Stanovení komplexačních konstant metodou částečného plnění a analýzou průtokem způsobené disperze a jejich srovnání s hodnotami stanovenými metodou afinitní kapilární elektroforézy / Determination of complexation constants by partial Filling method and Flow Induced Dispersion Analysis and their comparison with values determined by Affinity Capillary Electrophoresis

Partial Filling method (PF) and Flow Induced Dispersion Analysis (FIDA) were used for the determination of stability constants of model system of four profens (R-Flurbiprofen, S-Ibuprofen, S-Ketoprofen and S-Naproxen) complexing with β-cyclodextrin. When using PF method, only a part of capillary is filled with the selector and thus the analyte must migrate through the zone of selector first and then through the neat BGE (it has different mobilities in both zones). Dependency of the differences in migation time on the length of the selector zone is then studied. When FIDA is used, the analyte in pushed through the capillary by external pressure and dependency of the rate of peak dispersion of the analyte on the concentration of selector in capillary is observed. Moreover, PF experiments were theoretically studied by Simul 5 Complex computing programme. Values of stability constants obtained by both methods were compared with values obtained by frequently used Affinity Capillary electrophoresis (ACE) method. The comparison of stability constants values determined by PF method and FIDA shows that both investigated methods grants results comparable with those obtained by the ACE.

Title of Diploma Thesis: Physico-Chemical properties of drugs To test physico-chemical properties of new molecules is necessary during drug development. It could be helpful to understand or predict the pharmacokinetic parametres of a new drug in vivo/in vitro experiments. One of this parameters is a dissociation constant (pK). Dissociation constant is defined as " Number on pH scale, wherein is just fifty percent of molecule in a ionization condition". In real case this number can help us to know where in the gastro-intestinal tract (GIT) the drug will be absorbed. In GIT only molecules exhibiting pK from 3 to 11 could be absorbed. Out of this range it is not possible. In this work I would like to introduce the ways of experimental measurement of pK values. I was working with two methods to measure the pK values of water-soluble compounds. The spectrophotometric method and the potentiometric one. In the thesis we study a method for determining resonance energies - gen- eralization of the method of analytical continuation in the coupling constant, which is based on continuation of the coupling constant λ as a function of the momentum k. A formula for λ(k) is derived for spherically symmetric potential consisting of finite number of δ-functions and its Taylor series is studied. Taylor series of λ(k) and its asymptotic behavior is studied for sep- arable potential. Proper choice of added potential parameters is studied on examples. A method for determining λ(k) poles is described for spherically symmetric potential with added δ-function. It is tested whether the knowl- edge of λ(k) poles can be useful to improve the accuracy of the determination of the resonance parameters of the original potential.

Antrame skyrelyje aptartas semiparametrinis pagreitintų bandymų modelio atvejis. Trečias skyrelis skirtas apkrovos pasikeitimo atsitiktiniais momentais atvejo modeliavimui esant neparametriniam pagreitintų bandymų modeliui. Šio skyrelio tikslas - neparametrinio patikimumo funkcijos įverčio, ir pagreitinimo konstantos įverčio apskaičiavimas iš pagreitintų bandymų duomenų. Ketvirtame skyrelyje pateiktas apkrovos pasikeitimo atsitiktiniais momentais atvejo modeliavimas esant parametriniam pagreitintų bandymų modeliui. Thesis title: Physico-Chemical properties of drugs This thesis deals with measuring dissociation constants (the pKa value) and the lipophilicity of nitrogenous heterocycle derivatives produced at the Faculty of Pharmacy in Hradec Králové. Two methods were used to determine the pK value: potentiometric titration and spectrophotometry. Potentiometric titration almost invariably produced relatively exact results, however the results of pK value obtained by spectrophotometry were sometimes difficult to replicate. Dissociation constants were compared on the basis of the method applied, and also with the theoretically determined dissociation constant calculated by means of the ChemDraw Ultra 12.0 software. The results obtained by the two experimental methods turned out to be very similar, corresponding to the theoretical pK value. Lipophilicity is another essential property of drugs tested in the preclinical stage. It specifies the aqueous or lipophilic phase affinity of a drug, substantially affecting the drug's ADMET in organism. Humic acids are natural substances belonging to the group of humic substances. They arise mainly decomposition of plant residues. They are contained in soils, peat, sediments, young coal, water and even in the air. Humic acids are only partially soluble in water with increasing pH increases their solubility. Diploma thesis focuses on the use of chronopotentiometric titration of humic research. This method is mainly used for the determination of trace concentrations of analytes. This work is focused on the determination of acidity by potentiometric titration and the determination of dissociation constants using chronopotentiometry with measurement of pH of prepared samples.

Martin Beneš Determination of stability constants of charged cyclodextrin comlexes by capillary electrophoresis Abstract Stability constant characterizes binding interaction between an analyte and complexation agent. These interactions play very important role in separation processes of, in other way undistinguishable, compounds, e.g. enantiomers. The most widely used complexation agents are cyclodextrins. Affinity capillary electrophoresis (ACE) belongs to methods suitable for the determination of stability constants. The stability constant is determined from the dependence of the effective mobility of analyte on the increasing concentration of complexation agent in background electrolyte (BGE). If charged CDs are used, the attention must be paid not only to viscosity of the BGE and to the influence of Joule heating on the temperature in the capillary but also to the increasing ionic strength. The thermodynamic stability constants of R,R- and S,S-hydrobenzoin and R- and S-(3-brom- 2-methyl-1-propanol) with cationic modified β-cyclodextrin: 6-monodeoxy-6-mono(3- hydroxy)propylamino-β-cyclodextrin hydrochlorid (PABCD) were determined by affinity capillary electrophoresis. This work was the structure of resonant spruce wood devoted to. The tree-ring widths and summer wood measure were measured. By use of non - destructive methods their acoustic properties were measured - speed of sound, acoustic constant, logarithmic decrement of damping, acoustic wave resistance, wood - density and Young's modulus. Data were statistically analyzed and the wood structure coefficient of variation for each specimen was calculated. There were three different methodologies for verification of the wood structure variability effect formulated. Subsequently, it was found that it is possible to determine the effect on the acoustic properties of the wood macroscopic structure variability.

The theme of this thesis is a "Testing of reflective systems for distance measuring." Within the work was created reflective device of aluminum sheet and reflective foil. This device was tested with another reflective device that operates on the same principle. The aim of this study was to test devices under different climatic conditions. Standard deviations of the measured values and the values of additive constants for each total station and reflective devices were evaluated. In conclusion, it was suggested the use of these devices in geodetic work under land adjustment.
