Master´s thesis deal with the supporting steel structure of multi-storey multifunction center in Opava. The complex consists of three sub-objects of three and five storeys with a total floor plan dimensions 48 m × 82.27 m. Five-floor building is designed with ground plan dimensions 42 m × 30 m with the distance of the poles 6.0 m x 6.0 m. The median three-storey building is designed with ground plan measuring 42 meters x 22.27 meters to the distance of the poles 6m x 5m. Ultra three-storey building is designed with ground plan dimensions 42 m × 30 m with the distance of the poles 6m x 6m. Construction of the complex height is 5m.

The color scheme is in the range of black and white with the possibility of graphic designs and various viewpoints of the façade.

The purpose of the present study was to examine complex problem solving (CPS),emotional intelligence (EI) and creativity to further the knowledge aboutcompetencies that are important for the 21st century workplace. We hypothesizedthat CPS would be related to both creativity and EI. Furthermore, we hypothesizedthat age would show a negative relationship to CPS. 39 participants recruitedmostly from convenience sampling completed the CPS test, the creativity test andthe EI test at locations in Örebro and Stockholm. The results were analyzed withlinear and multiple regressions and showed that CPS significantly predictedcreativity and that CPS significantly predicted EI, with those two having a negativerelationship. A regression revealed that age significantly predicted CPS, with thosetwo having a negative relationship. It was theorized that working memory andintelligence were important factors explaining the regression of CPS and creativity.The unexpected negative relationship between CPS and EI was theoreticallyinvestigated, indicating that personality factors could have affected the results. / Syftet med den här studien var att undersöka komplex problemlösning (CPS),emotionell intelligens (EI) och kreativitet för att främja kunskapen om kompetensersom är viktiga för 2000-talets arbetsplats. Vår hypotes var att CPS skulle vararelaterat till både kreativitet och till EI. Dessutom var en hypotes att ålder skulleha en negativ relation till CPS. 39 deltagare rekryterade främst frånbekvämlighetsurval genomförde CPS-testet, kreativitets testet och EI-testet iÖrebro och i Stockholm. Resultaten analyserades med linjära och multiplaregressioner och visade att CPS signifikant predicerade kreativitet och att CPSsignifikant predicerade EI, med ett negativt förhållande. ARPC2 protein localization in a plant cell Kateřina Šlajcherová 1 Abstract Actin cytoskeleton is an ubiquitous structure which plays numerous irreplacable roles. Actin nucleation is, beside formins, performed by ARP2/3 complex (actin-related protein), comprising of seven subunits (ARP2, 3, C1-C5) and activated by protein SCAR/WAVE complex. ARP2/3 complex is attached to the membrane and branches existing microfilaments, apart from nucleating them de novo. ARP2/3 mutants in most organisms show severe defects. However, plant mutants exhibit only mild phenotype, for example, Arabidopsis thaliana ARPC2 mutant (dis2-1) has deformed trichomes and leaf epidermal cells, but its viability is not impaired. The aim of the thesis is to map ARPC2 localization within the cell and broaden our understanding of ARP2/3 complex role in plant cell morphogenesis. Tobacco ARPC2 (NtArpC2) subunit was visualized in Arabidopsis plants, using the GFP fusion protein as well as imunofluorescence and anti-ARPC2 antibody. Experiments were undertaken to collocalize the subunit with actin and microtubular cytoskeleton, with mitochondrions, endosomes and other membrane organelles. Systematika a fylogeografie diploidních zástupců druhového komplexu Arabidopsis arenosa / Systematics and phylogeography of diploid lineages of Arabidopsis arenosa complex

The Arabidopsis arenosa species complex is closely related to the model species Arabidopsis thaliana. Distribution range of the diploid lineage covers most parts of Central and South-eastern Europe with a remarkable diversity of ecological niches from sea level to alpine peaks. There is still little known about genetic relationships among the diploid populations of A. arenosa. The evolutionary history of the diploid cytotype across its entire range was explored by using 14 nuclear microsatellite loci. Five lineages occupying biogeographically distinct regions were identified in Arabidopsis arenosa complex. These lineages were labelled as Western Carpathians, Eastern Carpathians, Pannonian Lowland, Dinaric Alps and Baltic Shore (according to geographical regions where these lineages occur). Úloha vybraných podjednotek komplexu exocyst v odpovědi rostlin na patogena / The Role of selected exocyst subunits in response of plants to pathogen

