Matrix assisted laser desorption ionization for characterization of higher molecular thiols in biological samples

Among the heavy metal-binding ligands in cells the phytochelatins (PCs) and metallothioneins (MTs) are the best characterized. PCs and MTs are different classes of cysteine-rich, heavy metal-binding molecules. PCs are enzymatically synthesized peptides, whereas MTs are gene-encoded polypeptides. Several analytical methods have been used to analyze PCs, MTs and complexes of metal in different studies. Today, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDITOF MS) is renowned for its extreme ease of operation and requirement of inexpensive matrices for preparation of sample; more importantly, the instrumentation is capable to be fully automated. In this study, the optimal condition for the characterization of PC2 and Cd-PC2 complex by MALDI-TOF MS was determined. The human MT2A and MT3 genes were expressed in heterologous organisms (Escherichia coli) by cloning. Further, MT2 protein was isolated from rabbit liver. MTs were purified by fast protein liquid chromatography (FPLC). The present study demonstrates analytical approaches of employing MALDI-TOF MS for characterization MTs. Other electrochemical methods were used for confirmation of thiols complexes with metal. Moreover, easy-touse instruments for MTs isolation, coupled with simple analytical detection method in fully automatic mode, providing prototype for the construction of sensor was designed

Soil contamination as one of the influences on afforestation after precious metals mining at selected bearings in central Europe

Mining of precious metals poses a threat to the environment by heavy metal contamination. Soil contamination by selected heavy metals (As, Cd, Cu, Pb, Zn) and its effect on woody species used for afforestation was investigated at selected localities (Příbram-Březové Hory, Banská Štiavnica-Lintich, Olkusz-Bolesław). Concentrations of the heavy metals in the soil and assimilation apparatus were determined by the XRF method and other soil characteristics (pH, humus content, texture) were also determined. At all the localities, the pollution indicators have been exceeded, both for soils and for trees. In the case of trees, it was mainly zinc contamination and the maximum values of Zn in their assimilation apparatus on reclaimed areas have exceeded the lower limit of the pollution indicator 3 times in the case of Březové Hory, 11 times in the case of Lintich and 23 times in the case of Bolesław. The evidence of the heavy metal intoxication in the trees was also found. Based on the found situation and the planned use of reclaimed areas, management has been designed to improve the situation.

Soňa Procházková Department of Inorganic Chemistry, Faculty of Science, Charles University Supervisor: RNDr. Vojtěch Kubíček, Ph.D. Supervisor's email address: kubicek@natur.cuni.cz ABSTRACT Two types of α-aminophosphinates were prepared and studied: phosphinate analogues of iminodiacetic acid − aminobis(methyl(hydroxymethyl)phosphinic acid) (IDPhm ) and aminobis(methyl(2-carboxyethyl)phosphinic acid) (IDPce ), and bis(aminomethyl- phosphinates) − (hydroxy(fenyl)methylen)bis(aminomethylphosphinic acid) (PheOHABPin) and (methylen)bis(aminomethylphosphinic acid) (ABPin). All prepared compounds were fully characterized (by NMR, MS, elemental analysis). One crystal structure was determined by X-ray singlecrystal analysis. The diphosphinates were synthesized by Mannich type reaction, whereas the bis(aminomethylphosphinates) were prepared by multistep synthesis including reactive silylesters. Compounds were prepared in moderate or good yields Acid-base and coordination properties of ligands were investigated and compared by potentiometric and NMR titrations. The values of protonation constants of aminogroups, which determine stability of complexes, showed surprising results. An extremely low basicity of nitrogen atom was found in diphosphinates. Adverse reactions to metals is relatively frequent in sensitive individuals, and may lead to the development of various diseases, such as lichen planus and lichenoid reaction and was also described effect of amalgam to the pathology of autoimmune diseases. In sensitive patients with known reactions to metals leads to atrophy of the oral mucosa and discomfort (burning, cutting, excessive salivation). Adverse effects of dental materials are described as an allergic reaction type IV delayed hypersensitivity. It disrupt the immune system, increased vascular permeability, tissue is often damaged by chronic inflammation and it is also activated antigen- nonspecific component of immunity. Monocytes are the key cells of the immune system, which act as antigen presenting cells, in particular after conversion in the tissues into macrophages are directly involved in the activation of immune responses and their stimulation is therefore crucial for the development of adverse reactions to dental materials. In the thesis was measured cytokine production by monocytes after stimulation with metal salts. In this work, the crystallographic structure and its changes under thermal treatment of different systems consisting of metal oxide nanoparticles is studied. The principal method used throughout the thesis is x-ray powder diffraction enriched with grazing incidence small angle x-ray scattering when the nanoparticles form an ordered structure or with x-ray absorption spectroscopy when additional information on local crystallographic structure is required. For all the systems the preparation conditions were optimized according to the crystallographic data for further applications.

The aim of thesis is to identify and analyze the most significant changes based on the implementation the Industrial Emissions Directive. Theoretical part is focus to explanation basic terms of environmental legislation. The practical part is focused to evaluation of impacts on metal industry. It describes the transposition of the directive into national law. For the achievement of the goal was made analysis of the industry, customers and identification selected category to determine how to prepare for the environmental requirements. The modeling of economic impacts was made by the generation of value. The thesis contains analysis of the impact of new requirements on employment and competitiveness. The conclusion summarizes information of the impact Industrial Emissions Directive including risk and recommendations.

The goal of this project is explore and study the ways of metal detecting and create a metal detector for industrial use. When the device were inventing, a lot of methods were studied and the impulse detect method were chosen as the most perspective way of metal detecting, thanks to a huge electronics parts possibilities. After experiences with preceding device was necessary to redesign it. Main target of this thesis is to increase the computing power of detector. For this purpose will device need a new microcontroller. Device will communicate with PC trough a USB port. The device will have a battery which allows device to run for a few hours. Processing of measured data will be make even with DFT which will lead to better analyze of mass and type of conductor under device.
