Geometrické a algebraické vlastnosti diskrétních struktur / Geometric and algebraic properties of discrete structures

In the thesis we study two dimensional simplicial complexes and linear codes. We say that a linear code C over a field F is triangular representable if there exists a two dimensional simplicial complex ∆ such that C is a punctured code of the kernel ker ∆ of the incidence matrix of ∆ over F and dim C = dim ker ∆. We call this simplicial complex a geometric representation of C. We show that every linear code C over a primefield is triangular representable. In the case of finite primefields we construct a geometric representation such that the weight enumerator of C is obtained by a simple formula from the weight enumerator of the cycle space of ∆. Thus the geometric representation of C carries its weight enumerator. Our motivation comes from the theory of Pfaffian orientations of graphs which provides a polynomial algorithm for weight enumerator of the cut space of a graph of bounded genus. This algorithm uses geometric properties of an embedding of the graph into an orientable Riemann surface. Viewing the cut space of a graph as a linear code, the graph is thus a useful geometric representation of this linear code. The aim of this thesis is to describe the design of a habitable year-round holiday resort with rope and climbing center. The resort complex consists of individual cabins which vary in the equipment standard and in the construction itself. The central building includes a restaurant and a lounge for guests. As a part of the resort, there is also a rope and climbing center. That is why this thesis also deals with the structural wood protection features which are often applied for installations of this kind.

The aim of this thesis is to re-evaluate and re-discuss sealings that were uncovered during the excavations of the pyramid temple of King Raneferef in Abusir, resp. those which are kept in the collections of the Náprstek Museum in Prague. It deals with the general typology of cretulae and the method of their documentation and it tries to assess sealings under study in regard to the former typology. The second part of the work attempts to find details regarding the activity and authority of officials who were using seals in connection with the operation of the pyramid complex of King Raneferef. This was attempted by a search for recurrent seal designs attested on fragments of cretulae found in the area of the pyramid complex and by a study of the properties of their seal holders and their activity, areas where the fragments of sealings were found, titles that were attested on their sealings, and kinds of objects that were sealed by their seals.

Hybridizing species of the Daphnia longispina complex are key taxa in plankton communities of many European lakes. In several of these lakes, it has been documented that the taxonomic structure of the complex during the 20th century has substantially changed following human-mediated environmental changes, particularly eutrophication and re-oligotrophication and fish stock changes. We characterize these changes in Lago Maggiore (Italy/Switzerland), a southern pre-alpine lake, which also passed through the human-mediated environmental changes. Lago Maggiore is one of best and longest studied European lakes so a large number of historical Daphnia samples from regular monitoring are available. Because local Daphnia do not form dormant egg banks suitable for genetic analysis (being able to overwinter in the water column), we used a combination of morphology and geometric morphometrics (elliptic Fourier analysis) to evaluate the taxonomic and phenotypic changes in the Daphnia longispina complex in Lago Maggiore since the mid-20th century (1948-2012), and attampted to characterize the impact of environmental changes over the respective period on Daphnia body shape and size. The constitutive androstane receptor (CAR) plays an important role as a xenosensor in the organism and is therefore widely expressed in the kidney, liver, gallbladder and in the small intestine epithelium, where the biotransformation of xenobiotic occurs. CAR is also an important factor in the elimination of bile acids and bilirubin. CAR has also been shown to have an important role in regulation of glucose and lipid metabolism. Impairment of lipid and glucose metabolism is a common cause of cardiovascular and metabolic diseases such as atherosclerosis, type 2 diabetes, obesity and insulin resistance. These diseases are called metabolic syndrome and result in severe organ damage. CAR respective its complex with RXRα (retinoid X receptor α) has become a promising biological target for drug discovery for metabolic syndrome. The major aim of this study was a structural characterization of the CAR/RXRα protein complex together with the agonist IV676*HCl, which would describe in detail the interaction of this small molecule with the receptor. Obtained structural information would be used to design improved agonists.

Actin polymerization facilitated by the Arp2/3 complex plays a critical role in a wide range of cellular processes such as motility, endocytosis and cargo recycling. Activation and appropriate localization of the Arp2/3 complex is mediated by an interaction with the nucleation-promoting factor (NPF). WASH complex is the major endosomal NPF which plays a crucial role in the cargo recycling back to the trans-Golgi network (TGN) or plasma membrane. It is composed of five subunits: WASH1, SWIP, FAM21, CCDC53 and strumpellin. While WASH1 and FAM21 have been extensively studied, much less is known about strumpellin, a protein causally implicated in the onset of hereditary spastic paraplegia (HSP). This work focuses on strumpellin function in the cells, showing that only full-length protein incorporates into the WASH complex. In a strumpellin knock out cell line, we demonstrated that loss of strumpellin resulted in destabilization of the other WASH complex subunits. Still, an incomplete WASH complex without strumpellin was assembled. Cells also displayed enlarged endosomal subdomains and WASH complex nucleation activity on endosomes was largely diminished as assessed by loss of the actin patches. Finally, the absence of strumpellin was also accompanied by the accumulation of glucose transporter 1 (GLUT1)...

This thesis deals with implementation of detector QRS complex with use of wavelet transform. The first part is focused on formation and possibility to measure cardiac activity. The other part of thesis we will familiarise with the different possibilities of detection QRS complex and we intimately focus on wavelet transform that will be used for a project of detection QRS complex. The practical part of thesis focuses on the project of detector in programming language Matlab and his different setting. This projected detector has been tested with CSE database. Achieved results of projected detector are evaluated with the results of others authors.
