O objetivo deste trabalho foi avaliar os parâmetros de estabilidade e adaptabilidade de oito linhagens elites de milho (seis utilizadas como fêmea e duas como macho em cruzamentos de híbridos simples comerciais e experimentais) em oito ambientes pelo modelo de EBERHART & RUSSELL. Os ambientes foram caracterizados por duas localidades, duas épocas de semeadura (setembro e outubro) e dois sistemas de manejo de solo (convencional e plantio direto). Utilizou-se o delineamento em blocos casualizados com quatro repetições. Avaliaramse os caracteres de produção de grãos (kg ha-1), rendimento de sacos de sessenta mil sementes por hectare, altura de plantas, altura de espiga e comprimento de espiga. Constataram-se efeitos significativos para as linhagens, ambientes e interação linhagens x ambiente para todas as características avaliadas. Observou-se que todas as linhagens tiveram comportamento imprevisível para produção de grãos. De uma forma geral, observou-se uma maior produção de grãos no ambiente de Rolândia, semeado em setembro no sistema de plantio direto. O ambiente que propiciou a maior produção de grãos, necessariamente não é o ambiente com maior rendimento de sacos de 60 mil sementes por hectare. Os ambientes de Barretos se mostraram propícios para aumentar os parâmetros de altura de planta e altura de espiga. The objective of this work was to evaluate the parameters of stability and adaptability of eight Elite inbred lines of maize (six used as female and two used as male in single crosses of commercial and experimental hybrids) in eight environments by EBERHART & RUSSELL’s method. The environments were characterized by two locations, two planting dates (September and October) and two systems of soil tillage (no till and tillage). The experimental design used was a complete randomized block with four replications. It was observed the characters of grain yield (kg ha-1), weight of thousands kernels, yield of sixty thousand kernels bag ha-1, plant height, ear height and ear length. It was evidenced significant effects for inbred lines, environments and inbred lines x environment interaction for all traits evaluated. It was observed that all inbred lines showed unstable behaviour. In general, there was a higher grain yield in environment of Rolândia, planted in September in no till system. The environment with the highest grain yield, not necessarily is the environment with highest yield of 60 thousand kernels bags ha-1. Environments of Barretos showed conditions to increasing the parameters of plant height and ear height. Three of eight inbred lines showed an adaptability to unfavorable environments, while other inbred lines showed an adaptability to favorable environments. Os índices de seleção foram inicialmente propostos para a seleção simultânea de diversos caracteres e, por requerem a obtenção de estimativas de variâncias e covariâncias, são mais apropriados para programas de seleção recorrente. Posteriormente, desenvolveram-se outros índices, que por não requererem o conhecimento de tais estimativas, podem ser aplicados tanto em programas de seleção recorrente, como para a seleção de genótipos já fixados, como clones, linhagens e híbridos. Recentemente, Garcia (1998) desenvolveu um índice específico para a seleção de genótipos fixados, que se baseia no cálculo da distância de cada genótipo a um ideótipo. As principais vantagens são que este índice permite a aplicação de testes de comparações de médias, bem como a identificação de genótipos com performances aquém daquela exigida para os cultivares comerciais para um dado caráter. O objetivo deste trabalho foi avaliar a eficiência do índice de Garcia (1998) para a seleção de linhagens de soja na fase de pré-cultivares. O material experimental consistiu de 88 linhagens (Geração F7), derivadas pelo método S.S.D. e oriundas do cruzamento entre os genitores BR-80-14853 x PI-123439, e três testemunhas comerciais: IAC-12, IAC-Foscarin-31 e IAS-5. Os experimentos foram conduzidos no delineamento em blocos casualizados em seis anos agrícolas, com diferentes números de repetições por ano: 3 (1995/6), 4 (1996/7), 6 (1998/9), 6 (1999/0), 10 (2000/1) e 10 (2001/2). Os caracteres avaliados foram: produção de grãos (PG: g/parcela) altura das plantas na maturação (AM: cm/planta), e acamamento (AC: notas de 1 a 5). No cálculo das distâncias utilizaram-se a Distância Euclidiana e a Distância de Mahalanobis. Os resultados obtidos mostraram que os índices baseados tanto na Distância Euclidiana quanto na de Mahalanobis foram eficientes para a discriminação dos genótipos, embora os melhores resultados tenham sido obtidos com a Distância de Mahalanobis. Devido a isso, sugere-se o uso da Distância de Mahalanobis para identificar e selecionar pré-cultivares de soja e também, como proposto por Garcia (1998), que esta seleção seja acompanhada da avaliação de caracteres individuais, para evitar que genótipos promissores sejam descartados por apresentarem pequenos defeitos em caracteres de importância secundária. / Selection indices were first proposed for the selection of several traits at the same time, and because of the requirement of variances and covariances estimates, they are useful for recurrent selection