Master´s thesis covers introduction to the problems of materials with specific proprieties named liquid crystals. Theoretic part features primary fission these materials, theirs mechanical, electrotechnical and optical characteristics. Interesting chapter is description choice application including perhaps most interesting using liquid crystals - liquid crystal display. In this trend continues experimental part too, where there are first mentioned characteristics LCD, and after it alone metering capacitive characteristics display with active matrix. Main attention in experiment is however concentraded on evaluation frequency analysis of components complex permittivity regarding in plant temperature on submitted sample liquid crystals.

For this, it is used two algorithms created in professional computing interface MATLAB. The outputs from this master’s thesis are dendrograms, which divide QRS complexes into abnormal and normal clusters, and Pearson correlation coefficients.

The aim of this diploma thesis is to introduce methods of detection of the QRS complex and the subsequent detection of P waves. The intention is to create a program by specified method in the software Matlab which will be able to implement this method. The thesis describes the basic and important methods of detection and subsequent algorithm to detect P waves. Solution of the algorithm is tested on real data. It also describes the automatic signal evaluation and the results of this automatic function.

This thesis deals with the layout, construction, fire and thermal technical solution for residential building with business part. The residential complex is divided into four functional parts - underground garages, administrative, operational and residential part. This proposed building has four floors and a basement. The upper floor is sheltered by a flat roof. The constructional system is made of reinforced concrete columns and walls in the first two floors, as for the next floors the brick wall construction is used. The ceiling construction is a monolithic reinforced concrete panel with cross tension.

This Ph.D. thesis is focused on kinetic study of propene and hex-1-ene coordination polymerizations initiated by complex [(2-tBuC6H4)N=C(1,8-naphtalenediyl)C=N(2-tBuC6H4)]NiBr2 activated by simple organoaluminium compounds and on product properties. In first three parts proper activators are chosen in model polymerizations. The attention is paid to the reproducibility of polymerizations and analytical methods. The fourth part is focused on propene polymerization kinetics with the aim to define optimal reaction conditions, especially polymerization temperature and time. The four part concerns also properties of polymers. The fifth and sixth parts deal with hex-1-ene polymerization kinetics conducted at various activator/initiator molar ratios and various catalytic precursor concentrations, to find out kinetic orders with respect to catalyst and cocatalyst concentration. Last part is oriented on detail mechanistic investigation of nickel diimine complexes activation process. Results of measurements in presence or absence of monomer enabled to propose new interpretation of UV-vis spectra based on concrete structures of absorbing species.

Cytokinesis is the last step of cell cycle when two individual daughter cells separate in process called abscission. This process involves various cellular membrane structures such as endoplasmic reticulum or trans-Golgi network. Moreover, recent investigation has also highlighted an important role of recycling endosomes. The membrane dynamics appear to be important during cell division especially for the formation of new plasma membrane between two daughter cells. Numerous studies suggest that cytokinesis is tightly linked with highly sophisticated transmembrane shuttle that is controlled by Ras-superfamily members such as Rab and Ral proteins. Moreover, during last years has also been revealed the involvement of tethering factors which mediate the fusion of intracellular vesicles with the target plasma membrane. The best known tethering factor is the evolutionary conserved exocyst complex found in all eukaryotic cells. This protein complex is composed of eight subunits (Sec3, Sec5, Sec6, Sec8, Sec10, Sec15, Exo70 and Exo84) and was found to interact with members of Ras- superfamily suggesting its involvement in the regulation of cytokinesis. Plant cells are mostly immobile, therefore it is crucial for them to distinguish a direction of the signals coming into the cell and on the other hand they have to precisely target their own signals. To achieve this communication, plant cells use endomembrane system and secretory vesicles, which are recruited to the specific membrane domains. This ability is important for the plant defense against pathogenic microorganisms and it even forms a part of the innate plant immunity. Two complexes, the exocyst and SNARE, play a prominent role in the process of polarized secretion. In this work, we focused on a possible interaction between these two complexes in preinvasive defense and particularly, we studied the exocyst subunit EXO70B2 and SNARE protein SYP121. We obtained double mutant plants of EXO70B2 and SYP121 by utilizing the reverse genetics approach. These mutant plants did not show any obvious phenotype under standard conditions in comparison with Wt plants. However, we observed marked defects of secretory pathway in double mutant exo70B2/syp121 after infection by pathogenic fungi Blumeria graminis f. sp. hordei. Specifita vybraných podjednotek exocystu při vývoji trichomu / Specificity of selected exocyst subunits in trichome development

Trichomes are fine epidermal outgrowths covering aerial organs of most land plants. Although unicellular trichomes of Arabidopsis thaliana have long been used as a model system in plant cell and developmental biology, surprisingly little is known about the processes involved in cell wall biogenesis during the last stage of trichome maturation. A role of EXO70H4, a putative subunit of the vesicle tethering complex exocyst, in trichome maturation has recently been identified in our laboratory. Image analysis, histochemical detection and FT-IR spectroscopy methods were used in this study to analyze cell wall defects of the exo70H4 LOF mutant, revealing the mutation causes altered deposition of pectins and possibly also lignins and hemicelluloses. Transgenic lines with EXO70 paralogues driven by the EXO70H4 promoter were prepared and their analysis revealed that the closest paralogue EXO70H3, unlike EXO70A1 and EXO70B1, can complement the exo70H4 mutation. Based on the results, questions concerning trichome cell wall composition, the role of EXO70H4 in trichome maturation and functions of the plant exocyst complex are discussed. During the course of evolution the early land plants gained extensive innovations that can be seen in modern day plants. The polar growth is an ancient feature of eukaryotic cells and is one of preadaptations that helped plants in successful colonization of land. The polar growth in plants regulates not only the direction of cell expansion and structural properties of cell wall but especially also the orientation of cell division, and is governed by various factors, including the exocyst complex. The exocyst is a well conserved vesicle tethering multi-subunit complex involved in tethering of secretory vesicles to the target membrane. The essential role of the exocyst complex in regulation of various cellular processes in Angiosperms is now well documented. Here I present results of a doctoral project that contributed to phylogenetic analyses of the land plant exocyst complex and especially to uncovering functions of three moss exocyst subunits, namely EXO70 (isoform PpEXO70.3d), SEC6 and SEC3 (isoforms PpSEC3A and PpSEC3B) in the model organism Physcomitrella patens. The intracellular sperm injection (ICSI) technique is a very effective tool for the fertilization research. In the newly established laboratory at the Faculty of Science of the Charles University it was necessary to introduce this method and define the early developmental potential of fertilized oocytes. After fertilization oocytes were incubated to the blastocyst stage with a success comparable with other laboratories (17%). The ubiquitin-proteasome system which plays a major role in a protein degradation within cells is involved in a regulatory mechanism of sperm maturation. It is also responsible for a penetration of a vitelline membrane. In these processes ubiquitin residues are localized extracellulary. High level of sperm ubiquitination correlates with their low quality. Hypotetically it can be expected that the ubiqutination of impaired sperm cells can be used as a negative marker for their recognition and degradation by 26S proteasome complex localized. Experiments in this diploma thesis were designed based on the hypothesis that the executive part of the selective mechanism is the 26S proteasome.
