lead accumulators, including charging, discharging, battery components and individual degradation mechanisms. The main part of the thesis includes the preparation of electrochemical in situ XRD cells and following measurement of the growth of lead sulphate crystals on the OCV potential. Furthermore, the lead cell was subjected to the measurement of the cyclic voltammetry around the working point of the negative electrodes. From these measurements was evident gradual increase of the lead sulfate crystals. The analysis was supplemented with electron microscopy images. In addition, were evaluated the in-situ particle analysis of the LTO Sigma sample.

This work deals with problems of lithium-ion batteries. In the theoretical part are described electrochemical sources in general and their division. The main representatives of individual types of electrochemical sources are described in more detail. In the next part the thesis focuses specifically on lithium-ion accumulators, their history, electrochemical reactions taking place inside and materials of individual parts of which the accumulator consists. Next part focuses on electron microscopy and its division into scanning and transmission. Basic parts and the principle of operation are described. The practical part is focused on creating a small cell for examination under an electron microscope.

Využití tuhých elektrod na bázi uhlíku k elektrochemické oxidaci acikloviru / Utilization of carbon-based solid electrodes to electrochemical oxidation of acyclovir

Electrochemical oxidation of acyclovir at glassy-carbone electrode and boron-doped diamond electrode gives one pH-dependent oxidation signal; the corresponding electrode reaction is controlled by diffusion. It is followed by a second indistinctive signal in the pH range 2,0 - 9,0. Further, optimization of conditions was carried out for determination of acyclovir using DC and DP voltammetry. The calibration dependence on GC electrode in B−R buffer (pH = 6,0) is linear only in the concentration range from 2 to 10 μmol∙l−1 for DCV with limit of detection 0,38 μmol∙l−1; for DPV it is not linear. BDD electrode has to be activated to prevent its passivation. Anodic activation at +2,4 V for 15 s with simultaneous stirring of solution results in relative standard deviation of 0,9% for acyclovir concentration of 1∙10−4 mol∙l−1. Using DCV and DPV methods linear concentration dependences were obtained in 0,1 mol∙l-1 nitric acid and in pH 6,0 B−R buffer. The lowest limit of detection and limit of quantification (LOD = 0,47 μmol∙l−1 a LOQ = 1,55 μmol∙l−1) and linear range from 0,6 to 100 μmol∙l−1 was reached in pH 6,0 B−R buffer. The aim of this diploma thesis was to develop an electrochemical method for three phytosterols, namely stigmasterol β-sitosterol and ergosterol using voltammetry techniques on glassy carbon electrode and boron doped diamond electrode. The work builds on previous results, where it was shown that the electrochemical oxidation of phytosterols requires perchloric acid in acetonitrile. When this acid is used, a dehydration reaction occurs. Calibration dependences were measured here for all phytosterols in an environment with a water content of 0.43%. The stability of the signal with increasing distilled water content was further investigated. Then the calibration dependences in the increased water content were measured again. If we want to use HPLC-type separation techniques, we must first find out how the phytosterols will behave in this environment, and therefore the signal stability on the buffer content was investigated. The buffers used were phosphate and acetate buffer. Subsequently, the concentration dependence was measured in an environment with a higher content of phosphate buffer. Stanovení propylgallátu pomocí uhlíkové pastové elektrody / Determination of propyl gallate on carbon paste electrode

Propyl gallate (PG) is a significant synthetic antioxidant and preservative. Its determination has been studied at carbon paste electrode (CPE) using differential pulse voltammetry (DPV) and high performance liquid chromatography with electrochemical detection (HPLC-ED) and with UV spectrometric detection (HPLC-UV). Concentration dependences were measured in the media of Britton-Robinson buffer (pH 5) and methanol (20 %, v/v) by DPV and the limit of detection 0,6110-7 moldm-3 was obtained. Using HPLC with a mobile phase consisting of 0,01 moldm-3 phosphate buffer (pH 4) and methanol (50 %, v/v) with potential of working electrode E = +0,8 V and detection wavelength λ = 280 nm, concentration dependences were measured. Limit of detection was determined to 0,39 moldm-3 for HPLC-ED and 4,95 moldm-3 for HPLC-UV. After verification of the extraction procedure PG was determined in vegetable oil. The resulting value of 3,2 mgkg-1 corresponds with permited limits.