programs. Later on, other indices were proposed, which do not require the knowledge of these estimates, and thus are useful for both recurrent selection programs and for the selection of fixed genotypes, such as clones, inbred lines and hybrids. Recently, Garcia (1998) proposed a new index for the selection of fixed genotypes which is based on the distance of each genotype to and ideotype. The main advantages of this index is that multiple comparison tests can be done and also the identification of genotypes that perform below than those required by commercial cultivars for a given trait. The objective of this work was to evaluate the efficiency of Garcia (1998)´s index for the selection of pre-cultivars in soybeans. For the calculation of the distance of each genotype to the ideotype, the Euclidean and Mahalanobis methods were used. General results have shown that the index based on both Euclidean and Mahalanobis distances were efficient to discriminate the genotypes, but the best results were reached by using Mahalanobis distance. Consequently, in order to identify and select fixed genotypes of soybeans we suggest the use of Mahalanobis distance, and as proposed by Garcia (1998), that the selection be based on the index and also on the evaluation of each trait, in order to avoid the discard of superior genotypes showing small problems in traits of minor importance.

Genetic Improvement of Upper Half Mean Length and Short Fiber Content in Upland Cotton, Gosspium hirsutumBeyer, Benjamin 2012 August 1900 (has links)
Desired base upper half mean length (UHML) of upland cotton (G. hirsutum) in the U.S. has been set a 27.0 mm and is shorter than the standard set by the international community. Upland cotton genotypes from China, South Africa, West Africa, and the U.S. were test crossed to an extra long staple upland (ELSU) and a short staple upland (SSU) and selected genotypes that included both ELSU and MSU phenotypes were crossed in a half-diallel mating scheme to estimate general combing ability (GCA) effects and specific combining ability (SCA) effects. A recombinant inbred line (RIL) population was established to determine the narrow sense heritability (h^2) of AFIS short fiber content by weight (SFCw) and lower half mean length (LHML) and to estimate SFCw using HVI fiber properties.
Obsolete cultivars from China are not likely sources for UHML improvement, cultivars from Africa and the U.S. could harbor alleles not being used in current elite short staple cultivars or modern ELSU cultivars. Two ELSU lines used in this study derived through interspecific hybridization with G. barbadense could contain alleles for UHML improvement in modern ELSU cultivars developed without any apparent G. barbadense introgression. A third line D&PL 45-867, might contain alleles for UHML improvement in long staple upland cotton genotypes.
Narrow sense heritability estimates indicated a much higher heritability of LHML than AFIS SFCw. Correlation between AFIS SFCw and LHML did not agree with previous studies when using an ELSU X MSU cross. Further study is needed to understand this complex relationship.

QTLs for Energy Related Traits in a Sweet × Grain RIL Sorghum [Sorghum bicolor (L.) Moench] PopulationFelderhoff, Terry 2011 August 1900 (has links)
Recent initiatives for biofuel production have increased research and development of sweet sorghum. Currently, the initial major limitation to integrating sweet sorghum into existing production systems is the lack of sweet sorghum hybrids adapted to industrial production systems. Hybrid development is now underway, and the application of genetic markers can be used to define the genetic basis of sugar yield and its components, as well as reduce the time required to deliver new sweet sorghum hybrids to market. The purpose of this research was to further characterize the genetic components that influence sweet sorghum productivity, agronomics, and composition. Specifically, a grain x sweet sorghum recombinant inbred line (RIL) population developed for quantitative trait locus (QTL) analysis related to sugar production was evaluated for 24 phenotypic traits including brix, percent moisture, and biomass yield across four environments. The 185 F4 RILs were derived from the parents 'BTx3197' and 'Rio', which are pithy stalk grain and juicy stalk sweet sorghums respectively. Following screening, two genetic maps were constructed with 372 and 381 single nucleotide polymorphisms (SNPs) evaluated using an Illumina GoldenGate assay. Analysis of the data in QTL Cartographer revealed a major and previously reported QTL for soluble solids on chromosome 3, but in contrast to previous studies, this QTL co-localized with other QTLs that have a negative influence on biomass and seed production. Therefore, selection for this QTL may not be advantageous. Because only a few QTLs for percent moisture were found, the results indicated that the pithy stalk phenotype does not have a major effect on percent moisture as measured in this study. Thus, breeding for high or low moisture content will be more challenging than previously expected. The absence of dominance effects indicated that brix must be high in both parents to produce high brix in the hybrid.

