Utveckling av logistikflöde : En undersökning av införande av extern kittning av hydraulslang vid Komatsu Forest AB

Komatsu Forest AB tillverkar skogsmaskiner och aggregat. I monteringsprocessen är hydraulslangar en av komponenterna som hanteras och lagras per artikelnummer. Idag genomförs intern kittning då slangar plockas ur respektive lagerlösning mot en specifik monteringsoperation. Kittning innebär att delar och komponenter sorteras i set, så kallade kit, innan de levereras till monteringsoperationer. Genom extern kittning som utförs av leverantören kan eventuellt flödet av hydraulslang och utnyttjandet av ytan i anläggningen bli mer effektivt då kitten är färdiga när de anländer till anläggningen.   Syftet med arbetet är att identifiera det mest produktiva och kostnadseffektiva arbetssättet gällande hantering av hydraulslangar från beställning till montage på maskin. För att kunna jämföra alternativa arbetssätt med nuläget har en nulägesanalys av befintligt arbetssätt utförts genom intervjuer och observationer. Skapande av nya arbetssätt har utgått från tre principer som grundas i extern kittning: kitta alla slangar, kitta mest frekventa slangar och kitta minst frekventa slangar. Resultatet som genererats med de tre principerna har utvärderats för att avgöra hur olika typer av kittning påverkar hanteringstider och kostnader i anläggningen i förhållande till nuläget. De olika kittningsscenariona utvecklades för att kunna utesluta vissa typer av kittning som inte var lönsamma och för att försöka identifiera det mest fördelaktiga kittningsscenariot.   Ett kittningsscenario hittades som är effektivare än nuläget, sett till flöde och kostnad, nämligen kittning av samtliga slangar till en specifik maskin eller aggregat. Dessa slangar kan vara uppdelade på en eller flera pallar, där varje pall täcker in varsin del av monteringslinan. Antalet pallar som slangarna delas upp på påverkar hur stor kostnadsbesparingen i anläggningen blir, då besparingen minskar vid ökande antal pallar. Kostnadsbesparingen i anläggningen måste vägas mot ökade kostnader från leverantör för kittning, eventuell extern lagring och eventuell ökad leveransfrekvens.   Införande av kittning rekommenderas om Komatsu kan hantera kit i sitt datasystem och om monteringslinan går att dela upp i ett så pass lågt antal pallar att det är lönsamt. Tiden från att produktionsplanen är satt hos Komatsu till att kittet ska ut i monteringen måste även vara tillräcklig för att leverantören ska hinna plocka och leverera kitten. Om det brister i någon av ovanstående förutsättningar är nuvarande arbetssätt ett bättre alternativ än kittning. / Komatsu Forest AB manufacturers forest machines and heads. In the assembly process, hydraulic hoses are one of the components that are handled and stored by item number. Currently, internal kitting is performed as all hoses are picked out of each storage solution in preparation for a specific assembly operation. Kitting means that parts and components are sorted into sets, so called kits, before they are delivered to assembly operations. However, the flow of hoses and the use of the storage area might become more effective through external kitting performed by the supplier, as the kits will be ready when they arrive to the plant.   The purpose of the project is to identify the most productive and cost-effective work procedure when handling hydraulic hoses from order to assembly. In order to be able to compare alternative work procedures with the current procedure, an analysis of the current situation was executed on the basis of interviews and observations. The creation of new work procedures proceeded from three principles based on external kitting: kit all hoses, kit most frequent hoses and kit least frequent hoses. The generated result has been evaluated with respect to how different types of kitting affect handling times and costs in the plant in relation to the present situation. The different scenarios of kitting were further developed to be able to exclude some types of kitting as unprofitable and to be able to identify the most profitable way of kitting.   One kitting alternative was found that is more effective than the present situation, in terms of material flow and cost, namely to kit all hoses towards a specific machine or head. These hoses can be divided into one or several pallets, where each pallet caters to a part of the assembly line. The number of pallets that the hoses are divided into affects the cost savings in the plant, where the savings decrease with an increasing number of pallets. The cost savings in the plant have to be evaluated with respect to increasing costs from the supplier for kitting, possible external storage and possible increased frequency of delivery.   Kitting is recommended if it is possible to handle kits in Komatsu’s computer system and if the assembly line can be divided into a number of pallets so that kitting is a profitable option. The time from setting the production plan to the point at which the kit is required at the assembly line needs to be sufficient for the supplier to be able to make and deliver the kit. If one or more of the prerequisites above are not met, the current work procedure is a better alternative.