Využití bórem dopované diamantové filmové elektrody k voltametrické a ampérometrické detekci aminobifenylů / The utitilization of boron-doped diamond thin film electrode for the voltammetric and amperometric determination of amino derivatives of biphenyl

The aim to this work was to develop methods for the determination of 2 aminobiphenyl (2-AB), 3 aminobiphenyl (3-AB), and 4-aminobiphenyl (4-AB) in model mixtures. Concretely, the direct determination of the mixture of studied analytes has been tested using spectrophotometry and differential pulse voltammetry (DPV). Furthermore, separation and detection of 2-AB, 3-AB, and 4-AB have been performed using high performance liquid chromatography with electrochemical detection (HPLC-ED) with boron-doped diamond film thin electrode (BDDFE) in ,,wall-jet" configuration and using high performance liquid chromatography with UV detection (HPLC-UVD). It was found out that the spectrophotometric determination of 2-AB, 3-AB, and 4-AB is impossible in their mixture because of nearby values of local absorption maxima wavelengths of all three analytes studied. Upon the determination of 2-AB, 3-AB, and 4-AB in their mixture using DPV in BR buffer pH 2.0, the difference in peak potentials of 2-AB and 3-AB is too low for their determination in mixture. Upon the determination of mixture containing 2-AB and 4-AB in BR buffer pH 12.0, the limits of determination (LDs) were obtained in the concentration order of 10-6 mol.l-1 for 2-AB and 10-7 mol.l-1 for 4-AB. The entry part introduce into the problematic of rework processes by molding injection tools for thermoplastic. Aim is necessity of change current rework process usage by electro discharge machining (EDM) technology.The theoretical part of the thesis describes the physical essence of EDM technology, the system of production of cutting electrodes and related issues. The practical part of work compare the process of production of copper and graphite electrodes with focus on production time and costs. The production parameters of the EDM process were analyzed and the wear of copper and graphite electrodes during the production process was compared. Main result of the thesis is an evaluation of the research activity, interpretation of the results of this activity and a proposal of measures to shorten the production time of the cutting electrodes and the time of the EDM machining by these electrodes.

Mikroskopická pozorování struktur aktivní hmoty záporné elektrody olověného akumulátoru / Microscopic Observations of Active Mass Structures of the Negative Electrode of a Lead-Acid Battery

This doctoral dissertation deals with the study of the effect of additives on the properties of the negative electrode of a lead-acid battery. The work focuses on the investigation of the additive/base material interface for the negative electrode (so-called active mass) using atomic force microscopy (AFM). A lead sheet was used as the base material, on which the given additive was applied. The effects of carbon, TiO2, glass fibers and Indulin AT were specifically investigated. The negative electrode was cycled by a potentiostat in a defined manner and the morphology of the electrode surface was continuously scanned by AFM. This created a time sequence of images on which events on the surface of the electrode during its charging/discharging were captured. These records serve to better understand the effect of additives on the resulting properties of constructed lead-acid cells, specifically on the ability to form or dissolve PbSO4 crystals, which are formed during cell discharge and are responsible for the decrease in total capacity - gradual surface sulfation.

Thesis title: Evaluation of effectiveness of using myoelectric prosthesis by people with congenital or acquired amputation of the upper limb Aims: The aim of my thesis is to evaluate the effectiveness of myoelectric prosthesis using by people with upper limb amputation. Find out how effectively users are able to manage individual tasks with myoprosthesis and basic ADL activities. Methodology: The research group consisted of 6 people with upper limb amputation. In this thesis, there were used three methods of data collection. The first method that was applied was a survey, which was used to obtain anamnestic data by the research group. The second method of data collection was the observation of probands during the performance of individual tasks. The third chosen method was measurement by using a special test aimed at evaluation of the effectiveness while using and the involvement of myorpothesis in the locomotor chain against a healthy upper limb. Results: Based on observations and measurements, it was performed that all probands are able to functionally and effectively involve the myoprosthesis in the locomotor chain in the performance of individual tasks.
