Two experiments tested the efficacy of gonadotropin-releasing hormone
(GnRH) or estradiol cypionate (ECP) administration in combination with
controlled internal drug release (CIDR) inserts on pregnancy rate to fixed-time
artificial insemination (FTAI) in beef heifers. In the first experiment, heifers in the
two ECP groups received ECP in conjunction with the CIDR insert. The CIDR
insert was removed and PGF was administered on either d 7 or 9. Heifers
received ECP 24 h after CIDR removal and were inseminated 30 h after ECP.
Heifers in the two GnRH groups received GnRH in conjunction with the CIDR
insert. Heifers received PGF on either d 6 or 7, and the CIDR insert was
removed on d 7. Heifers were inseminated and received GnRH 48 h after CIDR
removal. Pregnancy rates to FTAI were higher for heifers treated with GnRH.
Pregnancy rate to FTAI was highest for heifers in the GnRH group that received
PGF on d 6 and was lowest for the group that received ECP in combination with
a 7-d CIDR insert. In the second experiment, heifers received GnRH in conjunction with the
CIDR insert. CIDR inserts were removed and PGF was administered on d 7.
Heifers were inseminated 60 h later, with one group receiving GnRH at
insemination. Pregnancy rate to FTAI was higher for heifers that received GnRH
at insemination.
The final experiment tested the efficacy of estradiol 17_ (E17) or GnRH in
combination with a CIDR insert on pregnancy rate to FTAI in Brahman cows.
Either E17 or GnRH was administered in conjunction with the CIDR insert.
CIDR inserts were removed on d 7. Cows in the GnRH group received PGF on
d 6 and GnRH at insemination which occurred 48 h after CIDR removal. Cows
in the E17 group received PGF on d 7, E17 24 h later, and were inseminated 30
h after E17 administration. Pregnancy rate to FTAI was higher for cows treated
with E17.
These data indicate acceptable pregnancy rates were obtained when the
CIDR insert was combined with either GnRH in beef heifers or with E17 in
Brahman cows.

Factors affecting estrogen excretion in dairy heifersTucker, Heather Ashley 14 August 2009 (has links)
Two studies were conducted to assess factors affecting estrogen excretion in dairy heifers. The objective of the first study was to quantify estrogenic activity in feces and urine during the estrous cycle. Ten non-pregnant Holstein heifers were fed the same diet for 28 d. Plasma, feces, and urine samples were collected daily. Plasma 17-β estradiol (17-β E2) was quantified with RIA and used to confirm day of estrous. Feces and urine samples from days -12, -6, -2, -1, 0, 1, 2, 6, 12 of the estrous cycle were analyzed with RIA and Yeast Estrogen Screen (YES) bioassay. Plasma 17-β E2 concentrations peaked on day of estrus, with feces and urine estrogenic excretion peaking a day after. The objective of the second study was to quantify variation in estrogenic activity in feces and urine due to increased dietary phytoestrogen content. Ten Holstein heifers were randomly assigned to treatment sequence in a two-period crossover design. Dietary treatments consisted of grass or red clover hay, and necessary supplements. Feces and urine samples were collected and pooled for analysis. Estrogenic activities of pooled samples were quantified using the YES bioassay. Estrogenic excretion in feces and urine was higher for heifers fed red clover hay. Fecal and urine samples from five heifers were analyzed using LC/MS/MS to quantify excretion of phytoestrogenic compounds. Heifers fed red clover hay excreted more equol than heifers fed grass. Identifying sources of variation in estrogenic activity of manure will aid in the development of practices to reduce environmental estrogen accumulation. / Master of Science

Effect of delaying insemination in beef heifers not expressing estrus by 48 hours after a 7-d CO-Synch plus controlled internal drug release timed artificial insemination protocolAlexander, Dayna C. January 1900 (has links)

