Étude de réseaux de jonctions Josephson à haute température critique / Study of High critical temperature Josephson junction arrays

La jonction Josephson (JJ) est le composé de base de nombreux circuits électroniques supraconducteurs (SQUID, détecteurs d’ondes millimétriques, logique RSFQ). Avec la découverte des supraconducteurs HTc (à haute température critique), d’intenses recherches ont été entreprises pour réaliser des JJ fonctionnant à la température de l’azote liquide (77 K) permettant l’utilisation d’une cryogénie compacte. Toutefois, la complexité de ces matériaux a longtemps rendu difficile le développement d’une technologie viable et simple à mettre en œuvre. Parmi les méthodes de fabrication de jonctions, celle utilisant l’irradiation par faisceau d’ions a atteint un niveau de maturité suffisamment important pour pouvoir envisager la production de circuits comportant plusieurs milliers de JJ à HTc.Le but de ma thèse repose sur la fabrication, la caractérisation et l’étude des propriétés électromagnétiques de réseaux de jonctions Josephson réalisés dans des films minces d’YBa2Cu3O7-d. par la méthode d’irradiation ionique. / The Josephson junction (JJ) is the basis of many compound superconducting electronic circuits (SQUID detectors millimeter wave RSFQ logic). With the discovery of HTS superconductors (high critical temperature), intensive research has been undertaken to make JJ operating at the temperature of liquid nitrogen (77 K) allowing the use of a compact cryogenics. However, the complexity of these materials has long hampered the development of a viable technology and simple to implement. Among the junctions manufacturing methods, one using the ion beam irradiation has reached a sufficiently high degree of maturity to be able to envisage the production of circuits having thousands of JJ to HTS.The aim of my thesis is based on the fabrication, characterization and study of the electromagnetic properties of Josephson arrays achieved in thin films of YBa2Cu3O7-d. by the ion irradiation method.

The Design and Manufacture of a Light Emitting Diode Package for General Lighting

Lighting technologies have evolved over the years to become higher quality, more efficient sources of light. LEDs are poised to become the market standard for general lighting because they are the most power efficient form of lighting and do not contain hazardous materials. Unfortunately, LEDs pose unique problems because advanced thermal management is required to remove the high heat fluxes generated by such relatively small devices. These problems have already been overcome with complex packaging and exotic materials, but high costs are preventing this technology from displacing current lighting technologies. The purpose of this study is to develop a low-cost LED lighting package capable of successfully managing heat. Several designs were created and analyzed based on cost, thermal performance, ease of manufacturing, and reliability. A unique design was created which meet these requirements. This design was eventually assembled as a prototype and initial testing was conducted. This thesis reviews the design process and eventual results of the LED package design.

Deep sequencing of pre-translational mRNPs reveals hidden flux through evolutionarily conserved AS-NMD pathways

Deep sequencing of mRNAs (RNA-Seq) is now the preferred method for transcriptome-wide quantification of gene expression. Yet many mRNA isoforms, such as those eliminated by nonsense-mediated decay (NMD), are inherently unstable. Thus a significant drawback of steady-state RNA-Seq is that it provides marginal information on the flux through alternative splicing pathways. Measurement of such flux necessitates capture of newly made species prior to mRNA decay. One means to capture nascent mRNAs is affinity purifying either the exon junction complex (EJC) or activated spliceosomes. Late-stage spliceosomes deposit the EJC upstream of exon-exon junctions, where it remains associated until the first round of translation. As most mRNA decay pathways are translation-dependent, these EJC- or spliceosome-associated, pre-translational mRNAs should provide an accurate record of the initial population of alternate mRNA isoforms. Previous work has analyzed the protein composition and structure of pre- translational mRNPs in detail. While in the Moore lab, my project has focused on exploring the diversity of mRNA isoforms contained within these complexes. As expected, known NMD isoforms are more highly represented in pre-translational mRNPs than in RNA-Seq libraries. To investigate whether pre-translational mRNPs contain novel mRNA isoforms, we created a bioinformatics pipeline that identified thousands of previously unannotated splicing events. Though many can be attributed to “splicing noise”, others are evolutionarily-conserved events that produce new AS-NMD isoforms likely involved in maintenance of protein homeostasis. Several of these occur in genes whose overexpression has been linked to poor cancer prognosis.

The microplasma discharges in the PN junction local defect micro-regions are as a rule, accompanied by the emission of light. This radiation from solar cell PN junctions was measured by means of a optical fibre connected to the optical input of a photomultiplier. By inching the fibre by means of computercontrolled X-Y plotter above the cell surface a 2-D image of the irradiation local regions has been created. It is seen that a cell of a superficial area of 100 square cm contains a large number of defects, which depends on applied reverse voltage. This method can be a convenient tool for study and diagnostics of optoelectronic devices.

Construction of northern bypass of Kuřim and usage of this bypass as a connection between R43 and Brno city, before whole R43 is build, is the assumption of this thesis. Thats why solution of the junction forsees the construction and operation in two phases.
