To understand molecular mechanisms of individual steps of virus infection is a prerequisite for successful design of specific and effective antiviral drugs. Polyomaviruses, replicating in the cell nucleus, travel from plasma membrane to the endoplasmic reticulum (ER) in endosomes. However, it is not clear how they deliver their DNA genomes from ER to the nucleus. In this thesis, we found that partially disassembled virions of the Murine polyomavirus (MPyV) interact with importin β1 at around 6 hours post infection. Mutational disruption of the nuclear localization signal (NLS) of the major capsid protein, VP1, and/or common NLS sequence of the minor capsid proteins VP2 and VP3 did not affect the structure and composition of virions, but it resulted in decreased viral infectivity (up to 80%). Virions are thus released from ER to cytosol and translocate to the nucleus via nucleopores. Mutation analyses of NLSs of individual capsid proteins showed that MPyV virions can utilize VP1 and VP2/VP3 NLSs in concert. However, one functional NLS, either that of VP1 or VP2/3 seems to be sufficient for the delivery of VP1-VP2/3 complexes into the nucleus, although none of these proteins is delivered into the nucleus separately. Recent knowledge suggests that the onset of cancer is modulated by the interplay of internal and external environmental factors along with numerous gene variants. Structural chromsomal aberrations in peripheral blood lymphocytes are considered as biomarkers of effect of genotoxic carcinogens and reflect elevated risk of cancer. Incomplete or deficient repair of double-strand breaks in DNA underlie chromosomal aberrations and the measurement of cytogenetic alterations may reflect interindividual differences in the response towards the mutagen. In this study the expected deficiences in the DNA repair capacity have been determined in incident oncological patients with breast, colorectal and urogenital cancers. The determination of chromosomal aberrations have been supplemented by the measurement of variants in genes involved in double-strand breaks repair (XRCC3, rs861539; RAD54L, rs1048771). Methodologically, we employed conventional cytogenetic analysis, cytogenetic analysis following the induction of chromocomal damage by bleomycin ("Challenge assay"), TaqMan discrimination analysis for the detection of allelic variants and statistical analyses. This thesis deals with the elaboration of The Permanent Magnet Synchronous Motor and the calculation of its parameters. In the first part, the construction and used permanent magnets of this motor are listed here. The next part of the thesis contains a short overview of the stator windings, the calculation of the winding factors and calculation of the basic parameters of the stator winding. The calculation of the magnetic induction in the air gap is also illustrated here. The result is recalculated by using the Carter factor and then the value of the first harmonics of air-gap flux density is determined. The result is compared with the analysis in the program FEMM. The next part of the thesis is creating a comprehensive overview of the machine using a replacement circuit diagram and phasor diagrams. Finally, the thesis contains model created in RMxprt and ANSYS Maxwell. The results of the model analysis are compared with the analytical calculation

The article describes how to produce energy necessary for sensor supply. These generators are used as a local source operating on vibratory principle. Mechanical vibrations that occur in moving machines, in nature etc. are used in order to gain required energy. So are kinesis principles and analysis employed in some of the avaible generators. It also contains patent and literary background research. Work is oriented to solving mechatronic perimeter, in the concrete micro - generator. Mechatronic perimeter piles from seat power control parts and mechanical parts, where common solving equation system leads to correct solving. Mentioned analyses be of consequence for usage optimization methods artificial intelligence. Optimization method are used on optimum solving proposal micro - generator. To inquest dynamism system was used program Simulink (part of MATLAB), generator is buckthorn for f = 17 [Hz] acceleration yam = 0,5g [ms-2]. As a algorithm is used SOMA – All to one.

This thesis examines the magnetic field and magnetic flow at the end of the stator synchronous machine. This problem is solid using nonlinear magnetic resistive network, which is simulating by PSpice software. A simple model of stator, consisting of stator plates, air gaps and the two-thumbs, which hold the stator together. All of this is replace magnetic nonlinear resistive network, which is then divided into elements. According to these element is a simplified model solid and analyzed.

Subsequently, are calculated short-circuit conditions in the main switching station. It follows an analysis of power lines. The last part dealt with the issue of reliability of sources in connection with random transition processes in the electrical network and the light of the operational reliability of electrical power equipment.

This thesis deals with the possibility of using the finite elements method for the analysing of synchronous permanent magnets machines. The aim of this work is to discuss the possibility of the machine design more effectively with using the finite elements method and Maxwell software for the simulations. On the created parametric model, simulations were performed according to master thesis assignment. The aim of this work is to determine induced voltage, iron losses, machine inductances and make a permanent magnets demagnetization analysis. From simulations results the machine efficiency is calculated. The parametric model was used for the optimization of selected parameters. From results of optimization new dimensions of permanent magnets was obtained. Machine efficiency was increased by new permanent magnet material and optimized design.

This diploma thesis is focused on an analysis and design of an AC machines windings. In the theoretical part of the thesis, differences between single-layer and double-layer windings are explained. Next, the properties of a stator winding of synchronous machines are described. Finally, the winding factor is determined by means of a standard calculation method and by means of a new progressive calculation method, too. In the experimental part of this thesis, the stator winding of a high-speed synchronous generator excited by permanent magnets on the rotor is designed. This generator is driven by a steam turbine.

Návrh a analýza synchronních strojů s permanentními magnety a soustředěným vinutím / Design and analysis of PMSM with concentrated winding

Permanent magnet synchronous machine (PMSM) are widely used in many industrial applications. This thesis starts with familiarization with the properties and structure of each type of synchronous machines with permanent magnets. Subsequently there are described properties and possible division of AC winding types. Afterward analytical designs of four machines with two different types of winding were realized. These designs were compared by means of simulations using the RMxprt program and their properties were subsequently verified by method of finite element using the Maxwell 2D program. Evaluation and comparison in terms of utility properties for all designs of machines were performed at the conclusion of this thesis.