Grieger / Synchronizing estrus before AI is an effective way to shorten the breeding season, and increasing the number of pregnancies per AI may lead to greater use and acceptance of synchronization protocols among beef producers. Our objective was to determine if pregnancy rates to fixed-time AI (FTAI) would be improved by delaying insemination in heifers not expressing estrus before FTAI in a 7-d CO-Synch + controlled internal drug release (CIDR) estrus-synchronization protocol. In Experiment 1, yearling beef heifers (n = 465) at three locations of commercial and purebred herds were treated with GnRH (Cystorelin 100 µg im) and a CIDR insert (1.38 g of progesterone) on Day 0. On Day 7 CIDR inserts were removed and all heifers received PGF[subscript]2α (Lutalyse 25 mg im) and were fitted with an estrus-detection patch (Estrotect; Rockway, Inc.). Heifers were assigned to three treatments based on estrus-detection patch color at 48 h after PGF[subscript]2α: (1). Estrus-Red 48 h (Red 48; n = 180), heifers that expressed estrus and were inseminated at 48 h; (2). Non-Estrus-Gray 48 h (Gray 48; n = 137) heifers that did not express estrus and were inseminated at 48 h; and (3). Non-Estrus Delayed- 56 h (Gray 56; n = 148), heifers that did not express estrus at 48 h, and were not inseminated until 56 h after PGF[subscript]2α. Pregnancy rate to AI was greatest (P < 0.0001) for Red 48 heifers (67.8%) compared with heifers in the Gray 48 (39.4%) and Gray 56 (42.6%) treatments. Heifers assigned to Gray 48 and Gray 56 achieved similar (P = 0.83) pregnancy rates. In Experiment 2, yearling beef heifers (n = 257) at two different locations were treated with the same 7-d CO-Synch protocol, but heifers were assigned to three different treatments based on estrus-detection patch color at 48 h after PGF[subscript]2α: (1). Estrus-Red 48 h (Red 48; n =95), heifers that expressed estrus and were inseminated at 48 h; (2). Non-Estrus-Gray 48 h (Gray 48; n = 84), heifers that did not express estrus but were inseminated at 48 h; and (3). Non-Estrus Delayed- 72 h (Gray 72; n = 78), heifers that did not express estrus at 48 h, and were not inseminated until 72 h after PGF[subscript]2α. Pregnancy rate to AI was greatest (P = 0.004) for Red 48 heifers (62.1%) compared with heifers in Gray 48 (40.5%), and Gray 72 (46.2%). No difference in pregnancy rates (P = 0.75) was detected between heifers assigned to treatments Gray 48 and Gray 72. Delaying insemination in heifers not expressing estrus by 48 h after PGF[subscript]2α did not improve pregnancy rates to AI.

A series of studies were guided by the principal hypothesis that it was necessary to characterize ovarian function during the seasons of the annual reproductive cycle in wapiti and that from this knowledge novel methods of exogenous control of ovarian function would be possible. To augment existing knowledge about endogenous control of ovarian function in wapiti studies were conducted to characterize ovarian follicle dynamics during the estrous season and to characterize ovarian follicular dynamics during the periods of transition into and out of the breeding season. The third study was designed to characterize ovarian follicle development and ovulation synchrony subsequent to an estrous synchronization protocol used commercially. To evaluate novel methods of exogenous control of ovarian function studies were conducted to determine if follicular wave emergence could be electively induced using hormonal or surgical treatments to evaluate novel ovarian superstimulatory treatment protocols. <p>It was concluded from the studies of ovarian function that follicle development during the breeding season was characterized by the regular and synchronous development of follicular waves and that 2, 3, or 4 waves occurred during each interovulatory interval. Transition into the breeding season was preceded by one short interovulatory interval (9 days) characterized by one wave of follicle development and a small, short-lived and hypo-functional corpus luteum. The last estrous cycle of the breeding season was similar to estrous cycles during the rut (21 days), but and transition to anestrus was marked by a failure of the dominant follicle to ovulate after luteal regression. The treatment protocol used commercially for estrous synchronization was effective, but unnecessarily long. It was concluded from the studies on exogenous control of ovarian function that follicular wave emergence could be electively induced using steroid hormones or follicle ablation and may be useful for estrus synchronization and superstimulatory protocols. The tested superstimulatory treatments were effective and had the advantage of reducing the treatment period by 6 days and the number of times the animals are handled by one third over a more conventional method. However, oocyte and embryo quality were not evaluated. <p>As a result of the studies conducted and one previous study during the anouvlatory season follicle and luteal dynamics are now known in wapiti for all seasons of the year and this knowledge will provide a template upon which other species of deer can be compared. Artificial Induction of Lactation in Nonbreeder Dairy CowsJewell, Tracy Michelle 31 July 2003 (has links)
Thirty-four cows (26 Holsteins and 8 Jerseys) were subjected to an estrous synchronization protocol administering 2 PGF2Æ Ã injections 11 d apart prior to beginning the lactation-induction protocol. Artificial induction of lactation yielded a 92% success rate for Holstein cows with success defined as achieving >9 kg milk/d, and a 88% success rate for Jersey cows with success defined as achieving > 5 kg milk/d. Mean accumulated milk yield for induced cows at 150 DIM was 65% of mean yield for nontreated cows. Mean peak milk yield for lactation- induced Holsteins and Jerseys was 32 kg/d and 20 kg/d, respectively. Mean serum and milk progesterone concentrations for samples collected during the first 6 d of lactation were not different between lactation-induced and nontreated cows. However, mean serum estradiol concentrations for induced cows were higher (P <0.05) in samples collected 3 and 5 DIM. Lactation-induced cows exhibited an increase in serum alpha-lactalbumin concentrations 2 d prior to initiation of milking, reaching values of ~260 ng/ml. Mean days-to-first service was greatly reduced in cows induced into lactation compared to nontreated cows, while mean services per conception was similar between induced and nontreated cows. Mean days to conception was lower for induced cows than for nontreated cows. By 150 DIM, pregnancy rate of induced cows was 70%, whereas nontreated cows averaged 56% pregnancy rate. / Master of Science