Avaliação do desequilíbrio de ligação e da origem genética em duplo-haplóides de milho /Barbosa, Mauricio Pires Machado. January 2009 (has links)
Resumo: Estudos de associação baseados em desequilíbrio de ligação (DL) são importantes ferramentas para construção de mapas de ligação e utilização em programas de melhoramento assistidos por marcadores. Em geral, são utilizadas populações segregantes ou linhagens isogênicas para composição destes programas. Com o objetivo de comparar o desequilíbrio de ligação em linhagens duplo-haplóides (DH) e linhagens obtidas por meio de autofecundação, duzentas e quarenta e cinco linhagens - cento e setenta e cinco convencionais e setenta DHs - foram submetidas à análise de marcadores do tipo Single Nucleotide Polymorphism (SNP), utilizando-se mil duzentos e trinta e quatro marcadores distribuídos pelos dez cromossomos. O resultado da regressão entre as distâncias mostrou um R2 de 0,6546 para linhagens duplo-haplóides e uma equação de tendência logarítmica, sendo y= -0,024ln(x) + 0,159. Para as linhagens convencionais, o R2 foi de 0,5727, com a equação que explica a tendência, sendo y = -0,008ln(x) + 0,0659. Os dados de DL foram analisados individualmente, por cromossomo; e, assim como na análise conjunta, individualmente, todos os cromossomos tiveram o mesmo comportamento quando se comparam o DL de linhagens DH e os convencionais, sendo que os valores de DL nas linhagens DH foram em geral mais altos que nas convencionais. Os resultados indicam que, para a obtenção de linhagens DH, a recombinação ocorre em blocos maiores quando comparado com as linhagens convencionais. / Abstract: Association studies based on linkage disequilibrium (LD) are important tools for linkage maps construction and to use in marker-assisted breeding programs. Typically, segregating populations or isogenic lines are used to compose them. With objective of compare the linkage disequilibrium on double haploid lines (DH) and lines obtained by self pollination (Conventional), two hundred and forty five inbred lines, where one hundred seventy five conventional and seventy DHs where submitted to the analysis of Single Nucleotide Polymorphism (SNP) molecular markers, using one thousand two hundred and thirty four markers random distributed over the ten chromosomes. The regression output among the distances showed R2 of 0.6546 for double haploids lines and one logarithmic trend equation, being y = -0.024ln(x) + 0.159 For conventional lines, R2 was 0.5727, with an equation that explains the trend, being y = -0.008ln(x) + 0.0659. LD data were analyzed by chromosomes individually and as such in the joint analysis, all individual chromosomes showed the same patternr when comparing the LD of DH lines versus conventional lines, being the LD of DH lines generally higher that the conventional lines. Etude des variations épigénétiques liées aux séquences répétées comme source de changements phénotypiques héritables chez Arabidopsis thaliana / Study of epigenetic changes associated with repeated sequences as a source of heritable phenotypic changes in Arabidopsis thalianaCortijo, Sandra 10 September 2012 (has links)
Des changements de méthylation de l’ADN peuvent affecter l’expression des gènes et pour certains être transmis au travers des générations. De telles « épimutations » qui concernent des groupes de cytosines à proximité ou dans les gènes sont donc une source potentielle de variation phénotypique héritable en absence de changements de la séquence de l’ADN. Chez les plantes la méthylation de l’ADN est cependant principalement observée au niveau des séquences répétées. Il reste à déterminer dans quelle mesure les changements de méthylation au niveau de ce type de séquences peuvent être héritées et affecter les phénotypes. Afin de répondre à ces questions, plus de 500 épiRIL (epigenetic Recombinant Inbred Lines) quasi-isogéniques a été générée chez Arabidopsis thaliana. Cette population a été obtenue par le croisement d’un parent sauvage et d’un parent mutant pour le gène DDM1 présentant une très forte réduction du taux de méthylation de l’ADN. Après un rétrocroisement de la F1 avec une plante sauvage, les individus sauvages pour le gène DDM1 ont été sélectionnés et propagées sur 6 générations par autofécondation. Nous avons montré par l’analyse du méthylome de plus de 100 épiRIL que l’hypométhylation induite par ddm1 présente selon les séquences affectées différents degrés de transmission au travers des générations. La réversion de l’hypométhylation concerne des régions associées à une abondance élevée en sRNA de 24 nt. Nous avons utilisé l’hypométhylation stablement