This thesis compares forward neural networks with algorithms using game theory on basis of board game Blokus. The theoretical introduction part describes the characteristics of neural networks and work with them. There is also outlined algorithm of game theory. The second part deals about the implementation of players based on the outlined principles and shortly descriptions GUI of application. In conclusion, the differences between the players are evaluated on the charts created on the performed tests.

Origin of mammalian somatic cells in the developing testes remains unclear. This origin could be explained by established cell culture derived from testes of Xenopus tropicalis juvenile male. The expression profile of the cell culture showed transcription of some pluripotency genes, somatic Sertoli and peritubular myoid cell markers and last but not least, the mesenchymal stem cell markers. Conversely, germ cell genes were downregulated. Immunocytochemical analysis revealed expression of Vimentin, Sox9 and α-smooth muscle actin, indicating that the testicular cell culture is a common mesenchymal progenitor of the Sertoli and peritubular myoid cells and that the cell culture did not arise from spermatogonial stem cells undergoing incomplete reprogramming in vitro. Testing of X. tropicalis cell culture during induction of differentiation in vitro revealed that these cells are probably multipotent with the ability to differentiate into adipocytes, chondroblasts and osteoblasts. The ability to derive multipotent stem cells from the juvenile testes opens new possibilities of using these cells for biotechnology and medicine. The screening mammography programme has been running succesfully in the Czech republic for 13 years. Due to its ability to uncover earlystages of breast cancer, it helps to fight this serious disease. Although the positive outcome of this programme in the Czech republic has been already proved, there is only approximately 50% of women who have been regularly treated with it.One of the major reasons for this fact is a fear of cancer induction caused by the use of X-rays during the mammography. The main goal of this thesis is to calculate the risk of breast cancer induction caused by the mammography and to compare benefits of the screening programme for a woman who goes for regular checkups and a woman who does not. The measurement of concrete doses was made in one screening centre center located in western bohemia region. The group of observed patients at the age of 45-59 was divided into three categories. The object of measurement was an average dose received during the screening mammography, plus an average dose received during an additional mammography, which may sometimes occure during the examination. These data was studied through a statistic investigation to prove that the received dose is not dependent on the patient´s age. In the theoretical part of this work main principals of the screening mammography programme and mammography itself are repeated. The methods of statistic investigation and the methodology for estimating breast cancer risks based on recommendations of ICRP Publication 103. In the practical part was proved, that the average dose received during the classical mammography does not depend on the age of the patient. This could not be proved for an additional mammography dose, because of not sufficient group of statistic data. The main hypothesis, that regular checkups using the mammographical screening programme is benefical for women , was verified.

The conclusion of this work contains technical-economical evaluation for the chosen options which expects the ten-year return with average production of 40 000 pieces.

This thesis deals with simulation’s possibilities of the electromagnetic fields in electrical machines. A finite element method is the final element technique. The content of this thesis is a model and mesh creation and electromagnetic simulation of synchronous machine. The stator and rotor pack termination effect on the synchronous machine.is analyzed by finite element method in ANSYS program. The type of the stator termination pack is described by four segments. The magnetic field distribution is deformed due to the material properties of used segments. In the next part a methodics of mesh creation is shown. The mesh quality (in term of elements number and elements shape) is important due to the solution accuracy and speed.

This thesis deals with design of optimal parameters of a relative displacement sensor, working on the electrodynamics principle. The sensor is based on the voltage induction to the coil placed in changing magnetic field. Thesis is divided into three parts: introductory, theoretical and practical. The introductory part provides basic relationships between displacement, velocity and acceleration sensors. The theoretical part contains physical concepts and describes relationships necessary for calculations and creating a model of permanent magnet in COMSOL Multiphysics. In thesis further presents patent research of electrodynamic sensors. The practical part describes the creation of the permanent magnet model and the subsequent calculation of the induced voltage in the coil in Matlab. A description of laboratory measurements follows, presenting the final comparison of the simulation and measurement results. There were found some differences, which might have been caused by incorrect calculation of the induced voltage. The final part of the thesis presents general recommendations for the design of sensor, resulting from simulations and measurements.

This work deals with the power supply of wireless sensors. When using a wireless sensor is desirable application of alternative energy sources, because the primary cells or batteries may reduce the extent or length of service of the sensor itself. Ambient energy can be used as a suitable alternative source. This energy must be in an appropriate form, which allows its conversion to electric energy. These appropriate, already used types include: solar en., temperature gradient en., en. of flowing liquids, vibration, etc. The advantage of vibrations is its presence in almost all mechanical systems. One of the possibilities for using the vibration of machine systems for power supply wireless sensors is using the vibration power generator with oscillating component. The generator must be designed so that its resonance frequency coincides with the frequency of vibration in the machine system. This method can be used only if the machinery system vibrations at least partially constant. Another option is to use the vibrations caused by, for example, transit transport, or different step acting factor. In this case, it is desirable that the generator is designed with variable resonant frequency, which can partly be achieved, for example, integrating several oscillating component in the body of generator. After the general analysis of the problem, graduation theses will be concerned with the possibility of use of energy from the short damped oscillation and step impulse. Emphasis is placed on experimental models and methods which are used for the assessment of enzyme induction. In addition to summarizing its well- established role as a xenobiotic-sensing receptor, PXR is also presented as a transcription factor with an important role in endogenous pathways. Furthermore, cell and animal models are evaluated in terms of expression and function of PXR and its target xenobiotic-metabolising enzymes. Primary human hepatocytes in 2D cultures are considered to be the gold standard of in vitro hepatic models. However, 3D technologies are expected to be increasingly used in the future. Apoptosis represents one of the cell death mechanisms which is realized after the application of taxanes in breast cancer cell lines. Apoptosis induction can be principally triggered either by outer or inner pathway. The aim of the diploma thesis is to contribute to the elucidation of role and mechanisms of the inner mitochondrial pathway of apoptosis induction after taxane application (paclitaxel and SB-T-1216) employing a model of breast carcinoma cell lines SK- BR-3 (nonfunctional p53, functional capase-3) and MCF-7 (functional p53, nonfunctional caspase-3). Specifically, we tested the effect of both employed taxanes on mitochondrial membrane potential, ROS level and the expression and localization of proteins regulating inner mitochondrial pathway. Taxane application resulted in mitochondrial membrane dissipation in SK-BR-3 cell line. However, this was not shown in MCF-7 cell line. We found no changes in Bax and Smac/DIABLO expression after taxane application in both tested cell lines. There was a decrease of Bid expression after taxane application in SK-BR-3 line, but not in MCF-7 line. Taxane application did not lead to the translocation of Bax and Bid (tBid) proteins from cytosol to mitochondria in both tested cell lines. The Diploma thesis focuses on the drop-out of beginning teachers. Theoretical part elaborates on the phenomenon of drop-out, it describes the reasons why the beginning teachers leave and why they are the group of teachers which is the most threatened by drop-out. Furthermore, this part describes problems that beginning teachers have to deal with and the most frequent reasons for leaving the profession, their rate and consequences of drop-out. The end of the theoretical part presents options and suggestions that would help solve this problem. The empirical part of the thesis is realized by an on-line questionnaire and in-depth interviews. The research focuses on four main aspects: 1 - the motivation to study the teaching and to practise it, 2 - the main reasons for new teachers to leave, 3 - the possibilities to prevent drop-out and 4 - whether the respondents continue to work with children or not. Last but not least, the research focuses on the possibilities that would help respondents to stay in the profession. The results of the research confirm the reasons for quiting as described by literature. The qualitative description of the reasons for drop-outs provides a deeper understanding of how they interact in an institutional, interactive and cultural level.