Canine Mast Cell Tumours: Characterization of Subcutaneous Tumours and Receptor Tyrosine Kinase Profiling

This work explored features of canine mast cell tumours (MCT) to improve prognosis and to discover potential therapeutic targets. Subcutaneous MCT - a subset of these tumours arising in the subcutis - are usually grouped with cutaneous MCT, but there is evidence that they may be clinically different. The first objective was to develop a grading scheme for subcutaneous MCT. Over 300 canine subcutaneous MCT were evaluated retrospectively and parameters were correlated with clinical outcomes, making this the largest retrospective survival study of these tumours to date. The results of the study showed that the majority of subcutaneous MCT had excellent outcomes and key prognostic markers were identified (mitotic index, surgical margins and degree of infiltration). A subset of the subcutaneous MCT from the retrospective study was further evaluated to assess the cellular localization of KIT - a receptor tyrosine kinase (RTK) which is dysregulated and constitutively activated in some cutaneous MCT - as well as Ki67, a proliferation marker. In addition, evaluation of mutations of c-KIT, the gene for KIT, was determined for each MCT. Cytoplasmic KIT localization and high Ki67 values were predictive of decreased survival time and time to local reoccurrence, but no c-KIT mutations were detected. The majority of canine MCT do not appear to depend solely upon KIT for tumour progression and few other RTK targets have been studied in canine MCT. Based on evidence that vascular endothelial growth factor receptors (VEGFR) and platelet-derived growth factor receptors (PDGFR) - may play a role in the progression of canine MCT; the expression and distribution of these RTK were evaluated. The results showed that canine MCT have unique expression profiles and activity of KIT, VEGFR2 and PDGFR. Two novel mast cell tumour cell lines were generated and used to assess signalling of KIT and VEGFR2 in vitro. Stimulatory and inhibitory responses were assessed and found to be different in both cell lines. Both had autophosphorylated VEGFR2 and an autocrine VEGF/VEGFR2 signalling pathway existed for both cell lines. La protéine SHP2 est une tyrosine phosphatase cytosolique impliquée dans la régulation des phénomènes de prolifération, migration et survie cellulaire. Elle contrôle l'état d'activation des voies de signalisation des récepteurs à tyrosine kinase faisant intervenir en particulier les protéines kinases MAPK/ERK, PI3K/AKT. Des mutations germinales et somatiques de SHP2 sont à l'origine de syndromes et de néoplasies. Au cours de ce travail, nous avons mis en évidence une nouveau partenaire pour SHP2: la protéine CDCP1. CDCP1 est une protéine transmembranaire dont la surexpression et la tyrosine phosphorylation par les kinases de la famille de SRC dans de nombreux carcinomes permet une augmentation des propriétés métastatiques des cellules. Nous avons de plus mis en évidence la régulation de la phosphorylation de CDCP1 et sa présence à la membrane cellulaire par SHP2. Dans une seconde partie de ce travail de thèse, notre attention s'est portée sur la protéine adaptatrice GAB2, partenaire et substrat principal de SHP2, dont la surexpression dans les mélanomes primaires et métastatiques commence à être décrite. Nous mettons en évidence la présence d'une boucle de régulation entre GAB2, dont l'expression est régulée par le facteur de transcription MITF et le récepteur KIT, dont l'activation, dépendante de l'expression de GAB2 régule l'activation de MITF et la motilité des cellules de mélanome.L'ensemble de ce travail met ainsi en évidence la présence d'un nouvel interacteur de SHP2, et d'une nouvelle boucle de régulation dans les cellules de mélanome. / SHP2 is a protein tyrosine phosphtase which regulates cell proliferation, mirgration and survival. It controls MAPK/AKT and PI3K/AKT signaling pathways activated by tyrosine kinase receptors. Germinal and somatic mutations in SHP2 lead to syndromes and neoplasia. We show a novel interaction between SHP2 and the protéine CDCP1. CDCP1 is a transmembrane protein which overexpression and tyrosine phosphorylation by SRC family kinases in carcinomas lead to an increase of the tumor cells metastatic properties. We also demonstrated that SHP2 regulates CDCP1 tyrosine phosphorylation level and its presence at the cell membrane.In a second part of this work, we focused on GAB2 anchor protein, which is one of the most important partner and substrate for SHP2. GAB2 is overexpressed in primary and metastatic melanoma. We show the presence of a regulation loop, involving GAB2 regulation by the transcription factor MITF, KIT receptor and its activation depending on GAB2, which controls MITF activation and melanoma cell motility.This thesis work demonstrate the presence of a novel partner for the protein SHP2 and of a new regulation pathway in melanoma cells.

This master’s thesis contains design of practice exercises for microcontroller MCF52233 firm Freescale. Exercises are focused on working with basic peripheries as counters and timers, measurement with A/D converters, PWM modulation or I2C bus. Exercises are interested in evaluation of matrix keyboard and communication with alphanumeric display controller. To these exercises is made a preparation of external peripherals, which is connected with evaluation kit M52233DEMO. This preparation contains all necessary peripheries, which are used for practice exercises – buttons, LED diodes, potentiometer and I2C periphery. Individual periphery usually shares connection to microcontroller with another periphery. Therefore preparation contains switches, which are necessary to set up for correct functionality of the peripherals. The board of external peripheral also includes ports for connecting matrix keyboards and alphanumeric display. Connecting the display requires an external power supply. Work deals with implementation of laboratory exercises using a software firm Freescale. Development environment for programming and basic debugging is in program CodeWarrior. More advanced possibility of reading information from a running program in the microcontroller offers program FreeMaster, which is used in the exercise of AD converters. Each lab exercise contains an introduction, task, instructions and the flowchart of realized exercise. Instructions are written so that it contains important steps to properly manage assignments. Individual exercises are designed to gradually increasing complexity of exercises students obtain basic programming habits and manage the complex exercise.