In the recent years, there has been a growing number of publications indicating at the involvement of plant secretory pathway in defense against phytopathogens. Specifically, roles of plant exocyst complex have been explored in deeper detail in current research. Yet, exactly how exocyst- mediated exocytosis contributes to secretion of antimicrobials and cell wall-based defense remains unclear. In the presented Dissertation, I provide both experimental evidence and devise further hypotheses on selected exocyst's subunits in plant immune reactions. Particularly, I show that EXO70B1 exocyst subunit interacts with immunity-related RIN4 protein. Cleavage of RIN4 by AvrRpt2 Pseudomonas syringae effector protease releases both RIN4 fragments and EXO70B1 from the plasma membrane when transiently expressed in Nicotiana benthamiana leaves. I speculate on how this might have an implication in regulation of polarized callose deposition. In a co-authored opinion paper, we also hypothesize that EXO70B1-mediated autophagic degradation of TN2 resistance protein prevents its hyperactivation and lesion mimic phenotype development. Roztoky Intrusive Center (33-28 Ma) is connected with tertiary volcanic activity in the territory of the Bohemian Massif (Czech Republic). It is located in the České středohoří volcanic complex on the tectonic line of the Ohře rift. The aim of this work was to petrologically describe local rocks and to verify the possible connection of the Roztoky Intrusive Center (RIC) with carbonatite magmatism, as could be suggested by the presence of alkaline rocks including their exotic types (essexites, monzodiorites, sodalitic syenites,…). It is assumed that there is a hidden carbonatite intrusion under the RIC caldera structure, which may also indicate geophysical manifestations of two hidden gravimetric minimas. In a detailed exploration of a drill core from the R-2 borehole from the 1960s, a petrographic rocks composition of a part of the caldera filling was described, including new find of xenolite of carbonatite in 286,5 m of R-2 borehole. By means of geochemical and micro-probe analyzes the well material including carbonatite was examined in detail. Carbonate mineralization with high REE contents was found in carbonatite, but also in the exocontact of the previously described silicocarbonatite (Rapprich et al. 2017). Diploma thesis "The collapses of distant worlds" deals with interpretations of the conquista of Mexico and Peru (i.e. the conquests of Aztec and Inca empires). The basic question, which all the works considering the conquista deal with, is: how could the arrival of small groups of Spanish conquerors disrupt great Indian empires in such a short time? My thesis has two main goals: to define the typology of interpretations of the conquista and offer my own interpretation which places emphasis on the role of mentalities and cultural complexes. I have distinguished four basic types of interpretations of the conquista: 1. the metaphysical ones; 2. the technological ones; 3. the environmental ones; 4. the mentalistic ones. I used the conception of mentalities (derived from the work of Lucien Lévy-Bruhl) and the system conception of cultur, where I worked with the term "cultural complex" especially. After my approach the conquista represented the clash between two different types of mentalities: the archaic one (which Indian societies featured) and the literary one (which was developed in European societies). The Munich agreement signed on the 29th of September 1938 has always been giving rise to strong emotions in Czech society. This event, frequently described as "about us, without us", is a synonym for humiliation, injustice and a symbol of betrayal for many. The historian Jan Tesař holds a non-traditional point of view on this matter. He opposes this established interpretation and sees it as a source of origin of the so-called Munich Complex. The proposed diploma thesis focuses on this phenomenon, which is examined in printed media. Firstly, it provides a brief historical context and introduces an overview of different interpretations of Munich events within Czech historiography as well as describes the arguments of Tesař himself. Furthermore, the Munich complex is conceptualized and the resulting concept includes variables necessary for the research part of the thesis. This section consists of qualitative and quantitative analysis of the content of selected representative media samples. Among examined aspects are arguments used in the press, patterns of interpretation and attributed meanings. The time scope for analysis ranges from 1948 to 2018. Extremely polymorphic genes of major histocompatibility complex (MHC) play a significant role in the function of immune system by recognizing heterogeneous particles, mainly pathogenic origin. Previous research on various vertebrate species indicates that MHC influences individual body odour and mate choice preferences. Many individuals tend to prefer MHC dissimilar partner so that warrants them an offspring resistant against wider spectrum of infections. Research on MHC-related mate preferences in humans, however, is inconclusive to date. Several studies indicate that women not taking hormonal contraceptives prefer the smell of MHC dissimilar partners while other studies have not come to this conclusion. This can be caused by the absence of potentially influencing factors like the menstrual cycle phase. The aim of this study was to test MHC-similarity mate choice preferences in odour, facial and vocal modalities. In particular, we focused on a potential effect of hormonal contraception. Furtermore, we tested preferential shifts across the menstrual cycle by comparing women's preferences in the follicular and the luteal phase in pill and non-pill users.