transmise dans les épiRIL induite par ddm1 afin de détecter des QTL (Quantitative Trait Loci) affectant le temps de floraison et la longueur de la racine primaire, deux caractères pour lesquels les variations observées dans les épiRIL présentent une héritabilité importante. En dernier lieu, nous avons recherché par différentes approches les variations causales de ces QTL. / Loss or gain of DNA methylation can affect gene expression and is sometimes transmitted across generations. Such epigenetic alterations, which concern clusters of cytosines located near or within genes, are thus a source of heritable phenotypic variation in the absence of DNA sequence change. In plants however, DNA methylation targets repeat elements predominantly and it remains unclear to which extent DNA methylation changes over repeat sequences can be inherited and affect phenotypes. To address these issues, a population of near-isogenic, epigenetic Recombinant Inbred Lines (epiRILs) was generated in Arabidopsis thaliana. These were derived from a cross between a wild type and an isogenic ddm1 mutant line, in which DNA methylation is compromised specifically over repeat elements. After backcrossing of the F1 and selection of the progeny homozygous for wild-type DDM1, the epiRILs were propagated through six rounds of selfing. Analysis of the methylomes of more than 100 epiRILs and of the parents, indicates that ddm1-induced hypomethylation exhibit different patterns of inheritance through generations. Reversion of ddm1-induced hypomethylation is observed for regions associated with high level of 24 nt siRNA. Based on these findings, stable ddm1-induced hypomethylated regions were used to map quantitative trait loci (QTL) for flowering time and primary root length, two complex traits for which substantial heritable variation is observed in the epiRIL population. We finally analysed these QTL by different approaches to find their causal variations.

Seed Coat Color in Flax (Linum usitatissimum L.) Conditioned by the b1 Locus, its Linkage with Simple Sequence Repeat Markers (SSRs) and its Association with Flower Shape, Flower Color, Fatty Acid Profile and Grain Yield2015 January 1900 (has links)
Previously seed coat color in flax has been used as a phenotypic marker for specialty quality traits and currently there is an increasing demand to use seed coat color in flax to market flax for human and animal nutrition uses. Seed coat color was studied to 1) understand the inheritance of seed coat color conditioned by the b1 locus, to 2) understand the relationship of other important flax traits with seed coat color as well as to 3) identify markers that are linked to seed coat color for future marker assisted selection of seed coat color. Spearman’s rank correlation and an allelism test was used to show the inheritance of the alleles at the b1 locus. Bulked segregant analysis (BSA) was used to identify putatively linked markers with the b1 locus, these were then screened on the CDC Bethune x M96006 recombinant inbred line population. Furthermore, the CDC Bethune x M96006 and CDC Bethune x USDA-ARS Crystal recombinant inbred line populations were used to identify any important flax traits that had a significant relationship with seed coat color. It was shown that seed coat color conditioned by the b1 locus was stably inherited and that b1vg and b1 are allelic to one another. The results of the BSA showed that there were 17 candidates for linkage but when these markers were screened on the population only the Lu456 from linkage group (LG) six was identified to have linkage (χ²=3.90; P<0.05) with the b1 locus. Additionally, it was shown that the b1 seed coat color allele of the b1 locus had a pleiotropic effect on flower color and flower shape and that seed coat color was associated with linolenic fatty acid content. None of the traits examined were found to be associated with the b1vg allele of this locus. These results show that the b1 locus is likely present on linkage group six, more marker coverage on linkage group six of markers that are polymorphic between the two seed coat color parents would increase the accuracy of detection. Lastly, this study showed that plant breeders should consider using the b1vg allele that conditions the variegated seed coat color to mark unique lines with important combinations of traits because it sorted independently for seed quality traits. Whereas, the yellow seed coat color conditioned by the b1 allele was found to be associated with higher linolenic fatty acid content and the semi-lethality of this allele would make it not suitable for use in parental lines.
