Hodnocení kvality ejakulátu beranů vybraných plemen

The aim of this thesis was to investigate the issues of ejaculate evaluation and analyse the quality of ejaculates via macroscopic and microscopic methods. From 1st January to 31st March 2015, 69 ejaculates were collected from 7 rams of the dual-purpose Zwartbles breed owned by Ing. Martin Hoška, PhD, 4 rams of the Suffolk breed and 1 ram of the Charollais breed from the School Agricultural Facility Žabčice of Mendel University in Brno. Macroscopic and microscopic analyses were performed immediately after sample collection, including the detection of the ejaculate volume as well as the sperm activity and concentration. For the evaluation of morphological parameters of the ram ejaculates, morphological smears were performed, which were transported to a laboratory of the Department of Animal Breeding, Faculty of Agronomy, Mendel University in Brno. Mainly changes in the sperm head, acrosome, midpiece and tail (torsion, DAG effect) were monitored. Further, the presence of immature and degenerated sperm was also monitored. The results show that the quality of ejaculate is influenced by the age of the rams and particularly by the frequency of the sampling and the size of the testicles whereas the difference between the breeds of distinct purpose was not significant.

The diploma thesis solves the problem of oxygen concentration in Muscat Moravian and Cerason wines. In the literary part attention is paid to oxygen as a chemical element, its occurrence in various forms or possibilities to originate. Further, there is explained the theme of oxygen in wine, its forms and importance for winemaking. Also, the oxidation and controlled addition of oxygen to the wine, or the importance of sulfur dioxide in winemaking is described in the thesis. The last chapter is devoted to types of methods to determine the concentration of oxygen in wines. The experimental part describes the materials used in the thesis and research procedures. In this part are listed the activities affecting the oxygen concentration in the wine that was determined before and after the activity. These processes were racking, clarifying, filtering and bottling wine. After filling the bottles, there was monitored the concentration of dissolved oxygen, headspace oxygen and total oxygen in wines. When oxygen concentration stagnated, bottles were opened and concentration of free and total sulfur dioxide was determined in the wines as well as selected chemical parameters.

1 Abstract The dissertation focused its attention on the principle of the concentration and its application in civil proceedings. The principle of the concentration and its issues represented after the amendment of Civil Procedure Code No. 99/1963 realized by Act No. 30/2000 Sb. and the by Act No. 7/2009 Sb. are highly actual topic in the civil practice and in the theory of civil procedure. The dissertation attempts to explain the positives and the negatives of the principle of the concentration in civil proceedings, seeks and finds its adequate place in the civil procedural law in order to achieve an effective and quick protection of endangered or infringed subjective private rights. The theoretical basis of the dissertation consists of definition of the essence of the principle of the concentration, its object and mainly its purpose along in the system of the principles regulating civil proceedings at all. The modern civil proceedings should be based on the principle of the concentration in order to ensure the effective protection of endangered or infringed subjective private rights provided by the courts. This Master's degree thesis deals with legislation on the "principle of concentration in the contentious procedure" and related legal institutes. Pursuant to the principle of concentration, the parties to the civil proceedings are obliged to concentrate and perform the prescribed pleadings within particular time period. These pleadings include among others: allegation of material facts, proposed evidence to prove these facts and objections to the procedural matters. After the passage of prescribed time, a factual and evidentiary "stop condition" occurs, i.e. the parties to the proceedings are not able to perform the pleadings with relevant consequences in law anymore. Legislation on the principle of concentration has been significantly amended by adoption of several Civil Procedure Code amendments, namely Act N. 30/2000 Coll. and Act N. 7/2009 Coll. The thesis aims to analyse and critically evaluate these amendments. The thesis consists of seven chapters. Marcela Vejsová, Ph.D. Title of diploma thesis: Evaluation of activity of potencial antifungal substances trought the use of microdilution broth method Background: The aim of this thesis was evaluation of antifungal activity of 70 substances trought the use of microdilution broth method. Substances for testing were synthesized at the Faculty of Pharmacy and they were divided into 5 groups based on chemical structure. Methods: The microdilution broth method was used for testing. The antifungal activity of substances was tested on 8 strains of yeasts and filamentous fungi: Candida albicans, C. tropicalis, C. glabrata, C. krusei, Trichosporon asahii, Aspergillus fumigatus, Absidia corymbifera a Trichophyton mentagrophytes. Results: The most active substances were in group of salicylanilide N- monosubstituted and N,N-disubstituted carbamates and thiocarbamates. The most effective substances were SAL-1L, SAL-1M, SAL-1N. They were effective on Trichophyton mentagrophytes already at concentration of 1,95 µmol.l-1 . Tyrosol has been formerly confirmed as a substance which shortens the lag phase of Candida albicans cell cycle and stimulates its conversion to the hyphal form, farnesol stimulates blastospores. This work covers the effect of tyrosol combined with two antimycotics, fungistatic fluconazole and fungicidal amphotericine B. Candida albicans strains used in the experiment were mostly isolated from samples taken from patients suffering from variously located Candida albicans infections and four laboratory standard strains were also used. The method used for the experiment was a modified broth dilution method using combinations of both tyrosol and antimycotics various concentrations, which were prepared by two-fold dilution. Using amphotericine B, 28 strains did not show any difference in MIC compared with the control sample, 6 strains showed mildly higher MIC and 3 strains shower mildly lower MIC. Various problems which relate to the obligation of assertion and obligation of evidence and corresponding burden of assertion and burden of evidence and activity of a court connected with them are the topic of my diploma thesis. The first part concentrates on the definitions of basic terms, which appear in the field of evidence, for example the evidence as a complex process, the subject of evidence, means of evidence and also key terms as burden of assertion and burden of evidence. The next part of the thesis is focused on the description of the institute, which was introduced to the legal system in 2009 and the aim of which is to make civil procedure faster and more effective.