A Comparison of Three Estrous Detection Management Schemes for Dairy HeifersLagerstedt, Ann 01 May 1990 (has links)
Heifers should represent the greatest genetic potential within a dairy herd. To maximize this potential, heifers must be inseminated to proven sires, a practice requiring management changes on many dairies. Holstein heifers (n=115) were allotted to one of three groups to compare alternatives to daily estrous detection that may facilitate the management of a program of artificial insemination for heifers. Group 1 heifers (Controls) received no treatment but were observed twice daily for signs of estrus. Group 2 heifers (2X-PGF) were synchronized with two injections of prostaglandin F2 alpha given 11 days apart and observed for estrus. Group 3 heifers (MGA+PGF) were synchronized with 9 days of melengestrol acetate feeding and an injection of prostaglandin F2 alpha 14 days after the last feeding and observed for estrus. Comparisons were made on the effectiveness of each treatment in estrous response and pregnancy rates and on the management requirements and economics associated with each program. Estrous response was significantly higher for the 2X-PGF group, while synchronized pregnancy and first-service conception rates did not differ. On an annual basis, the MGA+PGF group was calculated to require three-quarters of the labor input as compared to the 2X-PGF group and less than a fifth of the labor required for the Control group. A program of daily estrous detection was calculated to have an economic advantage over synchronization the frequency estrous synchronization programs was scheduled four times per year. was increased to six times per when When year, synchronization had a slight economic advantage over daily estrous detection when animal numbers were low and labor costs were high. The economic advantage of daily estrous detection is reduced when synchronization is performed more frequently throughout the year.

Comparison of short-term vs. long-term estrous synchronization protocols using CIDR devices in sheep and goats during and outside the natural breeding seasonHarl, Audra Whitney January 1900 (has links)

Grieger / Controlling reproductive cycles during active cyclicity and seasonal anestrous in small ruminants is critical for profitability. The objective of this study was to evaluate the effect on estrous response and interval to estrus of two CIDR protocols in sheep and goats during breeding and non-breeding seasons. In experiment 1, 133 ewes were randomly assigned to 1 of 3 treatments during the breeding season. In the CIDR-7 group, ewes received a CIDR insert for 7 d. In the CIDR-7 + PGF treatment, ewes received a CIDR insert for 7 d and 20 mg of prostaglandin-F[subscript]2[subscript]α (PGF[subscript]2[subscript]α) upon CIDR removal. Ewes in the CIDR-14 treatment received a CIDR insert for 14 d. Following CIDR removal all ewes were exposed to a ram every 12 h until breeding. There was a shorter interval from CIDR removal to estrus in the CIDR-14 treatment compared to the CIDR-7 and CIDR-7 + PGF treatments (P<0.05). There was no difference in number of ewes per treatment displaying estrus. In experiment 2, 54 ewes were randomly assigned to one of two treatment groups during the anestrous season. Ewes in CIDR-7 and CIDR-14 treatments received a CIDR insert for 7 d and 14 d, respectively. Upon CIDR removal ewes were exposed to a ram every 12 h until breeding. There was a significantly shorter interval from CIDR removal to estrus in CIDR-14 ewes when compared with CIDR-7 ewes (P<0.05). For experiment 3, 37 Boer does were randomly assigned to one of two treatments. In the CIDR-10 treatment, does received a CIDR insert for 10 d and 20 mg of PGF[subscript]2[subscript]α at time of CIDR removal. In the CIDR-19 treatment, does received a CIDR insert for 19 d. Upon CIDR removal, does were exposed to a buck fitted with a marking harness and chalk marks were recorded every 12 h. The number of does displaying estrus was not different (CIDR-7, 85%; CIDR-14, 95%). There was no difference in interval from CIDR removal to estrus between treatments. Results from experiments 1 and 2 supported the hypothesis that long-term protocols yield a shorter interval to estrus when compared with short-term protocols.

Para estudo de diferentes períodos de protocolos de indução do estro e de um novo dispositivo de progesterona para pequenos ruminantes (DPR - Tecnopec, Brasil) foram realizados dois experimentos em ovelhas da raça Texel durante a contraestação reprodutiva. Experimento 1: com o objetivo de comparar a eficiência de protocolos de indução do estro em períodos de nove versus doze dias e com a utilização de dispositivos novos versus usados através das taxas de gestação e manifestação de estro, foram utilizados 160 ovelhas Texel. Os animais foram divididos aleatoriamente em 4 grupos: protocolo de 9 dias com a utilização de um dispositivo feito em silicone novo (G9N, n=91), protocolo de 9 dias com a utilização de dispositivo usado (G9U, n=25), protocolo de 12 dias com a utilização de dispositivo novo (G12N, n=26) e protocolo de 12 dias com a utilização de dispositivo usado (G12U, n=18). Os dispositivos foram colocados no dia zero (D0) e retirados no dia nove (D9) nos grupos G9N e G9U e doze dias após (D12) nos grupos G12N e G12U. Junto com a aplicação de 0,125mg de cloprostenol (Estron, Agener, Brasil) e 500UI de eCG (Folligon®, Intervet, Netherland). Experimento 2: com o objetivo de comparar o Dispositivo Pequenos Ruminantes (DPR - Tecnopec, Brasil) versus esponja impregnada com medroxiprogesterona Progespon® (MSD, Brasil) no protocolo de 9 dias foram utilizadas 152 ovelhas Texel. Os animais foram divididos aleatoriamente em 2 grupos: protocolo de 9 dias com a utilização da esponja Progespon® (GP, n=61) e protocolo de 9 dias com a utilização do Dispositivo Pequenos Ruminantes (GDPR, n=91). O protocolo utilizado foi semelhante ao G9N do experimento 1 exceto pelo tipo de dispositivo utilizado. No experimento 1 e 2 as variáveis analisadas foram: taxas de manifestação de estro, concepção e prenhez e dispersão no intervalo de cio. O exame de ultrassonografia para detecção de prenhez (Chisson, KYLUMAX) foi realizado com 30 e 60 dias da retirada do dispositivo. O delineamento utilizado foi inteiramente casualizado e os dados analisados através do programa SAS System for Windows (SAS, 2000). Como resultados observou-se que não houve diferenças entre os protocolos de 9 e 12 dias, assim como entre os dois tipos de dispositivos em relação a taxa de manifestação de estro, concepção e prenhez. Houve maior taxa de manifestação de estro (P<0,05)) do dispositivo novo se comparado com o usado. Foi observado menor (P<0,05) dispersão na ocorrência de estro no protocolo de 9 dias utilizando o dispositivo novo. Conclui-se que os protocolos de 9 e 12 dias e ambos dispositivos são semelhantes quanto a eficácia de indução do estro em ovelhas Texel durante a contraestação reprodutiva e que o dispositivo novo apresenta maior taxa na manifestação de estro em comparação com o dispositivo usado. / Two experiments were conducted to study different periods of estrus induction and study of a new progesterone device for small ruminants (DPR - Tecnopec, Brazil) protocols in Texel ewes during the non-breeding season. Experiment 1: 160 Texel ewes were used in order to compare the efficiency of estrus induction protocols with the nine versus twelve days periods and the use of new versus used devices through the rates of estrus manifestation and pregnancy. The animals were randomly divided into 4 groups: 9 days protocol with the use of the new device (G9N, n = 91) and 9 days protocol with the use of used devices (G9U, n = 25) 12 days protocol with the use of the new device (G12N, n = 26) and 12 days protocol with use of used device (G12U, n = 18). The devices were placed on day zero (D0) and removed on day nine (D9) in the G9N and G9U groups and twelve days after (D12) in the groups G12U and G12N, together with the use of 0.125 mg of cloprostenol (Estron, Agener, Brazil) and 500 IU of eCG (Folligon®, Intervet, Netherland). Experiment 2: 152 Texel ewes were used in order to compare the Small Ruminants Device (DPR - Tecnopec, Brazil) versus Sponge impregnated with medroxyprogesterone Progespon® (MSD, Brazil) in the 9 days protocol. The animals were randomly divided into 2 groups: 9 days protocol using sponge Progespon® (GP, n = 61) and 9 days protocol using Small Ruminants Device (GDPR, n = 91). The protocol used was similar to that G9N of experiment 1 except by the type of device used. 312 sheep were used in the period of September to November 2012. The considered methodologies in experiment 1 were: rates of estrus manifestation, conception and pregnancy and dispersion in the heat interval. The ultrasound examination for detection of pregnancy (Chisson, KYLUMAX) was performed at 30 and 60 days after D9 and D12. The experimental design was completely randomized and analyzed using the SAS System for Windows (SAS, 2000) program. As results it was shown no differences between the protocols 9 and 12 days, as well as between the two types of devices in relation to the estrus manifestation, conception and pregnancy rate. There was a higher estrus manifestation rate (P <0.05) of the new device compared with the used one. It was observed lower (P <0.05) dispersion in the occurrence of estrus in the 9 days protocol. We conclude that the protocols 9 and 12 days and both devices are similar in efficacy to induce estrus in Texel ewes during the non-breeding season and the new device has the highest rate of estrus manifestation compared to the used device.
